Erforschung der Bildung und Verwertung von Huminen als Nebenprodukt der Lävulinsäuresynthese
The replacement of fossil raw materials is of great interest to the chemical industry. For this purpose, the synthesis of platform chemicals from biomass enables the production of industrially relevant chemicals while avoiding fossil raw material sources. One of the most important platform chemicals is levulinic acid (LA), which has so far been produced industrially using the Biofine process via an acidic reaction using sulfuric acid. However, this process uses catalysts that are harmful to the environment, as well as reactors and produces larger amounts of humins as a byproduct. The formation of these solids causes a significant reduction in the manufacturing efficiency of the sought-after platform chemical, especially as the further utilization of these humins is difficult due to their insoluble nature and unknown structure. A more detailed investigation and optimization of the LA synthesis, also regarding the formation, structure and further utilization of the humins produced, can ensure a more efficient and environmentally friendly production of the platform chemical.
First, the formation process and structure of the humins was examined in more detail. 28 different humins were produced through a variation of the converted biomass substrate (fructose, glucose, xylose, sucrose), the used solvent (water, 1:1 mixture of water/ethanol, 1:1 mixture of water/dimethyl sulfoxide (DMSO)) and the used acidic catalyst (sulfuric acid, para-toluene sulfonic acid (pTSA), acetic acid (AA)). The solid analysis of the humins indicated a primarily furan ring-based structure, which resulted from the polymerization of 5-hydroxymethylfurfural (5-HMF) or furfural with themselves, other intermediates in the reaction solution and also the organic solvents. In addition, three recurring fragments of the larger humin structure were also identified. The humins produced were then converted via selective catalytic oxidation (SCO) with the polyoxometalate H8[PV5Mo7O40] (HPA-5) to form low-molecular carboxylic acids in order to determine the viability of the produced humin structures for further valorization. The main products obtained from the SCO were CO2, formic acid (FA) and AA. In general, the greatest influence was exerted by the choice of solvent. The substrate conversion in the ethanolic solution resulted in a reduced humin yield, with a minimum humin yield of about 5 wt%. These humins also showed the best conversion rates via SCO, reaching conversionrates of up to 65 %. A comparison of solids analysis before and after SCO indicated a preferred conversion of aliphatic ether and ester bonds. The combination of AA as a catalyst and the water/ethanol solution produced the lowest humin yield of about 5 wt%, with the produced humin, while also possessing the best conversion rate of 65 %, and therefore shows great potential for the efficient production of LA.
The optimization of the LA synthesis was then addressed. The first step was to find an alternative to the conventionally used sulfuric acid. When comparing five different acidic catalysts (sulfuric acid, FA, H3[PMo12O40] (HPMo), H3[PW12O40] (HPW), H3[SiW12O40] (HSiW)) in an acidic conversion of fructose, the highest LA yield of about 61 mol% was achieved using the polyoxometalate HSiW. Using HSiW as an acidic catalyst, an optimization of the reaction conditions of the LA synthesis was then carried out via a design of experiment (DoE) reaction plan using a Box-Behnken design (used parameters: reaction temperature (T), reaction time (t), proportion of the organic solvent acetone (wt%(acetone)), substrate concentration (c(fructose)), which aimed at a minimized humin yield and a maximized LA yield. Here, the humin yield depended primarily on the solvent composition and temperature, while the LA yield was primarily influenced by the solvent composition and the substrate concentration. Within the parameters used (T = 140-180 °C, t = 1-5 h, wt%(acetone) = 0-80 %, c(fructose) = 0.1-0.4 mol*L-1), it was determined by interpolation of the synthesis results that high humin yields (over 40 wt%) were achieved by maximizing all parameters used. High LA yields (over 60 mol%), however, were achieved by maximizing either temperature or time while minimizing the remaining three parameters. Three parameter combinations were determined that focused on different optimization aspects. The most industrially relevant set of parameters was then successfully applied to conversions of the carbohydrates glucose, xylose, sucrose and cellobiose in order to successfully confirm the transferability of the results.
