    Does oxytocin directly regulate the atrial contractility in mouse? : Study using the isolated atrial preparations

    BulletinOxytocin is a peptide hormone released from a posterior pituitary gland and acts on myometrium and mammary gland by activation of oxytocin receptor. In addition, oxytocin acts as neurotransmitter and/or neuromodulator in the central nervous system and attenuates the stress-induced hypothalamic-pituitaryadrenocortical axis and sympathetic nervous-adrenal-medullary axis. Although oxytocin regulates the cardiovascular function through peripheral mechanisms in rats and humans, the effects of oxytocin in the mouse heart have never been examined. This study was performed to clarify whether oxytocin directly affects mouse atrial contractility or not. Amplitude and frequency of spontaneous atrial contraction were not changed significantly by bath applied oxytocin (1 nM - 10 μM) in both male and female mice. Oxytocin did not alter the carbachol-induced negative inotropic and negative chronotropic actions. In conclusion, it is not likely that oxytocin affects the mouse atrial contractility acting as hormone different from rats and humans. Action of oxytocin on autonomic nervous center in medulla oblongata has been suggested as function of oxytocin in the mouse heart.departmental bulletin pape

    Generation of floxed Pomk mice by the CRISPR-Cas9 method

    Bulletinタンパク質O-マンノースキナーゼ(POMK)はタンパク質のO-マンノシル型糖鎖の基幹部のマンノースをリン酸化する酵素である。本研究は,組織選択的Pomk コンディショナルノックアウトマウスの樹立に必要なfloxed Pomk マウスの作製を試みた。マウスPomk 遺伝子の第4エクソンを挟む位置にloxP 配列を配置した人工遺伝子をドナーDNA とし,C57BL/6 マウスの受精胚を材料にCRISPR/Cas9 法でゲノム編集を行った。得られた35 匹のF0 マウスの中で,少なくとも5匹の新生仔のゲノムは両側のアレルで目的の編集がなされた。これらを基に,戻し交配を繰り返すことで5系統のfloxed Pomk マウスを作製した。Protein O-mannose kinase (POMK) is a sole enzyme which is responsible for generating of phosphorylated Omannosyl trisaccharide. In this study, we have attempted to establish floxed Pomk mice which are required to cause tissue-specific knockout of Pomk gene. By the CRISPR/Cas9-driven genome editing, we then introduced two loxP sequences in the flanking region of 4th exon of the Pomk gene in embryos. PCR and subsequent DNA sequencing analysis indicated that genomes of five offspring were precisely edited. After repeating the process of back-crossing multiple times with C57BL/6 mice, we established five floxed Pomk lines.departmental bulletin pape
