APLIKASI SINBIOTIK UNTUK PENCEGAHAN INFEKSI INFECTIOUS MYONECROSIS VIRUS PADA UDANG VANAME (Litopenaeus vannamei) (Synbiotic Application for Prevention of Infectious Myonecrosis Virus Infection in White Shrimp (Litopenaeus vannamei))
This study aimed to evaluate the effectiveness of dietary synbiotic at different giving frequencies on growth, immune responses, and resistance of white shrimp infected by infectious myonecrosis virus (IMNV). Synbiotic used in this study was combination of probiotic Vibrio alginolyticus SKT-b and prebiotic oligosaccharides extracted from sweet potatoe (Ipomoea batatas L). Doses of probiotic and prebiotic used were 1% and 2% (w/w), respectively. The white shrimps (0.4930.035 g) were divided into five treatments consisting of A and B (without supplementation of synbiotic: (A) positive control; (B) negative control), C (daily synbiotic supplementation), D (twice a week synbiotic supplementation), and E (weekly synbiotic supplementation). After 30 days of feeding trial, white shrimps were infected by IMNV (except negative control). The results showed that daily growth rate of white shrimp on all synbiotic treatments (C, D, and E) ranged from 6.930.025-6.970.019% and had higher values than controls (A and B) (P0.05). Meanwhile, feed conversion value in C and D (1.540.142 and 1.580.117) were lower than controls (P0.05). Supplementation of synbiotic with different frequencies also affected survival rate of white shrimp after the challenge test with IMNV; daily synbiotic supplementation (C) resulted in a 50% higher survival rate than positive control (P0.05). This was associated with immune responses parameters values of synbiotic treatment (before and after the challenge test) which were better than positive control. In conclusion the addition of synbiotic in feed resulted in higher growth performances, immune responses,and resistance of white shrimp to IMNV infection
KAJIAN MOLEKULER LUMBA-LUMBA HIDUNG BOTOL (Tursiops sp.) ASAL LAUT JAWA BERDASAR URUTAN GEN CYTOCHROME C OXIDASE SUB-UNIT II (COX II) (Molecular Studies of Bottlenose Dolphins (Tursiops sp.) from Java Sea by Gene Sequences of Cytochrome C Oxidase Sub-Unit II (COX II))
The purpose of this study was to examine the molecular basis of Tursiops sp. originated from Java sea based on gene sequences ofCytochrome C Oxidase sub-unit II. Samples of blood were collected from five male bottlenose dolphins from PT. Wersut Seguni Indonesiacaptivity. Total deoxyribonucleic acid (DNA) was isolated from blood samples, then amplified by polymerase chain reaction (PCR), sequenced,and the data were analyzed using MEGA version 5.1 program. The amplification result of the PCR product obtained 824 base pairs (bp), andDNA sequencing obtained 684 nucleotides constituent of COX II genes that encode 228 amino acids making up the COX II. Nucleotide andamino acid sequence of COX II bottlenose dolphins originated from Java sea has a very high homology to Tursiops aduncus that are 99.13%and 100% respectively. Phylogram by neighbour joining method based on the nucleotide sequence of COX II gene indicating bottlenose dolphinsfrom Java sea belongs to group of Tursiops aduncus.Key words: bottlenose dolphin, COX II gene, nucleotide, Jawa Se
UJI TOKSISITAS AKUT EKSTRAK ETANOL DAUN MALAKA (Phyllantus emblica) TERHADAP MENCIT (Mus musculus). (Acute Toxicity Test of Ethanolic Extract of Malaka (Phyllantus emblica) Leaves on Mice (Mus musculus))
ABSTRACT The aim of this research was to asses the acute toxicity of ethanolic extract of malaka leaves using lethal dose 50 (LD50) on mice (Mus musculus). Twenty male mice weighing between 20-30 g were randomly divided into 4 groups (group K1-K4) of 5 mice each. All mice in group K1, K2, K3, and K4 were administered ethanolic extract of malaka leaves with the dose of 2, 4, 8, and 16 g/kg bw, respectively. Single dose of ethanolic extract of malaka leaves were given by oral gavage prior to clinical observation . The observation period was 14 days post administration, for sign of toxicity symptom, weight loss, and mortality. The result showed that no mortality was observed in the experimental animals during this study. Slight reduction of body weight was observed in group K2, K3, and K4, and no toxicity sign was found during fourteen days of observation. The LD50 of ethanolic extract of malaka leaves was higher than 16 g/kg body weight, thus, the substance was practically non toxic substance
