    Die Rolle regulatorischer T-Zellen in der Verbesserung des Verlaufs und Ausgangs zweiter Schwangerschaften

    Für einen positiven fetalen Ausgang der Schwangerschaft und einen gesunden Schwangerschaftsverlauf ist die adäquate Anpassung des mütterlichen Organismus an die Schwangerschaft auf mehreren Ebenen notwendig. Im Vordergrund stehen dabei neben der kardialen und metabolischen vor allem die immunologische Adaptation an diese besondere Situation. Dabei unabdingbar sind die regulatorischen T-Zellen (Tregs), die zusammen mit anderen Zellen eine große Rolle in der Induktion immunologischer Toleranz gegenüber dem Fetus spielen. In der durchgeführten Literaturrecherche konnte gezeigt werden, dass immunologisch assoziierte Schwangerschaftskomplikationen (Präeklampsie, FGR, Frühgeburtlichkeit und spontaner Abort) nach Auftreten in der ersten Schwangerschaft in der zweiten Schwangerschaft ein erhöhtes Risiko zeigten, erneut aufzutreten. Frauen mit unkomplizierter erster Schwangerschaft zeigten in der zweiten Schwangerschaft ein erniedrigtes Risiko. Das lässt die Vermutung eines immunologischen Schwangerschaftsgedächtnisses zu. Verantwortlich sind hierfür unter anderem vermutlich Gedächtnis-Tregs. Die Experimente im Mausmodell zeigten, dass die Anzahl Fetal-Antigen-spezifischer Tregs in der zweiten Trächtigkeit höher war. Außerdem konnte der Ausgang der Trächtigkeit bei Mäusen in ihrer ersten Trächtigkeit nach Transfer von trächtigkeitsgeprägten Tregs auf den Ausgang einer zweiten Trächtigkeit angehoben werden. Die Experimente unterstützen daher die These der Existenz von Treg- Gedächtniszellen, die durch die Schwangerschaft entstehen. Insbesondere in Hinblick auf Therapiemöglichkeiten, die im Rahmen der reproduktiven Immunologie möglich sein könnten, sind weitere Experimente notwendig.In order to achieve a healthy pregnancy and a good fetal outcome the maternal organism has to adapt adequately to the pregnancy on different levels. Besides the cardiac and metabolic adaptation, the focus lies on the immunological adaptation to this special situation. Here regulatory T cells (Tregs) are indispensable. Together with other cells, they play a pivotal role in the induction of tolerance towards the fetus. The literature research carried out showed that immunologically associated pregnancy complications (pre-eclampsia, FGR, premature birth and spontaneous abortion) showed an increased risk of reoccurring in the second pregnancy after the occurrence in the first pregnancy. Women with an uncomplicated first pregnancy showed a reduced risk in the second pregnancy. This allows the assumption of an immunological pregnancy memory. Aside other aspects, memory Tregs are probably responsible for this. The experiments in the mouse model showed that the number of fetal-antigen-specific Tregs was higher in the second pregnancy. In addition, the pregnancy outcome was better in mice in their first pregnancy after transfer of pregnancy-imprinted Tregs. The experiments therefore support the thesis of the existence of memory Tregs that are formed during pregnancy. Especially with regard to possible therapeutic options in the context of reproductive immunology, further experiments are necessary

    Determination of the Jet Energy Resolution and Measurements of Jet Substructure and the Top Quark Mass in Decays of Boosted Top Quarks at CMS

    This thesis presents two analyses using data recorded by the CMS detector in proton-proton collisions at the LHC. First, a determination of the jet transverse momentum resolution scale factors is performed, followed by the measurement of the jet mass in hadronic decays of boosted top quarks and by an extraction of the top quark mass. The first analysis determines the jet transverse momentum resolution scale factors for data collected during the LHC Run 2 and Run 3 data-taking periods. The width of the jet energy response is calibrated in simulation to match the width in data, exploiting the transverse momentum balance in QCD dijet events. In this thesis, this method has been extended to cover a jet transverse momentum from 200 GeV to 2000 GeV. A modified correction method for additional jet activity is introduced, refining the uncertainty treatment and the reliability of the calibration. The compatibility of simulation to data is studied, particularly in low transverse momentum ranges which are constrained due to the additional jet activity. The final calibration for data collected at a center-of-mass energy of 13 TeV has been performed, covering the years 2016, 2017, and 2018, corresponding to an integrated luminosity of 138 fb^-1. Moreover, the first calibrations for data collected at 13.6 TeV is presented for 2022 and 2023 corresponding to 62 fb^-1. The second analysis presents the measurement of the differential top quark pair production cross section as a function of the jet mass in decays of boosted top quarks, using data collected at 13 TeV corresponding to 138 fb^-1. At high momenta, the decay products of top quarks are highly Lorentz boosted and can be reconstructed within a single jet, which require a detailed understanding of the jet substructure. The reconstruction of the hadronic top quark decay in a single jet provides the opportunity to explore new energy regimes. Moreover, the jet mass distribution can be compared to analytic calculations, allowing for the extraction of the top quark mass in a well-defined mass scheme. In this work, the focus is set on the dominant uncertainties of an earlier analysis result, the calibration of the jet mass scale and the modeling of the final state radiation. The uncertainty of the jet mass scale is constrained by calibrating the jet mass scale to the reconstructed W boson mass. A refined modeling of the final state radiation is introduced, based on the N-subjettiness. This reduces the uncertainty of the jet mass scale by a factor of three and minimizes the final state radiation modeling uncertainty, making it a negligible source of uncertainties. The jet mass is unfolded to particle level and used to extract the top quark mass with (173.06 +- 0.84) GeV

    Late Antique and Mediaeval Egyptian Bindings as Archival Instruments: Defining, Ordering and Transmitting Knowledge in Egypt