Finally, based on the knowledge gained so far, a cyclic LA synthesis was designed in which FA is used as a catalyst for the production of LA and is then recovered via SCO from the humins formed as a byproduct. First, one of the previously optimized parameter sets (T = 180 °C, t = 1 h, wt%(acetone) = 0 %, c(fructose) = 0,1 mol*L-1) was adapted in a two-stage process for the usage of FA instead of HSiW in a larger reactor system. Using the modified reaction conditions, a humin fraction was formed, which was subsequently converted using SCO. Optimized conditions, using H5[PV2Mo10O40] (HPA-2) as a catalyst with the addition of methanol to suppress CO2 formation, were used here, producing a high yield of FA (about 28 carbon wt%) as the main product. The SCO catalyst could afterwards be separated and recycled using membrane filtration. The FA produced was then separated and concentrated by distillation, resulting in solution containing 87.00 wt % water, 11.65 wt% FA, 0.73 wt% succinic acid, 0.49 wt% AA and 0.13 wt% methyl acetate. However, a reaction solution whose composition was similar to the FA fraction obtained could be successfully used for a new LA synthesis, whereby the additional product residues had no visible influence on the structure of the humin produced. The cycle was thus successfully closed and the basis for a more environmentally friendly and efficient synthesis of LA was created
„Jobship als One-Click-Bewerbung 2.0?“ Eine empirische Analyse der Chancen und Grenzen eines wissenschaftlich basierten sowie innovativen Bewerbungs- und Personalauswahlkonzepts für MINT-Fachkräfte im digitalen Zeitalter
Im Rahmen der vorliegenden Dissertation wurde das Ziel verfolgt, ein wissenschaftlich fundiertes Bewerbungs- und Personalauswahlkonzept einschließlich der digitalen Anwendung „Jobship als One-Click-Bewerbung 2.0“ zu entwickeln, dessen Einsatz evidenzbasierte und antidiskriminierende Personalentscheidungen fördert (Forschungsfrage 1), die Passung zwischen Person und potenziellem Arbeitsumfeld möglichst ganzheitlich ermittelt (Forschungsfrage 2) sowie zu einer positiven Candidate- und Recruiter-Experience beiträgt (Forschungsfrage 3). Methodisch wurde ein qualitatives Forschungsdesign mit Methoden-Triangulation aus problemzentrierten Interviews, Expert:inneninterviews und einer Gruppendiskussion gewählt. Die bewusst heterogene Stichprobe umfasst 15 MINT-Fachkräfte und 15 HR-Expert:innen, die aktuelle Bewerbungs- und Personalauswahlprozesse aus Kandidat:innen- und Unternehmensperspektive analysierten. Mittels qualitativer Inhaltsanalyse nach Mayring (2023) wurden 6891 Äußerungen in ein deduktiv-induktiv entwickeltes Kategoriensystem überführt. Die empirischen Ergebnisse belegen deutliche Diskrepanzen zwischen wissenschaftlichen Empfehlungen und gelebter Praxis: Auswahlentscheidungen basieren vorwiegend auf un- bzw. halbstrukturierten Interviews, valide Testverfahren finden kaum Anwendung. „Jobship“ adressiert diese Lücke durch ein dreistufiges, gamifiziertes Self- und Online-Assessment, das bereits in der Orientierungsphase eine wissenschaftlich fundierte Passungs- und Eignungsprognose ermöglichen und damit sowohl Entscheidungsqualität als auch Candidate und Recruiter Experience verbessern könnte. Zugleich zeigen die Befunde Anforderungen an eine authentische und transparente Datenqualität auf, um unrealistische Erwartungen zu vermeiden. Zudem wird anhand der Ergebnisse deutlich, dass es im Kontext der einzelnen Forschungsfragen der Berücksichtigung einer Vielzahl weiterer – insbesondere zwischenmenschlicher – Faktoren bedarf – wie einer wertschätzenden, kontinuierlichen Kommunikation während des gesamten Bewerbungs- und Personalauswahlprozesses, personaldiagnostisch geschulter Personalverantwortlicher sowie ausreichender zeitlicher Ressourcen. Die Dissertation leistet hier einen zweifachen Erkenntnisbeitrag: Erstens erweitert sie die Personaldiagnostik um einen experience-orientierten, digitalisierten Ansatz; zweitens liefert sie praxisnahe Implikationen, die aufzeigen, wie evidenzbasierte, antidiskriminierende und zugleich bewerber:innenzentrierte Prozesse gestaltet werden können. Limitationen, insbesondere die begrenzte Generalisierbarkeit aufgrund des qualitativen Designs, sowie Anregungen für weiterführende quantitative Validierungs- und Längsschnittstudien werden abschließend diskutiert
The Relation of Multiple Sclerosis to Early Life and Adolescence Determinants: Insights from Observational Studies
This dissertation investigates the influence of potential risk factors on the development of multiple sclerosis (MS). Factors that occur in (early) childhood and adolescence are of particular interest. This work is based on a systematic review with meta-analysis and two observational studies, the “German National Cohort (NAKO Gesundheitsstudie, NAKO)” and the “Study on the Influence of Risk Factors on Disease Progression and the Development of Multiple Sclerosis (StERKE)”.
