    The therapeutic effects of bioactive compounds on colorectal cancer via PI3K/Akt/mTOR signaling pathway: A critical review

    Understanding the molecular signaling pathways of colorectal cancer (CRC) can be accepted as the first step in treatment strategy. Permanent mTOR signaling activation stimulates the CRC process via various biological processes. It supplies the survival of CRC stem cells, tumorigenesis, morbidity, and decreased response to drugs in CRC pathogenesis. Therefore, inhibition of the mTOR signaling by numerous bioactive components may be effective against CRC. The study aims to discuss the therapeutic capacity of various polyphenols, terpenoids, and alkaloids on CRC via the PI3K/Akt/mTOR pathway. The potential molecular effects of bioactive compounds on the mTOR pathway's upstream and downstream targets are examined. Each bioactive component causes various physiological processes, such as triggering free radical production, disruption of mitochondrial membrane potential, cell cycle arrest, inhibition of CRC stem cell migration, and suppression of glycolysis through mTOR signaling inhibition. As a result, carcinogenesis is inhibited by inducing apoptosis and autophagy. However, it should be noted that studies are primarily in vitro dose-dependent treatment researchers. This study raises awareness about the role of phenolic compounds in treating CRC, contributing to their future use as anticancer agents. These bioactive compounds have the potential to be developed into food supplementation to prevent and treat various cancer types including CRC. This review has the potential to lead to further development of clinical studies. In the future, mTOR inhibition by applying several bioactive agents using advanced drug delivery systems may contribute to CRC treatment with 3D cell culture and in vivo clinical studies.This article has funded in the scope of TUBITAK-ULAKBIM “Read and Publish” agreement

    Determination of carnosic acid by a novel HPLC-UV method in human plasma and application to a prototype pharmacokinetic study

    An HPLC method with UV detection was developed for the determination of carnosic acid in human plasma and applied to a pharmacokinetic study after oral administration of Rosemary extract to a healthy volunteer. Sample preparation depends on liquid-liquid extraction with hexane. Chromatographic separation was achieved with C18 column (150 mm × 4.6 mm × 5 μm), at 25°C with isocratic elution, mobile phase composed of solution A (methanol), and solution B (2% o-phosphoric acid in water) (90:10, v/v) at flow rate of 1.0 mL/min. The analyte was detected at 230 nm. The retention time is 4.20 ± 0.03 min. The method was validated in terms of accuracy, precision, specificity, robustness and detection and quantification limits, in accordance with European Medicines Agency guidelines. LOD and LOQ were found to be 0.075 and 0.25 ng/mL, respectively. The method was applied to the analysis of carnosic acid in human plasma with good recovery as 91.7%. The plasma concentration-time profile and pharmacokinetic parameters: AUC0-t, AUC0-∞, Cmax, tmax, t1/2 were calculated according to the assays. The method can certainly be used for routine analysis of carnosic acid in human plasma after oral administration of Rosemary extract, and for phase I clinical studies and bioavailability-bioequivalance studies as well.Bezmialem Vakıf University -- 2020020

    Introduction: Family medicine encompasses a unique set of competencies and characteristics aimed at providing primary care with a patient-centered focus. In this paper, we aimed to present a case study demonstrating how family practice competencies were utilized in the management of central retinal vein occlusion (CRVO). Case: A 48-year-old diabetic man, seeking a driving license health report, presented without complaints. Even though he self-reported normal vision, a comprehensive examination revealed a visual loss in his left eye, leading to a diagnosis of CRVO. Despite the absence of typical risk factors such as hypertension or hyperlipidemia, detailed examination and comprehensive evaluation of the patient offered the patient a chance for diagnosis and treatment. Discussion: CRVO often manifests as painless unilateral vision loss. Family physicians play a crucial role in early detection through comprehensive assessments, even in asymptomatic patients. Rapidly diagnosis and prompt referral to ophthalmologists allow for timely intervention, as seen in this case where intravitreal bevacizumab injections led to improved visual acuity. Conclusion: This case highlights the importance of family physicians detecting signs of disease, in patients without obvious symptoms. Family physicians contribute to the early diagnosis and effective management of many diseases with their comprehensive approach and patient-oriented care.Giriş: Aile hekimliği, hasta merkezli, birinci basamak sağlık hizmeti sunan, benzersiz bir dizi yeterlilik ve özelliği sahip bir tıp disiplinidir. Bu vaka takdiminde, aile hekimliği çekirdek yeterliliklerinin kullanılarak santral retinal ven tıkanıklığının (CRVO) tanısı ve yönetiminin tartışıldığı bir vaka sunmayı amaçladık. Vaka: Ehliyet sağlık raporu almak isteyen 48 yaşındaki diyabet tanılı erkek hasta şikayetsiz olarak başvurdu. Görüşünün normal olduğunu bildirmesine rağmen, kapsamlı muayene sonucunda sol gözde görme kaybı olduğu tespit edildi. Hastaya ileri değerlendirme ile CRVO tanısı konuldu. Hipertansiyon veya hiperlipidemi gibi tipik risk faktörlerinin bulunmamasına rağmen hastanın ayrıntılı muayenesi ve kapsamlı değerlendirilmesi hastaya tanı ve tedavi şansı sunmuştur. Tartışma: CRVO sıklıkla ağrısız tek taraflı görme kaybı olarak kendini gösterir. Aile hekimleri, asemptomatik hastalarda bile kapsamlı değerlendirmeler yaparak erken teşhisin sağlanmasında önemli bir rol oynamaktadır. Hızlı tanı ve oftalmologlara hızlı sevk; intravitreal bevacizumab enjeksiyonlarının görme keskinliğinde iyileşme sağladığı bu vakada görüldüğü gibi zamanında müdahaleye olanak tanır. Sonuç: Bu vaka, aile hekimlerinin, belirgin semptomları olmayan hastalarda bile, hastalıkların bulgularını tespit etmelerinin önemini vurgulamaktadır.     Diagnostic role of opsonic activity in Acinetobacter baumannii ventilator-associated pneumonia