    This research explores the archival role of Coptic bindings in defining, organising, and transmitting knowledge in Late Antique and Medieval Egypt (3rd–13th centuries). Therefore, it examined archiving as a cultural practice, extending beyond the documentary content of the bound texts. The research recognizes that bindings serve more than just a mechanical protective function; they also play a crucial archival role, transmitting messages beyond the written text. It explores how the materials and techniques used in creating bindings provide insights into the cultural context surrounding the texts they encase. Bindings can be seen as cultural artefacts that convey messages encoded by collective knowledge, reflecting the cultural identity of the society that produced them. One of the key achievements of the research is its clarification that the term ‘Coptic’, when applied to bindings, is fundamentally incorrect. The term mistakenly suggests a connection to Christianity and the Coptic language, which the binding and the text it contains may never have had. Despite this inaccuracy, the research maintains it since its use has been deeply rooted in literature since 1911, following its adoption by Hugo Ibscher, an influential conservator of the Berliner Papyrussammlung. However, in this context, the term ‘Coptic’ is retained with its original meaning of ‘Egyptian’. The research successfully addressed the challenge posed by the fragmentary state in which Coptic bindings are often preserved. This fragmentation is primarily due to a historical focus on recovering ancient texts repurposed to stiffen leather covers. The research developed a standardized survey method that ensures consistent and comprehensive binding descriptions. This method can be applied to all bindings and binding fragments, whether directly examined, described in the literature, or observed in photographic images. The survey uses controlled terminology and a standard structure to facilitate the comparative study of different binding traditions and highlight their similarities and differences. The survey resulted in constructing a database that is eventually publishable as a web application where the data gathered on Coptic bookbinding can be stored. The research established that the defining feature of binding is its sewing technique, and the expression ‘Coptic bindings’ extends beyond Christian texts in Coptic. According to this definition, 294 bindings and binding fragments were identified. Despite the challenges and delays caused by COVID-19 restrictions, the research applied the developed survey method to 147 directly examined artefacts. These included Coptic bindings, binding fragments, and fragments ultimately found to be unrelated to bindings. Through direct contact with various collections, the research assessed the current condition of bindings known from the literature, determining which are still preserved and which have been lost, and identifying any newly discovered bindings. Thanks to a direct examination of the bindings and unpublished photographic documentation, the research could develop a typological classification of Coptic bookbinding that is purely technological, based on the sewing techniques used to bind the leaves or quires together. However, their historical evolution emerges by grouping the bindings according to their common characteristics. It became clear how the Coptic technique from the eighth century evolved, assimilating features of Islamic bindings, thus reflecting the process of Arabisation of Egyptian society. This research made it possible to assess the similarities and differences between Coptic and Ethiopian bindings, clarifying that while the two binding traditions share a similar sewing technique, after the Arab conquest, they departed from each other. The Coptic binding technique eventually adopted characteristics of Islamic bindings, while Ethiopian bookbinding preserved the ancient methods for binding Christian texts. In Ethiopia, Islamic and Christian book cultures coexist, yet they remain distinct, with each tradition identifiable by the unique forms of their books. The research represents the first comprehensive study to explore the meanings of Coptic bindings beyond the text itself, focusing on their archival function. Although the study found no direct correlation between cover design, decoration, and the content of the text, it did reveal that decoration often serves as a sign of provenance and ownership in Coptic bindings. Ultimately, the materials and techniques used in the binding offer valuable insights into the context of the text’s production and use. In summary, the research shed light on lesser-known aspects of Coptic book production, specifically bindings, the evolution of the technique, the relation with the Ethiopian technique and its archival function, transmitting additional meaning beyond the text.Diese Forschungsarbeit untersucht die archivarische Rolle koptischer Einbände bei der Definition, Organisation und Weitergabe von Wissen im spätantiken und mittelalterlichen Ägypten (3.–13. Jahrhundert). Dabei geht diese Untersuchung davon aus, dass Einbände eine mechanische Schutz- und Archivierungsfunktion haben, da sie Informationen über den geschriebenen Text hinaus vermitteln können. Es wird untersucht, wie die bei der Herstellung von Einbänden verwendeten Materialien und Techniken Informationen über den kulturellen Kontext der darin enthaltenen Texte vermitteln. Sie gehören zu einem kollektiven Wissen, das Teil der kulturellen Identität der Gesellschaft ist, die sie hergestellt hat. Daher sind koptische Einbände Ausdruck der ägyptischen kulturellen Identität zwischen dem dritten und dreizehnten Jahrhundert. Eines der wichtigsten Forschungsergebnisse dieser Untersuchung ist die Klarstellung, dass der Begriff ‘koptisch’ in Bezug auf Einbände grundsätzlich falsch ist. Der Begriff suggeriert fälschlicherweise eine Verbindung zum Christentum und zur koptischen Sprache, die die Einbände und die darin enthaltenen Texte möglicherweise nie hatten. Trotz dieser Ungenauigkeit wird der Begriff in der Forschung beibehalten, da er seit 1911, nach seiner Übernahme durch Hugo Ibscher, einen einflussreichen Konservator der Berliner Papyrussammlung, tief in der Literatur verwurzelt ist. Im Zusammenhang dieser Untersuchung wird der Begriff ‘koptisch’ jedoch in seiner ursprünglichen Bedeutung von ‘ägyptisch’ beibehalten. Diese Methode kann auf alle Einbände und Einbandfragmente angewendet werden, unabhängig davon, ob sie direkt untersucht, in der Literatur beschrieben oder auf Fotografien beobachtet werden. Die Erhebung verwendet eine kontrollierte Terminologie und eine Standardstruktur, um die vergleichende Untersuchung verschiedener Einbandtraditionen zu ermöglichen und ihre Gemeinsamkeiten und Unterschiede sichtbar zu machen. Die Erhebung führte zur Erstellung einer Datenbank, die schließlich als Webanwendung veröffentlicht werden kann, in der die gesammelten Daten zur koptischen Einbandtradition gespeichert werden können. In dieser Untersuchung ist das bestimmende Merkmal der Einbände die Hefttechnik, und die Bezeichnung ‘koptische Einbände’ geht über christliche Texte in koptischer Sprache hinaus. Nach dieser Definition wurden 294 Einbände und Einbandfragmente identifiziert. Durch direkten Kontakt mit verschiedenen Sammlungen wurde im Rahmen der Forschung der aktuelle Zustand von Einbänden bewertet, die aus der Literatur bekannt sind, wobei festgestellt wurde, welche noch erhalten sind und welche verloren gegangen sind, und neu entdeckte Einbände identifiziert wurden. Dank einer direkten Untersuchung der Einbände und einer unveröffentlichten fotografischen Dokumentation konnte im Rahmen der Forschung eine typologische Klassifizierung der koptischen Einbandkunst entwickelt werden. Die Klassifizierung ist technisch und basiert auf den verwendeten Hefttechnik, die die Blätter oder Lagen zusammenhalten, und identifiziert die historische Entwicklung der Technik. Es wurde deutlich, dass die koptische Technik ab dem 8. Jahrhundert begann, Merkmale islamischer Einbände zu übernehmen, was den Prozess der Arabisierung der ägyptischen Gesellschaft widerspiegelt. In dieser Forschungsarbeit wurden auch die Ähnlichkeiten und Unterschiede zwischen koptischen und äthiopischen Einbänden untersucht. Die beiden Einbandtraditionen teilen zunächst eine ähnliche Hefttechnik entwickelten sich aber nach der islamischen Eroberung auseinander. Die koptische Einbandtechnik übernahm schließlich Merkmale islamischer Einbände, während die äthiopische Tradition die alten Methoden beibehielt, die für das Einbinden christlicher Texte verwendet wurden. In Äthiopien existieren islamische und christliche Buchkulturen nebeneinander, bleiben aber dennoch unterschiedlich wobei jede Tradition an den einzigartigen Formen ihrer Bücher erkennbar ist. Diese Forschungsarbeit ist die erste Untersuchung der Archivfunktion koptischer Einbände, die sich auf die Bedeutungen konzentriert, die sie über den Text selbst hinaus vermitteln. Die Untersuchung fand keine direkte Korrelation zwischen der Gestaltung des Einbands, der Verzierung und dem Inhalt des Textes, aber sie zeigte, dass die Verzierung oft ein Zeichen für Herkunft und Besitz ist. Damit geben die verwendeten Materialien und Techniken Einblicke in den Kontext der Textproduktion und -verwendung. Zusammenfassend beleuchtete diese Untersuchung weniger bekannte Aspekte der koptischen Buchproduktion, nämlich die Einbände. Diese Forschungsarbeit identifizierte die Entwicklung der Technik, die Beziehung zur äthiopischen Technik und ihre Archivierungsfunktion, die darin besteht, über den Text hinaus zusätzliche Bedeutung zu vermitteln