The results of this study clearly show the multifactorial aetiology of MS, which on the one hand has a genetic component and on the other hand also appears to be influenced by environmental and lifestyle factors, as well as the resulting potential for prevention. We were able to reproduce
the results reported in the literature on already established risk factors such as an EBV infection, obesity in childhood and adolescence, and vitamin D deficiency. In addition, we were able to generate numerous new findings concerning MS aetiology. We observed associations between the cumulative number of common childhood infections (chickenpox, mumps, rubella, pertussis and measles), the cumulative number of stressful life events (death of a partner, death of a close person (other than partner), serious illness of a close person), and being the first-born child of a mother who was ≥ 30 years old at childbirth and an increased risk of MS. We observed a protective effect of PA during teenage years and having been breastfed on the risk of developing MS later in life.
This work complements the existing literature in that i) MS prevention should begin in early childhood and adolescence, ii) due to the overlap with other chronic diseases in terms of potential risk factors, it should be based on prevention programs, such as smoking cessation programs or the promotion of PA, for these diseases, and iii) there is still a great need for studies that focus on MS-specific factors such as disease severity, but also the sex distribution in the prevalence of MS between men and women.Im Rahmen dieser Dissertation wird der Einfluss potenzieller Risikofaktoren auf die Entstehung der Multiplen Sklerose (MS) untersucht. Von besonderem Interesse sind hierbei vor allem Faktoren, die in der (frühen) Kindheit und Jugend auftreten. Grundlage dieser Arbeit bilden sowohl ein systematisches Review mit Metaanalyse als auch zwei Beobachtungsstudien, die „NAKO Gesundheitsstudie (NAKO)“ und die „Studie zum Einfluss von Risikofaktoren auf den Krankheitsverlauf und die Entstehung der Multiplen Sklerose (StERKE)“.
Die Ergebnisse dieser Arbeit zeigen deutlich die multifaktorielle Ätiologie der MS, die einerseits eine genetische Komponente aufzuweisen, aber auch maßgeblich durch Umwelt- und Lebensstilfaktoren beeinflusst zu sein scheint sowie das sich daraus ableitende Präventionspotenzial. So war es uns möglich, die in der Literatur berichteten Ergebnisse zu bereits etablierten Risikofaktoren wie z.B. einer EBV-Infektion, Übergewicht in der Kindheit und Jugend und einem Vitamin D-Mangel zu reproduzieren. Darüber hinaus konnten wir zahlreiche neue Erkenntnisse in Hinblick auf die MS-Ätiologie generieren. Wir beobachteten Assoziationen zwischen der kumulativen Anzahl häufiger Infektionserkrankungen (Windpocken, Mumps, Röteln, Pertussis und Masern) im Kindesalter, der kumulativen Anzahl belastender Lebensereignisse (Tod des/r Partners/Partnerin, Tod einer nahestehenden Person, schwere Erkrankung einer nahestehenden Person), sowie dem Umstand das Kind einer Erstgebärenden zu sein, die bei der Geburt ≥ 30 Jahre alt war, und einem erhöhten MS-Risiko. Für körperliche Aktivität in der Jugend sowie im Säuglingsalter gestillt worden zu sein beobachteten wir einen protektiven Effekt auf das Risiko, im weiteren Leben an MS zu erkranken.
