    Time below range alone is insufficient to identify severe hypoglycaemia risk in type 1 diabetes—the critical role of hypoglycaemia awareness: results from the SFDT1 study

    International audienceAbstract Aims/hypothesis Severe hypoglycaemia events (SHE) remain frequent in people with type 1 diabetes despite advanced diabetes technologies. We examined whether time below range (TBR) 3.9 mmol/l (70 mg/dl; TBR70) or 3.0 mmol/l (54 mg/dl; TBR54) is associated with future SHE risk and whether impaired awareness of hypoglycaemia (IAH) modifies this relationship. Methods We analysed data from participants in the Study of the French-speaking Society of Type 1 Diabetes (SFDT1) who used continuous glucose monitoring. IAH was assessed using the Gold Score (≤2, no IAH; 3, undetermined; ≥4, IAH). SHE frequency was self-reported 12 months after inclusion. We analysed associations between TBR and SHE using logistic regression models adjusted for age, sex, social vulnerability and insulin treatment, including TBR–IAH interactions. We performed spline analyses to explore non-linear patterns. Results One-year incidence of SHE was 11.7% among 848 participants (mean ± SD age 41.6 ± 13.3 years; 53.8% female sex, HbA 1c 57.2 ± 10.9 mmol/mol [7.4 ± 1.0%]). Incidence by TBR70 was 12.1% for ≤1%, 10.2% for 1.1–3.9%, 10.6% for 4–6%, and 14.6% for >6%. Only those with TBR70 >6% and IAH had a significantly higher SHE risk (OR 3.32 [95% CI 1.40, 7.82]) compared with TBR70 ≤1% and no IAH. For TBR54, SHE incidence was 11.0% and 13.3% for categories <1% and ≥1%, respectively. Similarly, only individuals with TBR54≥1% and IAH had increased SHE risk (OR 2.99 [95% CI 1.46, 5.92]). Spline analysis showed low, stable SHE risk across TBR70 values in participants without IAH, with a non-linear pattern only in those with IAH. Conclusions/interpretation TBR alone is not discriminative for high-risk SHE but combining TBR with hypoglycaemia awareness status identifies those at the highest risk for both TBR70 and TBR54.     International audienceAchieving an economically viable transformation of plant cell walls into bioproducts requires a comprehensive understanding of enzymatic deconstruction. Microscale quantitative analysis offers a relevant approach to enhance our understanding of cell wall hydrolysis, but becomes challenging under high deconstruction conditions. This study comprehensively addresses the challenges of quantifying the impact of extensive enzymatic deconstruction on plant cell wall at microscale. Investigation of highly deconstructed spruce wood provided spatial profiles of cell walls during hydrolysis with remarkable precision. A distinct cell wall autofluorescence distribution pattern marking enzymatic hydrolysis along with an asynchronous impact of hydrolysis on cell wall structure, with cell wall volume reduction preceding cell wall accessible surface area decrease, were revealed. This study provides novel insights into enzymatic deconstruction of cell wall at under-investigated cell scale, and a robust computational pipeline applicable to diverse biomass species and pretreatment types for assessing hydrolysis impact and efficiency

    PBK-TD modelling of the gonadotropic axis: Case study with two azole fungicides in female zebrafish

    International audienceEndocrine disruptors (EDs) can disrupt the gonadotropic axis, which consists of the Hypothalamus-Pituitary-Gonads (HPG), notably by altering aromatase (cyp19a), a key enzyme regulating the endocrine system and reproductive function in fish. The effects of EDs can be predicted by integrating both toxicokinetic (TK) and toxicodynamic (TD) processes in order to relate adverse outcomes to external exposures. In this study, we developed a physiologically based kinetic-toxicodynamic model to simulate the disruption of the HPG axis (PBK-TD, hereafter named PBK-HPG) in female zebrafish exposed to either of two aromatase inhibitors, imazalil or prochloraz. The model was calibrated using Bayesian methods and supported by novel experimental data, including measurements of vitellogenin, 17β-estradiol, and 11-ketotestosterone levels, along with in vivo monitoring of the cyp19a1a gene in transgenic cyp19a1a-GFP ebrafish. Seamless integration of a PBK model within a TD model of the HPG-axis, provided the link between external exposure and internal levels of imazalil and prochloraz in key organs, allowing for mechanistic predictions of their inhibitory effects on gonadal aromatase. Our PBK-HPG model accurately predicted both baseline homeostasis and the effects of aromatase inhibition, with all endocrine endpoints including reproductive disruption, i.e., decreased egg production, falling within a twofold range of both experimental and literature data. Therefore, our PBK-HPG model could further support the development of a mechanistic qAOP with TK considerations. The model offers significant potential for improving environmental risk assessments of EDs and possibly other stressors across species

    International audienceBetween January and May 2023, I participated in a Fulbright program at the University of Hawaii at Mānoa, where I explored regenerative tourism. Building on a decade of research into commodification of Hawaiian culture this program helped me shift focus to regenerative practices that benefit local communities and ecosystems. Through interviews with stakeholders in Hawaii's tourism industry, I gained insights into the practical and theoretical aspects of regenerative tourism, deepening my understanding of its challenges and potential for global application.Entre janvier et mai 2023, j'ai participé à un programme Fulbright à l'Université d'Hawaï à Mānoa, où j'ai exploré le tourisme régénérateur. S'appuyant sur dix années de recherche sur la marchandisation de la culture hawaïenne, ce programme m'a permis de me concentrer sur des pratiques régénératrices bénéfiques pour les communautés et les écosystèmes locaux. Grâce à des entretiens avec des acteurs du secteur touristique hawaïen, j'ai approfondi mes connaissances sur les aspects pratiques et théoriques du tourisme régénérateur, approfondissant ainsi ma compréhension de ses défis et de son potentiel d'application à l'échelle mondiale

Centré sur l'héritage, ce séminaire coordonné par Arnaud Coutant, Professeur de droit public, a réuni les contributions des étudiants du Master 2 Droit du vin et des spiritueux de la faculté de droit. Ont participé à cet ouvrage Veronika Alekhine, Camille Delattre Mendez, Julie Desimeur, Naïs Gublin, Lena Lamotte, Margot Limon, Alexis Peccavet et Julie Zanaska. Les contributions ont porté sur des films, des séries télévisées, des bandes dessinées et des romans comportant des références juridiques à l'héritage en matière de droit de l'alcool