    Structure Elucidation and Synthesis of Volatile Semiochemicals - Chemical Communication in Insects, Plants and Microorganisms

    Diese Dissertation widmet sich der Identifizierung und Strukturaufklärung von flüchtigen Signalstoffen durch Analyse von Naturextrakten und Synthese von Referenzverbindungen. Langkettige Acetogenine wurden als Botenstoffe von Lepidopteren und von einer Erzwespe gefunden: (2R)-Butyl-(7Z)-dodecenoat und (2R)-Butyl-(7Z)-tetradecenoat sind Komponenten des Sexuallockstoffs von weiblichen Pflaumenmotten, Illiberis rotundata. Die Miniermotte Tischeria ekebladella verwendet ein dreikomponentiges Pheromon aus (3Z,6Z,9Z)-Tricosa-3,6,9-trien, (3Z,6Z,9Z,19Z)-Tricosa-3,6,9,19-tetraen und (3Z,6Z,9Z)-Nonacosa-3,6,9-trien. Die Struktur des neuen Tetraens wurde durch E/Z-selektive Synthese von acht Doppelbindungspositions¬isomeren aufgeklärt. Bei der Mandelsamenwespe Eurytoma amygdali wurden die als Sexualpheromone für Hymenopteren ungewöhnlichen Polyene (6Z,9Z)-Tricosadien, (6Z,9Z)-Pentacosadien, (3Z,6Z,9Z)-Tricosa-3,6,9-trien und (3Z,6Z,9Z)-Pentacosatrien identifiziert. Unverzweigte Spiroacetale wurden als Signalstoffe sowohl bei Insekten als auch bei Pflanzen gefunden. (5S,7S)-Conophthorin und Chalcogran dienen als Duftstoffe der Kaffeekirschen von Coffea arabica dem spezialisierten Kaffeekirschenbohrer Hypothenemus hampei bei der Wirtwahl. Die Täuschblume Ceropegia dolichophylla hingegen imitiert die Pheromone von Hymenopteren, um parasitische Fliegen als Bestäuber anzulocken. Im Blütenduft wurden neben ebenfalls biologisch aktiven n-Alkenen neun EAD-aktive Spiroacetale identifiziert, darunter die neuen Verbindungen 2,8-Diethyl-1,7-dioxaspiro[5.5]undecan, 7-Ethyl-2-propyl-1,6-dioxaspiro-[4.5]decan, 8-Methyl-2-propyl-1,7-dioxaspiro[5.5]undec-3-en und 2-Ethyl-8-methyl-1,7-dioxaspiro[5.6]dodecan. Bockkäfer der Gattung Megacyllene sowie Callisphyris apicicornis produzieren ähnliche Düfte offenbar zum Schutz durch chemische Mimikry von Wespen. Bei den Käfern wurden acht bekannte Spiroacetale sowie das neue (2E,7E)-2-Methyl-7-propyl-1,6-dioxaspiro-[4.6]undecan durch Synthese identifiziert. Männchen der Goldrutengallfliege Eurosta solidaginis nutzen ein Bouquet aus C9-Spiroacetalen vermutlich als Sexualpheromon. Dessen Hauptkomponente, (E,S)-Conophthorin, löst bei der Wirtspflanze, Solidago altissima, ein Priming der Abwehrmechanismen aus. Pflanzenduftstoffe sind häufig terpenoiden Ursprungs und dienen Herbivoren zur Wirtfindung. Der Thujenborkenkäfer, Phloeosinus aubei, nutzt dazu Monoterpenoide seines Wirts, Thuja occidentalis. Die Weibchen produzieren (-)-α-Pinen und dessen Oxidationsprodukt (-)-Myrtenol als Aggregationspheromon. Spezifische terpenoide Signalstoffe müssen sich von ubiquitären pflanzlichen Terpenen unterscheiden. Bei Rüsselkäfern der Gattung Conotrachelus, bei Pseudopiazurus obesus und bei Pissodes castaneus wurden (1R,2S)-Grandisol, (1R,2S)-Grandisal, (1R,2S)-Grandisolsäure und die neuen Naturstoffe (1R,6R)-2,2,6-Trimethyl-3-oxabicyclo[4.2.0]octan-4-on und (1R,2S,6R)-2-Hydroxymethyl-2,6-dimethyl-3-oxabicyclo[4.2.0]¬octan als Sexual- bzw. Aggregationspheromone der Männchen identifiziert. Gynen der Amazonenameise Polyergus breviceps verwenden als Sexualpheromon das mutmaßliche Bisnorterpenoid (R)-3-Ethyl-4-methylpentanol in Kombination mit dem Polyketid Methyl-6-methylsalicylat. Palmlilien (Yucca spp.) produzieren zur Attraktion symbiontischer Bestäuber Blütendüfte, die bisher einzigartige Derivate des Tetrakisnorsesquiterpens E-DMNT enthalten. Die Strukturen der neuen Naturstoffe (Z,R)-3-Allyliden-5-(2-methylprop-1-enyl)dihydro-3H-furan-2-on, (Z)-2-Allyliden-6-methylhept-5-en-1-ol, (Z)- und (E)-Allyliden-6-methylhept-5-enal sowie 2-(4-Methylpent-3-en-1-yl)cyclopent-3-enon wurden durch Synthese aufgeklärt. Der Hefepilz Candida albicans emittiert Farnesol und (3R,6E)-2,3-Dihydrofarnesol als Allomon gegen mikrobielle Konkurrenten. Bei der Termite Hodotermes mossambicus wurden anhand der Massenspektren ein Homoterpenalkohol mit Farnesol-Substruktur und der korrespondierende Aldehyd als Komponenten des Sexualpheromons der Männchen vermutet. Für beide mussten drei mögliche Strukturisomerenpostulate nach Synthese verworfen werden. Für Biotests bei Formosa-Termiten, Coptotermes formosanus, wurden 3-, 11- und 13-Methylheptacosan enantioselektiv dargestellt. Durch Synthese mehrerer Konfigurations- und Stereoisomere wurden die neuen, vermutlich propanogeninen Naturstoffe (2E,4E)-syn,syn-4,6,8,10-Tetramethyltrideca-2,4-dien und (2E,4E)- syn,syn-4,6,8,10-Tetramethyltrideca-2,4-dien-1-ol als Sexualpheromonkomponenten von Weibchen der parasitoiden Erzwespe Trichogramma turkestanica identifiziert. Bei Raubwanzen der Gattung Triatoma wurden 2-alkyl-substituierte 4-Ethyl-5-methyl-1,3-dioxolane als eine neue Klasse flüchtiger Verbindungen bei Insekten identifiziert. Das Hauptprodukt in Drüsenextrakten war (4S,5S)-2,2,4-Triethyl-5-methyl-1,3-dioxolan. Die Carbonyl-Komponenten der Acetale sind vermutlich Aminosäure-Derivate. Diese sind als Biosynthesebausteine besonders bei Bakterien häufig. Bei dem phytopathogenen Bakterium Xanthomonas euvesicatoria wurden homologe Reihen von unverzweigten, iso- und anteiso-verzweigten Methylketonen und Methylcarbinolen sowie alkyl-substituierte Pyrazine identifiziert. 