Diese Arbeit ergänzt die bestehende Literatur dahingeheng, dass i) die MS-Prävention bereits im frühen Kindes- und Jugendalter beginnen sollte, ii) sich diese bedingt durch die Überlappung mit anderen chronischen Erkrankungen in Hinblick auf potenzielle Risikofaktoren, an Präventionsprogrammen, wie z.B. Rauchentwöhnungprogrammen oder aber der Förderung körperlicher Aktivität, für diese Erkrankungen orientieren sollte und iii) es weiterhin einen hohen Bedarf an Studien gibt, die vor allem MS-spezifische Faktoren wie z.B. die Erkrankungsschwere, aber auch die Geschlechtsverteilung in der Prävalenz der MS zwischen Männern und Frauen berücksichtigen

Sino-EU Cooperation in the Field of Science and Technology (S&T) - Strengths and Limits of Rational Choice Analysis
The dissertation attempts to explain how did the cost-benefit calculations of the interests of China and the interests of the EU influenced the institutionalization of their S&T cooperation. After a comparison between the EU and China’s respective Science and Technology Policies, this dissertation assesses the institutionalization of EU-China S&T cooperation which is the dependent variable, by looking at the degree, intensity, and effectiveness of the institutionalization in three typical cases: Galileo project, The Europe-China Clean Energy Centre project (EC2), and the Dragon program. These three cases explain interrelated issues from different angles, creating a holistic image of the field of S&T cooperation. The cost-benefit explanations are insufficient for the institutionalization of Sino-EU S&T cooperation; other independent variables, such as path dependency and critical junctures, are also discussed in this dissertation. The thesis operates with six hypotheses based on rational choice institutionalism and historical institutionalism, which are discussed in depth, with some hypotheses verified, others falsified, and some requiring revision. While the main analysis concludes in January 2021, the epilogue examines developments until the year 2024, which may prompt some readers to question my findings, thereby highlighting the need to constantly reassess theoretical frameworks and results in light of evolving international dynamics.Diese Dissertation versucht zu erklären, wie die Kosten-Nutzen-Rechnungen der Interessen Chinas und der Interessen der EU die Institutionalisierung ihrer wissenschaftlich-technischen Zusammenarbeit beeinflusst haben. Die Arbeit beginnt mit einem Vergleich der jeweiligen Wissenschafts- und Technologiepolitik von EU und China. Sie bewertet die Institutionalisierung der W&T-Zusammenarbeit zwischen der EU und China, welche die abhängige Variable darstellt, indem sie den Grad, die Intensität und die Effektivität der Institutionalisierung in drei typischen Fällen untersucht: Dem Galileo-Projekt, dem Europe-China Clean Energy Centre (EC2) Projekt und dem Dragon-Programm. Mit Hilfe dieser drei Fallstudien werden die miteinander verbundenen Aspekte aus verschiedenen Blickwinkeln erklärt, wodurch ein ganzheitliches Bild des Bereichs der W&T-Zusammenarbeit entsteht. Funktionalisierung von Kohlenstoffträgern für PtNi-Nanopartikel als Brennstoffzellenkatalysator
To enhance efficiency, catalysts are employed in fuel cells to facilitate the anode and cathode reactions. The most common catalyst is composed of platinum-based nanoparticles, distributed on a carbon black (CB) support. This composite catalyst degrades during usage, gradually lowering the fuel cell performance. Hence, the objective of this work was to decrease catalyst degradation by increasing the particle-support interaction through modification of the carbon black surface with functional groups. The functionalisation on the carbon black Vulcan XC72R was carried out in two steps: a chemical oxidation, followed by a condensation reaction of the produced COOH groups with diamines (ethylenediamine (EDA), triethylenetetramine (TETA), phenylenediamine (PDA)) or a dithiol (butanedithiol (BDT)).
The chemical oxidation was conducted with a three-to-one mixture of concentrated sulphuric and nitric acid for varying durations. The successful oxidation was indicated by an increase of the O and H content with roughly a third of the O content originating from COOH groups and the remainder attributed to a variety of oxide groups. The degree of oxidation increased with longer oxidation times, accompanied with a reduction of the BET surface area. Short oxidation times (1-4 hours) provided a sufficient amount of COOH groups while maintaining a large surface area.
