    The Development of Ascaridia galli Infective Eggs by In Vitro Culture

    The aim of this study was to determine the survival of embrionated eggs of Ascaridia galli. Adult female worms were obtained from lumen of intestine of native chickens in a slaughter house. Eggs obtained from the uteri of adult female worms were incubated in distilled water at room temperature for 20-31 days in order to develop A. galli infective eggs. The eggs were counted using stereomicroscope. The result showed that the amount of A. galli eggs were 1,045,478 and the amount of embrionated eggs were 935,300 (89.46%).Keywords: Ascaridia galli, embrionated egg

    PROFIL HORMON ESTROGEN DAN PROGESTERON PADA SIKLUS BERAHI KAMBING LOKAL (Estrogen and Progesterone Profile in Aceh Local Goat Estrous Cycle )

    The aim of this research was to find out the normal estrogen and progesterone profile in local goat estrous cycle. Five female Aceh local goats which the criteria clinically in good condition, have been giving birth before, 1.5-3.0 years old, and showed minimal twice regularly estrous cycle. All samples were synchronized with 0.5 ml cloprostenol (Estrone, Bioveta) using randomly infection pattern. The data obtained were analyzed descriptively. Blood serum was collected for examination of estrogen and progesterone content on 0, 7, 14, and 21 day of estrous cycle (day 0 of estrous cycle). Hormone analysis was conducted using ELISA. The results showed that estrogen and progesterone profile on day 0, 7, 14, and 21 were 62.21; 28.61; 29.84; 79.89 pg/ml and 0.0; 8.6; 14.7; 0.0 ng/ml respectively.Keywords: estrogen, progesterone, estrous cycle, Aceh local goat

    Effect of Heat Stress on Body Weight Gain, Heterophile-Lymphocite Ratio and Body Temperature in Broiler

    Increasing in ambient temperature inside the cage could lead to heat stress in broilers. This research was conducted to find out effects of heat stress on body weight gain, heterophile-lymphocite ratio and body temperature in chicken broiler. Twenty broilers aged 20 days (strain Cobb) were randomly divided into 2 groups. The first group was treated with no heat stress, the second one was caged in 331 0C temperature for 4 hours per day for 14 days. The results indicated that heat stress reduced body weight gain, increased body temperature, and changed behavior, but no effect on feed conversion ratio (FCR) and heterophile-lymphocyte ratio. It suggested that the heat stress caused detrimental effects on broiler chicken

    KONSENTRASI PROTEIN DAN PENENTUAN BERAT MOLEKUL EKSKRETORI/SEKRETORI L3 Ascaridia galli

    Penelitian ini bertujuan menentukan konsentrasi dan berat molekul protein ekskretori/sekretori larva (L3) Ascaridia galli (A. galli). Larva L3 diperoleh dari usus halus 100 ayam tujuh hari setelah pemberian dosis 6000 L2 melalui esofagus ayam. Sebanyak 510 L3 dikultur secara in vitro dalam setiap ml medium Rosswell Park Memorial Institute (RPMI 1640), pH 6,8, tanpa merah fenol dalam inkubator pada temperatur 37 0C dan 5% CO2 selama 3 hari. Ke dalam medium ditambahkan 100 unit ml-1 penisilin G, 100 g ml-1 streptomisin, 5 g ml-1 gentamisin dan 0,25 g ml-1 kanamisin. Ekskretori/sekretori dipreparasi dari produk metabolisme L3 yang dilepaskan ke dalam medium kultur. Untuk mendapatkan protein ekskretori/sekretori, medium kultur dipekatkan dengan vivaspin 30.000 MWCO, dan kuantitas protein dihitung dengan metode Bradford. Berat molekul protein ekskretori/sekretori divisualisasikan dengan sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS PAGE). Hasil penelitian menunjukkan bahwa konsentrasi protein ekskretori/sekretori adalah 0,595 mg/ml dengan berat molekul 28 kDa

    PENGARUH LAMA MATURASI DAN LAMA INKUBASI FERTILISASI TERHADAP ANGKA FERTILITAS OOSIT SAPI PERANAKAN ONGOLE SECARA IN VITRO

    Penelitian ini dilakukan untuk mengetahui pengaruh lama maturasi dan lama inkubasi fertilisasi terhadap angka fertilitas oosit sapi peranakan Ongole (PO) secara in vitro. Oosit diaspirasi dari folikel ovarium yang berdiameter 2-6 mm. Oosit dirandom untuk penelitian menggunakan rancangan blok acak sempurna (RCBD), dengan lama maturasi 20, 22 dan 24 jam sebagai blok dan lama inkubasi fertilisasi 4, 5 dan 6 jam sebagai perlakuan. Hasil penelitian menunjukkan angka fertilitas oosit sapi PO dengan lama maturasi 20 jam dan lama inkubasi fertilisasi 4, 5 dan 6 jam masing-masing adalah 48,86%+3,88, 48,08%+2,22 dan 51,96%+ 2,27. Angka fertilitas oosit sapi PO dengan lama maturasi 22 jam dan lama inkubasi fertilisasi 4, 5 dan 6 jam masing-masing adalah 50,14%+4,18, 51,14%+4,35 dan 52,27%+2,26. Angka fertilitas oosit sapi PO dengan lama maturasi 24 jam dan lama inkubasi fertilisasi 4, 5 dan 6 jam masingmasing adalah 51,21%+4,49, 53,49%+2,43 dan 50,96%+4,18. Tidak ada perbedaan yang nyata (P0,05) angka fertilitas antara lama maturasi 20, 22 dan 24 jam dan tidak ada perbedaan yang nyata (P0,05) angka fertilitas antara lama inkubasi fertilisasi 4, 5 dan 6 jam

