    Comparison of Selenic Acid and Pyruvic Acid-Loaded Silver Nanocarriers Impact on Colorectal Cancer Viability

    Colorectal cancer (CRC) is a leading cause of morbidity and death worldwide. As current cancer drugs are ineffective, new solutions are being sought in other fields, including nanoscience. Similarly, silver nanoparticles play an important role in the pharmaceutical industry as they act as anti-cancer agents with less harmful effects and are usually 1 to 100 nm in size. Selenic acid (SA) and pyruvic acid (PA) are involved in various metabolic pathways in cancer. For this reason, we decided to detect their influence on colorectal cancer using silver-based (Ag) nanocarriers. DLS, Zetasizer, SEM and UV-Vis analyses were used to characterize AgSA and AgPA. A UV spectrophotometer was used to analyze the release of the NPs. MTT analyses were used to measure the viability of HCT116 and HUVEC cells, and IC50 values were calculated using GraphPad Prism. The indicated dosage and particle size of AgSA NPs proved to be suitable for cytotoxicity. Moreover, injection of these nanoparticles into non-cancer cells proved safe due to their minimal toxicity. In contrast, the AgPA NPs have no cytotoxicity and induce proliferation of HCT116 cells. Finally, only the synthesised AgSA nanoparticles could be used for advanced cancer therapy, which is both inexpensive and has minimal side effects

    Endokrin hastalık tanısı almış çocukların tipik gelişim gösteremeyebildikleri; sosyalduygusal sorunlarla birlikte dil, akademik ve bilişsel becerilerde güçlük yaşayabildikleri bilinmektedir Bu çalışmada endokrinolojik hastalığı olan çocuklar, normal gelişim gösteren akranları ile dil, biliş, matematiksel beceri ve psikosoyal özellikler açısından karşılaştırılmıştır. Bu çalışmanın amacı okul başarısızlığı yaşayan endokrin hastalığı bulunan çocukların, dil, biliş ve matematiksel beceri alanlarına yönelik olası terapötik ihtiyaçlarının ortaya konabilmesidir. Bu yolla dil ve konuşma bozukluklarının erken dönemde değerlendirilmesi ve müdahalenin planlanması mümkün hale gelebilecektir. Çalışmaya 15 endokrin hastalığı tanısı almış çocuk ve 15 sağlıklı gelişim gösteren çocuk katılmıştır. Katılımcılara Türkçe Okul Çağı Dil Gelişimi Testi (TODİL), Türkçe Anlamsız Sözcük Tekrarı Testi (TAST), Türkçe Çok Dilliler Cümle Tekrarı Testi (LITMUS-TR), Wechsler Çocuklar için Zeka Ölçeği (WISC-R), Problem Çözme Testi (PÇT), Çocuklarda Anksiyete ve Depresyon Ölçeği-Yenilenmiş (ÇADÖ-Y), Coopersmith Benlik Saygısı Envanteri, 4-18 Yaş Çocuk ve Gençlerde Davranış Değerlendirme Ölçeği uygulanmıştır. Verilerin analizinde SPSS (25. versiyon) paket programı kullanılmıştır. Analizlerde anlamlılık değeri olarak p<,005 değeri dikkate alınmıştır. Araştırmadan elde edilen veriler endokrin hastalığı olan çocukların sağlıklı akranlarına göre dil, biliş ve matematiksel becerilerde daha düşük performans gösterdiklerini, psikolojik ölçekler ile değerlendirilen sosyal duygusal durum açısından ise farklılaşmadıklarını göstermektedir