The amine and thiol groups were introduced by either direct treatment of an oxidised carbon with a diamine, or via an acyl chloride intermediate step, followed by diamine/dithiol treatment. The latter synthesis route demonstrated higher conversion rates, but entailed increased sulphur impurities. The functionalisations with EDA and TETA were the most successful with EDA mainly displaying monofunctional and TETA displaying bridged functionalisations.
To obtain the nanoparticle-support catalysts, pre-synthesised colloidal PtNi or Pt nanoparticles were deposited on the modified carbon blacks via a precipitation method, adjusted according to the changing polarity of the substrates and nanoparticles. The catalytic activity was investigated using electrochemical half-cell measurements of a working electrode containing a catalyst thin-film. The thin-film morphology, produced by drying a catalyst suspension (ink) on the working electrode, is influenced by the drying conditions. Stationary drying resulted in the formation of a coffee-ring. Flow-restricted drying in a container with a small hole was able to produce more uniform thin-films. However, the influence of the thin-film morphology was negligible towards the electrochemical active surface area (ECSA).
The ECSA was employed to quantify the catalytic activity of the synthesised catalysts before and after an accelerated stress test (AST) to assess the degradation rate. The investigated samples comprised either PtNi or Pt nanoparticles, supported one of the following CB supports: unfunctionalised Vulcan XC72R, oxidised CB, EDA-functionalised, TETA-functionalised. As a reference, the synthesised catalysts were compared with the commercial catalyst (Pt on CB).
Of the prepared catalysts, PtNi catalysts demonstrated higher catalytic activity than Pt catalysts. For both particle types, the Vulcan XC72R-supported catalysts displayed the highest ECSAs, and the highest degree of degradation. The lowest degradation was achieved for PtNi with the EDA-functionalised support and for Pt with the TETA-functionalised support, which also displayed initial ECSAs, comparable to Vulcan XC72R. This may be attributed the preferred interaction of the PtNi with primary amine groups (EDA) and of the Pt particles with secondary amine groups (bridged binding of TETA). Furthermore, the reduced relative degradation rate may be attributed to a removal of the carbon fine structures in the oxidation step. Compared to all prepared catalysts, the commercial catalyst HiSPEC 3000 displayed a higher initial ECSA, but also a higher degree of degradation.
To conclude, the amine-functionalisation of the carbon supports for PtNi and Pt nanoparticles successfully improved the electrochemical stability of the catalysts while maintaining ECSA values comparable to the unfunctionalised carbon support

The learning paths of experts from different fields of action in the areas of experiential education and outdoor training (activities) with regard to their professionalisation of safety and prevention measures - A qualitative study based on explorative expert interviews to reconstruct learning experiences and routines of experts from the fields of alpine, whitewater/canyoning, kayaking & rafting, caving, paddling, cooperation exercises, bivouac & trekking
Erlebnispädagogik und Outdoor-Training haben sich als Aktivitäten etabliert, die von professionell ausgebildeten Trainern angeboten werden. Die Aktivitäten beruhen in der Regel auf einem erlebnisorientierten Ansatz. Es werden spezifische, herausfordernde und oft naturnahe Lernsettings genutzt, um die Teilnehmer zu fördern. Gerade im Alpenraum (in der DACH-Region) sind die geografischen Naturräume prädestiniert für diesen erlebnisorientierten Umgang mit der Natur. Die besonderen Kräfte von Naturgewalten im Gebirge bringen gleichwohl herausfordernde Gefahren mit sich, woraus sich eine ebenso besondere Herangehensweise an die Risiko- und Unfallprävention ergibt. Meine Forschungsarbeit untersucht die langfristige Entwicklung der Professionalisierungsprozesse von Menschen, die sich in der Natur bewegen und dort Gruppen leiten oder führen. Dabei liegt der Fokus auf der Prävention und der Anwendung von Sicherheitsmaßnahmen in Naturerlebnisfeldern wie in Höhlen, auf Bergen, auf dem Wasser, aber auch in künstlichen Erlebnisfeldern wie Klettersteigen und Hochseilgärten.