    Development of Selection Method for Genetic Potential and Adjusment of Nutritional Requirement to Stimulate Gene Expression of Local Meat Chicken

    This study aimed to evaluate the genetic potential of local meat chickens (LMC) to get the candidate of parent stock through the development of the selection method applied, as well as economical and practical approach and adjusment of nutritional requirement, especially protein and energy balance. Research was divided into 2 groups, where the female and male parent was reared for 2 months and the offspring (F1) was reared for 4 months and selected. The parent was mated by the method of artificial insemination (AI). Method of selection was used on the orientation of the meat chicken production parameters with a positive correlation with the gene expression Diet treatments were formulated by optimal nutrition standards in order to drive maximum primary production of local meat chicken, contained 17, 19, and 21% of protein. The result showed that feeding diet with 19% protein can stimulate optimum growth and food conversion of ALPU male, whereas female growth LMC in line with the increasing availability of protein in the diet, wherein feeding 21% of protein result in optimal growth until age 4 months

    Adhesi merupakan tahap inisiasi dari proses kolonisasi bakteri. Kemampuan adhesi ini diperantarai oleh keberadaan hemaglutinin pada permukaan bakteri. Tujuan penelitian ini adalah melihat kemampuan hemaglutinin pada Escherichia coli dalam proses adhesi. Uji adhesi pada sel epitel pipi dilakukan dengan menggunakan metode Velentine-Weiggand. Hasil penelitian menunjukkan bahwa Escherichia coli yang memiliki hemaglutinin menunjukkan kemampuan adhesi yang jauh lebih baik yakni rata-rata 67,6 bakteri/sel dibandingkan dengan yang tidak memiliki hemaglutinin rata-rata 7,2 bakteri/sel.     Characterization of Pregnancy Specific Protein B (PSPB) from Placenta Foetalis (Cotyledon) Friesian Holstein Cows

    This research was aimed to find out a substance that was useful in early pregnancy diagnosis of Friesian Holstein cows. Pregnancy Specific Protein B (PSPB) was isolated, purified and partially characterized from cotyledon Friesian Holstein cows. Characterization of PSPB protein was conducted using SDS-PAGE and Western Blot. Antisera were developed against PSPB and by immunohistochemical techniques the protein was localized to the binucleated cells of the cotyledon. The estimated molecular size of Friesian Holstein cows PSPB was 59.88 kDa with the concentration of PSPB protein in cotyledon was 6480 ng/ml. PSPB protein could induce anti-PSPB antibody with the value of optical density (OD) were 0.1790.0102 (before immunization); 1.4660.3288 (3rd week after immunization); 1.9360.4754 (1st week after booster;) and 2.2560.4842 (2nd week after booster)

    Mineral Level Status of Copper, Magnesium, and Iron in Local Goat Serum Related to Keeping Pattern System

    This research was conducted to find out the mineral level status of Cu, Mg, and Fe in blood serum of local goats which are reared with different methods. Blood serum was taken from 15 male and 15 female goats, 15-18 month old. The goats were divided into three groups which consist of 5 males and 5 females. Three methods of caging in which most commonly conducted by local people were selected as treatments, i.e. 1) Goats are caged only at night, while in daytime (around 10.00-12.00 WIB) they are discharged freely therefore they could foraging; 2) Goat are caged at night, while in daytime (around 10.00-18.00 WIB) they are bound with rope at which they could foraging around and, the rope would be removed several times in that particular daytime; 3) Goats are released freely all day long, but they are provided with kind of place as a cage, under the house or under the tree. Blood serum was taken from every unit sample. This research revealed that there was no significant different among methods of caging on the level rates of Cu, Mg, and Fe in local goat serum. The level rates of Cu, Mg, and Fe in goats serum that was found from the research were in normal range

    Correlation of Aspergillus flavus with the Concentration of Aflatoxin B1 in Ikan Kayu

    The objective of this research was to know the contamination of Aspergillus flavus in ikan kayu and the number of aflatoxin B1 (AFB1) produced. Also the corelation the number of A. flavus with AFB1. Samples were taken from traditional markets in Banda Aceh and Lhokseumawe. The sample were used to isolation and identify A. flavus also detect the present of AFB1 by ELISA (Enzym Linked ImmunosorbantAssay). The result shows that all samples show the present of A. flavus with the concentration 1 x 102-61 x 102 Cfu/g and the concentration of AFB1 between 0.9-38.7 ppb. In conclusion the present A. flavus in ikan kayu has a corelation with the production of AFB1