3-Alkyl-2-methoxypyrazine sind für den charakteristischen Geruch von Kaffeekirschen verantwortlich.This thesis focuses on the identification and structural elucidation of natural organic volatiles through the analysis of natural extracts and the synthesis of reference compounds. Long-chain acetogenins have been found as pheromones in Lepidoptera and a chalcid wasp: (2R)-butyl (7Z)-dodecenoate and (2R)-butyl (7Z)-tetradecenoate are components of the sex attractant of the female Plum Moth, Illiberis rotundata. The leaf miner Tischeria ekebladella uses a three-component pheromone of (3Z,6Z,9Z)-tricosa-3,6,9-triene, (3Z,6Z,9Z,19Z)-tricosa-3,6,9,19-tetraene and (3Z,6Z,9Z)-nonacosa-3,6,9-triene. The structure of the novel tetraene was elucidated by E/Z-selective synthesis of eight double bond isomers. In the Almond Wasp, Eurytoma amygdali, (6Z,9Z)-tricosadiene, (6Z,9Z)-pentacosadiene, (3Z,6Z,9Z)-tricosa-3,6,9-triene and (3Z,6Z,9Z)-pentacosatriene were identified. These polyenes would be unusual as sexual pheromones in Hymenoptera. Unbranched spiroacetals have been found as semiochemicals in both insects and plants. (5S,7S)-conophthorin and chalcogran from coffee berries of Coffea arabica are used by the specialized coffee cherry borer Hypothenemus hampei for host selection. In contrast, the deceptive plant Ceropegia dolichophylla mimics Hymenoptera pheromones to attract parasitic flies as pollinators. In addition to biologically active n-alkenes, nine EAD-active spiroacetals were identified in the floral scent, including the novel compounds 2,8-diethyl-1,7-dioxaspiro[5.5]undecane, 7-ethyl-2-propyl-1,6-dioxaspiro[4.5]decane, 8-methyl-2-propyl-1,7-dioxaspiro[5.5]undec-3-ene and 2-ethyl-8-methyl-1,7-dioxaspiro[5.6]dodecane. Longhorn beetles of the genus Megacyllene and Callisphyris apicicornis produce similar odours, apparently for protection by chemical mimicry of wasps. In the beetles, eight known spiroacetals and the novel (2E,7E)-2-methyl-7-propyl-1,6-dioxaspiro[4.6]undecane were identified by synthesis. Males of the Goldenrod Gall Fly, Eurosta solidaginis, probably use a bouquet of C9-spiroacetals as a sex pheromone. Its main component, (E,S)-conophthorin, elicits the priming of the chemical defense in the host plant Solidago altissima. Plant volatiles are often terpenoids and are used by herbivores for host selection. The Cedar Bark Beetle, Phloeosinus aubei, uses monoterpenoids from its host, Thuja occidentalis, for this purpose. The females produce (-)-α-pinene and its oxidation product (-)-myrtenol as an aggregation pheromone. Specific terpenoid semiochemicals must differ from the ubiquitous plant terpenes. In weevils of the genus Conotrachelus, in Pseudopiazurus obesus and in Pissodes castaneus, (1R,2S)-grandisol and its derivatives (1R,2S)-grandisal, (1R,2S)-grandisoic acid and the novel natural products (1R,6R)-2,2,6-trimethyl-3-oxabicyclo[4.2.0]octan-4-one and (1R,2S,6R)-2-hydroxymethyl-2,6-dimethyl-3-oxabicyclo[4.2.0]octane were identified as male sex and aggregation pheromones, respectively. Gynes of the social parasitic ant Polyergus breviceps use a probable bisnorterpenoid, (R)-3-ethyl-4-methylpentanol, in combination with the polyketide methyl-6-methylsalicylate as a sex pheromone. Yucca plants produce floral scents containing unique derivatives of the tetrakisnorsesquiterpene E-DMNT to attract symbiotic pollinators. The structures of the novel natural products (Z,R)-3-allylidene-5-(2-methylprop-1-enyl)dihydro-3H-furan-2-one, (Z)-2-allylidene-6-methylhept-5-en-1-ol, (Z)- and (E)-allylidene-6-methylhept-5-enal and 2-(4-methylpent-3-en-1-yl)cyclopent-3-enone were elucidated by synthesis. The human pathogenic yeast Candida albicans emits farnesol and (3R,6E)-2,3-dihydrofarnesol to suppress microbial competitors. In the termite Hodotermes mossambicus, based on the mass spectra, a homoterpene alcohol with farnesol substructure and the corresponding aldehyde were postulated to be components of the male sex pheromone. For both, three possible structural isomer postulates had to be rejected after synthesis. For behavioral tests in Formosa termites, Coptotermes formosanus, 3-, 11- and 13-methylheptacosane were synthesized enantioselectively. By synthesis of several configurational and stereoisomers, the new putative propanogeninic natural products (2E,4E)-syn,syn-4,6,8,10-tetramethyltrideca-2,4-diene and (2E,4E)-syn,syn-4,6,8,10-tetramethyltrideca-2,4-dien-1-ol were identified as sex pheromone components of the females of the parasitoid chalcid wasp Trichogramma turkestanica. In metasternal glands of Triatoma predatory bugs, 2-alkyl-substituted 4-ethyl-5-methyl-1,3-dioxolanes were identified as a new class of insect volatiles. The main compound was (4S,5S)-2,2,4-triethyl-5-methyl-1,3-dioxolane. The carbonyl components of the acetals are probably derived from amino acids. These are also widely used as biosynthetic building blocks in bacteria. In the phytopathogenic bacterium Xanthomonas euvesicatoria, homologous series of unbranched, iso- and anteiso-branched methylketones and methylcarbinols as well as alkyl-substituted pyrazines have been identified. 3-Alkyl-2-methoxypyrazines are responsible for the characteristic scent of coffee berries