In der deutschsprachigen Bibliografie der Outdoorsportarten existiert, vor allem in den jeweiligen Spezialbereichen, eine Auswahl an Sicherheitsempfehlungen. Besonders in Richtung Erlebnispädagogik und Outdoor-Training wurde jedoch auch auf angelsächsische Literatur zurückgegriffen, weil dort durch die jahrzehntelange Praxiserfahrung in diesem Bereich viel publiziert worden ist. Aus diesem Grunde gibt es eine einschlägige angelsächsische Bibliografie zu Präventions- und Sicherheitsmaßnahmen, die auch die Erlebnispädagogik und das Outdoor-Training in der DACH-Region beeinflusst haben. Die vorherrschenden Empfehlungen basieren vor allem auf dem Urteilsvermögen der Leitungsakteure.
Das stellt die Forschungslücke dar, denn es wurde zwar viel über Urteilskraft und Urteilsvermögen publiziert, jedoch wurde in der Forschung nicht dargelegt, wie Wahrnehmungs-, Entscheidungs- und Handlungsfähigkeiten entwickelt werden. Was machen Experten anders als Novizen? Meine Forschungshypothese ist, dass es individuelle Unterschiede geben muss, aus denen sich qualifiziertes Urteilsvermögen entwickelt. Es gibt viele Risiken (z. B. Naturgewalt, Teilnehmerverhalten, Materi-albruch etc.), bei denen Leitungsakteure vor der Herausforderung stehen, Risiken wahrzunehmen und entsprechende Entscheidungen treffen zu müssen, um Sicherheit zu gewährleisten.
Mit dieser Studie möchte ich dazu beitragen, das Wissen über die Lernwege von Experten mit jahrzehntelanger Praxiserfahrung in der Erlebnispädagogik und im Outdoor-Training (Outdoor-Sport) zu erweitern. Dabei liegt der Fokus auf der Sensibi-lisierung durch prägende Ereignisse. In meiner Forschungsarbeit gehe ich nicht nur der Frage nach, ob es schwierige Erlebnisse gab, sondern auch der Frage, welche biografisch lebensgeschichtlichen Ereignisse einen Anlass gegeben haben, dass die Experten ihr Bewusstsein über Sicherheitsaspekte verändert haben. Meine Forschungshypothese beruht auf der Annahme, dass Menschen durch prägende Erlebnisse, welche von emotionaler Bedeutung waren, für Gefahrensituationen sensibilisiert wurden. Der Prozess der Sensibilisierung baut auf emotionalen und kognitiven Fähigkeiten auf, auf deren Basis sich die Routinen der Experten entwickeln und modifizieren. Durch Sensibilisierungsprozesse bildet sich ein Urteilsvermögen, das Wahrnehmungs-, Entscheidungs- und Handlungskompetenzen bezüglich Gefahrenmöglichkeiten umfasst. Um die individuellen Erfahrungen zu untersuchen, habe ich in dieser Arbeit auf Basis der biografischen Lernwege Sensibilisierungssituationen rekonstruiert.
Damit ich die Erfahrungen und Verarbeitungsprozesse lebensgeschichtlich erforschen konnte, habe ich mit Handlungsakteuren qualitative explorative Experteninterviews geführt. Diese Akteure verfügen über jahrzehntelange Praxiserfahrung in unterschiedlichen Bereichen (Höhle, Alpin, Wasser und Kooperationsübungen). Mit den Interviews wollte ich herausfinden, wie sie als erfahrene Leitungsakteure ihre Wahrnehmungs-, Entscheidungs- und Handlungsroutinen entwickelt haben. Als Grundlage für diese Befragungen diente die Interviewstrategie zum Führen qualitativer Inter-views nach Richartz (2008). Ebenso wurde der Leitfaden nach den Faustregeln von Richartz (2008) entwickelt, um sicherzustellen, dass bei den Befragten Narrationen erzeugt werden, welche durch gezielt erzählauffordernde Fragen episodisches Wissen hervorholen sollten (Richartz, 2008, 23).
