An immunotherapeutic method for COVID-19 patients: a soluble ACE2-Anti-CD16 VHH to block SARS-CoV-2 Spike protein
The third outbreak of coronavirus (CoV) infection (after SARS-CoV and MERS-CoV) caused by a novel CoV (SARS-CoV-2) of the genus Beta-coronavirus has become a global pandemic. CoVs are enveloped viruses whose proteins include spike (S), membrane (M), and envelope (E) which are embedded in the viral envelope. The glycosylated S protein, which forms homo-trimeric spikes on the surface of the viral particle, mediates viral entry into host cells. SARS-CoV-2, like SARS-CoV, uses the Angiotensin-Converting Enzyme 2 (ACE2) cell surface protein for cellular entry. An attractive anti-viral approach is targeting virus entry into cells, for which three strategies are suggested: 1) direct targeting of the viral glycoprotein; 2) targeting the viral receptor on the cell surface; and 3) using soluble (s) ACE2 that binds to S protein thereby neutralizing the virus. In this article, the advantages and disadvantages of these strategies are explained. Moreover, we propose that fusion of the sACE2 to anti-CD16 to produce a bi-specific molecule could be a promising anti-viral strategy
Levels and ecological and health risk assessment of PM2.5-bound heavy metals in the northern part of the Persian Gulf
Bushehr, a port along the northern part of the Persian Gulf, has repeatedly encountered dust storms in recent years but there is not been a comprehensive study on the PM2.5 contents in this region. The present study reports the characteristics and health risks of atmospheric PM2.5-bound heavy metals (HMs) in Bushehr from December 2016 to September 2017. A total of 46 samples were analyzed, and a high volume air sampler equipped with quartz fiber filters was used for sampling. Inductively coupled plasma optical emission spectroscopy (ICP-OES) was also used for HMs analyses. Risk assessment and hazard index (HI) of these metals were computed by using USEPA’s exposure parameters. The results showed that the average 24-h mass concentration of PM2.5 ranged from 22.09 to 292.45 μg/m3. The results also indicated that 95.65 and 82.61% of the samples were higher than WHO and EPA guidelines for 24-h PM2.5. Also there was no statistically significant relationship between wind direction and PM2.5.The average concentration levels of seven measured metals (Cd, Co, Cr, Fe, Ni, Pb, and V) in the PM2.5 samples were in the range of 6.03 ng/m3 to 1335.94 ng/m3, and the order of their concentration was Fe > Ni > Pb > Cr > Cd > V > Co. Principal component analysis (PCA) showed that PM2.5-bound heavy metals were categorized in three groups. The ecological risk level of calculated metals was very significant, and the major contribution of the ecological risk was related to Cd. The highest HQ in children and adults was related to Cr, and overall HI in children was higher than adults. Also the RI values of Cr in both groups of children and adults were indicated high risk of developing cancer in human
Toxoplasma gondii ROP38 protein: Bioinformatics analysis for vaccine design improvement against toxoplasmosis
Rhoptry proteins (ROPs) play a significant role in various stages of Toxoplasma gondii (T. gondii) life cycle, being critical for both invasion and intracellular survival. ROP38 is a key manipulator of host gene expression and has a function in tachyzoite to bradyzoite conversion. In this study, we've employed various bioinformatics online tools for immunogenicity prediction of ROP38 protein, comprising physico-chemical, antigenic and allergenic profiles, transmembrane domain, subcellular localization, post-translational modification (PTM) sites, secondary and 3D structure, B-cell, MHC-binding and cytotoxic T-lymphocyte (CTL) epitopes. The findings showed 54 PTM sites without a transmembrane domain. Also, ROP38 was proved a non-allergenic and antigenic protein. The protein had Sec signal peptide (Sec/SPI) with 0.8762 likelihood. The secondary structure included 52.68% random coil, 29.57% alpha helix and 17.74% extended strand. Based on Ramachandran plot output for refined model, 95.3%, 3.4%, and 1.4% of amino acid residues were incorporated in the favored, allowed, and outlier regions, respectively. B-cell epitopes TFPGDDIQTSS (67–72) and KAKNKWGRTRYTLQG (207–221) as well as T-cell epitope LSPVGFFTAL (6–15) possessed the highest antigenic index in the protein sequence. This paper is a premise for further researches, and provides insights for the development of a suitable vaccine against toxoplasmosis. More empirical studies are required using the ROP38 alone or in combination with other antigens/epitopes in the future
Perceived discrimination and subjective well-being among Middle Eastern migrants in Australia: The moderating role of perceived social support
Background: Middle Eastern (ME) migrants are vulnerable to developing mental health problems due to pre-migration and post-migration traumas and stresses. The evidence on the subjective well-being of ME migrants and its contributing factors is limited and inconclusive. Objectives: The aim of this study is to examine the moderating role of perceived social support in the association between perceived discrimination and subjective well-being, after controlling for socio-demographic factors. Method(s): This cross-sectional survey study was conducted in Queensland, Australia. A total of 382 first-generation young adult ME migrants, aged between 20 to 39 years, completed a self-administered questionnaire. To analyse data, a series of three hierarchical multiple regression (HMR) analyses were conducted, one for each of the subjective well-being components (i.e., positive affect, negative affect, and