    Kekemelik, erken çocukluk döneminde başlayan tekrarlar, uzatmalar ve bloklar şeklinde görülen akıcılık bozukluğudur. Yoğunlaştırılmış dikkat bir ya da birden fazla uyaran varken uyaranlardan yalnız birine odaklanabilmektir. Bu çalışmanın amacı yoğunlaştırılmış dikkat eğitimi ile kekeme bireylerin konuşmaya odaklanmalarını engelleyen etkenleri görmezden gelerek dikkat düzeylerini artırmaktır. Bu çalışmaya 9 yaş ile 9 yaş 11 ay arasında toplam 24 çocuk (12 kekeme çocuk, 12 akıcı konuşan çocuk) katıldı. Tüm katılımcıların çalışma öncesi ve sonrası d2 dikkat testleri ile Kekemelik Şiddetini Ölçme Aracı-4 (KEŞİDA-4) ile değerledirmeleri yapıldı. Kekemeliği olan 12 çocuğa yoğunlaştırılmış dikkat eğitimi verildi. Araştırmada, demografik değişenler bakımından öğrencilerin kekemelik şiddeti ve dikkat düzeyleri karşılaştırıldı. Kekeme bireylere uygulanan dikkat eğitimi öncesi ve sonrasında yapılan d2 dikkat testine (EK-4) göre psikomotor ve kavrama hızlarında artma, dikkatsizlik ve yönergeye uyma problemlemlerinde azalmalar görüldü ve sonuçlar istastistiksel olarak anlamlıydı (p<0.05). KEŞİDA-4 ile yapılan kekemelik değerlendirmesinde ise öntest ve sontest sonuçlarına göre çocukların kekemelik şiddetlerinde azalma tespit edildi (p<0.05). Kekemeliği etkileyen birçok faktör vardır ve dikkat becerisi bu faktörler arasında önemli bir yere sahiptir. Çalışmamızda elde edilen bulgulara göre kekeme bireylere uygulanan yoğunlaştırılmış dikkat eğitiminin kekemelik şiddetinde azalmalara ve psikomotor hızda artmalara neden oduğu görüldü.     A Longitudinal Study in Turkiye of Host Ability to Produce Antibodies following a Third Homologous BNT162b2 Vaccination

    Obesity is a multifaceted, complex condition that has negative impacts on one's health. There are conflicting reports regarding the COVID-19 vaccine's ability to induce antibody formation in obese people. Our study aimed to determine anti-S-RBD IgG and surrogate neutralizing antibody (snAb) levels before and after the third Pfizer-BioNTech (BNT162b2) vaccination (at 15, 60, 90, and 120 days) in normal-weight adults, overweight, and obese individuals without any comorbidity or previous SARS-CoV-2 infection history, but it did not evaluate the response to the first two doses. In this longitudinal prospective study in Istanbul, Turkey, a total of 323 consecutive adult individuals (141 normal weight, 108 overweight, and 74 patients with obesity) were included. Peripheral blood samples were collected. Anti-S-RBD IgG and surrogate neutralizing antibody levels were detected using the ELISA method. After the third dose of BNT162b2 vaccination, obese patients had significantly lower levels of snAb against SARS-CoV-2 compared with normal-weight controls, but the levels otherwise did not differ between the study groups. Across all individuals in our cohort, titers peaked about a month after this third vaccination and then gradually faded. Anti-S-RBD IgG and snAb IH% levels against SARS-CoV-2 were not correlated with IL-6 and TNF-α levels. In conclusion, anti-S-RBD IgG titers and snAb IH% levels against SARS-CoV-2 were determined longitudinally for 120 days after the third homologous BNT162b2 vaccination. Although there were no significant differences in anti-S-RBD IgG, we found significant differences in the snAb IH% levels against SARS-CoV-2 between obese and healthy control subjects

    Medical image diagnosis based on adaptive Hybrid Quantum CNN

    Hybrid quantum systems have shown promise in image classification by combining the strengths of both classical and quantum algorithms. These systems leverage the parallel processing power of quantum computers to perform complex computations while utilizing classical algorithms to handle the vast amounts of data involved in imaging. The hybrid approach is intended to improve accuracy and speed compared to traditional classical methods. Further research and development in this area can revolutionize the way medical images are classified and help improve patient diagnosis and treatment. The use of Conventional Neural Networks (CNN) for the classification and diagnosis of medical images using big datasets requires, in most cases, the use of special high-performance computing machines, which are very expensive and hard to access by most researchers. A new form of Machine Learning (ML), Quantum machine learning (QML), is being introduced as an emerging strategy to overcome this problem. A hybrid quantum-classical CNN uses both quantum and classical convolution layers designed to use a parameterized quantum circuit. This means that the computing model utilizes a quantum circuits approach to construct QML algorithms, which are then used to transform the quantum state to extract image hidden features. This computational acceleration is expected to achieve better algorithm performance than classical CNNs. This study intends to evaluate the performance of a Hybrid Quantum CNN (HQCNN) against a conventional CNN. This is followed by some optimizer modifications for both proposed and classical CNN methods to investigate the possible further improvement of their performance. The optimizer modification is based on forcing the optimizer to be directly adaptive to model accuracy. The optimizer adaptiveness is based on the development of an optimizer with a loss base adaptive momentum. Several algorithms are developed to achieve the above-mentioned goals, including CNN, QCNN, CNN with the adaptive optimizer, and QCNN with the Adaptive optimizer. The four algorithms are tested against a Kaggle brain dataset containing over 7000 samples. The test results show the hybrid quantum circuit algorithm outperformed the conventional CNN algorithm. The performance of both algorithms was further improved by using a fully adaptive SGD optimizer