    In this study, we investigated the diagnostic value of opsonic activity against Acinetobacter baumannii in Ventilator-Associated Pneumonia (VAP) among 50 patients, compared to 102 negative and positive controls. Out of the 50 patients, only 33 (66 %) were diagnosed with VAP using the Clinical Pulmonary Infection Score (CPIS). The opsonic activity assay demonstrated three key findings: (i) 95 % sensitivity and 91.7 % specificity, with a Receiver Operating Characteristic (ROC) area of 0.976 for distinguishing A. baumannii culture positives from negatives; (ii) 95 % sensitivity and 78.7 % specificity, with a 0.915 ROC area, in differentiating VAP/blood culture positive patients from colonized/negative groups; (iii) An ROC area of 0.553 for VAP and colonization, as identified by CPIS alone, indicating an indeterminate threshold. These results highlight that CPIS, microbiological, and clinical evaluations were not correlated, suggesting that opsonic activity against A. baumannii could be a potential VAP diagnostic tool, with the need for large-scale validations.Istanbul University Scientific Research Projects -- 4010

    The effect of nutritional knowledge level of female students at Iğdır University on hedonic hunger

    Duyguların, insanların yeme alışkanlıklarını etkilediği ifade edilmektedir. Sağlık açısından zararlı yeme davranışlarının tespit edilmesi ve bu davranışların gelişiminin engellenmesi, fiziksel, duygusal ve sosyal problemleri azaltarak bireylerin yaşam kalitesini artırdığı bilimsel çalışmalarla kanıtlanmıştır. Bu çalışma Mart-Nisan 2024 tarihleri arasında Iğdır Üniversitesi’nde okumakta olan 18-35 yaş arasındaki kız öğrenciler dahil edilmiştir. Katılımcı sayısı tahmini olarak 384 kişi olarak planlamıştır. Beslenme Bilgi Düzeyinin Hedonik Açlık üzerine etkisi amaçlanmıştır. Bu araştırmanın veri toplanmasında anket yöntemi kullanılmıştır. Çalışma araştırmaya katılmayı kabul eden gönüllü bireyler üzerinde yürütülmüştür. Anketler yüz yüze görüşme yöntemiyle yapılmıştır. DİA olumlu ve olumsuz durumlar ile iştahlı biri olma arasında istatistiksel açıdan anlamlı farklılık saptanmıştır (p<0,05). İştahlı olduğunu belirten bireylerin olumlu durum ortalaması (52,70±14,13) p <0,012, iştahsız olanlardan (48,87±15,20) p<0,001 anlamlı şekilde daha yüksek bulunmuştur. Duygusal iştah anketi olumsuz durumlarda, alkol kullanan kişilerin ortalaması 50,25±14,23 iken, düzenli alkol kullanmayan kişilerin ortalaması 51,16±14,73 olarak bulunmuştur. Beslenme bilgi düzeyi ölçeğinde ise, temel beslenme bilgisi düzenli alkol kullanan kişilerin ortalaması 47,25±6,81, kullanmayan kişilerin ise 52,25±7,27 olarak bulunmuştur. Her iki durumda da Mann-Whitney U Testi sonuçları p<0,05 olarak bulunmuştur, ancak, iki koşulda da anlamlı bir fark saptanmamıştır.The study aims to explore the influence of nutritional knowledge on hedonic hunger, focusing on identifying harmful eating behaviors and preventing their development to enhance individuals' quality of life by reducing physical, emotional, and social problems. Conducted between March and April 2024, the research included female students aged 18-35 enrolled at Iğdır University, with an estimated participation of 384 individuals. Face-to-face interviews were conducted for data collection. The relationship between participants' nutritional knowledge level and hedonic hunger was not identified. The Emotional Appetite Survey revealed a statistically significant difference (p<0.05) between positive and negative situations and the inclination towards being hungry. Participants who indicated being hungry had a significantly higher average in positive situations (52.70±14.13) compared to those not feeling hungry (48.87±15.20). In negative situations, individuals who consumed alcohol had an average score of 50.25±14.23, whereas those who did not consume alcohol regularly scored an average of 51.16±14.73. Similarly, in the Nutrition Knowledge Scale, individuals who consumed alcohol regularly scored an average of 47.25±6.81, whereas non-consumers scored 52.25±7.27. In both cases, Mann-Whitney U Test results indicated a statistically significant difference (p<0.05)

