    Drug Addiction and Stress-Response Genetic Variability: Association Study in African Americans

    Stress is a significant risk factor in the development of drug addictions and in addiction relapse susceptibility. This hypothesis-driven study was designed to determine if specific SNPs in genes related to stress response are associated with heroin and/or cocaine addiction in African Americans. The analysis included 27 genes (124 SNPs) and was performed independently for each addiction. The sample consisted of former heroin addicts in methadone maintenance treatment (n = 314), cocaine addicts (n = 281), and controls (n = 208). Fourteen SNPs showed nominally significant association with heroin addiction (p < 0.05), including the African-specific, missense SNP rs5376 (Asn334Ser) in the galanin receptor type 1 gene (GALR1) and the functional FKBP5 intronic SNP rs1360780. Thirteen SNPs showed association with cocaine addiction, including the synonymous SNPs rs237902, in the oxytocin receptor gene (OXTR), and rs5374 in GALR1. No signal remained significant after correction for multiple testing. Four additional SNPs (GALR1 rs2717162, AVP rs2282018, CRHBP rs1875999, and NR3C2 rs1040288) were associated with both addictions and may indicate common liability. The study provides preliminary evidence for novel association of variants in several stress-related genes with heroin and/or cocaine addictions and may enhance the understanding of the interaction between stress and addictions

    Domain-specific hedonic deficits towards social affective but not monetary incentives in social anhedonia

    Anticipatory and consummatory dissociation of hedonic experience may manifest as anhedonia in schizophrenia. However, it is unclear if this temporal dissociation of pleasure experience is also relevant in other symptoms like social anhedonia in the schizophrenia disorder spectrum. The present study applied two incentive delay tasks involving different incentive types (money vs. social affective images) to a sample of 28 participants with elevated social anhedonia (SocAnh) and 38 healthy controls from a population of 476 college students. The results showed that the SocAnh group had comparable anticipatory sensitivity and consummatory pleasure towards monetary incentives as the controls; but they exhibited significant decrease in both anticipatory sensitivity and consummatory experience to positive social affective images. These findings demonstrate the presence of a domain-specific deficit in people with social anhedonia towards social affective information, and suggest that incentive types could confound the findings on the dissociation of anticipatory vs. consummatory hedonic capacities

    Structural and Diffusion Property Alterations in Unaffected Siblings of Patients with Obsessive-Compulsive Disorder

    Disrupted white matter integrity and abnormal cortical thickness are widely reported in the pathophysiology of obsessive-compulsive disorder (OCD). However, the relationship between alterations in white matter connectivity and cortical thickness in OCD is unclear. In addition, the heritability of this relationship is poorly understood. To investigate the relationship of white matter microstructure with cortical thickness, we measure fractional anisotropy (FA) of white matter in 30 OCD patients, 19 unaffected siblings and 30 matched healthy controls. Then, we take those regions of significantly altered FA in OCD patients compared with healthy controls to perform fiber tracking. Next, we calculate the fiber quantity in the same tracts. Lastly, we compare cortical thickness in the target regions of those tracts. Patients with OCD exhibited decreased FA in cingulum, arcuate fibers near the superior parietal lobule, inferior longitudinal fasciculus near the right superior temporal gyrus and uncinate fasciculus. Siblings showed reduced FA in arcuate fibers near the superior parietal lobule and anterior limb of internal capsule. Significant reductions in both fiber quantities and cortical thickness in OCD patients and their unaffected siblings were also observed in the projected brain areas when using the arcuate fibers near the left superior parietal lobule as the starting points. Reduced FA in the left superior parietal lobule was observed not only in patients with OCD but also in their unaffected siblings. Originated from the superior parietal lobule, the number of fibers was also found to be decreased and the corresponding cortical regions were thinner relative to controls. The linkage between disrupted white matter integrity and the abnormal cortical thickness may be a vulnerability marker for OCD

    Temporal and spectral profiles of stimulus-stimulus and stimulus-response conflict processing

    The ability to detect and resolve conflict is an essential function of cognitive control. Laboratory studies often use stimulus-response-compatibility (SRC) tasks to examine conflict processing in order to elucidate the mechanism and modular organization of cognitive control. Inspired by two influential theories regarding cognitive control, the conflict monitoring theory (Botvinick, Braver, Barch, Carter, & Cohen, 2001) and dimensional overlap taxonomy (Kornblum, Hasbroucq, & Osman, 1990), we explored the temporal and spectral similarities and differences between processing of stimulus-stimulus (S-S) and stimulus-response (S-R) conflicts with event related potential (ERP) and time-frequency measures. We predicted that processing of S-S conflict starts earlier than that of S-R conflict and that the two types of conflict may involve different frequency bands. Participants were asked to perform two parallel SRC tasks, both combining the Stroop task (involving S-S conflict) and Simon task (involving S-R conflict). ERP results showed pronounced SRC effects (incongruent vs. congruent) on N2 and P3 components for both S-S and S-R conflicts. In both tasks, SRC effects of S-S conflict took place earlier than those of S-R conflict. Time-frequency analysis revealed that both types of SRC effects modulated theta and alpha bands, while S-R conflict effects additionally modulated power in the beta band. These results indicated that although S-S and S-R conflict processing shared considerable ERP and time-frequency properties, they differed in temporal and spectral dynamics. We suggest that the modular organization of cognitive control should take both commonality and distinction of S-S and S-R conflict processing into consideration. (c) 2013 Elsevier Inc.     Like or Dislike? Affective Preference Modulates Neural Response to Others' Gains and Losses

