Genetic evaluation for milk and semen variables on Egyptian buffaloes
2584 lactation records of Egyptian buffaloes calving between1992 to 2010 kept at Natafe El Gadded farm were studied. Several variables Animal model (SVAM) was used. Milk variables are milk production (MP), days in milk (DM), and calving interval (CI) and semen quality are ejaculate volume (EV) life of sperm (LS) and total Motility (TM). The overall means of MP, LL, CI, EV, LS, and TM were 2025 kg, 203 d, 447 d, 3.25 ml, 65.91% and 58.40%, respectively. Heritabilities were 0.23, 0.13, 0.02, 0.08, 0.27 and 0.24, for milk production, lactation length, current calving interval, ejaculate volume, live of sperm and total motility, respectively. Genetic association among MP and each of DM and CI were positive and were 0.80, 0.04, respectively, while among DM and CI was negative (-0.54). Genetic association among EV and each of LS and TM were .04 and -0.03, respectively, while among LS and TM was (1.0). In conclusion, our findings provide useful information on the heritability of milk production, life of sperm and mass motility traits in Egyptian buffaloes and relationships among them and should assist in selection for improvement of milk yield and semen traits in Egyptian buffaloes and bulls
Peptidoglycan recognition protein-1 gene polymorphism and its association with mastitis in Murrah buffalo
A wide variety of environmental or contagious microorganisms implicated in mastitis, impede the economic growth of dairy sector. Identification of polymorphism in candidate gene of host’s immune system and to rule out mastitis resistant allelic form of candidate gene usually remains prime focal point of research. Bovine peptidoglycan recognition protein-1 (PGLYRP-1), exclusively present in the granules of polymorphonuclear leukocytes has direct microbicidal properties. The present study was carried out to find the association between PGLYRP-1 polymorphic alleles with mastitis. Milk samples for somatic cell count and blood samples for PCR-RFLP analysis of PGLYRP-1 gene were collected from 20 mastitis negative and 20 mastitis positive Murrah buffaloes. There was significant difference in somatic cell count of mastitis and mastitis free animals. All amplified PCR products of ~862 bp size of partial region of PGLYRP-1 gene were subjected to each restriction enzyme (HincII or TaqαΙ or ApaI). Polymorphism in the partial region of PGLYRP-1 gene had not been established using PCR-RFLP as uniformity in pattern of digested fragments was seen. Target sequence PGLYRP-1 gene of Murrah buffalo was cloned and sequenced. BLAST analysis revealed sequence identity of PGLYRP-1 of Murrah buffalo with Bos taurus (JN085441.1) sequence at NCBI was 96%, 96% with Bos indicus (JN085440.1) and 96% with Bos indicus X Bos taurus (EU746454.1). In phylogenetic tree, the target sequence of PGLYRP-1 gene of Bubalus bubalis are found more closely related to Bos taurus than to Bos indicus
Performance of lactating buffaloes on soybean hulls based concentrate mixture
Eighteen lactating buffaloes were fed for 90 days to assess the effect of utilization of soybean hulls replacing maize grains in the concentrate mixture of lactating buffaloes. The buffaloes were divided into three groups of six buffalo each and fed as (CG) with maize based concentrate mixture, (25SH) concentrate mixture with 25% replacement of maize with soybean hulls and (50SH) fed on concentrate mixture with 50% replacement of maize with soybean hulls. The DMI and DMI percent BW showed nonsignificant differences. The NDF digestibility was significantly (P<0.05) better in soya hulls groups, all other nutrient digestibility did not vary significantly. The milk production and composition did not alter on soya hulls feeding, except milk protein. The rumen liquor profile did not alter due to supplementation of soybean hulls. The milk economics was better in 50SH group. It was concluded that soybean hulls can replace corn up to 50% as an energy source in the concentrate mixture of lactating buffaloes for economical milk production
Evaluation of homoeopathic remedy for haematogalactia in lactating dairy buffaloes