    Auswirkung der organspezifischen Überexpression der Polypeptid-N-Acetylgalactosaminyltransferase-2 auf die Organmorphologie und den Metabolismus des murinen Pankreas

    Die organspezifische Überexpression von GalNT2 im murinen Pankreas führte zu deutlichen morphologischen Veränderungen sowie Auffälligkeiten auf molekularer Ebene. Es zeigte sich mit dem Älterwerden der transgenen Tiere bis zu 15 Wochen das Fortbestehen physiologischer exo- und endokriner Zellreihen neben dem zunehmend größer werdenden Anteil an Fettgewebe und Fibrose. Unsere Versuche haben zudem gezeigt, dass sich die Überexpression von GalNT2 auf molekularer Ebene sowohl auf die Proteinhomöostase, Inflammation sowie den Lipidstoffwechsel auswirkt. Es fanden sich Anzeichen einer vermehrten Proteinakkumulation sowie zellulärem Stress. Die entzündlichen Reaktionen könnten in Zusammenhang zur Fibrosereaktion der Pankreata stehen. Insgesamt sprachen die Ergebnisse eher für eine Infiltration des pankreatischen Gewebes durch Adipozyten als eine Transdifferenzierung aus Azinuszellen. In Vergleichen zu ähnlichen Phänotypen konnten wir die Vermutung aufstellen, dass die feingeweblichen Veränderungen in Verbindung zu Alterationen von Prox-1 stehen könnten. Wir bewerteten die beobachteten Prozesse jedoch als Zusammenspiel multipler Faktoren, wodurch es sich empfiehlt, in künftigen Arbeiten die einzelnen Mechanismen weiter zu untersuchen. Zusammenfassend konnte in dieser Arbeit ein weiterer Beitrag zum Verständnis der Glykobiologie im murinen Pankreas geschaffen werden. Dies dient somit als Ausgangspunkt weiterer wissenschaftlicher Forschung in Bezug auf Pathophysiologie der pankreatischen Lipomatose, Fibrose und Proteinhomöostase.This work demonstrates the impact of overexpression of GalNT2 in murine pancreas and outlines the importance of glycobiology for organ morphology and protein homeostasis. The mice model of overexpressed GalNT2 in pancreas could show, that throughout different stages of age a development of increased adipocytes as well as fibrotic tissue of the pancreatic organ arises. There were indications for an increased level of protein accumulation as a potential sign of an inflammatory process. So far we could not determine evidence for a transdifferentiation of pancreatic exocrine tissue into adipocytes or fibroblasts. Therefore we assume an infiltration of previously mentioned cell types into the murine pancreas of our mice model. We concluded that the investigated morphological changes underlie a multifactorial process. In summary further investigation can be performed based on the current findings to evaluate the complex influence of GalNT2 for biochemical signalling

    Seasonal influence of maternal effects and abiotic factors on early life stages of Atlantic herring Clupea harengus

