STUDI PENETAPAN KADAR LOSARTAN DENGAN METODE SPEKTROFOTOMETRI DAN HIGH PERFORMANCE LIQUID CHROMATOGRAPHY (HPLC) SERTA APLIKASINYA PADA TRANSPOR TRANSDERMAL in vitro
The development of drug delivery system needs a usefull determination method ofdrug in many kind of samples. This research was purposed to develop a determinationmethod of losartan, an angiotension receptor antagonist II, from the sample of in vitrotransdermal transport. Three methods were studied, i.e. normal spectrophotometric, 1stderivative spectrophotometric, and HPLC. The spectrophotometric method wasconducted using Spectrophotometer Shimadzu tipe UV 1700 controlled by UV Probesoftware (Shimadzu), while HPLC method was performanced by Shimadzu HPLCcontrolled by LC Solution software (Shimadzu). Lichrospher RP 18 250-4 (5 µm) wasused as stationary phase and acetonitril-acetic buffer 0,01 M pH 4 (60:40) was used asmobile phase. Chromatogram was recorded using UV 223 nm and 254 nm as adetector. Some parameters of determination method performance were calculated, i.e.LOD, LOQ, recovery, systemic error, and random error. The results shown thatspectrofotometric methods did not have an enough performance parameters to use intransdermal transport of losartan, while HPLC method had LOD and LOQ 27,329 and91,098 ng/ml using UV 223 nm as detector and 36,178 and 120,590 ng/ml using UV 254nm as detector. Detector UV 223 nm was selected. This HPLC method had recovery106,405% and random error 3,71%, and could be used to determine the losartanconcentration in sample from transdermal transport in vitro
ISOLASI DAN UJI PENANGKAPAN RADIKAL BEBAS DPPH OLEH ISOLAT-1, FRAKSI ETIL ASETAT, DAN EKSTRAK ETANOL AKAR PASAK BUMI (Eurycoma longifolia Jack)
The free radical was produced when undertaking processes metabolit or opposedinfection and when the body dissolved food. The unstable free radical could beneutralised with antioxidant. The root of the pasak bumi (Eurycoma longifolia Jack)was known contained the compound flavonoid, that it was known had the activity asantioxidant. This research aimed at knowing the activity capacity of the capture of thefree radical the water fraction and the faction ethyl acetate fraction the extract ethanolof the root of the pasak bumi by using the DPPH method.The root of the pasak bumi was extracted with ethanol 70% used maceration. Theethanolic extract was dissolved in ethyl acetate, fractination with ethyl acetate so as tobe received by the fraction ethyl acetate. The concentration of the ethanolic extract andthe fraction of ethyl acetate that was used were 2; 4; 8; 16 ug/mL and isolat-1 that is:0.8; 1.6; 3.2; and 6.4 ug/mL.Results of the research showed that all the treatments had the activity asscavenging the free radical. Results of the analysis statistika with the Kruskal Wallismethod in the level of the confident 95% that was followed by the test Mann Whitneyshowed the existence of the difference of the activity of the capture of the free radicalwho was significant between respectively the treatment group. The conclusion from thisresearch was the value ES50 ethanalic extract (15.64 ug/mL) bigger than the factionethyl acetate fraction and higher (13.948 ug/mL) value of ethanalic extract was thanisolat 1 (3.961 g/mL)
GAMBARAN PASIEN GERIATRI MELAKUKAN SWAMEDIKASI DI KABUPATEN SLEMAN
The increase in life expectancy caused geriatric population is also increasing. Geriatric patient is identicalto the decline in physiological function that can lead to various diseases. Self medication in geriatric requirespecial attention from the pharmacist. The purpose of this study was to determine the description of geriatric thatdoes self medication and assosation factors in geriatric patients doing the self medication in Sleman. This researchwas conducted using a survey to determine the presence of pharmacy in Sleman. Determination of pharmaciesconducted with a purposive sampling technique and determination of areas of research subjects or respondentswith accidental sampling. Subjects were geriatric patients with age> 60 years who did self medication inpharmacies. The data were analyzed by using chi-square statistic to determine which are the most influentialassociation factors. The research subjects in this study was 38 respondents. The pattern of disease that occur ingeriatric patients who performs a self-medication is a chronic 12 respondens, non-respondens and 16 non chronicrespondens ten respondens is unknown. Factors that association geriatric patients that is doing self medication isown of health insurance. The results of the chi square test showed that the sex factor with OR 1,11 ( p >0,05),ownership asurance (OR = 1,841,p < 0,05,CI : 1,184 – 2,184) is a factor of self association with geriatric patientsmedications at the pharmacy of district Sleman