DIVERSITAS GENETIK DAN HAPLOGROUP KAMBING GEMBRONG BERSTATUS KRITIS DI KABUPATEN KARANGASEM, BALI (Genetic Diversity and Haplogroup of Endangered Gembrong Goat In Karangasem, Bali)
This study was conducted to evaluate the genetic diversity and phylogeny of Gembrong goat. For this purpose, 21 goats from endangered breed in Karangasem Bali were used. Molecular analysis of genetic diversity and phylogeography used hypervariable segment 1 of mitochondrial DNA control region. The result showed that genetic variability of Gembrong goat was homogeneous with only one different sites, namely the substitution pyrimidines of C T (transitional). Phylogeny analysis results showed maternal origin of Gembrong goat is lineage (subhaplogroup) B1 with frequency of 100%. Haplogroup B were known has been domesticated from wild goat in western Asia, then headed to south Asia and infiltrated to southeast Asia, including Gembrong goat in Bali, Indonesia. As a conclusion, genetic diversity of Gembrong goat from remaining population in Karangasem very low and originate from lineages/haplogroup B1 with a frequency of 100%
VERIFIKASI UJI CEPAT KOMERSIAL Escherichia coli PADA CONTOH UJI DAGING SAPI BEKU (Verification of Escherichia coli Commercial Rapid Test Kit on Frozen Meat)
This study was aimed to assess the performance and suitability of commercial rapid test compared to the conventional test through verification process. This study used frozen meat from laboratory routine samples which divided into five groups, those were: natural group, low bacteria level group, medium bacteria level group, high bacteria level group, and control group, each sample test performed 9 replicates. All samples were tested for E. coli by conventional method (SNI 2897:2008) and commercial rapid test method. E. coli test result from both methods was calculated as precision (relative standard deviation), sensitivity, specificity, false negative, false positive, and kappa. The results showed that the commercial kit test had good precision with relative standard deviations score was 0.103. The sensitivity, specificity, false negative, false positive, and kappa score were 94.44%, 100%, 5.56%, 0%, and 0.872, respectively, indicates an equal performance with conventional method. The t student test showed that commercial rapid test method and conventional method had suitability on natural group, low bacteria levels group and medium bacteria level group
RESISTENSI Salmonella spp. TERHADAP BEBERAPA ANTIBIOTIK PADA DAGING ITIK DI KABUPATEN BOGOR YANG DAPAT MEMENGARUHI KESEHATAN KONSUMEN (Resistance of Salmonella spp. to Several Antibiotics from Duck Meat in Bogor District that Could Influence Consumer Health)
This study aimed to determine the presence of Salmonella spp. in duck meat and to identify the resistance of Salmonella spp. against several antibiotics in duck meat associated with food safety. Total of 52 meat samples of ducks was collected from 5 subdistricts in Bogor District, i.e., Cariu-Jonggol, Gunung Sindur, Klapanunggal, Parung Panjang, and Ciomas based on the assumption of 95% confidence level, 50% predicted prevalence, and 10% standard error. The results showed that three samples (5.8%) were positive Salmonella spp. The majority of Salmonella spp. isolated from duck meat showed resistance against erythromycin (66.7%), streptomycin (33.3%), and chloramphenicol (33.3%). Nevertheless, Salmonella spp. was still sensitive againts enrofloxacin, trimethoprim sulfamethoxazole, cephalothin, ampicillin, nalidixid acid, tetracycline, and gentamicin
ISOLASI DAN IDENTIFIKASI VIRUS AVIAN INFLUENZA PADA BERBAGAI SPESIES UNGGAS SECARA SEROLOGIS DAN MOLEKULER (Isolation and Identification of Avian Influenza in Different Species of Poultry by Means of Serological and Molecular Methods)
