    Effect of Ethylene Glycol Concentration and Length of Exposure on In Vitro Fertility of Bovine Oocyte

    The purpose of the study was to determine the influence of the concentration of ethylene glycol (EG) and length of exposure to levels of bovine oocytes in vitro fertility. This research was conducted using a completely randomized design factorial 5x3 with 7 replicates. The first factor was the concentration of cryoprotectants EG 10, 20, 30, 40, and 50%. The second factor was the length of exposure 1, 3, and 5 minutes. The results showed that the concentration of EG and the length of exposure were effect on fertilized oocytes (P0.05). The level of oocytes in vitro fertility after vitrification in 30% EG and long exposure to 3 minutes did not different (P0.05) compared to fresh oocytes, while the EG treatment 10, 20, 40, and 50% significantly showed a lower fertility rate than the 30% EG (P0.05). It was concluded that the concentration of EG and length of exposure were effect on fertilized oocytes. The highest oocytes in vitro fertility level were found in 30% EG concentration with 3 minutes exposure time

    Microscopical Study of Pancreatic Tissue of Babirusa (Babyrousa babyrussa) Using Conventional and Immunohistochemical Methods

    This study utilized conventional and immunohistochemical methods, to describe the morphology and distribution of glucagon and insulin cells in the pancreas of adult male babirusa (Babyrousa babyrussa). The results of research showed that the endocrine portion (Langerhans islet) spread evenly on the acinar portion of pancreas. Observation using immunohistochemical method showed that glucagon cells were distributed on the perifer, whereas insulin cells in the center of the Langerhans islet of pancreas. In general, the number of insulin cells was higher than glucagon cells. It is interesting to note, in the pancreas babirusa also found that neonatal elements similar to the Langerhans islet, but have larger sizes (ranging between 200 -700 mm). The picture is similar to the large Langerhans islet was reported in ruminant. These cells give a negative reaction to insulin and glucagon antisera

    This study aimed to verify the effect of ethanol extract of jaloh leaves at various doses on parasitemia percentage of in mice (Mus musculus), wich inoculated Plasmodium berghei. This study used 12 male mice (Mus musculus) strains Balb / C with average body weight of 30.2 grams of 2-month-old. This research using Completely Randomized Design (CRD) with four treatments and three replications. All the mice were infected with Plasmodium berghei prior to treatment on three days post infected the mice in group 2,3 and 4 were given with extract ethanol jaloh leaves at dose 50, 100, 150 mg/kgbw inspectively. The mice in group 1 (K0) were not given anything and consident as control.Observations parasitemia percentage using microscopy on preparations stained thin blood smear Giemsa. The parasitemia percentage data were analyzed using one way ANOVA. On the average (SD) percentage of parasitemia during the three days in four treatments (K0, K1, K2, and K3) on the first day were 24.871.86; 24.331.53; 24.201.06; and 24.931.01. On the second day were 22.672.52; 23.001.00; 22.801.20; and 24.132.01; and on the third day were 24.771.66; 22.973.76; 21.131.90; and 24.271.42. Statistical analysis showed that ethanol extract of the jaloh leaves for KO, KI, KII, and KIV respectively treatment did not affect the percentage of parasitemia in mice (Mus musculus) inoculated Plasmodium berghei

    Histopathological Changes of Erythrocyte and Quantity of immature Erythrocytes in Tegal Duckling (Anas Javanica) Induced by Lead (Pb)

    The aim of this research is to find out the histopathological changes of erythrocyte and the quantity of immature erythrocyte of tegal duckling treated with lead (Pb). Twenty five young female ducks with the age of 7 days and 60.55.07 g body weight alloted into 4 treatment groups and 1 control group (Ko) with 5 replication each. The treatment group fed with K1 (5.7), K2 (11.4), K3 (17.1), and K4 (22.8) mg Pb/kg body weight for 10 days, respectively. Young ducks showed clinical symptoms such as the color of feces were brown to black, green color of diarrhea, anemic and decreasing body weight. Histopathologically, erythrocyte showed that immature and abnormal in shape (like teardrops, dumb bell, and bottles). The result of variant analysis followed by linier regression shows that the higher the dosage of Pb in diet the higher the quantity of immature erythrocyte andabnormal erythrocyte shape found (P0.01)

    The Effects of Sulphaquinoxalin in Broiler Chicken: Clinical Symptom and Anatomical Pathology

    The clinical symptom and anatomical pathology effect of sulphaquinoxalin administration were studied in 12 male -seven days-old Hubbard broiler chickens with the weight of 85.57.83 g. The chickens were randomly dividedinto 2 groups of 6 each. The P0 group as control group just orally received aquadest and the P groups were treated 1 orally with 0.1% sulphaquinoxalin which dissolved in drinking water. The sulphaquinoxalin was administrated forseven days. At the end of experiment all chickens were sacrificed for necropsy. The results indicated that less appetite and drinking water as well as decrease of body weight (P0.01). Ptechial haemorrhagic was seen in thorax andfemur muscle, mucosa of duodenum, liver, kidney, heart, lung, and spleen. The liver, kidney, heart, and spleen were swollen

    Effect of Storage Time of Epididymis at 5 oC on Spermatozoa Quality of Aceh Local Goat