EFEK EKSTRAK ETANOL RIMPANG TEMULAWAK (Curcuma xanthorrhiza Roxb) TERHADAP MEMORI SPASIAL TIKUS MODEL DEMENSIA YANG DIINDUKSI TRIMETHYLTIN
Curcuma xanthorrhiza Roxb is one of the plants containing curcumin. Curcumin has anti-amyloidogenic,antioxidant, and anti-inflammatory activity that can prevent dementia. The aim of the research was to know theeffect of Curcuma xanthorrhiza Roxb rhizome ethanol extract (CREE) to spatial memory on dementia model ratinduced by trimethyltin (TMT). This research use 30 rats, divided into 6 groups, each group consisted of 5 rats. Allgroupswere given trimethyltin intraperitoneal lyexcep tgroup I.Group I (healthy controls) and group II (TMT/illness control) were given a solution of 1% CMC-Na orally. Group III (drug control) were given citicoline 200mg/kgBW orally. Group IV, V, and VI were given CREE orally dose 50 mg/kgBW, 100 mg/kgBW, and 150mg/kgBW respectively. Data of spatial memory were obtained by Morris water maze test consisted of acquisitiontrial, and a probe trial. Data from the test then statistically analyzed by one-way repeated measure ANOVA.Morris water maze test showed that there were significant differences (p<0,05) between TMT group comparedwith CREE dose 50 mg/kgBW, 100 mg/kgBW, and 150 mg/kgBW group. But there was no significant difference(p>0,05) between CREE dose100 mg/kgBW group anddrug control group.Based on the research results can beconcluded that ethanol extract Curcuma xanthorrhiza Roxb rhizome at dose of 100 mg/kgBW can improvespatial memory on dementia model rat induced by trimethyltin
EFEK ULCEROGENIC DISPERSI PADAT IBUPROFEN-POLIVINILPIROLIDON (PVP) PADA TIKUS PUTIH JANTAN
Ibuprofen is an anti inflammatory, analgesic, and antipyretic agent. It has low solubility and bioavailability.The administration of ibuprofen in a long time and high dose can increased of side effect at gastrointestinal (pepticulcer). This study was aimed to determine the influence its solid dispersions formation on ulcerogenic effect ofibuprofen buprofenis compared with ibuprofen alone and physical mixture. Solid dispersion of Ibuprofen-PVP(1:1) was prepared by solvent method. The ulcerogenic test was conducted by observing is white rat, dividedrandomly into 5 groups are healthy group (aquadest), suspending agent control (PVP 1%), drug (ibuprofen) at adose of 360 mg/kg body weight, group solid dispersion (ibuprofen-PVP) and group physical mixture(ibuprofen-PVP). Rats gastric was observed of formation ulcer and evaluated by scoring then followed thecalculation by the index ulcer. The results showed that the number of ulcer index of ibuprofen is 6,67. Number ofulcer index of solid dispersion was 4,00 and physical mixture was 3,34. The ulcerogenic effect of solid dispersionand physical mixture difference was significant (p<0,05) compared with ibuprofen alone. Ulcerogenic effectadministration of solid dispersion was smaller than ibuprofen alone and statistically no different (p>0,05)compared with physical mixture
BIOAVAILABILITAS TABLET IBUPROFEN PADA PEMBERIAN BERSAMAAN DENGAN EKSTRAK AIR HERBA PEGAGAN (Centella asiatica (L) Urban) PADA KELINCI JANTAN
The tendency to use herbal drug simultaneously with synthetic drug may result inan interaction. Based on research, aqueous extract of Centella asiatica (L) Urbancontains saponin (asiaticoside and madecassoside) compounds that inhibit CYP450and is a safe herbal alternative that is used for anti-inflammatory treatment. The aim ofthis study was to investigate the effect of aqueous extract of Centella asiatica (L) Urbanto the ibuprofen tablet bioavailability in male rabbits. Both compounds are metabolizedby CYP450 enzymes in the same way with different mechanism. The cross over designwith the same subject was used in this study. The samples consisted of 400 mg ibuprofenwith 0%b/v, 25%b/v, 50%b/v dan 100%b/v aqueous extract of Centella asiatica (L)Urban respectively. The blood were taken after 0; 0,5; 1; 1,5; 2; 2,5; 3; 3,5; 4; 6; 8; 10hours after samples treatment, then the ibuprofen plasma concentration weredetermined spectrophotometrically. The results then were used to evaluat thebioavailability parameters: tmaks, Cpmaks, and AUC and were analyzed using ANOVAtest in the same way as well as the 95% trust standart. The results showed, the use ofibuprofen tablet stimultaneously with aqueous extract of Centella asiatica (L) Urbanincreased Cpmaks dan AUC, but there were no influence to the tmaks value. The use ofibuprofen tablet stimultaneously with 50 %b/v and 100 %b/v aqueous extract ofCentella asiatica (L) Urban had a significant influence to the bioavailability ofibuprofen
