Effects of Probiotic Bacteria on Central Neuronal Activation in Experimental Colitis
Background: Brain–gut axis dysregulation is observed in inflammatory bowel disease. However, the effect of altered gut flora on neuroimmunomodulation and its role in the pathogenesis of inflammatory bowel disease are unknown. The aims of this study are to determine (i) whether colitis modifies the expression of c-fos, a marker of general neuronal activation in the brain and (ii) whether this activation could be modulated by probiotic bacteria.
Methods: In this study, 28 Sprague–Dawley rats were divided into 4 groups: colitis-probiotic group, non-colitis-fed-control group receiving probiotic Lactobacillus delbrueckii subsp. Bulgaricus B3 strain for 7 days, colitis group, and sham group receiving only sodium chloride. Colitis was induced by intracolonic administration of trinitrobenzene sulfonic acid–ethanol. The expression of c-fos was detected by immunohistochemistry in the brain tissue. Cytokines and inflammatory mediators were analyzed in the plasma. Histological scores and oxidative status were analyzed in the colon samples.
Results: The inflammatory response was accompanied by increased levels of cytokines, lipid peroxidation activities, c-fos expression in the medial nucleus of the amygdala, and decreased levels of antioxidant enzymes in the colitis (P < .001). Probiotic treatment reversed those effects. Also, histopathologic scores were significantly lower in the probiotic-treated groups compared to the colitis group (P = .035). In contrast, the expression of c-fos was significantly increased in the paraventricular nucleus of hypothalamus in the probiotic-treated rats (P < .001).
Conclusion: Colitis and intestinal inflammation are associated with the activation of neurons in the limbic system creating stress-like effects in the brain. Probiotics diversely modulate limbic response and hypothalamic axis activity in addition to protective effects in inflammation
The correlation between nursing students' healthy lifestyle behaviors, cardiovascular disease risk factors' knowledge level, and obsession symptoms
BACKGROUND: Nursing students have a role in the awareness of risk factors in the development
of healthy lifestyle behaviors. Nursing students’ awareness of disease risk factors plays a role in
developing healthy lifestyle behaviors. Therefore, it was aimed to determine the relationship between
the effect of nursing students’ healthy lifestyle behaviors and cardiovascular disease risk factors’
knowledge levels and obsessive–compulsive symptoms.
MATERIALS AND METHODS: This was a descriptive cross‑sectional study. Two hundred and
twenty‑four students studying in the nursing departments of two foundation universities participated
in the study between April and October 2020. Sociodemographic information form, Health Promoting
Lifestyle Profile II (HPLP‑II), Cardiovascular Disease Risk Factors Knowledge Level (CARRF‑KL),
and Maudsley Obsessive–Compulsive Inventory (MOCI) were applied to the students.
RESULTS: It was found that the nursing students’ healthy lifestyle behaviors (123.53 ± 25.78) and
cardiovascular risk factors’ knowledge level (21.08 ±± 2.70) were high. Obsessive–compulsive
symptoms (16.12 ± 6.22) were moderate. In the correlation of sociodemographic characteristics, it
was found that age correlated with the nutrition subdimension of CARRF‑KL and HPLP‑II, income
level correlated with HPLP‑II, place of residence correlated with HPLP‑II, CARRF‑KL, and MOCI, and
having a health problem correlated with health responsibility. Income status, place of residence, and
presence of health problems were found to be correlated with HPLP‑II in all subdimensions in the
regression analysis. In the regression analysis of HPLP‑II with CARRF‑KL and MOCI scales, it was
found that it was significantly related to interpersonal relations, spiritual growth, stress management,
and total score dimensions.