satisfaction with life). The interaction between perceived social support and perceived discrimination for the prediction of subjective well-being components was further explored using simple slope analysis. Results: Gender was found to be a significant predictor of positive and negative affect. Education was found to predict satisfaction with life, but not positive and negative affect. Married migrants were found to have significantly higher life satisfaction, positive affect and lower negative affect. Perceived social support had a moderating role in the association between perceived discrimination and subjective well-being. Conclusion: Considering the buffering role of perceived social support against the adverse effects of perceived discrimination on subjective well-being, organizations and individuals working with ME migrants should acknowledge the significance of social support and improve their capacity to deliver effective and appropriate social support services to migrants experiencing discrimination
Prospective evaluation of phosphorus-32 radiation synovectomy in patients with severe and chronic rheumatoid arthritis unresponsive to conventional medical treatment
OBJECTIVES: To determine the efficacy of Phosphorus-32 radiation synovectomy in the treatment of patients with severe rheumatoid arthritis (RA) unresponsive to systematic or local medical therapy. METHODS: Twenty-three knees in 16 patients with severe chronic RA were prospectively treated by intra-articular installation of Phosphorus-32 and evaluated at 1, 3, 6, and 12 months. The Western Ontario and McMaster Universities Arthritis index (WOMAC), visual analog scale (VAS), Health Assessment Questionnaire (HAQ), pain scale, and grade of joint effusion and tenderness were assessed. At the 12-month time interval, the treated knees were classified into four categories: excellent response (no symptoms), good response (significant reduction of symptoms), moderate response (slight decrease), and poor response (no change or worsening). RESULTS: Excellent response or good response was observed in 13/23 (56.5%) of the treated knees, moderate response in 2/23 (8.7%) of the treated knees, and poor response in 8/23 (34.8%) of the treated knees. The grades of effusion and tenderness were significantly decreased one-month posttreatment (P 0.05). There was a significant improvement in the HAQ, WOMAC, VAS, and pain scores at all time points compared with baseline, including at the 12-month time interval (P < 0.05). CONCLUSION: Phosphorus-32 radiosynovectomy is an effective therapeutic treatment for patients with RA of the knee unresponsive to conventional pharmacotherapy
Worldwide bottled water occurrence of emerging contaminants: A review of the recent scientific literature
Contaminants of emerging concern (CECs) have recently been detected in bottled water and have brought about discussions on possible risks for human health. However, a systematic review of CECs in bottled water is currently lacking due to the relatively new introduction and/or detection of these pollutants. Hence, this paper reviews the existing studies on the presence of six major groups of emerging contaminants including microplastics (MPs), pharmaceuticals and personal care products (PPCPs), bisphenol A (BPA), phthalates, alkylphenols (APs), and perfluoroalkyl and polyfluoroalkyl substances (PFASs) in bottled water from different countries. Also, the findings related to CECs’ levels, their possible sources, and their risks are summarized. The gathered data indicate that MPs within the size range of 1−5 μm are the most predominant and potentially toxic classes of MPs in bottled water. In addition, PPCPs, PFASs, APs, and BPA occur in concentration levels of ng/L, while phthalates occur in the μg/L level in bottled water. The bottle type plays an important role in the contamination level. As expected, water in plastic bottles with plastic caps is more polluted than in glass bottles. However, other sources of contamination such as contact materials during cleaning, bottling, and storage are not negligible. Based on the gathered data in this review, the CEC levels except for MPs (no threshold values) in bottled water of most countries do not raise a safety concern for the human. However, the occurrence of individual CECs and their association in bottled water need more accurate data to understand their own/synergistic effects on human health
Rational design of hypoallergenic vaccines: Blocking ige-binding to polcalcin using allergen-specific igg antibodies
Chenopodium album polcalcin (Che a 3) is characterized as a major cause of cross-reactivity inallergic patients to the Chenopodiaceae family. Therefore, the present study was conducted to develop a hypoallergenic Che a 3 derivatives as the candidate vaccine for type 1 allergy. Four derivatives were generated from Che a 3. The first was a mosaic peptide derivative computationally identified in Che a 3 which was coupled to keyhole limpet hemocyanin (KLH). The second one was a mutant Che a 3, and the other two derivatives included N-and C-Terminal halves of Che a 3 that both coupled to KLH. The IgE-binding capacity of Che a 3 and its derivatives and also their ability to induce there combinant Che a 3 (rChe a 3)-specific IgG antibody, were determined using the enzyme-linked immune sorbent assay (ELISA). Moreover, the lymphopro liferative capacity of rChe a 3 or its derivatives and their pro-inflammatory cytokine response interleukin (IL)-5 and IL-13 were measured in the human peripheral blood mononuclear cells (PBMCs). Among all derivatives, the N-Terminal half peptide and mosaic peptide exhibited the lowest IgEbinding capacity. In addition, in comparison to other antigens, KLH-coupled mosaic peptide induced the highest level of the recombinant Che a 3 (rChe a 3)-specific IgG antibody and ther Che a 3 specific-blocking IgG antibody in mice. Moreover, the mosaic peptide lacked lymphopro liferative capacity and down-regulated expression of pro-Allergic IL-5 and IL-13 cytokines. Therefore, a peptide-carrier fusion vaccine, composed of the B-cell epitope coupled to the carrier, could be considered as one of the promising hypoallergenic vaccines to treat patients with allergy to low molecular weight allergens such as Che