    Türk üniversitelerinde hemşirelik alanında yürütülen liderlik konulu lisansüstü tezlerin analizi

    Objective: This study aimed to analyze different variables of graduate theses related to leadership conducted in the field of nursing in Turkish Universities. Methods: This is a bibliometric and descriptive study. The search for the theses was carried out in May 2022 in the National Thesis Center of the Council of Higher Education using “leadership” and “nursing” as descriptors. Results: A total of 48 theses were included. Most theses (45.83%) were published between 2017 and 2022. 66.67% of the theses were conducted at public universities. Almost all the theses were produced by the departments of nursing, nursing management, or nursing fundamentals and management. In most of the theses, the samples consisted of staff nurses. Quantitative approaches were present in all the theses and scales were the most used data collection instruments (56.25%). The most discussed topics were “leadership behaviors” (29.17%), “leadership styles” (20.83%), and “transformational leadership” (14.58%). Conclusion: It is suggested that leadership be more widely discussed across different nursing departments and featured in a greater number of doctoral theses, primarily through qualitative approaches. Nursing students should be considered more as research samples, as it is important to investigate future nurses’ opinions and knowledge about leadership.Amaç: Bu çalışma, Türk üniversitelerinde hemşirelik alanında gerçekleştirilen liderlikle ilgili lisansüstü tezlerine ilişkin farklı değişkenleri analiz etmeyi amaçlamaktadır. Yöntem: Bu çalışma bibliyometrik ve tanımlayıcıdır. Tezlerin aramasında Yükseköğretim Kurulu (YÖK) Ulusal Tez Merkezi’nde detaylı arama sayfasında tez adı kısmında “liderlik” anahtar kelimesi ve konu bölümünde “hemşirelik” seçildi. Bulgular: Toplamda 48 tez dahil edilmiştir. Tezlerin çoğu (%45,83) 2017-2022 yılları arasında yayımlanmıştır. Tezlerin %66,67’si devlet üniversitelerinde yapılmıştır. Neredeyse tüm tezler hemşirelik, hemşirelik yönetimi veya hemşirelik esasları ve yönetimi departmanları tarafından üretilmiştir. Tezlerin çoğunda örneklemi hemşireler oluşturmuştur. Tezlerin hepsinde nicel yaklaşımlar bulunup ve en çok kullanılan veri toplama aracı ölçekler olmuştur (%56,25). En çok tartışılan konular “liderlik davranışları” (%29,17), “liderlik tarzları” (%20,83) ve “dönüşümcü liderlik” (%14,58) olmuştur. Sonuç: Liderliğin farklı hemşirelik departmanlarında ve daha çok doktora tezlerinde ağırlıklı olarak nitel yaklaşımlar aracılığıyla geniş çapta tartışılması önerilmektedir. Geleceğin hemşirelerinin liderlik konusundaki görüş ve bilgilerinin araştırılması önemli olduğundan, hemşirelik öğrencileri daha çok araştırma örneklemi olarak ele alınmalıdır

    Characterizing the amniotic fluid-derived stem cells and optimizing the passage number for targeted applications