CONCLUSION: Healthy lifestyle behaviors of nursing students are related to cardiovascular risk
factors’ knowledge level and obsession symptoms. In addition, some demographic characteristics
affect healthy lifestyle behaviors
S100 Protein Family in Rheumatological Diseases
Inflammation in joint tissues is one of the causes of early mortality in rheumatological diseases. In fact, inflammation process is followed by cell infiltration, synovial tissue proliferation, cartilage and bone destruction. Therefore, various studies have been done to understand the pathogenesis and biochemical mechanisms of inflammation. Specifically, relationship between S100 protein familyand inflammation in some type of rheumatologic diseases constitute the basis of this review. Scientific evidences emphasis a crucial role for S100 proteins in acute and chronic inflammatory diseases and also have been proposed as useful biomarkers of certain types of rheumatological diseases.Eklem dokularındaki inflamasyon, romatolojik hastalıklarda erken ölüm nedenlerinden biridir.Aslında inflamasyon sürecini hücre infiltrasyonu, sinovyal doku proliferasyonu, kıkırdak ve kemik yıkımı takip eder. Bu nedenle inflamasyonun patogenezini ve biyokimyasal mekanizmalarını anlamak için çeşitli çalışmalar yapılmıştır. Spesifik olarak, bazı romatolojik hastalıklarda S100 protein ailesi ile inflamasyon arasındaki ilişki bu derlemenin temelini oluşturmaktadır. Bilimsel kanıtlar, akut ve kronik inflamatuar hastalıklarda S100 proteinlerinin çok önemli bir rolü olduğunu göstermektedir ve ayrıca bu protein ailesinin üyeleri belirli romatolojik hastalıklarda yararlı biyobelirteçleri olarak önerilmiştirler
Does YouTube™ provide reliable information on oral candidiasis?
Objective: To evaluate the quality, source, usefulness and/or reliability, visibility, and popularity of Youlube (TM) videos on oral candidiasis.
Materials and Methods: A YouTube (TM) search for "oral candidiasis" was performed, setting the English language. 133 of the 351 videos watched were included in the study. Two periodontologists scored the videos for visibility, popularity, quality, utility, and reliability. Videos' quality was evaluated using the DISCERN and the Global Quality Scale (GQS) tools, by categorizing them through quality assessment, source, duration, views, likes, and dislikes of each video were noted.
Results: 82.7% of the videos were moderately or very useful, and the GQS scores were mostly good (41.4%-Score 4) and excellent (42.1%-Score 5). Significant relationships were obtained between Source of Upload, Video Type, Total Discern, and GQS variables and Usefulness scores (p <0.05). The 53.4% of the video sources were Healthcare professionals and their video usefulness scores were mostly "moderately useful" (47.9%) and "very useful" (%42.3). There was a significant positive correlation between the usefulness scores of the videos and the number of likes (p = 0.004), comments (p = 0.019), and the viewing rate (p = 0.006).
Conclusions: The videos that rank high in searches for oral candidiasis on YouTube (TM) are mostly useful and comprehensive videos uploaded for educational purposes
Expression of salivary LINC01206, LINC01209, LINC01994, and ABCC5-AS1 may serve as diagnostic tools in laryngeal cancer
Çalışmanın amacı: lncRNA'ların, memelilerde bakteriyel enfeksiyona karşı bağışıklık tepkisinin önemli bir epigenetik düzenleyicisi olarak görev yaptığı görülmektedir. Ancak enfeksiyon sırasında yalnızca patojenik veya faydalı potansiyel sergileyen bir lncRNA henüz tanımlanmamıştır. Üstelik, ekspresyonu belirli bir patojene yanıt olarak değişen spesifik lncRNA'ların olup olmadığı veya lncRNA'ların esas olarak farklı stres uyaranlarına karşı temel hücresel bağışıklık yanıtlarında yer alıp almadığı hala tam olarak bilinmemektedir. Bu çalışma, larenks kanseri hastalarında tükürük lncRNA'ları