PENENTUAN KONSENTRASI SODIUM DODECYL SULFATE DALAM PENGENCER RINGER LAKTAT-KUNING TELUR UNTUK PRESERVASI SEMEN AYAM PELUNG (Determination of Sodium Dodecyl Sulfate Concentration in Ringer Lactate-Egg Yolk Extender for Pelung Rooster Semen Preservation)
The objective of this experiment was to determine the addition of sodium dodecyl sulfate (SDS) to Ringer Lactate-Egg Yolk (RL-EY) extender on pelung chicken semen preservation. Semen was collected three times a week from three pelung chickens. Collected semen was evaluated macroscopically and microscopically. Only ejaculates of at least 70% sperm motility was then divided into three equal microtubes. Each of them diluted with RL-EY with ratio 90%:10%, then added with 0.00, 0.025, and 0.05% SDS respectively. The liquid semen then stored at 5 C for 72 hours. Sperm motility and viability were observed every 12 hours. The addition of 0.025% SDS showed higher spermatozoa motility and viability (72.081.44% and 80.821.30%) which were significantly higher (P0.05) than 0.00 and 0.05% SDS addition at 24 hours of storage. There was no differences on the spermatozoa motility and viability between 0.00 and 0.05% SDS addition. The decrease of spermatozoa motility and viability was observed in 0.025% SDS addition (4.170.56% and 4.650.59%, respectively) that significantly lower compared to 0% and 0.05% SDS addition at 24 hours of storage. In conclusion, 0.025% SDS addition in a Ringer Lactate -Egg Yolk extender maintained pelung spermatozoa motility and viability better than 0.00 and 5% SDS addition
DEKONTAMINASI BAKTERI Escherichia coli DAN Staphylococcus aureus PADA SARANG BURUNG WALET DENGAN PERLAKUAN PEMANASAN (Decontamination of Escherichia coli and Staphylococcus aureus in Edible Birds Nest Using Heat Treatment)
This research was aimed to examine the heating effect at 70 C for 3.5 seconds on Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) decontamination in edible birds nest. This study used 40 clean edible birds nest samples. Samples were divided into two groups, first group was contaminated with E. coli and second group was contaminated with S. aureus. Each group was divided into two treatments. The first treatment was directly tested for microbiological examination and the second treatment was heating at temperature 70 C for 3.5 seconds prior to microbiological examination. The results showed that both of bacteria E. coli and S. aureus had been destroyed by heating treatment. Heating process at 70 C for 3.5 seconds was effective for decontamination of both E.coli and S.aureus
PREVALENSI DAN FAKTOR RISIKO INFEKSI Toxocara cati PADA KUCING PELIHARAAN DI KOTA BOGOR (Prevalence and Risk Factors of Toxocara cati Infection in Bogor Pet Cats)
The aims of this research were to estimate the prevalence and to determine the risk factors related to Toxocara cati (T. cati) infection in pet cats in Bogor. This study was cross sectional study using two types of data; laboratory examination results of feces samples and interview results of the cat owner using a structured questionnaires including characteristics of pet owner and pet care management. The data obtained from this study was analyzed using chi-square test. The results showed that 85 of 243 cats were positively infected by T. cati with 35% prevalence. The significant risk factor associated with the T. cati infection were: sex, sand provided, deworming and type of feed. Pet care management in Kota Bogor were well managed, however the prevalence of T. cati infection was still high. Therefore it can be a potential public health problem especially on cat owners
PENGGANTIAN BOVINE SERUM ALBUMIN PADA PENGENCER CEP-2 DENGAN SERUM DARAH SAPI DAN PUTIH TELUR TERHADAP KUALITAS SEMEN CAIR SAPI LIMOUSIN SELAMA PENDINGINAN (The Substitution of Bovine Serum Albumin with Cattle Blood Serum and Egg White in CEP-2 Diluent on the Quality of Limousin Bull Liquid Semen during Refrigerating)
