    Evaluation of project management education in terms of academic stress factors

    İnşaat sektörünün beklentilerini karşılayabilmek amacıyla, yönetim içerikli dersler üniversitelerde; mühendislik yönetimi, yapım yönetimi ve proje yönetimi (PM) dersleri ile inşaat mühendisliği ve mimarlık disiplinleri eğitim planlarına dâhil edilmiştir. Yüksek lisans veya lisans düzeyindeki PM bilgi alanlarını içeren derslerin amacı; öğrencilerin teknik, yönetsel ve kişisel becerilerini geliştirmesine ve sektörde çalışmaya hazır insan kaynağının sağlanmasına yöneliktir. Proje ekibinde stres yönetimi proje yöneticilerinin sorumluluk alanlarından biri olmakla birlikte, proje ekibindeki insan kaynağı gelişiminin ve projenin başarısı için de önem taşımaktadır. Bu bağlamda, mimarlık fakültelerinde lisans düzeyinde verilen PM eğitiminde, öğrencilerin akademik stres faktörleri karşısında, gösterdikleri tepkileri anlamlandırabilmek, hem kişisel gelişim açısından hem de dersin içeriğinin, dersi veren öğretim elemanlarının gelişimi açısından önem arz etmektedir. Araştırmanın tasarımında, Türkiye ve dünya üzerindeki PM eğitimi ve akademik stres faktörlerine yönelik teorik altyapının hazırlanmasının ardından, ilgili konu başlıkları belirlenmiş ve 15 soruluk çevrimiçi bir anket çalışması hazırlanmıştır. Araştırmacıların üç akademik dönem boyunca PM dersi verdikleri İstanbul’da bir vakıf üniversitesinde düzenledikleri pilot anket çalışmasını takiben, PM dersi alan öğrencilerden 99 adet geri bildirim elde edilmiştir. Akademik stres faktörlerinin evreni, bu ders ile ilgili çalışma kapsamında belirlenen üç akademik stres faktörü için sınırlandırılmıştır, dersin öğretim elemanı, dersin dili, ders teslimleri için bir PM yazılımı kullanılmasına yönelik öğrenci görüşleri sorulmuş ve cinsiyet faktörünün stres anında gösterilen tepkiler açısından farklılığı da araştırılmıştır. Elde edilen veriler, 5'li Likert ölçeği üzerinden sorgulanmış, SPSS (stastical package for the social sciences) paket programı yardımıyla işlenmiş ve araştırma kapsamında önerilen hipotezler sınanmıştır. Son olarak, çalışma bulgularına yönelik tartışma ve öneriler sunularak çalışma tamamlanmıştır.The construction industry's aim is to meet expectations by including management-related courses in universities; engineering management, construction management, and project management (PM) courses, as well as building engineering and architecture disciplines' educational plans. The purpose of the PM knowledge areas included in graduate or undergraduate level courses is to develop students' technical, managerial, and personal skills and to provide a human resource ready to work in the industry. Stress management is one of the responsibilities of project managers in a project team, but human resource development and project success are also important. In this context, in the PM education given at the undergraduate level in architecture faculties, it is important to understand the responses of students to academic stress factors, both for personal development and for the development of the content of the course and the development of the instructors. The study design, after preparing the theoretical background for PM education and academic stress factors in Turkey and the world, identified the relevant topics and prepared a 15-question online survey study. After the pilot survey study conducted by the researchers in an Istanbul-based foundation university where they taught PM courses for three academic periods, 99 student feedbacks were obtained. The scope of the academic stress factors in this study was limited to three academic stress factors that were determined within the scope of the study: the instructor of the course, the language of the course, and the use of a PM software for course submissions. The study also examined the difference in the responses shown during stress according to the gender factor. The data collected was analyzed using the SPSS (Statistical Package for the Social Sciences) package program and the hypotheses proposed in the study were tested.     Exploring the molecular mechanism of linagliptin in osteosarcoma cell lines for anti-cancer activity