    The North Sea and Baltic Sea rank among the fastest warming seas in the world, exhibiting the most pronounced warming rates during spring. This rapid increase in temperature may disrupt the life cycle scheduling of spring-spawning fish that depend on temperature cues for reproduction. Atlantic herring Clupea harengus is one of the most abundant and widely distributed small pelagic fish across the northern European region as well as a crucial prey species for piscivorous fish and mammals. In recent decades, elevated water temperatures prior to or during spring have been associated with reduced recruitment across various stocks of spring-spawning stocks. In particular, low recruitment rates in the western Baltic spring-spawning (WBSS) herring, compounded by overexploitation, have resulted in a steady decline in stock size since the 2000s. Long-term larval sampling (Rügen herring larvae survey) has been conducted annually in the main spawning and nursery ground of the WBSS herring, Greifswald Bay, where samples were taken weekly at various stations during spawning seasons and larvae were counted and measured. The survey revealed that the contribution of early-season cohorts to recruitment was low while the survival of larvae from late-season cohorts alone has been insufficient to rebuild the stock size. Although field evidence implies larval cohorts from different times of the season have different survival rates, few studies have examined the explanatory factors. Seasonal differences in survival may be correlated with maternal and/or abiotic factors which also change with the progression of the season. In herring, larger, older females tend to spawn earlier than smaller, younger females, with first-time recruit females spawning last. Larger females also allocate more energy to reproduction and may produce larger and/or higher-quality eggs compared to smaller females. Moreover, larvae hatching at different times of the season encounter different environments such as different water temperatures. WBSS herring initiate spawning at a seawater temperature of ca. 4°C during spring while late-season females spawn ca. two months later. This translates to a temperature difference of more than 6°C between early- and late-season larval cohorts. As temperature governs the growth, development and metabolic rates of individuals according to their body size, larger and smaller larvae may use energy more (or less) efficiently at different temperatures. The present thesis aims to disentangle maternal and environmental effects from seasonal effects on the survival of herring and size-at-age at early life stages. My co-authors and I used field data on WBSS spawners to define the early, mid and late phases of the spawning season. We randomly selected 6–8 females from each seasonal timing and incubated their progeny in separate chambers throughout the egg (Chapter 2) and yolk-sac larval stages (Chapter 3) at early- (7°) and late-season (13°C) temperatures. The relative contribution of seasonal timing, females and incubation temperature to survival and mean size-at-ages was quantified. Finally, multi-year data on the size-at-ages and size-at-stages of larvae from WBSS as well as a winter- (Downs) and an autumn-spawning (Gulf of Riga) herring stocks were compiled. We quantified the variation in larval size through ages and ontogeny and examined the contribution of seasonal effects on the variation in larval growth and development (Chapter 4). In WBSS herring (Chapters 2 and 3), survival of progeny from eggs until the end of the yolk sac larval stage was high (>85%) and no seasonal but small among-female differences were detected. Mean sizes of females, eggs and post-yolk larvae all decreased with the progression of the spawning season (R2 = 0.31, 0.39 and 0.27, respectively). Compared with seasonal timing, individual females contributed equally (R2 = 0.37) to the variances in mean egg size and 1.5 times more to that in mean post-yolk larval size. Although progeny size differed among different females, these maternal effects were, however, unrelated to female size, age or condition. Incubation temperature influenced the shape but not the yolk utilisation efficiency of yolk-sac larvae. In feeding larvae across herring stocks (Chapter 4), the (relative) variation in size-at-age increased after hatching to a maximum (10–15%) whereas the variation in size-at-stage decreased to a minimum (6%) prior to flexion, marking a threshold size required to enter the flexion stage. Larvae from colder months had a relatively shorter pre-flexion stage due to a larger initial size. In conclusion, the present thesis identified the seasonal timing of spawning as a good predictor of egg and larval size, with size-at-ages of the early-season cohort being consistently larger than those of the late-season cohort in WBSS herring. Larvae with a larger initial size reached the threshold size for flexion more easily than smaller individuals. Although early-season cohorts had the advantage of a larger size, their low survival at the nursery grounds implies that other extrinsic processes such as seasonal match-mismatch with prey likely play more important roles in survival. While future studies should further examine the mechanisms of the seasonal variation in prey abundance and biotic dynamics, management should implement plans to cease fishing until this stock has recovered and reduce nutrient input into coastal nursery grounds.Die Nord- und die Ostsee gehören zu den sich am schnellsten erwärmenden Meeren der Welt, wobei die Erwärmung im Frühjahr am stärksten ist. Diese rasche Erwärmung kann den Lebenszyklus von Fischen stören, die im Frühjahr laichen und deren Fortpflanzung von der Temperatur abhängig ist. Der Atlantische Hering Clupea harengus ist einer der häufigsten und am weitesten verbreiteten kleinen pelagischen Fische in Nordeuropa und eine wichtige Beute von Raubfischen und Säugetieren. In den letzten Jahrzehnten wurden höhere Wassertemperaturen vor oder während des Frühjahrs mit einer geringeren Rekrutierung in verschiedenen frühjahrslaichenden Heringspopulationen in Verbindung gebracht. Die geringe Rekrutierung beim frühjahrslaichenden Hering der westlichen Ostsee (WBSS) und die Überfischung dieses Bestands haben seit den 2000er Jahren zu einem stetigen Rückgang der Bestandsgröße geführt. Im wichtigsten Laich- und Aufwuchsgebiet (Greifswalder Bodden) des WBSS-Herings wurden langfristige, jährlich Larvenbeprobungen (Rügen-Heringslarven-Survey) durchgeführt, bei denen wöchentlich während der Laichzeit an verschiedenen Stationen Proben entnommen und die Larven gezählt und gemessen wurden. Die Erhebung hat gezeigt, dass der Beitrag der frühsaisonalen Kohorten zur Rekrutierung gering war, während das Überleben der Larven aus den spätsaisonalen Kohorten allein nicht ausreichte, um die Bestandsgröße wiederherzustellen. Obwohl Feldstudien darauf hindeuten, dass Larvenkohorten aus verschiedenen Saisonzeiten unterschiedliche Überlebensraten aufweisen, haben nur wenige Studien die erklärenden Faktoren untersucht. Saisonale Unterschiede in der Überlebensrate können mit mütterlichen und/oder abiotischen Faktoren zusammenhängen, die sich auch mit dem Verlauf der Saison ändern. Bei Heringen neigen größere, ältere Weibchen dazu, früher zu laichen als kleinere, jüngere Weibchen, wobei erstgebärende Weibchen zuletzt laichen. Größere Weibchen wenden auch mehr Energie für die Fortpflanzung auf und können im Vergleich zu kleineren Weibchen größere und/oder hochwertigere Eier produzieren. Darüber hinaus treffen die zu verschiedenen Zeiten der Saison schlüpfenden Larven auf unterschiedliche Umgebungsbedingungen wie z. B. unterschiedliche Wassertemperaturen. WBSS-Heringe beginnen im Frühjahr bei einer Meerwassertemperatur von ca. 4°C mit dem Laichen, während die Weibchen in der Spätsaison ca. zwei Monate später laichen. Dies bedeutet einen Temperaturunterschied von mehr als 6 °C zwischen den frühen und späten Larvenjahrgängen. Da die Temperatur das Wachstum, die Entwicklung und den Stoffwechsel von Individuen in Abhängigkeit von ihrer Körpergröße steuert, können größere und kleinere Larven bei unterschiedlichen Temperaturen mehr oder weniger effizient Energie verbrauchen. Die vorliegende Dissertation zielt darauf ab, mütterliche Einflüsse und Umwelteinflüsse auf das Überleben und die Größe im Alter der frühen Lebensstadien des Herings von saisonalen Effekten zu trennen. Meine Co-Autoren und ich haben Felddaten von WBSS-Laichern verwendet, um die frühen, mittleren und späten Phasen der Laichzeit zu definieren. Wir wählten nach dem Zufallsprinzip 6-8 Weibchen aus jeder Phase aus und bebrüteten ihre Nachkommen während des gesamten Ei- (Kapitel 2) und Dottersack-Larvenstadiums (Kapitel 3) in getrennten Kammern bei den Temperaturen der frühen (7°) und späten Saison (13°C). Der relative Beitrag des saisonalen Zeitpunkts, der Weibchen und der Inkubationstemperatur zum Überleben und zur durchschnittlichen Größe im Alter wurde quantifiziert. Schließlich wurden mehrjährige Daten über die Größe im Alter und die Größe im Stadium der Larven der WBSS-Heringe sowie einer im Winter laichenden (Downs) und einer im Herbst laichenden (Rigaischen Meerbusen) Heringspopulation zusammengestellt. Wir quantifizierten die Variation der Larvengröße über die Alters- und Ontogenesephasen und untersuchten den Beitrag saisonaler Effekte zur Variation des Larvenwachstums und der Larvenentwicklung (Kapitel 4). Beim WBSS-Hering (Kapitel 2 und 3) war die Überlebensrate von den Eiern bis zum Ende des Dottersack-Larvenstadiums hoch (>85 %), und es wurden keine saisonalen, jedoch geringe Unterschiede zwischen den Weibchen festgestellt. Die durchschnittliche Größe von Weibchen, Eiern und Dottersacklarven nahm mit dem Fortschreiten der Laichzeit ab (R2 = 0,31, 0,39 bzw. 0,27). Im Vergleich zum saisonalen Zeitpunkt trugen die einzelnen Weibchen in gleichem Maße (R2 = 0.37) zur Varianz der mittleren Eigröße und 1,5 Mal mehr zur mittleren Größe der Larven nach dem Dottersackstadium bei. Die mütterlichen Auswirkungen auf die Größe der Nachkommenschaft waren jedoch unabhängig von der Größe, dem Alter oder dem Zustand der Weibchen. Die Inkubationstemperatur beeinflusste die Form, allerdings nicht die Dotterverwertung der Dottersacklarven. Bei der Fütterung von Larven aus verschiedenen Heringsbeständen (Kapitel 4) nahm die (relative) Variation der Größe im Alter nach dem Schlüpfen bis zu einem Maximum (10-15 %) zu, während die Variation der Größe im Stadium vor der Flexion bis zu einem Minimum (6 %) abnahm, was eine Mindestgröße markiert, die für den Eintritt in das Flexionsstadium erforderlich ist. Larven aus kälteren Monaten hatten aufgrund ihrer größeren Ausgangsgröße ein relativ kürzeres Stadium vor der Flexion. Zusammenfassend wurde in dieser Dissertation festgestellt, dass der saisonale Laichzeitpunkt ein guter Prädiktor für die Größe von Eiern und Larven ist, wobei die Größe im Alter der frühsaisonalen Kohorte beim WBSS-Hering durchweg größer ist, als die der spätsaisonalen Kohorte. Larven mit einer größeren Ausgangsgröße erreichten die Mindestgröße für die Flexion leichter als kleinere Individuen. Obwohl die frühsaisonalen Kohorten den Vorteil einer größeren Größe hatten, deutet ihr geringes Überleben in den Aufzuchtgebieten darauf hin, dass andere extrinsische Prozesse, wie z. B. die saisonale Nichtübereinstimmung mit der Beute, wahrscheinlich eine wichtigere Rolle für das Überleben spielen. Während zukünftige Studien die Mechanismen der jahreszeitlichen Schwankungen des Beuteaufkommens und der biotischen Dynamik weiter untersuchen sollten, müsste das Management Pläne zur Einstellung der Fischerei umsetzen, bis sich dieser Bestand erholt hat, und den Nährstoffeintrag in die küstennahen Aufwuchsgebiete reduzieren