    Araştırma ile lumbal disk hernili hastalara yönelik yüksek frekanslı diatermik akım tedavisi ve lumbal stabilizasyon egzersiz tedavisinin karşılaştırılması amaçlandı. Tüm katılımcıların demografik bilgileri sorgulandıktan sonra tedavi öncesinde ve tedavinin bitiminde aynı fizyoterapist tarafından fonksiyonel durumları Oswestry Bel Ağrısı Engellilik Anketi ile, ağrı seviyeleri Visual Analog Skalası (VAS) ile ve eklem hareket açıklıkları gonyometre ile değerlendirilmiştir. Çalışmamızda kapasitif ve rezistif diatermik akım tedavisinin ağrı, fonksiyonel durum ve eklem hareket açıklığı üzerine olan etkilerini araştırmak amacıyla 6 aydan uzun süreli bel ağrısı mevcut olan ancak nörolojik defisiti olmayan 30 hasta değerlendirildi. Hastalar randomize olarak 15' er kişilik 2 gruba ayrıldı. Deney grubuna lumbal stabilizasyon egzersizleri ve yüksek frekanslı diatermik akım, kontrol grubuna lumbal stabilizasyon egzersizleri uygulandı. Tüm katılımcılar tedavi öncesi ve tedavinin bitiminde değerlendirildi. Vizüel Analog Skala ile değerlendirilen ağrı parametresinde gruplar arasında istatistiksel olarak anlamlı bir fark bulunmadı (p>0,05). Oswestry Bel Ağrısı Engellilik Skalası'nın yürüme dışındaki alt parametrelerinin tümünde ve toplam skorlarının fark değerlerinde gruplar arasında istatistiksel olarak anlamlı bir fark bulunmadı (p>0,05). Eklem hareket açıklığı ölçümlerinin sonuçlarına göre deney grubunda kontrol grubuna kıyasla tedavi sonrası sadece gövde fleksiyonu eklem hareket ölçümündeki fark istatistiksel olarak anlamlı şekilde daha yüksekti (p0,05). Sonuç olarak; LDH'li hastalarda kapasitif ve resistif diatermik akım tedaviye dahil edildiğinde rehabilitasyon sürecine katkı sağlayan, kolay uygulanabilir ve etkili bir tedavi yöntemidir.The aim of the study was to compare high frequency diathermic current therapy and lumbar stabilization exercise therapy for patients with lumbar disc herniation. After questioning the demographic information of all participants, their functional status was assessed with the Oswestry Low Back Pain Disability Questionnaire, pain levels with the Visual Analog Scale (VAS) and range of motion with a goniometer by the same physiotherapist before and after the treatment. To investigate the effects of capacitive and resistive diathermic current therapy on pain, functional status and range of motion, 30 patients with low back pain for more than 6 months but without neurologic deficit were evaluated. The patients were randomly divided into 2 groups of 15 patients each. The experimental group received lumbar stabilization exercises and high frequency diathermic current, while the control group received lumbar stabilization exercises. All participants were evaluated before and at the end of the treatment. There was no statistically significant difference between the groups in the pain parameter evaluated with the Visual Analog Scale (p>0.05). There was no statistically significant difference between the groups in all sub-parameters of the Oswestry Low Back Pain Disability Scale except walking and in the difference values of the total scores (p>0.05). According to the results of range of motion measurements, only the difference in the trunk flexion joint motion measurement was statistically significantly higher in the experimental group compared to the control group after treatment (p0.05). In conclusion; When capacitive and resistive diathermic current is included in the treatment of patients with LDH, it is an easily applicable and effective treatment method that contributes to the rehabilitation process

    Assessment of Epstein-Barr virus, Candida albicans, and some periodontal pathogens in rheumatoid arthritis patients with periodontitis

    OBJECTIVE: To comparatively investigate the periodontal results and microbial load in subgingival biofilm samples (SBS) in rheumatoid arthritis subjects and healthy volunteers.METHODS: One hundred twenty subjects were classified into different cohorts: healthy (H-C); periodontitis with good systemic health (H-P); rheumatoid arthritis (RA) and good periodontal health (RA-C); and periodontitis with RA (RA-P). The periodontal parameters were recorded, and SBS were collected to determine periodontal pathogens including Epstein-Barr Virus (EBV) and Candida albicans using reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: Subjects that had greater disease course, determined by moderate or high disease activity scores 28 (DAS28), suffered from worse oral health conditions (higher plaque index, gingival index, bleeding on probing, probing depth, and excessive clinical attachment loss) than those with low DAS28 scores. A higher prevalence of Treponema denticola (T. denticola) was observed in the RA-P group. Cyclic citrullinated peptide was associated with the occurrence of T. denticola and Campylo-bacter rectus. DAS28 using C-reactive protein (DAS28-CRP) had a significant association with Capnocytophaga gingivalis and EBV. The duration of the RA disease was associated with the presence of T. denticola.CONCLUSION: Subgingival microbial difference could reliably discriminate RA from healthy individuals. Especially, T. denti-cola and EBV may play a key role in periodontitis associated with RA

