    This dataset includes all the data integrated in the publication in Lemos et al., 2024, BBA-MCR and contains all data necessary to replicate the published figures. The project investigates the nature and the properties of the metabolic enzyme PKM2 in the regulation of IP3R-mediated Ca2+ signaling. Using both chemical and genetic methods, we provide evidence that the catalytical active, tetrameric form of PKM2 preferentially suppresses IP3R-mediated Ca2+ release. Moreover, our data strongly suggests that this inhibitory effect necessitates an additional cellular factor, which is most likely GRP75.     This data depository contains all the data used in the following publication: Characterization of a droplet microfluidic platform for the enrichment of live and functional clones using a genotype-based read-out The following data is provided: - dPCR folder: contains .tiff images of the dPCR experiments to characterize the effect of the copy number on the sensitivity (Table 1 in the paper). The file name corresponds to the sample. Abbreviations: HC = High-copy number, LC = low-copy number, SC = single-copy number, EP = End-point, SP = starting point. The number of positive droplets can be derived from the FAM image, while the total number of droplets can be obtained from the VIC image. For the HC sample, we only have starting and end-point FAM images. - FACS folder: contains all FACS data. The 20221116_Flowjo_Analyzed_data pdf file contains the analyzed data in FlowJo which was used to determine the fraction of positive droplets. The raw FACS data files (.fsc) have been made available in the FASC_data_files folder. The FACS_sort_parameters folder contains a summary of the sort per sample and per run and was used to obtain how many target droplets were sorted per sample. - Imagestream folder. Contains the raw Imagestream data of the sample before and after enrichment. (Presort vs Postsort). Imagestream data can be opened and analyzed using FSC Express. - Data_tables: Excel file containing the calculations and raw data for generating the tables in the paper. - Data_supplementary_figures: Python file (Jupiter Notebook). Contains the data and code for generating the supplementary figures

    Musculoskeletal disorders present significant health and economic challenges on a global scale. Current intraoperative imaging techniques, including computed tomography (CT) and radiography, involve high radiation exposure and limited soft tissue visualization. Ultrasound (US) offers a non-invasive, real-time alternative but is highly observer-dependent and underutilized intraoperatively. US enhanced by artificial intelligence shows high potential for observer-independent pattern recognition and robot-assisted applications in orthopedics. Given the limited availability of in-vivo imaging data, we introduce a comprehensive dataset from a comparative collection of handheld US (HUS) and robot-assisted ultrasound (RUS) lumbar spine imaging in 63 healthy volunteers. This dataset includes demographic data, paired CT, HUS, RUS imaging, synchronized tracking data for HUS and RUS, and 3D-CT-segmentations. It establishes a robust baseline for machine learning algorithms by focusing on healthy individuals, circumventing the limitations of simulations and pathological anatomy. To our knowledge, this extensive collection is the first healthy anatomy dataset for the lumbar spine that includes paired CT, HUS, and RUS imaging, supporting advancements in computer- and robotic-assisted diagnostic and intraoperative techniques for musculoskeletal disorders

Their combination is analysed for (i) rotor position, (ii) rotor acceleration, (iii) unbalanced magnetic pull, (iv) bearing acceleration, and (v) stator currents in the presence of the following mechanical faults: (i) no-fault case, (ii) a stator outer race misalignment, (iii) a rotor mass unbalance, (iv) a bearing outer race spall fault, (v) a bearing inner race spall fault, (vi) a bearing ball spall fault. In this study an accelerated storage experiment (60°C, 70% relative humidity) was performed with samples consisting of native or toasted wheat bran mixed with vitamin A-enriched oil.