Low-power density radiations emitted from common wi-fi routers influence sperm concentration and sperm histomorphometric parameters: A new horizon on male infertility treatment
Background: Male infertility is defined as an inability to impregnate a fertile fe-male; it is a widespread problem which is usually caused by some male factors such as low quantity and quality of sperm, specifically oligospermia and azoospermia. Objective: This study aimed to evaluate the bio-positive effects of low power density Wi-Fi radiation on the reproductive system of infertile and healthy mice. Materials and Methods: In this experimental study, thirty adult male Balb/c mice were randomly divided into 5 groups. Groups oligospermic-sham (OS), oligo-spermic-exposure 1 (OE1) and oligospermic-exposure 2 (OE2) received Busulfan, 10 mg/kg, intraperitoneally, but the control-sham (CS) and control-exposure (CE) groups left without Busulfan therapy. Groups CE, OE1 and OE2 were exposed to 2.4 GHz Wi-Fi radiation while, the CS and OS were sham exposed to Wi-Fi radiation without energizing the Wi-Fi router. The right and left testes and right epididymis were dissected out and histopathological, histomorphologic changes and the quality of the sperms were analyzed. Results: Low power density Wi-Fi radiation significantly increased sperm concentration in the CE group compared to that in CS, while enhancement of spermatid cells was not significant. Sperm concentration in OE2 was more than that in OE1 as the spermatid cells enhanced. Conclusion: Findings revealed that radiation hormesis induced by low power density Wi-Fi radiation have biological beneficial effects on mouse sperm concentration and sperm histomorphometric parameters
Production and Characterization of Monoclonal Antibody against Vit v1: A Grape Allergen Belonging to Lipid Transfer Protein Family
Allergy to non-specific lipidtransfer protein (nsLTP), the major allergen of grape (Vit v1), is considered as one of the most common fruit allergies in Iran. Therefore, a specific monoclonal antibody (mAb) can be used for the characterization and assessment of. Accordingly, this study aimed to generate and characterize a mAb against Vit v1 with a diagnostic purpose. To this end, Vit v1 allergen (9 kDa) was extracted using a modified Bjorksten extraction method. Natural Vit v1-immunized mouse splenocytes were fused with SP2/0Ag-14 myeloma cells for generating hybridoma cells. Specific antibody-secreting Hybridoma cells were selected using ELISA. Finally, anti-Vit v1 mAb was characterized by western blotting, ELISA, and isotyping methods. In the current study, a 9 kDa (Vit v1) protein was attained fromcrude and fresh juice of grape extracts and the isotype of desired anti-Vit v1 mAb was determined as IgM with K light chain. In addition, The ELISA results demonstrated that anti-Vit v1 mAb was specified against natural Vit v1 in the grape cultivar and related LTP allergens, such as Pla or 3 (p<0.0001). In the present study, a specific mAb was produced for detecting the LTP allergen. This mAb with a confirmed specificity can be utilized for evaluating the LTP allergens and their allergenicity in different grape cultivars. © 2020 Tehran University of Medical Science
Development and implementation of water safety plans for groundwater resources in the southernmost city of West Azerbaijan Province, Iran
The transfer of water from the source to the consumption point is always associated with the possibility of contamination in any of its various components. To resolve this problem, the World Health Organization has considered a water safety plan. The purpose of this study is to implement water safety plan in the water supply system of Bukan city. This study was performed on Bukan’s water supply system in 2019–20 using a software to guarantee the quality of the water safety plan and the WHO and IWA guidelines. The software checklists were prepared and after confirming the validity of the translation and its facial and content validity, it was completed based on the records of the Water and Sewerage Company and interviews with experts. Out of a total of 440 points of full-application of the program and 392 points for the reviewed phases, 183.6 points were acquired and 43.7% of WSP-coordinated implementation was observed. The highest percentage of WSP-coordinated implementation (75.2%) was assigned to the validation stage with the highest point, and the support program stage had the lowest percentage of performance (1.1%). Among the major components of the water supply system, the final consumption point received the most attention from the system. Given the lifespan of the introduction and use of WSP in the world, it was expected that better results would be obtained from evaluating the implementation and progress of this approach in Bukan’s water supply system. However, the implementation rate of this program in this city compared to other cities in Iran, showed that according to the implementation time (one year), the obtained results are relatively convincing and good and the water supply system has a moderate level of safety