In der Auswertung habe ich mittels qualitativer Inhaltsanalyse individuelle Unterschiede, Kontraste, Gemeinsamkeiten und Veränderungsprozesse hinsichtlich ihrer Routinen und Strategien herausgefiltert. Das Ergebnis meiner Dissertationsarbeit ist die Darstellung von Sensibilisierungsprozessen zur Einschätzung von Risiken durch erfahrene Leitungsakteure. Dies basiert auf der Rekonstruktion der Lernerfahrungen und Routinen von Experten aus den Bereichen Outdoor-Training, Outdoor-Aktivitäten und Erlebnispädagogik. Die Sensibilisierungsprozesse beruhen auf bewältigten Erlebnissen, bei denen Emotionen Verarbeitungsprozesse auslösten. Herausfordernde und gefährliche Situationen wurden nicht alle selbst erlebt. Es reichte den Leitungspersonen aus der Praxis ebenso aus, nur davon zu hören, berichtet zu bekommen oder davon gelesen zu haben. Sensibilisierung erfolgt allerdings immer über Emotionen, welche bei den Befragten ausgelöst wurden. Durch negative Emotionen gab es eine kognitive Suchbewegung, welche zu bewussten Strategien der Risikominimierung geführt hat. Vor allem die Emotion der Besorgnis bewirkte durch das Gefühl, Verantwortung für die Teilnehmer zu tragen, eine Sensibilisierung. Lernanlässe führten mittels dieser längerfristigen Sensibilisierungsprozesse dazu, dass die befragten Leitungspersonen aus der Praxis ihre Routinen überprüften, für wirksam befanden oder veränderten und optimierten. Dies hat sich in meiner Arbeit als zentraler Mechanismus für die Ausbildung von qualifiziertem Urteilsvermögen mit einer gelingenden Unfallprävention im Outdoor-Bereich erwiesen.Experiential education and outdoor training have been established as activities offered by professionally trained trainers. The activities on offer are generally based on an experience-orientated approach, which is why specific settings are used to encourage participants by offering a challenging learning setting that is often close to nature. Especially in the Alpine region (DACH region), the geographical natural areas are predestined for this experience-orientated approach to nature. The unique natural forces in the mountains nevertheless bring about challenging dangers, which necessitate a special approach to risk and accident prevention.For this reason, my research focuses on the long-term development of professionalisation processes for people who are active in nature and lead or guide groups. This includes the prevention and application of safety measures in nature experience fields such as caves, mountains, and water, as well as in artificial experience fields like climbing routes and high ropes courses.
The German-language bibliography of outdoor sports includes a selection of safety recommendations, particularly in specialised areas. However, Anglo-Saxon literature has also been utilised, especially in experiential education and outdoor training, due to the wealth of publications resulting from decades of practical experience in this area. Consequently, relevant Anglo-Saxon literature on prevention and safety measures has influenced experiential education and outdoor training in the DACH region. The prevailing recommendations are primarily based on the judgement of the leaders.This is the gap in the research. While much has been published on judgement and discernment, the development of perceptual, decision-making, and action skills has not been outlined. What do experts do differently from novices? My research hypothesis is that individual differences must exist from which qualified judgement develops. There are many potential risks (e.g., natural forces, participant behaviour, material failure) where managers face the challenge of perceiving risks and making appropriate decisions to ensure safety.
With this study, I aim to contribute to expanding knowledge about the learning paths of experts with decades of practical experience in experiential education and outdoor training (outdoor sports), particularly in relation to their sensitisation through formative events. My research work not only investigates whether there were challenging experiences but also which biographical and life-historical events led the experts to change their awareness of safety aspects. My research hypothesis is that people were sensitised to dangerous situations by formative experiences that were of emotional significance. The process of sensitisation builds on emotional and cognitive abilities that develop and modify the experts’ routines. Sensitisation processes develop a capacity for judgement, which includes perceptual, decision-making, and action skills concerning potential dangers. To investigate individual experiences, I have reconstructed sensitisation situations in this thesis based on biographical learning paths.
To explore the experiences and processing procedures from a life history perspective, I conducted qualitative explorative expert interviews with individuals who have decades of practical experience in different areas (cave, alpine, water & cooperation exercises). These interviews aimed to understand how they have developed their perception, decision-making, and action routines as experienced leaders. The interview strategy for conducting qualitative interviews was based on Richartz (2008). The guidelines were also developed according to Richartz’s (2008) rules of thumb to ensure that narratives were generated in the interviewees, which were intended to extract episodic knowledge by asking specific questions that prompted narratives (Richartz, 2008, 23).
