    Get PDF
The objective of this study is to observe the relationship of development of adrenocorticotropic hormone (ACTH) cells of adenohypophysisadrenal cortex axis during pre and postnatal period of longtailed monkey (Macaca fascicularis). Five fetuses aged 70, 85, 100, 120, and 150 days of gestation and 2new born aged 10 and 105 days old were used as research materials. The sections of the pituitary gland stained immunohistochemically using human ACTH antiserum and the adrenal glands stained by Hematoxylin-Eosin procedures. At the fetus 120 days old, the ACTH cells and blood vessel of fetaladenohypophysis developed well and its similar that found in older age. This condition suggested distributing the ACTH to whole body optimally. At the same time, the adrenal cortex cells of fetus 120 days old were active in proliferation and differentiation processes to form transition zone that lies between definitive and fetal zones, and central vein also developed in adrenal gland. In fetus aged 150 days, the cortex adrenal structure was similar with found in older fetus. The adenohypophysis ACTH cells developed earlier than adrenal cortex and their secretion suggested influencing the development of adrenal cortex cells of long-tailed monkey

This research was conducted to prove the ability of betel nut extract (Areca catechu) in causingdegradation of sperm motility on rats (Rattus norvegicus) strain Wistar. This research used 2-3 monthsage, 200-250 g body weight of male white rats Rattus norvegicus strain Wistar. The rats were dividedinto 5 groups in equal number, 3 rats each group. They were a control group without treatment and 4treatment groups which were given different doses of betel nut extract for 7 days, i.e., 1, 2, 3, and 4 g/200g body weight respectively. The result showed that increasing doses of betel nut extract can result indegradation of sperm motility significantly (P0.05).Keywords: Areca catechu, Rattus norvegicus, sperm motilit

IDENTIFICATION OF COPPER-INDUCIBLE GENES IN SWORDTAIL FISH (Xiphophorus spp.) USING DIFFERENTIAL DISPLAY

    Get PDF
Differential display technique was used to identify differentially expressed gene ofswordtail fish (Xiphophorus spp.) that induced by 1 g/ml copper for 24 hours. From eightprimers that have been used for differential display, seven of them showed the differentialgenes which were not present in the control. The reamplification of these fragments rangingfrom below 100 to 200 bp. But only two fragments were successfully cloned and sequenced, those are H-AP10 and H-AP11. Sequence analysis showed that both of the sequences revealedhigh similarity with Homo sapiens stress-associated endoplasmic reticulum protein, a signalgenerated that induces apoptosis. Keywords: differential display, swordtail fish, copper pollution, mRNA expression, biomarke

The study was done to investigate the glycogen distribution in the stomach of long tailed macaques (Macaca fascicularis). The study used eight individuals of long tailed macaques consist of 6 fetuses (55, 70, 85, 100, 120, and 150 days pregnancy) and 2 infants (10 and 105 days). The gastrict tract were histologically stained using Hematoxyllin-Eosin (HE), and histochemically stained using Periodic Acid Schiff (PAS), to detect glycogen. The results revealed that the histogenesis pattern of stomach glandullar cells are developed in line with the age of the fetuses and infant. We also found that glycogen existance are increase in line with the age in the glandullar stomach. From the present study we concluded that the glycogen distribution in gastrict glandular cells were influenced by the age in the fetal period, while in the infant was under influence of the digestion activities, and the activities of the gastrict glandular cells was already seen at 55 days gestation

Cytoskeleton of Bovine Oocytes after Vitrification Using Ethylene Glycol Cryoprotectant

    Get PDF
The objective of this research is to know the influence of vitrification method toward thecondition of bovine oocytes cytoskeleton based on microtubules using the immunohistochemistrytechnique. Oocytes classified in two treatments such as fresh oocytes (control) and oocytes withvitrification treatments of 10, 20, 30, 40, and 50% of ethylene glycol (EG). Data were analyzed bydescriptive analyzing. The results of this research indicated that vitrification treatments were affected tomicrotubules organization of oocytes. Concentration of EG 10, 20, 40, and 50% of EG could not repaircytoskeleton after vitrification, but cytoskeleton structure could be protected by EG 30%

The experiment was conducted to study the effect of ethylene glycol (EG) on disturbance of spermatogenesis process of the male mice. Sixteen mice at the age of 2 months were randomly into 4 groups of 4 each. The mice of control group were only given drinking water without EG. Group KPI, KP2, and KP3 were given EG 5, 10, and 15 ml/liter drinking water, respectively. On day 31 of post treatment, all mice were killed for their testis. The testis were collected and processed for microscopic examination and haematoxylin-eosin staining. The result showed that EG given orally for 30 days on male mice have negative effect on the process of spermatogenesis as the numbers of spermatogonia, spermatocyte and Sertoli cells decreased significantly (P0.01

    Get PDF
This research was aimed to know histopathology change of white mouse ovary as result of EG intoxication. Eight white mice randomly divided into three treatment groups and one control group. Treatment of P1 group was given 10 ml EG/L drinking water, P2 was given 20 ml EG/L drinking water, and P3 was given 30 ml EG/L drinking water for two weeks. Control group was given aquadest. Then, white mice were euthanized and ovaries sample were taken for histology observation. The result for treatment groups (P1 and P2) had shown ovary hyperemic (congest). But, the change is worst in P2 rather than P1. Changes in P3 treatment group had shown ovary hyperemic (congest) and leukocyte infiltration in follicular antrum and follicular necrosis causing lost of ovum cell (cariolysis). Poisoned of EG in white mouse can cause inflammation and necrosis of ovary follicle thus the formed of ovum cell was failed (ovogenesis)

Antibody to Enteropathogenic Eschericia coli (EPEC) K1.1. can be produced in egg yolk by the application of inactivated bacterial cells intravenously in layer chicken. The presence of specific antibody in sera and egg can bedetected with immunodiffusion techniques (Agar Gel Precipitation Test/AGPT), expressed by the occurrence of specific precipitation reaction between antibody and homolog antigen and no cross reaction of this antibody with antigen of Salmonella sp. and Klebsiella sp. The presence of specific antibody previously can be detected in sera, then 1 week after this the antibody start to be able be detected in egg yolk. The antibody is still present in sera as well as in egg yolk in large amount for 7 weeks and decrease significantly in the 8th week. This results indicated that theegg can be used in the producing specific antibody in the large quantity