This study aims to determine the effect of the substitution of bovine serum albumin (BSA) with cattle blood serum and egg white in the diluent of Cauda epididymal Plasma 2 (CEP-2) on sperm quality of Limousin cattle during cooling at 3-5 C. The research material used was rejected Limousin bull sperm (motility of 50-60%) from Artificial Insemination Centre Singosari, Malang. This research was a laboratory experiment using a randomized block design which was composed of six treatments with 10 replications, those were T0 as controls ((90% CEP-2 with BSA + 10% egg yolk); T1 (83.84% CEP-2 + 6.16% cattle blood serum + 10% egg yolk); T2 (81.84% CEP-2 + 8.16% cattle blood serum + 10% egg yolk); dan T3 (90% CEP-2 + 0,4% egg white + 10% egg yolk); T4 (90% CEP-2 + 0.8% egg white + 10% egg yolk); and T5 (90% CEP-2 without BSA + 10% egg yolk). Parameters measured were the percentage of motility, viability, and abnormality of sperms. Results of research after 48 hours of storage showed that the percentage of sperm motility in T0, T1, T2, T3, T4, and T5 were 40.505.90, 3636.16, 34.006.58, 40.503.69, 38.503.37, and 38.504.12, respectively, while the percentage of sperms viability were 75.164.30, 70.502.88, 73.802.80, 74.803.30, 75.133.13, and 74.034.13, respectively, and the percentage of sperms abnormality were 10.142.34, 10.621.34, 11.332.00, 10.942.82, 10.021.95, and 10.781.96, respectively. PEMBERIAN LIMBAH PERKEBUNAN SINGKONG YANG DISUPLEMENTASI SENG DAN KOBALT TERHADAP PERFORMA REPRODUKSI DOMBA BETINA (Utilization of Cassava Plantation Waste Supplemented with Zinc and Cobalton Reproduction Performance of Ewe)
The study aimed to evaluate the utilization of cassava plantation waste suplemented with zinc and cobalt on ewe reproduction performance. Six pregnacy ewes were allocated at experimentrations, i.e. K1: control (50% fresh grass + 50% concentrate) and K2: 50% fresh grass + 50% cassava plantation waste + 30 ppm zinc + 5 ppm cobalt. The results showed that the concentration of zinc and cobalt in blood of ewe in group K1 and K1 were 7.03 vs 8.22 ppm and 0.48 vs 0.44 ppm, respectively, while in faeces were 147,72 vs 158.59 ppm and 8.3 vs 9.8 ppm. In addition, the concentration of alkaline phosphatase and progresterone in blood were 116 vs 196 u/l and 0.56 vs 0.59 mg/ml, respectively. The average of offsprings weight on 24 day postpartum in K1 and K2 was 3.48 vs 3.16kg. In conclusion, cassava plantation waste could be used as feed for pregnancy and lactating ewe. Supplementation of 30 ppm zinc is efectively increase the concentration of zinc and alkaline phosphatase in blood.Key words: cobalt, reproduction, cassava, zinc, ew
EFEK PENAMBAHAN UMBI BUNGA DAHLIA SEBAGAI SUMBER INULIN TERHADAP KECERNAAN PROTEIN DAN PRODUKTIVITAS AYAM LOKAL PERSILANGAN (Effect of Dahlia Tuber supplementation as Inulin Source on Protein Digestibility and Productivity of Crossbred Local Chicken)
The research was aimed to evaluate feeding effect of powder and extract of dahlia tuber as a sources of inulin prebiotic on protein digestibility and productivity of crossbred local chicken. A total of 280 unsex local crossbred chicken with the age of 22 days and average weight of 180.461.21 g was assigned in a completely randomized design with 7 treatments and 4 replications (10 birds each). Dietary treatments consist of T0 (basal ration), T1 (T0 + 0.4% dahlia powder), T2 (T0 + 0.8% dahlia powder), T3 (T0 + 1.2% dahlia powder), T4 (T0 + 0.39% dahlia extract), T5 (T0 + 0.78% dahlia extract), T6 (T0 + 1.17% extract dahlia). Treatment was conducted for 8 consecutive weeks. Variables observed were protein digestibility, short chain fatty acid (SCFA), muscle protein mass, muscle calsium mass, and body weight. The data were analyzed using ANOVA and followed by Duncan test at the level of 5%. The results showed that feeding inulin in the form of powder or extract of dahlia tuber significantly (P0.05) increased protein digestibility, SCFA level, and body weight. Treatments T2 until T6 significantly (P0.05) increased muscle calsium mass but did not increase muscle protein mass. In conclusion, the used of 1.2% dahlia powder and 1.17% extract dahlia tuber as an inulin source increase protein digestibility and productivity of crossbred local chicken.Key words: inulin, dahlia tuber, crossbred local chicke
POTENSI TRANSDIFERENSIASI SEL FIBROBLAS MENJADI SEL SARAF SECARA IN VITRO (Transdifferentiation Potency of Fibroblast Cell to Neuron Cell in Vitro)