    Prothrombotische Varianten des von Willebrand Faktors (VWF)

    The von Willebrand factor (VWF) circulates as a multimeric, multidomain glycoprotein in the bloodstream and initiates hemostasis at sites of vascular injuries. Through shear mediated mechanical activation, the structurally exposed binding site for the platelet receptor GPIbα in the A1 domain of VWF enables reversible platelet adhesion. This interaction exhibits a shear-dependent, biphasic ‘catch-slip’ binding behavior, which leads to signal transduction and intracellular calcium mobilization at increased shear forces. The subsequent intermediate activation of the previously inactive GPIIb/IIIa receptor enables binding to agonists, such as the VWF C4 domain and fibrin, and cross linking of platelets. However, full activation of GPIIb/IIIa, and thus stable irreversible aggregation, only occurs after platelet activation via extracellular calcium influx in presence of prolonged increased shear rates. Recently, two single nucleotide polymorphisms (SNPs) p.Phe2561Tyr and p.Pro2555Arg in the C4 domain of VWF were identified in vitro as gain-of-function (GOF) variants. While variant p.Phe2561Tyr shows a higher sensitivity to shear forces, the variant p.Pro2555Arg forms larger platelet aggregates. The prevalence of variant p.Phe2561Tyr was associated with a significantly increased risk of myocardial infarction in young women. In this study, a SNP was identified in another cohort, in a patient with myocardial infarction. Together with 8 other SNPs in the C4 and C6 domain of VWF this SNP was classified as a GOF variant in the course of an in vitro characterization of the recombinant protein using light transmission aggregometry and microfluidic experiments. Additionally, 9 loss-of-function (LOF) variants in C2 and C4 domain were identified which, when recombinantly expressed in HEK293F cells, either showed a secretion or multimerization defect, or exhibited reduced activity in shear-dependent assays. The identified GOF variants were phenotypically similar to variants p.Pro2555Arg or p.Phe2561Tyr. While aggregate size in microfluidic experiments showed a strong dependence on the GPIIb/IIIa interaction, the increased shear sensitivity was demonstrated to be GPIIb/IIIa-independent. In vivo studies in mice underlined a prothrombotic effect of variant p.Pro2555Arg and revealed a tendency to unstable thrombi formation and thus a possible increased risk of thromboembolism for variant p.Phe2561Tyr. Taking into account all the data collected in the present study and in agreement with previously published results, a new mechanism can be postulated that could be responsible for the GOF of shear-sensitive variants: Variant p.Phe2561Tyr and similar variants appear to impair the stability of the dimeric stem region, resulting in the formation of more elongated multimers. This reduces the shear force required for platelet adhesion via GPIbα, which accelerates the formation of the GPIbα-VWF-A1 ‘catch-bond’, allowing intermediate activation of GPIIb/IIIa to be achieved at lower shear rates. Due to the pre-activated GPIIb/IIIa receptor, the platelet aggregates are cross-linked via GPIIb/IIIa VWF C4 interactions at lower shear rates. However, since the required tensile force for complete activation is not present, platelet activation might be incomplete and the formed aggregates may be unstable, allowing it to be torn away. This could therefore increase the risk of thromboembolism, which is in agreement with the increased risk of myocardial infarction described for p.Phe2561Tyr. On the other hand, variants such as p.Pro2555Arg appear to promote vascular occlusion due to increased GPIIb/IIIa-mediated cross-linking of platelets. The present study has thus contributed significantly to generating new, clinically relevant insights into the GOF mechanism that could account for the prothrombotic effect of VWF GOF variants