ile tükürük bakteriyel patojenleri arasındaki ilişkiyi değerlendirmeyi amaçlamaktadır. Yöntemler: LINC01206, LINC01209, LINC01994 ve ABCC5-AS1, laringeal karsinomalı 35 hastanın ve 25 sağlıklı kontrolün tükürük numunelerindeki 13 aday lncRNA arasında analiz edildi. Hem ifadeleri hem de oral bakteri üyelerinin kantitatif miktarı (Rothia mucilaginosa, Streptococcus spp., Prevotella oris, Veillonella dispar, Neisseria subflava ve Peptostreptococcus stomatis) qPCR kullanılarak analiz edildi. Bu lncRNA'ların ve bakteriyel patojenlerin tanısal biyobelirteçler olarak yararlı olup olmadığını belirlemek için bunların klinikopatolojik ve demografik özelliklerle ilişkileri analiz edildi. Bulgular: Çalışma grubu kontrol grubu ile karşılaştırıldığında LINC01206, LINC01209, LINC01994 ve ABCC5-AS1 ekspresyonu sırasıyla 2,84 kat, 2,33 kat, 4,46 kat ve 2,27 kat daha düşüktü (p 0,05). Sonuç: Bu sonuçlar larenks kanserinin altında yatan moleküler mekanizmaya yeni bakış açıları sağlayabilir ve lncRNA/mikrobiyom uygulamaları gelecekte larenks kanseri gelişimini önlemek için yeni ve alternatif bir yöntem oluşturabilir.Purpose of the study: lncRNAs appear to act as an important epigenetic regulator of the immune response to bacterial infection in mammals. However, a lncRNA that only exhibits pathogenic or beneficial potential during infection has not yet been described. Moreover, it is still not fully known whether there are specific lncRNAs whose expression changes in response to a particular pathogen or whether lncRNAs are mainly involved in basic cellular immune responses to different stress stimuli. This study aims to assess association between salivary lncRNAs and salivary bacterial pathogens in laryngeal cancer patients. Methods: LINC01206, LINC01209, LINC01994, and ABCC5-AS1 were analyzed among 13 candidate lncRNAs in the saliva samples of 35 patients with laryngeal carcinoma and 25 healthy control. Both their expressions and the quantitative amount of oral bacteria members (Rothia mucilaginosa, Streptococcus spp., Prevotella oris, Veillonella dispar, Neisseria subflava, and Peptostreptococcus stomatis) were analyzed using qPCR. To determine whether these lncRNAs and bacterial pathogens are useful as diagnostic biomarkers, their association with clinicopathological and demographic characteristics was analyzed. Results: When the study group compared with the control group, expression of LINC01206, LINC01209, LINC01994, and ABCC5-AS1 were 2.84-fold, 2.33-fold, 4.46-fold, and 2.27-fold lower, respectively (p 0.05). Conclusion: These results may provide novel insights into the molecular mechanism underlying laryngeal cancer and lncRNA/microbiome applications may constitute a new and alternative method to prevent development of laryngeal cancer in the future
Antitumor activity of irinotecan with ellagic acid in C6 glioma cells
OBJECTIVE: Irinotecan-based combination chemotherapies in malignant gliomas need to be examined. The aim of this study was to investigate the
synergetic effect of ellagic acid, a natural polyphenolic antioxidant compound, with irinotecan, an inhibitor of topoisomerase I enzyme, on the growth,
cadherin switch, and angiogenic processes of a glioma cell line.
METHODS: A combination of 100 μM ellagic acid and 100 μM irinotecan was applied to rat C6 glioma cells for 24th, 48th, and 72nd h. The cell
proliferation was evaluated by 5-bromo-2ʹ-deoxyuridine immunocytochemistry. The expression levels of vascular endothelial growth factor, E-cadherin,
and N-cadherin were measured using real-time polymerase chain reaction and their immunoreactivities using immunocytochemistry.
RESULTS: The treatment of irinotecan with combining ellagic acid enhanced antitumor activity and the synergistic effect of these reduced the cell
proliferation of C6 glioma by inhibiting the cadherin switch and promoting the antiangiogenic processes.