    Acute bilateral extensor pollicis longus tendon rupture following bilateral displaced distal radius fracture: A case report

    Extensor pollicis longus tendon ruptures are rarely reported after traumatic events, especially after distal radius fractures. Its classical presentation is a late-onset and unilateral rupture after a non-displaced distal radius fracture. In this case report, we present the treatment of a patient with acute bilateral extensor pollicis longus tendon ruptures after bilateral displaced distal radius fractures. The patient was treated with bilateral open reduction and volar plate fixation via a volar Henry approach for distal radius fractures and bilateral exten-sor indicis proprius tendon transfer for extensor pollicis longus tendon ruptures. We achieved satisfactory results, and no complications were reported with these treatment approaches. However, this case report demonstrates that extensor pollicis longus tendon rupture may occur at the time of fracture; thus, clinicians should be aware of this issue

    The Role of Glycogen Synthase Kinase-3β in the Zinc-Mediated Neuroprotective Effect of Metformin in Rats with Glutamate Neurotoxicity

    Metformin has been suggested to have protective effects on the central nervous system, but the mechanism is unknown. The similarity between the effects of metformin and the inhibition of glycogen synthase kinase (GSK)-3β suggests that metformin may inhibit GSK-3β. In addition, zinc is an important element that inhibits GSK-3β by phosphorylation. In this study, we investigated whether the effects of metformin on neuroprotection and neuronal survival were mediated by zinc-dependent inhibition of GSK-3β in rats with glutamate-induced neurotoxicity. Forty adult male rats were divided into 5 groups: control, glutamate, metformin + glutamate, zinc deficiency + glutamate, and zinc deficiency + metformin + glutamate. Zinc deficiency was induced with a zinc-poor pellet. Metformin was orally administered for 35 days. D-glutamic acid was intraperitoneally administered on the 35th day. On the 38th day, neurodegeneration was examined histopathologically, and the effects on neuronal protection and survival were evaluated via intracellular S-100β immunohistochemical staining. The findings were examined in relation to nonphosphorylated (active) GSK-3β levels and oxidative stress parameters in brain tissue and blood. Neurodegeneration was increased (p < 0.05) in rats fed a zinc-deficient diet. Active GSK-3β levels were increased in groups with neurodegeneration (p < 0.01). Decreased neurodegeneration, increased neuronal survival (p < 0.01), decreased active GSK-3β (p < 0.01) levels and oxidative stress parameters, and increased antioxidant parameters were observed in groups treated with metformin (p < 0.01). Metformin had fewer protective effects on rats fed a zinc-deficient diet. Metformin may exert neuroprotective effects and increase S-100β-mediated neuronal survival by zinc-dependent inhibition of GSK-3β during glutamate neurotoxicity

    Adaptive Reuse Potential of Cultural Heritage: The Case of Istanbul Drapers' Bazaar

    The main objective of the study is to investigate the possibilities of adaptive reuse for the protection of cultural heritage buildings. It aims to set Istanbul Drapers' Bazaar (IDB), as an example of the buildings with similar characteristics and importance and for the cities hosting those structures upon an application of the AdaptSTAR method, which was developed to assess the conversion potential of buildings. Focused on a bazaar complex, an important example of modern architectural heritage, the present study contributes to the healthy transfer of the building to future generations and the improvement of its international recognition by investigating it in the context of convertibility. In this study, a mixed-mode research methodology including a two-stage approach was adopted. The first stage includes detailed qualitative interviews conducted with the architect and the administrative unit manager of the bazaar complex. The second stage consists of a survey conducted with a group of 45 experts, composed of scholars and professionals in Turkey. The results of the study suggest that it is necessary to develop IDB, especially in physical and technological terms and its function should be reconsidered. In particular, it was seen that economic issues constituted one of the major obstacles via-a-vis the continuity of use of large-scale buildings. Furthermore, it was realized that the artworks across the building group increased the importance of preserving the buildings and contributed to the possibility of their new functions being envisaged in the culture-art axis