    The objective of this study was to evaluate the effect of storage time of epididymis at 5 oC on spermatozoa quality of Aceh local goat. This research was conducted at Reproduction Laboratory, FKH Unsyiah from May to June 2009. Nine epididymis from local goat at the age of 1.5-2.0 years were used in this study. The epididymis were collected from abattoir in Banda Aceh. The experimental design used in this study was completely randomized design with 3 treatments and 3 replications. Spermatozoa quality which consist of sperm concentration, motility, amount of life sperm, and sperm abnormality were examined after the collection of epididymis on day 0 (H-0), day 1 (H-1), and day 3 (H-3) post storage at 5 oC. Data were analyzed using one-way analyses of variance (ANOVA). The result showed that no significant difference (P0.05) seen in the spermatozoa quality after different storage time. The average of spermatozoa concentration on H-0, H-1, and H-3 were 318x107/ml, 282x107/ml, and 241x107/ml respectively. On the average, the percentage of spermatozoa motility on H-0, H-1, and H-3 were 82,272,75; 80,252,83; and 78,070,92%, respectively. Life spermatozoa observed on H-0, H-1, and H-3 were 82,292,71; 80,631,87; and 80,093,31%, respectively. Observation on spermatozoa abnormality showed that the average of spermatozoa abnormality on H-0, H-1, and H-3 were 7,230,27; 8,210,55; and 10,753,14%, respectively. It could be concluded that the spermatozoa quality were not affected by the storage time at temperature of 5 oC

    Antimicrobial Activity and LC Determination of Ethanol Crude Extract of 50 Pliek U, an Achehnese Traditional Fermented Food

    Antimicrobial activity of pliek u crude extracts was evaluated against five bacterial species (Staphylococcus aureus, Bacillus cereus, Escherichia coli, Salmonella enterica serovar Enteritidis and Pseudomonas aeruginosa,)and one fungal species (Candida albicans). Pliek u has been consumed as spices and hot sauce and poultry feed. These foods were collected from household industry at Reudep village in Aceh Besar, Province of Aceh. The ethanol extract of pliek u were obtained by standard method. The antimicrobial activity was detected using paper disc method. The concentration of ethanol crude extract of pliek u (EEP) was determined with the dilution method. The lethality initial test has been detected by using Artemia salina L. bioassay to determine the toxic concentration based on the LC value of EEP. The ethanol crude extract (EEP) was active against all microbial strains. EEP showed 50 antimicrobial activity at a minimum inhibitory concentration (MIC) and a minimum microbicidal concentration (MMC) at 2.5-10 mg/ml and 10-20 mg/ml, respectively. The lethality concentration EEP gave the LC value of 3.36 50 mg/ml. It was concluded that ethanol crude extract (EEP) shows significant antimicrobial activity and it is not toxic

    Study of Toxoplasmosis Infection in Human and Related to Animal in Banda Aceh

    The research has been done to know prevalence of toxoplasmosis in livestock, to search the source of infection that have potency to cause toxoplasmosis in human and to make a map of toxoplasmosis infection area in Banda Aceh. The method used in this research was serologic test, Card Aglutination Test (CATT). Data of fertile women in this study are collected through questioner. The result of this research showed that the toxoplasmosis prevalency number of society in Banda Aceh was 3.15%, while at livestock were varied respectively: goats 40%, chickens 25%, cattles 23%, duck 20%, cats 16%, buffalos 15%, and sheeps 10%. The potencial sources of to coures toxoplasmosis in human are: goat, chicken, cattle, duck, cat, buffalo, and sheep. The area found toxoplasmosis in humans and animal were: Baiturrahman, Kuta Raja, Ulee Kareng, Kuta Alam, and Syiah Kuala.     The Use of Sour Soup (Annona murricata) Seed Powder as Acaricide on Cow and Goat

    This research was aimed to study curative effect of sour soup seed powder on cattle invested with ticks and goat infected with scabies. This study was using 12 cattle invested with ticks and 12 goats with scabies. The cattle divided into 4 groups (S1, S2, S3 and S4) while goats were divided into 3 groups (K1, K2, and K3) equally. For cattle with ticks group S1 received water (control group), while group S2, S3, and S4 received 1%, 5%, and 10% sour soup powder respectively. Ticks that fell to the ground and not engorged were collected and identified. Statistical analysis showed that all concentrations of sour soup were effective in paralyzing and or killing ticks of the genera Boophilus sp. and Dermacentor sp. but were not effective against Rhipicepalus sp. For goats with scabies, groups K1, K2, and K3 received 1, 5 and 10% sour soup powder respectively mixed with water applied to whole area of infected and uninfected skin surrounding infected area. Number of mites per cm2 before and after treatment was counted. Statistical analysis showed that 1, 5, and 10% sour soup powder effective in reducing the number of scabies mites on day 1 and 7 after treatment and were significantly different from those number of mites before treatment (P0.01). Statistical analysis also showed that no significant difference among concentration of sour soup seed powder in decreasing the number of mites (P0.05)

    Characterization of Inhibin from Culture and Non Culture of Granulose Cells for Monoclonal Antibody of Inhibin Production

    This study has long-term objectives to obtain immunogenic prototype that can be used to induce multiple ovulation in goats. Working steps of this study were begun with the collection of ovarium from goats, collection of granulose cells, culture of granulose and characterization of molecular weight and isoelectric point (pI) of inhibin protein of granulose cells obtained from culture and non-culture of granulose cells, and followed by preparation of monoclonal antibody toward inhibin. The results showed that inhibin isolated either from culture or non-culture of granulose cells produced a 32 kDa band. Molecular weight of inhibin was measured by Western Blot. The 32 kDa band of SDS PAGE product appeared on Western Blot result was inhibin molecules produced by granulose cells collected fom culture and non-culture of granulose cells that can be identified by Mab-inhibin. Product of IEF gel electrophoresis suggested that inhibin molecule collected from culture of granulose cells has no charge at isoelectric points ranging from 5-6, depends on its total amino acid composition