The objective of the current study was to determine the comparative efficacies of homeopathic and allopathic therapeutic regimens to cure heamatogalactia in buffaloes. The study comprised 40 sub-clinically mastitis lactating buffaloes manifesting blood in the milk from one or two teats which were divided into two equal groups i.e. B1 and B2. Milk from the affected quarters identified subclinical mastitis of 1+ grade detected by Surf Field Mastitis Test (SFMT) and their cultural examination determined staphylococcal and Streptocococcal bacterial infection. Hamamalis-Q and Echinatia-Q were administered orally 10 drops twice /day one hr. apart for 7 days to the animals of Group B1 while allopathic drugs, i.e., Milfone-C 300 ml I/V, Inj. Adrenaline 1% sol. 5 ml I/V, formaline 5 ml orally and Inj. Penbiotic 5 gm were administered I/M to B2 Group animals daily for 7 days. Milk from the Group B1 animals became normal in 3 to 5 days. On the 8th day, milk culture indicated the absence of microbes and SFMT became negative validating the wholesomeness of milk while it took 7 days in Group B2 to recover only 11 animals with 9 animals remaining obstinate. Albeit all animals in both groups became negative on culture and SFMT basis on the 8th day, the problem persisted in a few animals of the Group B2. It was concluded that homeopathically treated animals recovered 100% in 7 days while allopathic treated animals recovered 55% only till the 8th day, showing the superior efficacy of homeopathic therapeutic regimen as compared to allopathic therapy to cure haematogalactia in dairy buffaloes
Molecular characterization of β-lactoglobulin (βLG) gene in Indian buffalo (Bubalus bubalis)
The aim of the study was to characterized the β-lactoglobulin gene and protein in Indian buffalo. The β-lactoglobulin (Bu_βLG) is a major whey milk protein and cause an allergic reaction in infants and children. In the present study, the full open reading frame (ORF) of buffalo β-lactoglobulin (Bu_βLG) gene was characterized, which consisted of a 543 bp sequence with 180 amino acid residues. The phylogenetic tree showed that the cattle, yak, and buffalo βLG gene formed one cluster and buffalo is more closer (96%) to an exotic cow (Bos taurus). ProtParam analysis showed that Bu_βLG protein was acidic (pI, 4.93), thermo-tolerant, and hydrophobic. The Bu_βLG 3-D model was generated by I-TASSER, which revealed a more stabilized nature of the predicted structure. Further, the Ramachandran plot validated that the 3-D model of Bu_βLG protein, which was of good quality. The presence of four ligand-binding sites for retinoic acid, oleic acid, vitamin D3, and benzophenone in Bu_βLG suggests that this protein bind to several fatty acids and ions. IEDB analysis displayed the seven and six epitope sites in the Bu_βLG protein for B-cell and T-cell, respectively. Thus, the epitope sites in the βLG protein may have certain immunological roles, which can be used to reduce the allergenicity of βLG protein for improving the buffalo milk quality
Clinical evaluation of onlay double layer polyester mesh for repairing large umbilical hernias in buffalo calves
Umbilical hernias are a common issue in buffalo calves and can vary in their causes and treatment. While herniorrhaphy is a viable option for smaller hernias, larger hernias with diameters greater than three finger widths typically require the use of prosthetic materials for a successful and tension-free repair. This study aimed to evaluate the effectiveness of using a double layer polyester mesh to repair umbilical hernias in ten buffalo calves with hernial rings sizes ranging from ⩽11 cm in width. The study found that the use of a double layer of polyester mesh was successful in repairing hernias without any complications. Ultrasonographic examination was performed daily to assess the healing progress of the hernias. The use of a mesh allowed for tension-free repair and reduced the risk of recurrence. This technique may be a useful alternative for repairing larger umbilical hernias in buffalo calves. Overall, this study highlights the importance of using appropriate techniques for repairing umbilical hernias in buffalo calves, particularly when dealing with larger hernias. The use of prosthetic materials, such as a double-layer polyester mesh, can provide a successful and tension-free repair, reducing the risk of complications and recurrence
Detection of anti-3AB3 non-structural protein antibodies in foot-and-mouth disease virus vaccinated buffaloes at a semi-organized farm of Madhya Pradesh in India