PENETAPAN KADAR Pb PADA SHAMPOO BERBAGAI MERK DENGAN METODE SPEKTROFOTOMETRI SERAPAN ATOM
The source of lead pollution (Pb) can come from cosmetic product including shampoo. The purpose of thisresearch are to know wether there is Pb in the shampoo cosmetic based on different brands and colours of theshampoo. The sample of the shampoos that used in this research are A, B, C, and D. Each of the samples replicated5 times, then analyzed with atomic absorbtion specktrophotometry. Then, one of the sample are taken from onebrand with different colour and replicated 5 times. The result of this research shows that from the different brandof shampoos contain Pb. The average level of Pb that obtained from analysis are, brand A (0,4838 ppm 0,0538ppm), brand B (0,6935 ppm 0,3131 ppm), brand C (0,8354 ppm 0,0405 ppm), and brand D ( 0,9517 ppm0,0811 ppm). Whereas for the same shampoo brand but different in colour had average level of Pb: shampoo I (redcolour)= (0,5428 0,0192) ppm, Shampoo II (yellow colour) = (0,3606 0,0255) ppm, Shampoo III (blue colour)= (0,4385 0,0062) ppm, and Shampoo IV (green colour) = (0,1427 0,0133) ppm
RESIDU PESTISIDA ALDRIN DAN DIELDRIN PADA SAMPEL TANAH DAN AIR DI DESA SRIGADING KECAMATAN SANDEN KABUPATEN BANTUL
Aldrin and dieldrin pesticide residue in soil and water samples from Srigading Village, Sanden District,Bantul Regency has been investigated. The purpose of this study were to analyze, identificate and evaluatepyrethroid pesticide residues in soil and water samples. Soil sample was extracted using shaker with acetonesolvent. Water samples was extracted using separator funnel with 15% (v/v) dichloromethane/n-hexanesolvent.Clean-up was couducted using chromatography column using florisil and determination of pesticideresidue in the samples was carried out by gas chromatography apparatus equipped with electron capture detector(GC-ECD). The result showed that pesticide residues in soil sample in the range level: aldrin (4,8-64,8 ppb) anddieldrin (not detected-6,0 ppb). In water sample pesticide residues in the range level: aldrin (not detected-1,0 ppb)dan dieldrin (-not detected1,2 ppb). The quality or water sample taken from onion field is laid under the standardof water that has been settled
IMPACT OF LIMITED ORAL ANTITUBERCULOSIS INFORMATION TO THE TUBERCULOSIS PATIENTS' COMPLIANCE AND THEIR QUALITY OF LIFE
The study about impact of limited oral antituberculosis information delivered bypharmacist to patient compliance and their quality of life in two public health centers inYogyakarta has been carried out. This study was performed using cohort eventmonitoring design, with 14 pulmonary TB patients and positive Acid Resistant Basilwere treated with oral antituberculosis (OAT) for at least 1 month and were older than15 years old. We observed the compliance of one group of patients using oralantituberculosis who were serviced with OAT information and the other group withoutOAT information service. The patients’ compliance was checked by counting theremaining OAT and by interviewing the patients, meanwhile patient’s quality of lifewere measured by SF-36 instrument. The correlation between limited OAT informationservice and patient compliance were analysed using Chi Square and quality of patient’slife were analysed using Independent t-Test with 95% confidence level. It was shownthat characteristics of patients such as age, gender, education, duration of illness, andDirectly Observed Treatment involvement did not influence patient compliance. Therewas no significant correlation between limited OAT information service to patientcompliance and patients’ quality of life (P>0.05). There is no impact of limited OATinformation service to the TB patients’ compliance and their quality of life. Wesuggested that the further and deeply information service delivered by pharmacists inthe public helath centers in Yogyakarta should be carried out
SINTESIS SILIKA MCM-41 DAN UJI KAPASITAS ADSORPSI TERHADAP METILEN BIRU
Synthesis of MCM-41 with hydrothermal time variation 18, 24, 36, 48 hoursusing templating agent cetiltrimetilammonium bromide (CTAB) has been done. Theresults showed that the time of hydrothermal influence the intensity and the index field(ao). Synthesis of MCM-41 with a time of hydrothermal 24 hours produce the highestintensity which indicates that MCM-41 has the best crystallinity.