    Previous studies have demonstrated that the brain responds differentially to others' gains and losses relative to one's own, moderated by social context factors such as competition and interpersonal relationships. In the current study, we tested the hypothesis that the neural response to others' outcomes could be modulated by a short-term induced affective preference. We engaged 17 men and 18 women in a social-exchange game, in which two confederates played fairly or unfairly. Both men and women rated the fair player as likable and the unfair players as unlikable. Afterwards, ERPs were recorded while participants observed each confederates playing a gambling game individually. This study examines feedback related negativity (FRN), an ERP component sensitive to negative feedback. ANOVA showed a significant interaction in which females but not males displayed stronger FRNs when observing likable players' outcomes compared to unlikable ones'. However, males did not respond differently under either circumstance. These findings suggest that, at least in females, the neural response is influenced by a short-term induced affective preference

    Integration of animal behaviors under stresses with different time courses

    We used animal models of "forced swim stress" and "chronic unpredictable stress", and tried to reveal whether a passive coping style of high flotation behavior in forced swim stress predicts anhedonia behavior after chronic unpredictable stress, and whether the dopamine system regulates floating and anhedonia behaviors. Our results confirmed that depression-prone rats use "floating behavior" as a coping strategy in forced swim stress and more readily suffer from anhedonia during chronic unpredictable stress. Intraperitoneal injection or nucleus accumbens microinjection of the dopamine 2/3 receptor subtype agonist ropinirole reduced floating behaviors in depression-prone animals, but increased sucrose preference in rats showing anhedonia. These data indicate that floating behavior is a defensive mode that is preferred by susceptible individuals under conditions of acute stress. Simultaneously, these animals more readily experienced anhedonia under long-term stress; that is, they were more readily affected by depression. Our results suggest that dopamine 2/3 receptor subtypes in the nucleus accumbens play an important role in floating behaviors and anhedonia

    Tai Chi Chuan optimizes the functional organization of the intrinsic human brain architecture in older adults

    Whether Tai Chi Chuan (TCC) can influence the intrinsic functional architecture of the human brain remains unclear. To examine TCC-associated changes in functional connectomes, resting-state functional magnetic resonance images were acquired from 40 older individuals including 22 experienced TCC practitioners (experts) and 18 demographically matched TCC-naive healthy controls, and their local functional homogeneities across the cortical mantle were compared. Compared to the controls, the TCC experts had significantly greater and more experience-dependent functional homogeneity in the right post-central gyrus (PosCG) and less functional homogeneity in the left anterior cingulate cortex (ACC) and the right dorsal lateral prefrontal cortex. Increased functional homogeneity in the PosCG was correlated with TCC experience. Intriguingly, decreases in functional homogeneity (improved functional specialization) in the left ACC and increases in functional homogeneity (improved functional integration) in the right PosCG both predicted performance gains on attention network behavior tests. These findings provide evidence for the functional plasticity of the brains intrinsic architecture toward optimizing locally functional organization, with great implications for understanding the effects of TCC on cognition, behavior and health in aging population

    Reading Is Fundamentally Similar Across Disparate Writing Systems: A Systematic Characterization of How Words and Characters Influence Eye Movements in Chinese Reading

    While much previous work on reading in languages with alphabetic scripts has suggested that reading is word-based, reading in Chinese has been argued to be less reliant on words. This is primarily because in the Chinese writing system words are not spatially segmented, and characters are themselves complex visual objects. Here, we present a systematic characterization of the effects of a wide range of word and character properties on eye movements in Chinese reading, using a set of mixed-effects regression models. The results reveal a rich pattern of effects of the properties of the current, previous, and next words on a range of reading measures, which is strikingly similar to the pattern of effects of word properties reported in spaced alphabetic languages. This finding provides evidence that reading shares a word-based core and may be fundamentally similar across languages with highly dissimilar scripts. We show that these findings are robust to the inclusion of character properties in the regression models and are equally reliable when dependent measures are defined in terms of characters rather than words, providing strong evidence that word properties have effects in Chinese reading above and beyond characters. This systematic characterization of the effects of word and character properties in Chinese advances our knowledge of the processes underlying reading and informs the future development of models of reading. More generally, however, this work suggests that differences in script may not alter the fundamental nature of reading