In the evaluation, I employed qualitative content analysis to identify individual differences, contrasts, similarities, and change processes in their routines and strategies. The outcome of my dissertation is the presentation of sensitisation processes for risk assessment by management actors from practice, based on the reconstruction of the learning experiences and routines of experts in outdoor training, outdoor activities and experiential education. The sensitisation processes are rooted in mastered experiences where emotions triggered processing processes. Not all challenging and dangerous situations were experienced by the participants; it was sufficient for them to have heard about, been told about, or read about such incidents. However, sensitisation always occurs through emotions triggered in the interviewees. Negative emotions prompted cognitive search movements, leading to conscious risk minimisation strategies. The emotion of concern, in particular, has led to sensitisation through the feeling of responsibility for the participants. These longer-term sensitisation processes have resulted in the interviewed practitioners reviewing their routines, finding them effective, or modifying and optimising them accordingly. In my study, this has proven to be a central mechanism for developing qualified judgement, ensuring successful accident prevention in the outdoor sector
Die Funktion einer E3 Ubiquitin-Ligase in der Bildung von Membran-Membran-Kontaktflächen
Die pflanzliche Immunantwort ist präzise reguliert, sodass selbst geringfügige Veränderungen die Fitness einer Pflanze erheblich beeinflussen können. In diesem Zusammenhang spielen Ubiquitin-Ligasen, wie die SENESZENZ-ASSOZIIERTE E3 UBIQUITIN-LIGASE 1 (SAUL1), eine zentrale Rolle bei der Regulation von stressinduzierten Reaktionen in Arabidopsis thaliana. Während Temperaturen unter 23 °C eine Autoimmunantwort in saul1-1 knock-out Pflanzen induzieren, zeigen diese Pflanzen, durch eine konstitutive basale Immunantwort, bei 25 °C eine erhöhte Resistenz gegenüber bakteriellen Infektionen. Der genaue Mechanismus, durch den SAUL1 die pflanzliche Immunantwort reguliert, ist jedoch bisher ungeklärt. In dieser Arbeit wurden strukturelle Modifikationen am SAUL1-Protein vorgenommen, um die Funktionalität der rekombinanten Proteine zu untersuchen. Insbesondere wurde die
Affinität der Armadillo-like Domäne 7-11 zur Plasmamembran untersucht, indem verkürzte Teildomänen generiert und untersucht wurden. Dabei wurde gezeigt, dass die vollständige ARM7-11 Domäne für die Lokalisation von SAUL1 an der Plasmamembran notwendig ist. Darüber hinaus wurden gezielte Modifikationen in der positiv geladenen Region dieser Domäne durchgeführt. Hierbei wurden die drei Arginine R736, R737 und R775 durch die ungeladene Aminosäure Alanin, das positiv geladene Lysin oder die negativ geladene Glutaminsäure ausgetauscht. R736 trägt eine entscheidende Rolle bei der Lipid-Lipid-Interaktion von SAUL1. Im Vergleich zu GFP-SAUL1 war GFP-SAUL1R736A nicht in der Lage Membranflecken zu bilden. Dieses Phänomen wurde mithilfe von einfach, zweifach und dreifach punktmutierten SAUL1 in A. thaliana Mesophyllprotoplasten untersucht. Dabei konnte eine Dynamik der Membranflecken, ihre Induzierbarkeit sowie eine scheinbar konzentrationsabhängige Bildung von SAUL1-Vesikeln festgestellt werden. Für in vivo Experimente wurden transgene Arabidopsis thaliana Pflanzen mit den generierten Arginin zu Alanin Punktmutanten erstellt und sowohl phänotypisch als auch mikroskopisch untersucht. Das rekombinante Protein SAUL1R736A wurde exprimiert, aufgereinigt und mittels CD-Spektroskopie mit dem Wildtyp-Protein SAUL1 verglichen, wobei keine strukturellen Unterschiede festgestellt wurden. Anschließend wurden beide rekombinanten Proteine für Protein-Lipid-Interaktionsexperimente verwendet, bei denen eine Affinität von SAUL1 zu spezifischen Plasmamembranlipiden sowie Lipiden der Phosphatidylinositol-Familie nachgewiesen werden konnte