    Background: Finding new applications for widely used current drugs is a fast and effective technique for discovering new anticancer chemicals. Osteosarcoma (OS), the most prevalent form of bone cancer, has several side effects that significantly lower patients' quality of life. This study aims to systematically examine the anti-cancer activity of linagliptin (LG) in the osteosarcoma cell line Saos-2. Methods: MTT assays and flow cytometry were used to assess cell viability and apoptosis, respectively. qPCR array experiments were carried out to determine target gene expressions and explain the molecular mechanism of LG's action. Results: Linagliptin treatment significantly decreased the viability of Saos-2 cells and hFOB1.19 cells (p < 0.001). The treatment also induced increased apoptotic effects in both Saos-2 cells (p < 0.001) and hFOB1.19 cells (p < 0.05). qPCR assays were conducted to assess cancer pathway analysis after applying specific quantities of LG to Saos-2 and hFOB1.19 cells. Conclusion: The findings of this study demonstrate that LG inhibits the proliferation of Saos-2 cells and induces cell death. LG supports cell death by suppressing the expression of specific genes involved in cancer pathways

    The way to smart healthcare: A comprehensive research on technology assessment methodologies for digital health systems

    Digitalization of healthcare is not a new concept; however, nowadays the need for change has become obvious. The rapid increases in technological advancements in health and information technologies enable them to converge and develop in harmony with each other. Digital technologies are ubiquitous and aim to improve healthcare systems and allow new business models. From this point of view, selection and prioritization of them in order to keep up with the technological developments become vital. However, the allocation of resources to meet increasing demands is a multifaceted problem. In this framework, this study aims to analyze the way to smart healthcare. Therefore, initially, current technological implementations in health are reviewed. Afterward, the structure of health technology assessment models used in the selection of technology is detailed. Finally, the managerial strategies for administrative levels are presented in order to form the basis for further studies in this field

    Long-term endodontic management of previously traumatized immature maxillary central incisors 8-year follow-up

    Introduction: Although pulpal prognosis is good for uncomplicated crown fractures of open-apex incisors, the pulp can become necrotic and require endodontic intervention. This case report presents endodontic management of previously traumatized immature incisor. Case report: An 8-year-old girl was admitted with an abscessed and painful maxillary central incisor with an enamel-dentin fracture restored previously with composite resin. Radiographic examination showed periapical radiolucency with an open apex, therefore regenerative endodontic treatment was performed. After 15 months, periapical pathology was also observed in the maxillary right central incisor, thus regenerative treatment was applied. Retreatment was required in both teeth after 3 and 4 years due to new periapical lesion. The teeth were treated with hydraulic filling technique involving MTA sealer and single-cone gutta-percha. After 36 months follow-up, there was complete periapical healing. Discussion: In the case of failed regenerative endodontic treatment, conventional methods can provide predictable outcomes. Single cone obturation technique with MTA has shown promising results for necrotic immature anterior teeth with apical periodontitis. Conclusion: The present case report shows successful management of traumatized immature with the single cone filling technique after unsuccesful management using regenerative endodontic therapy