    Mesenchymal stem cells are considered potent sources that can be used for tissue regeneration and treatment of various diseases and syndromes. Among the mesenchymal stem cells, amniotic fluid-derived stem cells come forward as they possess some pluripotent properties and there are no serious ethical concerns upon their derivation. Therefore, gaining a better understanding of the nature of amniotic fluid stem cells is important. Although it is known that stem cells show slightly different characteristics between passages, scientists often consider only cell numbers and the proliferation pattern of the cells when deciding the passage to use in their studies. In this study, it was aimed to characterize rat amniotic fluid-derived stem cells for their mesenchymal and pluripotent features and make inter-passage comparisons by real-time qPCR to reveal their distinctions between different passages, and eventually help decide the appropriate passage numbers to be used in future research. The outcomes of the study showed that using rat amniotic fluid-derived stem cells at P5 could be beneficial for mesodermal differentiation studies and using them in earlier passages may be more favorable for the studies requiring better ectodermal differentiation properties

    Diagnostic criteria of computed tomography and cone-beam computed tomography in temporomandibular joint imaging

    Temporomandibular eklem (TME) ve çevre yapılardaki anomalilerin teşhisinde özellikle kemik yapıların incelenmesinde yüksek uzaysal çözünürlüğe sahip olan bilgisayarlı tomografi (BT) ve konik ışınlı bilgisayarlı tomografi (KIBT)'nin karşılaştırmalı çalışmalarda en iyi görüntüleme yöntemleri oldukları gösterilmiştir. Son yıllarda BT'ye göre daha düşük radyasyon dozu, daha kısa tarama zamanı, düşük maliyet, kolay kullanım, görüntülerin multiplanar kesitlerde detaylı analizine ve rekonstrüksiyonuna imkan vermesi nedeniyle KIBT, TME kemik yapılarının incelenmesinde daha yaygın olarak kullanılmaktadır. Bu yazıda TME görüntülemede BT ve KIBT'ın tanı kriterleri ile ilgili genel bilgiler sunulmaktadır.Comparative studies have shown that computed tomography (CT) and cone beam computed tomography (CBCT), which have high spatial resolution, are the best imaging modalities in the diagnosis of temporomandibular joint (TMJ) and surrounding anomalies, especially in the examination of bone structures. In recent years, CBCT has been used more widely in the examination of TMJ bone structures due to its lower radiation dose, shorter scanning time, low cost, ease of use, and detailed analysis and rearrangement of images in multiplanar sections compared to CT. In this article, up-to-date information on the diagnostic criteria of CT and CBCT in TMJ imaging is presented

    Development of an HPLC-UV method for the simultaneous determination of allantoin and D-panthenol in cosmetic products containing Aloe vera extracts

    A simple, fast and selective analytical method has been developed for the simultaneous determination of allantoin and D-panthenol in cosmetic products containing Aloe vera extracts. The proposed method depends on reversed-phase liquid chromatography with isocratic flow profile of the mobile phase composed of acetonitrile–10 mM phosphoric acid (pH 2.5) (85:15, v/v), with a C18 column at 30 8C. The analytes were detected with UV–vis. detector at 210 nm. The injection volume was 20 μL. The linearity ranges were found to be 0.2–20 and 0.1–10 μg mL 1 for allantoin and D-panthenol, respec tively. LOD values were found to be 0.07 μg mL 1 and 0.03 μg mL 1 , LOQ values were found to be 0.2 and 0.1 μg mL 1 for allantoin and D-panthenol, respectively. No interference was observed from concomitants. The developed method was applied to the analysis of 10 different type cosmetic products. It is foreseen that the method will be able to be used in order to carry out routine analysis, quality control and standardization in cosmetic products containing allantoin and D-panthenol

    Sharps injuries among nurses: Challenges and implications in practice

    Nurses are essential healthcare workers for the delivery of quality care. Therefore, these professionals must feel secure in the workplace. However, nursing is among the professions that suffer the most from workplace accidents. The most common occurrences involve sharps injuries. The objectives of this review were to explore and discuss aspects related to sharps injuries among nurses based on information available in the literature. This is an integrative review. The search for original articles of primary research was conducted in the first week of January 2024, in the Web of Science (WoS) Core Collection database. The review was framed around the following question: “What is the literature’s perspective on sharps injuries among nurses?” A total of 12 articles were found, of which seven were included. The selected articles were evaluated using the critical appraisal tool for cross-sectional analytical studies from the Joanna Briggs Institute. It was identified that the occurrence of sharps injuries among nurses is correlated with both organizational characteristics and specific individual factors of the nurses, such as scarcity of material and personnel resources, lack of knowledge and training, stress, and emotional exhaustion. The most common accidents involve needles, ampoules, and catheters. Effective measures to prevent sharps injuries among nurses should be taken. The implementation of efficient notification systems where nurses can report accidents is a measure that should be developed and properly monitored in healthcare institutions. It is understood, therefore, that nurse managers and healthcare administrators play fundamental roles in the development of better working conditions, with an emphasis on creating safe environments and implementing continuing education programs that enable nurses to learn and update information on prevention measures and safe use of materials and equipment