The aim of this study was to examine the potency of fibroblast cells transdifferentiated to neuron cells in vitro. Newborn rat neuron conditioned medium (NBRN CM) was collected from neuron cells cultured with mDMEM without serum for 48 hours. Fibroblast cells were collected from fetal rat muscle treated with trypsin. Fibroblast cells were culture with 3 kind of culture medium: mDMEM + 0.01 mM -mercaptoethanol; mDMEM + 50% NBRN-CM and mDMEM + 0.01 mM -mercaptoethanol + 50% NBRN CM . As control, cells was cultured with mDMEM +10% newborn calf serum (NBCS). The addition of NBRN CM into culture medium resulted in 12.97% newborn cells in fibroblast culture medium passage I. Newborn rat neuron conditioned medium in fibroblast culture medium resulted 12.97% neuron cells at passage 1. The percentage was increased (14.60%) when - mercaptoethanol added into medium. The same result was found at passage 3 (12.67%; 13.17%). It showed that fibroblast cells has potency to transdifferentiated into neuron cells when cultured with NBRN CM. POTENSI EKSTRAK ETANOL LABU SIAM (Sechium edule) UNTUK PERBAIKAN KERUSAKAN SEL BETA PANKREAS DAN KADAR NITROGEN OKSIDA PADA TIKUS YANG MENGALAMI DIABETES MELITUS (The Potenty of Sechium edule Ethanolic Extract to Repair Beta Pancreas Cells and Nitrogen Oxide Concentration in Streptozotocin-induced Diabetic Rat)
The objectives of this research were to examine the effect of Sechium edule ethanolic extract to repair beta pancreas cells damage and tomeasure nitrogen oxide (NO) concentration in streptozotocin-induced rats. Type I diabetic rats were obtained by intraperioneally injected therats with multiple low dose streptozotocin (MLD-STZ) for 5 days. In this research, rats were assigned into 5 treatment groups: untreated rats ingroup 1 were considered as negative control, while MLD-STZ induced type 1 diabetic rats in group 2 were considered as positive control. Inanother 3 groups, type 1 diabetic rats were orally treated with three doses of Sechium edule ethanolic extract (14, 28, 42 mg/kg bw) respectivelyfor 7 consecutive days. Repairing of beta pancreas cells were analyzed descriptively whereas concentration of NO were analyzed using one wayANOVA and continued with LSD test (= 0.01). The result showed that the administration of Sechium edule ethanolic extract significantlydecreased NO concentration (P0.01) up to 67.9 M and repaired beta pancreas cells damage. Dose of 42 mg/kg bw of Sechium edule ethanolicextract is the effective dose for diabetic rat therapy. Key words: Sechium edule extract, diabetic mellitus, rat, beta pancreas cell
DAYA TAHAN HIDUP Toxoplasma gondii DALAM SUSU KAMBING SETELAH PASTEURISASI SUHU TINGGI WAKTU SINGKAT
Tujuan dari penelitian adalah menetapkan daya tahan hidup takizoit Toxoplasma gondii galur RH dalam susu kambing setelah dipasteurisasi pada suhu tinggi dalam waktu singkat. Dalam penelitian ini digunakan metode in vivo dan mencit diinfeksi dengan takizoit Toxoplasma gondii galur RH secara intraperitoneal dengan konsentrasi 2,76x106 takizoit/ekor. Mencit dibagi atas tiga kelompok perlakuan, yaitu susu pasteurisasi dan takizoit yang dipanaskan pada suhu 72 C selama 15 detik (P), susu pasteurisasi dan takizoit tanpa dipanaskan sebagai kontrol positif (KP), dan susu pasteurisasi tanpa takizoit sebagai kontrol negatif (KN). Hasil penelitian menunjukkan bahwa tidak ditemukan takizoit di dalam cairan peritoneal pada P dan KN. Takizoit Toxoplasma gondii galur RH ditemukan pada KP yang mempunyai jumlah konsentrasi hampir sama sebelum dan setelah infeksi
PENGARUH PEMBERIAN MINYAK JINTAN HITAM (Nigella sativa L.) TERHADAP PENINGKATAN SPERMATOGENESIS TIKUS WISTAR YANG TERPAPAR ASAP ROKOK
Penelitian ini bertujuan mengetahui efek minyak jintan hitam (Nigella sativa L.) terhadap spermatogenesis tikus Wistar yang terpapar asap rokok. Jenis penelitian adalah eksperimental murni dengan subyek 24 ekor tikus yang terpapar asap rokok selama 21 hari dan dirandomisasi dalam 4 kelompok yaitu kelompok kontrol dan kelompok perlakuan dengan pemberian minyak jintan masing-masing 0,1; 0,2; dan 0,3 ml. Penegakan hipotesis dilakukan dengan menggunakan Krusskal-Wallis dan Mann Whitney dengan batas derajat kemaknaan P0,05, interval kepercayaan 95%. Pemberian minyak jintan hitam dapat memperbaiki derajat spermatogenesis