    Bilme eylemi noktasında belki de en çok kullanılan duyunun görme duyusu olduğu ifadeedilebilir. Görsel olanla kurulan ilgi bu bilme eylemini içermektedir. Görsel olan ister bir gösterge, temsil ister bir sanat yapıtı olsun bireyle, izleyiciyle direkt bağlamlar oluşturur.Görsel bir sanat olan sinema içerisinde; mekân, karakter, kurgu, kıyafet, nesne vb. farklı tasarım noktaları barındıran bir organizasyon bütünlüğü sunar. Bu noktada tasarım öğeleri sinema eserinin gerçekliği oluşturan parametrelerine dönüşür.Bu çalışma tam olarak bu parametrelerden bir tanesi olan nesneyle kimlik ilişkisine odaklanmaktadır. Sinema eseri içerisinde kullanılan tasarlanmış nesnelerin salt olarak bir nesne olmaktan ötesini barındırdıkları fikri bu çalışmayı imkânlı kılmıştır. Eser sahibinin söylemek istediği bazı söylemlerin nesneler üzerinden anlamlandığı anlar tartışmanın konusunu oluşturmaktadır. Bu söylemler gösterge bilimsel analizle irdelenmiştir.Bu bağlamda ilgili düşüncenin araştırılması için John Ronald Reuel Tolkien’in Yüzüklerin Efendisi serisi çalışmanın temel araştırma alanını oluşturmakta, bu alan içerisinde özellikle kimlik ve karakterle ilişki kuran aynı zamanda beden ve kurguyla uzamlar oluşturan öğeler olarak savaş enstrümanları temel nesneler olarak ele alınmıştır.Çalışmanın strüktürü; ilgili enstrümanların fonksiyonel, ergonomik, estetik vb. gibi tasarımsal ve sanatsal özelliklerinin kimlik ve karakterle bağdaştırılması aynı zamanda eser içerisindeki söylemlerinin keşfedilmesiyle oluşmaktadır. Sinema ve tasarım ara kesitinde var olan çalışma üretimle temsili, nesneyle kimliği, formla karakteri, olguyla hikâyeyi vb. buluşturan noktalara temas etmektedir. Sonuçta ilgili yapıtlarda var olan tasarlanmış nesnelerin birer kimlik anlatı aracı olduğu, hikâyede var olan bazı kaideleri doldurduğu ortaya konulmuş, sinema ve sinematografik anlatım içerisinde tasarım nesnesinin yeri ve pozisyonu keşfedilmeye çalışılmış, ürün-kimlik bağlamları oluşturulmuştur.The phenomena of displaying, watching and viewing, which are as old as mankind, have existed in different forms and methods throughout the history; evolving, developing, and continuing to exist alongside humans. This development not only changed and improved the essence of the phenomenon with various techniques but also renewed the perception of the viewer along with it. In terms of knowing, perhaps, the most frequently employed faculty is the sense of sight. Although the distribution among the employment of different senses is known to be quite balanced in the early periods of human history, in today’s world, the sense of sight is clearly the most frequently employed faculty. The relevance established with the visual should fundamentally be handled in this context. Whether the visual is a sign, a performance, or a work of art, it forms direct and indirect contexts with the individual and the viewer.In terms of knowing and perceiving visuality, plenty of different techniques and methods have been used in the course of history. Today, cinema exists in a central position in terms of viewing and knowing despite its relatively short history compared to several other forms of visual art due to its technological requirements. This position exists in a reality that majorly impacts the form of both narration and perception. The cinema, perhaps the most effective visual art in terms of conveying messages.As in many other fields, the phenomenon of design can also be considered among the leading actors in the field of cinema. The design can be read in every aspect of the cinema’s existence as a form of organization within its own essence. Beyond the simple act of problem solving, this organization involves providing an aesthetic and ergonomic solution to the problem. It forms meaning and context. At this point, the creation process comes into existence from its own essence and evolves from an idea to reality.Cinema offers an organizational integrity that includes different design points such as space, character, fiction, object, etc. At this point, these design elements turn into parameters that comprise the reality of the cinematic work. Cinema and design exist as two different representations of the same movement. The same complex structure can be observed in many areas of art either in mental or productional terms. In particular, art fields that contain fiction, action patterns, character, and planning exist in this contextuality, just like design.This study focuses precisely on the relationship between meta and identity, which constitutes one of these parameters. The study came to life with the idea that the designed objects used in the cinema works are more than just simple objects. The moments when certain discourses of the author are interpreted through metas make up the subject of the discussion. In this context, J.R.R. Tolkien’s The Lord of the Rings trilogy constitutes the main research area of the study in order to investigate the related thought. In this field, war instruments are considered as basic objects, especially as elements that relate to identity and character, but also create spaces with body and fiction. However, other books and movies such as The Hobbit and The Silmarillion can be expressed as additional areas that are mentioned when necessary. The study interprets the information obtained from the scenes from a perspective of design, making detailed and comparative analyzes of the war instruments used in the scenes. In this regard, the instruments have been depicted one by one, which enabled to reveal and perceive their differences. The study objectively analyzes the concepts such as the stories of the instruments, the background of the people who own them, their lives and deeds, their heroism, etc., and then examines their structures, places, narration styles, and positions within the work.The study is structured in a way that associates the design and artistic features of the relevant instruments such as their functional, ergonomic, and aesthetic qualities with identity and character, as well as in a way that discovers their discourses within the work. The study, which exists in the intersection of cinema and design, touches on the points that unite production and representation, meta and identity, form and character, phenomenon and story, etc. This discussion, which creates contexts between instruments, fiction, characters, and stories, concludes that the objects used in cinema works are, beyond being just functional objects, a means of representation, markers of the characters’ identities, and carriers of past details that the audience is unaware of and unable to see in the fil