Foot-and-mouth disease (FMD), one of the most important viral diseases of cloven-hoofed animals in India, is caused by FMD virus (FMDV) which belongs to Aphthovirus in the family Picornaviridae. There are three serotypes of FMDV (O, A and Asia 1) circulating amongst the livestock population of India. FMD is characterized by the formation of vesicles especially over the tongue and in between the interdigital space. FMD-affected animals with tongue lesions are reluctant to feed and subsequently yield less milk as well as affected animals never regain their production status causing huge economic losses to the animal owners. In the present study, random whole blood samples from FMD-vaccinated 38 adult buffaloes were collected in sterile containers of a semi-organized buffalo farm of Madhya Pradesh in India. Purified FMD vaccines only elicit antibodies (that are protective) against structural proteins of FMDV while natural FMDV infection invokes antibodies against both structural and non-structural proteins. Serum samples were employed in recombinant 3AB3 non-structural protein-based enzyme-linked immunosorbent assay (3AB3 NSP-ELISA kit provided by ICAR-Directorate of FMD, Mukteshwar) for differentiation of FMD-infected and vaccinated animals (DIVA). A total of 10.53% (4/38) serum samples tested positive in DIVA. Largely the vaccinated animals remained protected as no clinical signs of the disease were observed reiterating the importance of regular FMDV vaccination in animals at semi-organized dairy farms
The effects of solute carrier family 27 member 1 (SLC27A1) genotypes on fat content and major fatty acids in colostrum and milk from Murrah and “Murrah × Carabao” crossbred buffaloes
This study analyzed the effect of Solute carrier family 27 member 1 (SLC27A1) genotypes on fat content and major fatty acids (lauric acid C12:0, myristic acid C14:0, palmitic acid C16:0, stearic acid C18:0, and oleic acid C18:1n-9) in milk and colostrum of 46 Murrah and “Murrah × Carabao” crossbred buffaloes at the Philippine Carabao Center - University of the Philippines Los Baños dairy herd. The SLC27A1 genotypes (CC and CT) were determined by the polymerase chain reaction-restricted fragment length polymorphism (PCR-RFLP) method using DNA extracted from hair follicles. This study found polymorphism in the SLC27A1 gene that were consistent with Hardy Weinberg’s law of equilibrium (HWE), with a polymorphic information content (PIC) and heterozygosity (H) estimate of 0.2915 and 0.3542, respectively, for Murrah; and 0.3219 and 0.4032, respectively for “Murrah × Carabao” crossbreeds. In Murrah buffaloes, the CT genotype was significantly associated (P<0.05) with higher colostrum yield and milk fat content, compared to CC. In “Murrah × Carabao” crossbreds, CT was associated with higher C12:0, C14:0, and C16:0, but lower C18:1n-9 in colostrum; and lower C16:0 in milk compared to CC. This study showed polymorphisms in SLC27A1 genotypes and their significant effects on colostrum yield and milk fat content in Murrah buffaloes and some major fatty acids in colostrum and milk from “Murrah × Carabao” crosses
Impact of OTAs’ Digital Services on Customer Satisfaction in Thailand
The availability of digital technology has prompted many adaptations in the hotel industry, particularly among online travel agents (OTAs). Better services provided by OTAs might improve customer satisfaction. This research aims to examines the effect of OTAs’ Digital Services (DS), electronic service quality (ESQ), and electronic recovery service quality (ERSQ) on consumer satisfaction (CSAT). To better understand customer satisfaction via online booking, research survey of 415 persons in Thailand who have used OTAs to book hotels was conducted. For data analysis, structural equation modeling (SEM) was used. The study covers the metrics and conditions that could lead to consumer satisfaction. According to the study's findings, ESQ and ERSQ are the factors that would directly affect consumer satisfaction while DS can rather effect consumer satisfaction indirectly. Based on the findings, OTAs service providers should enforce quality and simplicity on their platforms, such as swift booking transactions, precise booking detail, and booking experience when searching for rooms. Customer satisfaction would progressively spread information, lowering marketing costs while increasing reliability