    Attosecond pulse generation with tailored sub-cycle fields

    Observing ultrafast molecular processes on the attosecond timescale of electrons is essential for understanding chemical and physical phenomena. Attosecond pulses offer the temporal resolution needed to capture electronic dynamics. Their tunability from the extreme ultraviolet (EUV) to soft X-ray regions enables probing both valence and deeper K- and L-shell electrons, with the latter providing site- and element-specificity. This thesis explores the generation of isolated attosecond pulses (IAPs) in the EUV and soft X-ray spectral regions, extending up to the water window (284–543 eV). Tailored optical fields, capable of reaching sub-cycle durations—i.e., shorter than their main oscillation period—are investigated as drivers of high-harmonic generation (HHG) to produce these IAPs. The tailored optical fields are delivered by a parametric waveform synthesizer (PWS) pumped by a 1-kHz Ti:Sa laser system. In the PWS, two carrier-envelope-phase (CEP)-stable, few-cycle pulses—one in the near-infrared (NIR, 0.65–1 μm) and the other in the infrared (IR, 1.2–2.2 μm)—are coherently combined. Their synthesis produces fields centered at 1.4 μm, with full-width at half-maximum (FWHM) durations as short as 2.8 fs (0.6 optical cycles), and with 0.5 mJ of energy. By controlling the relative delay between the pulses and applying a common phase shift, waveforms can be tailored with sub-fs precision. These sub-cycle fields are then tightly focused into a thin gas-cell filled with argon, neon, or helium to drive HHG. Attosecond streaking measurements directly characterize the sub-cycle fields, verifying their successful synthesis, and confirm the generation of EUV IAPs with FWHM durations ranging from 80 to 240 as, depending on the waveform. Additionally, waveform tailoring enables the generation of both narrowband and broadband IAPs, with spectra spanning from 30 to 110 eV in argon and up to 200 eV in neon, while maintaining pressures below 0.5 bar. Single-atom response simulations show that IAPs result from the confinement of HHG within a single cycle, while tunability is achieved via the precise control over the electron trajectories contributing to the high-harmonic emission. Extension into the soft X-ray water window range is achieved using neon or helium at pressures up to 10 bar, producing photon energies up to 350 eV and 450 eV, respectively. A strong CEP dependence of the spectra suggests IAP emission, a finding supported by macroscopic, on-axis HHG simulations. At low plasma levels, IAPs arise from HHG confinement to a single cycle, while at higher levels, plasma defocusing likely contributes to their generation. Finally, absolute conversion efficiency measurements show that tailored waveforms increase HHG efficiency with respect to IR-driven HHG, though high plasma levels can reduce this gain. An eightfold efficiency increase is measured in helium in the water window range. Complete characterization of the driving field directly at the HHG target allows identifying the waveform responsible for the efficiency enhancement.Die Beobachtung ultraschneller molekularer Prozesse auf der Attosekunden-Zeitskala von Elektronen ist entscheidend für das Verständnis chemischer und physikalischer Phänomene. Ihre Einstellbarkeit vom extremen Ultraviolett (EUV) bis hin zu weichen Röntgenbereichen ermöglicht das Abtasten sowohl von Valenzelektronen als auch von tiefer liegenden K- und L-Schalen-Elektronen, wobei letztere orts- und elementspezifische Informationen liefern. Diese Arbeit untersucht die Erzeugung isolierter Attosekunden-Pulse (IAPs) im EUV- und weichen Röntgenspektralbereich, die bis zum Wasserfenster (284–543 eV) reichen. Maßgeschneiderte optische Felder, die Sub-Zyklus-Dauern erreichen können—d.h. kürzer als ihre Hauptoszillationsperiode—werden als Treiber für die Erzeugung hoher Harmonischer (HHG) zur Erzeugung dieser IAPs untersucht. Die maßgeschneiderten optischen Felder werden von einem Parametric Waveform Synthesizer (PWS) geliefert, der von einem 1-kHz Ti:Sa-Lasersystem gepumpt wird. Im PWS werden zwei carrier-envelope-phase (CEP)-stabile, few-cycle-Pulse—einer im nahen Infrarot (NIR, 0.65–1 μm) und der andere im Infrarot (IR, 1.2–2.2 μm)—kohärent kombiniert. Ihre Synthese erzeugt Felder, die bei 1.4 μm zentriert sind, mit Vollbreiten bei halbem Maximum (FWHM) von nur 2.8 fs (0.6 optische Zyklen) und einer Energie von 0.5 mJ. Durch Steuerung der relativen Verzögerung zwischen den Pulsen und Anwendung einer gemeinsamen Phasenverschiebung können die Wellenformen mit Sub-Fs-Präzision angepasst werden. Diese Sub-Zyklus-Felder werden dann stark in eine dünne Gaszelle fokussiert, die mit Argon, Neon oder Helium gefüllt ist, um HHG anzutreiben. Attosecond-Streaking-Messungen charakterisieren die Sub-Zyklus-Felder direkt, bestätigen ihre erfolgreiche Synthese und zeigen die Erzeugung von EUV-IAPs mit FWHM-Dauern von 80 bis 240 as, abhängig von der Wellenform. Darüber hinaus ermöglicht die Wellenformanpassung die Erzeugung sowohl von schmalbandigen als auch breitbandigen IAPs, mit Spektren, die von 30 bis 110 eV in Argon und bis zu 200 eV in Neon reichen, während die Drücke unter 0.5 bar gehalten werden. Einzelatomantwort-Simulationen zeigen, dass IAPs durch die Beschränkung der HHG auf einen einzigen Zyklus erzeugt werden, während die Einstellbarkeit durch die präzise Kontrolle der Elektrontrajektorien erreicht wird, die zur hochharmonischen Emission beitragen. Die Erweiterung in den Wasserfensterbereich der weichen Röntgenstrahlung wird mit Neon oder Helium bei Drücken von bis zu 10 bar erreicht, wobei Photonenergien von bis zu 350 eV bzw. 450 eV erzeugt werden. Eine starke CEP-Abhängigkeit der Spektren deutet auf die Emission von IAPs hin, ein Befund, der durch makroskopische, on-axis HHG-Simulationen unterstützt wird. Bei niedrigen Plasmaniveaus entstehen IAPs durch die Beschränkung der HHG auf einen einzelnen Zyklus, während bei höheren Plasmaniveaus wahrscheinlich die Plasmadefokussierung zu ihrer Erzeugung beiträgt. Schließlich zeigen absolute Konversionseffizienz-Messungen, dass maßgeschneiderte Wellenformen die HHG-Effizienz im Vergleich zu IR-getriebener HHG erhöhen, obwohl hohe Plasmaniveaus diesen Gewinn verringern können. Im Wasserfensterbereich wird eine achtfache Effizienzsteigerung in Helium gemessen. Eine vollständige Charakterisierung des Antriebsfeldes direkt am HHG-Ziel ermöglicht die Identifizierung der Wellenform, die für die Effizienzsteigerung verantwortlich ist