    İnsanın doğada var olma serüveni evrim teorisinden, sanat ve mimarlık gibi kültürel etkinliklere kadar uzanan oldukça kapsamlı ve karmaşık bir süreçtir. Paleolitik çağ döneminde hayatta kalma amacıyla; avlanma, yiyecek toplama ve barınaklar inşa etme eylemleri ile başlayan bu deneysel yolculuk, ortaya çıkan problemler neticesinde ilkel alet tasarımlarının da gelişimini sağlamıştır. Özellikle Holosen çağ ve üst Paleolitik dönemden itibaren; taş, kemik, kil ve diğer malzeme kullanımı ile başlayan tasarım evrimi sonrasında resim, heykel gibi üretimlere de yansımıştır. İnsanların doğayı taklit etmesinin nedeni; doğadaki renkleri, geometriyi ve hareket yeteneğini anlamaya çalışırken, tasarımı işlevsel ve estetik açıdan güzel olan şeylere dönüştürme isteğidir. Bu bağlamda, insanın kendinden başka ve tür açısından kendine en yakın hayvan iskeletlerinin form ve işlevi ile kurduğu bağ; gözlem, merak ve deneysellik kesişiminde halen güncel tasarım ihtiyaçlarını karşılamaktadır. Doğanın ve evrenin sırlarını keşfetmek, teknolojiyi geliştirmek ve dünyanın geleceğini şekillendirmek için yapılan tüm üretimler sayesinde bugün insanlar; farklı kültürler, diller ve geleneklerle bir arada modern dünyada yaşayabilmektedirler. İnsanın doğadan ilham alma ihtiyacını ve yeteneğini anlayabilmek için, tarihsel süreç içerisinde ilkel medeniyetler ve medeni kabul edilen kültürlerin; tıp, sanat ve mimarlık alanlarında hayvan form ve iskeleti ile benzerlik gösteren, ürünlerini incelemek yol gösterici olacaktır. Günümüzde modern mimarların projelerinde, yapı tasarımı ve strüktürünü; biyomimetik felsefe ile hayvan iskeleti üzerinden, geometri ve işleve nasıl dönüştürdükleri bu nedenle çalışma kapsamında değerlendirilmiş başlıca konular arasındadır.The journey of human existence in nature is a comprehensive and complex process that ranges from the theory of evolution to cultural activities such as art and architecture. This experimental journey, which began with hunting, food gathering, and shelter construction activities for survival in the Paleolithic era, also ensured the development of primitive tool designs as a result of the problems encountered. Especially from the Holocene era and Upper Paleolithic period onwards, the design evolution that started with the use of materials such as stone, bone, clay, and others also reflected on productions such as painting and sculpture. The reason why humans imitate nature is the desire to transform what they understand about natural colors, geometry, and motion into functional and aesthetically beautiful designs. In this context, the connection established by humans with the form and function of animal skeletons, which are closest to themselves in terms of species other than themselves, still meets current design needs at the intersection of observation, curiosity, and experimentation. Thanks to all the productions made to discover the secrets of nature and the universe, develop technology and shape the future of the world, people today can live together in the modern world with different cultures, languages, and traditions. To understand human's need and ability to take inspiration from nature, examining the products of primitive civilizations and cultures considered civilized, which show similarities with animal form and skeleton in the fields of medicine, art, and architecture, can be guiding in the historical process. In the projects of modern architects today, how they transform the geometry and function of the structure and design of the building through the animal skeleton with the biomimetic philosophy is therefore among the main topics evaluated in this study