CONCLUSIONS: Further research is required to prove a negative relationship between C6 glial cell proliferation and irinotecan with ellagic acid
application. Our preliminary data suggest that even with the extremely short-term application, irinotecan with ellagic acid may affect glioma cells at
the level of gene and protein expressio

Nowadays, reducing energy consumption is an important target for energy-intensive manufacturing systems due to many reasons such as global warming, legal obligations and lowering company expenses. Therefore, this paper focuses on energy-based scheduling problem in manufacturing systems. A mixed-integer nonlinear programming (MINLP) model is developed for a single machine scheduling problem with the sequence-dependent setup times and different arrival times in order to minimize the total energy consumption and the total tardiness. An energy-based genetic optimization (EGOP) method is proposed by adopting the genetic algorithm (GA) approach, which is a heuristic method to solve the problem. The objective values and the computation times are compared with the analytical solution and the General Algebraic Modeling System (GAMS) solution so as to evaluate the performance of the proposed method. As a result, it is seen that the proposed EGOP method provides effective results
Relationship between health science students' gender-related attitudes and perception of honor
Amaç: Bu çalışmanın amacı, çeşitli sağlık bilimleri fakültelerinde öğrenim gören öğrencilerin (n=427) cinsiyet ve namus algılarını ve bu algılar arasındaki ilişkiyi belirlemektir. Tasarım ve Yöntemler: Veri toplama aracı olarak tanıtıcı bilgi formu, Cinsiyet Rolleri Tutum Ölçeği (GRAS) ve Kadınlara İlişkin “Namus” Anlayışına İlişkin Tutum Ölçeği (ASWRCH) kullanılmıştır. Bulgular: Öğrencilerin GRAS ve ASWRCH ortalama puanlarının sırasıyla 159,7±23,1 ve 104,1±15,5 olması öğrencilerin eşitlikçi bir tutuma ve onur algısına sahip olduklarını göstermektedir. Sonuç: Sağlıklı/hasta bireylere hizmet vermek üzere eğitim gören sağlık bilimleri fakültesi öğrencilerinin cinsiyet ve namus algısı açısından güçlü eşitlikçi tutumlara sahip oldukları belirlendi. Uygulamaya Etkileri: Sağlık bilimleri öğrencilerinin cinsiyet ve namus algısı açısından eşitlikçi bir yaklaşımla yetiştirilmesi, onların kişisel ve mesleki yaşamlarında eşitlikçi bir tutum benimsemelerini, toplumsal değişimi başlatma ve sürdürmede değişimin aktörleri olmalarını sağlayacaktır.Purposes: This study aimed to determine the perceptions of gender and honor of students (n = 427) in various health science faculties and the relationship between those perceptions. Design and Methods: An introductory information form, the Gender Roles Attitude Scale (GRAS), and the Attitude Scale for Women-Related Conception of “Honor” (ASWRCH) were used as data collection tools. Findings: The students' average scores on GRAS and ASWRCH were 159.7 ± 23.1 and 104.1 ± 15.5, respectively, demonstrating that students had an egalitarian attitude and perception of honor. Conclusions: Students from health science faculties, trained to provide service for healthy/sick individuals, were found to have strong egalitarian attitudes in terms of gender and perception of honor. Practice Implications: Raising health sciences students, with an egalitarian approach in terms of gender and honor perception will enable them to adopt an egalitarian attitude in their personal and professional lives and to become agents of change in initiating and sustaining social change
Therapeutic effect of finasteride through its antiandrogenic and antioxidant role in a propionic acid-induced autism model: Demonstrated by behavioral tests, histological findings and MR spectroscopy
Background: Autism is a clinically defined neurodevelopmental disorder with unknown origin characterized by significant social, communication and behavioral challenges. Although it can be a lifelong condition, treatments can help alleviate symptoms and enhance a patient's quality of life. Purpose: We aimed to assess the therapeutic potential of finasteride in autism with biochemical markers, histopathological evaluation, behavioral tests and radiological imaging. Materials and methods: Propionic acid (PPA) was injected intraperitoneally into 20 out of 30 rats for 5 days to establish an autism model. Rats were randomly assigned into four groups: control group (no procedure was applied, n = 10), placebo group (intraperitoneal PPA + 1 ml/kg/day % 0.9 NaCl saline was given via oral gavage for 15 days, n = 10) and treated group (intraperitoneal PPA + 5 mg/kg/day of finasteride was given via oral gavage for 15 days, n = 10). After 4 days of behavioral tests, magnetic resonance spectroscopy (MRS) was performed for measuring creatine and lactate levels. All animals were sacrificed for histopathological examination and biochemical analysis of brain tissue. Results: MDA, NF kappa B, TNF-alpha, IL-2, IL-17A and lactate levels in brain homogenates were significantly increased in the placebo group compared to the control group, while Nfr2 levels were decreased; and the levels of all biochemical markers were reversed by finasteride treatment. A significant improvement was observed in autism like behaviors in rats treated with finasteride compared to the placebo group. Further, the creatine and lactate levels in corpus striatum in MRS, the neuronal counts and glial activity of the hippocampus and cerebellum were closer to the control group in the finasteride-treated group compared to the placebo group. Conclusion: Finasteride led significant improvement in autism-like symptoms with its antioxidant effect through Nrf2 modulation in addition to its anti androgen effect