The purpose of this research was to identify avian influenza (AI) virus using serological and molecular methods on poultry which suspected as AI infected in Aceh province. This study used 37 samples of tracheal and cloacal swabs and organs from various species of poultry that were collected from several districts/cities in Aceh. Samples were collected and put into transport media and stored at 4 C before sending to the laboratory. Samples were inoculated in specific pathogen-free of embryonated chicken egg with the age of 9-11 days for further serological and molecular examination. From 37 samples which infected to embryonated chicken egg then followed by hemagglutinin agglutination test/hemagglutinin inhibition revealed that 7 samples were positively infected with AI virus. The amplification result of specific matrix gene primer was followed by electrophoresis on 2% agarose gel which were obtained in the form of a deoxyribonucleic acid (DNA) band at 276 bp for matrix gene and 1.725 bp for H5 gene for all isolates test. In conclusion, the virus which caused the death of various types of poultry in Aceh province is avian influenza A virus subtype H5.Key words: avian influenza virus, H5N1, serologic, matrix, heamaglutini
PEMETAAN GENETIK VIRUS RABIES PADA ANJING SEBAGAI DASAR PENETAPAN PENGENDALIAN PENYAKIT (Genetic Mapping of Rabies Virus in Dogs as a Basis for Disease Control)
The purpose of this study was to determine the genetic characteristics of rabies virus based on phylogenetic relationship among rabies virus in various regions in Indonesia. The amino acid sequence of the nucleoprotein gene of rabies virus isolate from Banten (RV/Banten-01/dog/2007), Makasar (RV/MKS-26/dog/2010), Bukit Tinggi (RV/BKT-52/dog/2009 and RV/BKT-58/dog/2009), Medan (RV/Medan-27/dog/2007) and Bali (RV/Bali-1/dog/2009; RV/Bali-2/dog/2009; RV/Bali-3/dog/2009), Indonesia was determined. These isolates showed a high degree of homology among Indonesian isolates which reached 100%. Meanwhile, the level of homology between rabies virus isolates from cats rabies virus isolates from dogs reached 97%. Results of phylogenetic analysis using the amino acid sequences of the N genes showed that all of Indonesian rabies virus isolates were closely related to rabies viruses from China than those from Thailand, Laos, Burma, and Vietnam which geografically closer to Indonesia. Data obtained from the phylogenetic analysis is expected to trace the source of rabies spread and the possibility to create a vaccines which more suitable with rabies virus that spreads in Indonesia. Based on the phylogenetic relationship analysis using the amino acid sequence of the rabies virus N protein gene showed that all of rabies virus isolated from Indonesian regions share a high homology with others ranging from 97-100%.Key words: sequencing, rabies, nucleoprotein gene (N), homolog
PREVALENSI DAN FAKTOR RISIKO KOKSIDIOSIS PADA SAPI PERAH DI KABUPATEN BANDUNG (Prevalence and Risk Factor of Coccidiosis in Dairy Cattle in Bandung District)
This study aimed to determine the prevalence and associated risk factors of coccidiosis. Samples were obtained from 400 dairy cattle (196 calves aged 6 months, 37 calves aged 6-12 months, and 167 calves aged 12 months). Feces samples were collected, examined and counted for prevalence and number of oocyst per gram faeces (OPG) by McMaster technique. A questionnaire was design to record information about animal health and husbandry, individually. Risk factors associated with the prevalence of Eimeria in cattle were analyzed by logistic regression model. The overall prevalence and the average of OPG of Eimeria in cattle was 179 (44.75%) and 286.75, while highest prevalence of Eimeria was observed in calves aged less than 6 months. Cattle aged more than 12 months showed significantly different relationship (P0.05) to the prevalence of Eimeria infection compare to calves aged less than 6 months and aged 6-12 months. The presence of an immature immune system in younger calves resulting in their higher susceptibility to coccidiosis. Among management and animal health practices, floor type and treatment of cattle influence the prevalence of Eimeria in cattle