    Kök ve kanal morfolojisinin kapsamlı bir şekilde bilinmesi ve anlaşılabilmesi, endodontik tedavide başarının temel taşlarından biridir. Bu amaçla geçmişten günümüze, boyama ve şeffaflaştırma, radyografik görüntüleme, konik ışınlı bilgisayarlı tomografi ve mikro bilgisayarlı tomografi gibi görüntüleme ve araştırma teknikleri kullanılmıştır. Bu tekniklerin kullanıldığı pek çok çalışma, dental morfoloji veri tabanına önemli katkıda bulunmuş, sadece nitel değil nicel veriler de sunmuş ve böylece kök kanal preparasyon sınırlarını da tanımlayabilmiştir. Böylece endodontistler ve endodontik tedavi yapan hekimler, tedavi esnasında karşılaşabileceği kök kanal varyasyonları hakkında bilgi sahibi haline gelmiştir. Ayrıca, kavite preparasyonundan, tedavinin sonlandığı minör apikal foramene kadar olan tedavi sınırlarını çizebilmiştir. Böylece, bu bölüm kök kanal morfolojisi incelenmenin, laboratuvar ve klinik araştırma yöntemlerini sunmayı amaçlamıştır.Comprehensive knowledge and understanding of root and canal morphology are keystones of success in endodontic treatment. For this purpose, imaging and research techniques such as staining and transparency, radiographic imaging, cone beam computed tomography and micro computed tomography have been used from the past to the present. Many studies using these techniques have contributed significantly to the dental morphology database, providing qualitative and quantitative data, thus defining root canal preparation boundaries. Thus, endodontists and endodontic physicians have become aware of the root canal variations they may encounter during treatment. Moreover, they can draw the treatment boundaries from the cavity preparation to the minor apical foramen, where the treatment ends.     Evaluation of the diagnostic performance of a SARS-CoV-2 and Influenza A/B combo rapid antigen test in respiratory samples

    This study aimed to evaluate the performance characteristics of a rapid antigen test developed to detect SARS-CoV-2 (COVID-19), influenza A virus (IAV), and influenza B virus (IBV) (flu) compared with those of the real-time reverse transcription-polymerase chain reaction (rRT-PCR) method. One hundred SARS-CoV-2, one hundred IAV, and twenty-four IBV patients whose diagnoses were confirmed by clinical and laboratory methods were included in the patient group. Seventy-six patients, who were negative for all respiratory tract viruses, were included as the control group. The Panbio™ COVID-19/Flu A&B Rapid Panel test kit was used in the assays. The sensitivity values of the kit were 97.5%, 97.9%, and 33.33% for SARS-CoV-2, IAV, and IBV, respectively, in samples with a viral load below 20 Ct values. The sensitivity values of the kit were 16.7%, 36.5%, and 11.11% for SARS-CoV-2, IAV, and IBV, respectively, in samples with a viral load above 20 Ct. The kit’s specificity was 100%. In conclusion, this kit demonstrated high sensitivity to SARS-CoV-2 and IAV for viral loads below 20 Ct values, but the sensitivity values were not compatible with PCR positivity for lower viral loads over 20 Ct values. Rapid antigen tests may be preferred as a routine screening tool in communal environments, especially in symptomatic individuals, when diagnosing SARS-CoV-2, IAV, and IBV with high caution

    Determination of content, cooking loss, and bioaccessibility of thiamine in red meats using in vitro simulated human gastrointestinal digestion system

    Meats are considered a valuable part of the human diet and a best source of B-group vita-mins. Thus, investigations that measure the losses by cooking and bioaccessibilities of micronutrients are crucial to knowing the vitamin amounts in meats. The aim of this research was to investigate the content and loss of thiamine in meats by four different cooking practices, boiling, roasting, grilling, and frying, and to determine the bioaccessibility of thiamine in cooked meats by a simulated in vitro human gastrointestinal digestion system. High-performance liquid chroma-tography was used to detect the contents of thiamine in each raw and cooked meat. In raw meats, the meas-ured thiamine amounts varied from 58.67 to 159 μg/100 g. After cooking, the amounts of thiamine ranged between 21 – 93 μg/100 g. While the lowest thi-amine loss was determined in the boiled mutton thigh (8%), the highest loss was found in roasted veal thigh and mutton sleeve with 64%. The bioaccessibilities of thiamine in meats after digestion ranged from 12 to 83 μg/100 g. The highest thiamine bioaccessibility was in the grilled veal chop with 91%, while the lowest bi-oaccessibility was found in the roasted mutton sleeve with 12%. The bioaccessibility of vitamins may be re-markably influenced by factors such as stability, pH of the gastrointestinal tract, and temperature. Deter-mining the bioaccessibility of thiamine in meats after cooking and digestion is important for the correct evaluation of the daily intake values of these vitamins.İstanbul Sabahattin Zaim Universit
