    IN VITRO REGENERATION OF Aloe vera L. IN A NEW PLANT TISSUE CULTURE MEDIUM

    A thesis Paper Submitted to the faculty of Agriculture, Sher-e-Bangla Agricultural University, Dhaka in partial fulfillment of the requirements for the degree of MASTER OF SCIENCEThe experiment was conducted at the Biotechnology Laboratory, Department of Biotechnology, Sher-e-Bangla Agricultural University, Dhaka, Bangladesh during the period from July 2021 to June 2022, to evaluate the regeneration of Aloe vera in a new tissue culture medium (SAU Tissue Culture Medium or Hoque medium) which was marked as Treatment-3. MS (1962) medium was used as Treatment-1 and MS Powder (Duchefa, The Netharland) was used as Treatment2. It was observed that, the highest percent (90%) of shoot regeneration was observed in the SAU Tissue Culture Medium. The same treatment showed maximum number of shoot in all the study period. The highest number of leaves were found in MS powder (Duchefa, The Netherland) medium and it was lowest in MS (1962) medium. Longest length of root (7.00) cm was observed in SAU Tissue Culture Medium and it was shortage (5.10) cm in MS (1962) Medium. Those three treatments were supplemented with same dose of phytohormone (1.00mg/L KIN + 2.00 mg/L NAA) to find out the ability of media in respect of hormonal condition. It showed that there was no significant difference in respect of days to shoot initiation and percent of shoot regeneration among the SAU Tissue Culture Medium and MS powder (Duchefa, The Netharland) when supllimented with phytohormone. Number of shoot per explant and number of leaves also showed non-significant difference between the Treatment-3 (SAU Tissue Culture Medium) and Treatment-2{MS powder (Duchefa, The Netherland)}. Percent of root induction and length of root also showed similar result among the Treatment-2 and Treatment-3. The MS (1962) medium showed lowest result for all the parameter under studied. The study highlighted the potentiality of SAU Tissue Culture Medium on In vitro regeneration of Aloe vera. It has indicated that the newly developed SAU Tissue Culture Medium (Hoque Medium) has excellent result on plantlet regeneration in Aloe vera. Hence, the formulation of SAU Tissue Culture Medium is a successful innovation in the field of tissue culture technology which can be applied for any other plant species

    PREVALENCE OF BLOOD PROTOZOANS OF MURINE RODENTS IN DHAKA CITY, BANGLADESH

Blood borne protozoans play a crucial role in this regards. The present study was performed to determine the prevalence of blood protozoan parasites of murine rodents in Dhaka city, Bangladesh. A total of 80 rodents with four rodent species were captured, namely, Rattus norvegicus (n=28), Rattus rattus (n=14), Bandicota bengalensis (n=30) and Mus musculus (n=8). The rodents were live captured by using traps from stationary/grocery shops (n=25), local rent houses (n=20), houses from slum areas (n=15) and fish markets (n=20). The overall prevalence of this study was 50%. The highest infection rate was found in R. norvegicus (57.14%) followed by B. bengalensis (53.33%), M. musculus (37.5%) and R. rattus (35.71%). Among the different areas, the highest prevalence was recorded in slum areas (66.67%) followed by fish markets (50%), stationary/grocery shops (48%), and local rent houses (40%). Five blood protozoa detected from the rodents which were Plasmodium spp. (31.25%), Anaplasma marginale (21.25%), Anaplasma centrale (17.5%), Babesia spp. (10%), and Trypanosoma spp. (3.75%). The ratio of single and mixed infection was equal (50%). Among the observed blood protozoa, Trypanosoma spp. and Babesia spp. have a great public health significance. Therefore, proper attention is recommended to prevent rodent born protozoal zoonosis through integrated control program

    EFFECT OF LEAF CLIPPING ON PRODUCTIVITY UNDER DIFFERENT POPULATION DENSITIES OF HYBRID MAIZE

    A Thesis Submitted to the Faculty of Agriculture Sher-e-Bangla Agricultural University, Dhaka, in partial fulfillment of the requirements for the degree of MASTER OF SCIENCE IN AGRONOMYA field experiment was conducted at Sher-e-Bangla Agricultural University, Dhaka during the period from October-2021 to February-2022 in Rabi season, to study the effect of leaf clipping at reproductive stage on productivity under different population densities of hybrid maize. The experiment consisted of two factors, and followed Randomized Complete Block Design (RCBD) with three replications. Factor A: Population density (4 levels) viz: P 1 = 60 cm × 15 cm (1,10,000 plants ha -1 ), P = 60 cm × 20 cm (83,000 plants ha 1 -1 ), P 3 = 60 cm × 25 cm (66,500 plants ha ) and Factor B: Leaf clipping (4 levels) viz: C -1 ) and P 1 4 2 = 60 cm × 30 cm (55,500 plants ha = Control (no clipping), C = Clipping of uppermost 2 leaves, C 3 = Clipping of uppermost 4 leaves and C = Clipping of uppermost 6 leaves. Experimental result revealed that, in case of different population densities of maize, the cob length (15.64 cm), cob diameter (4.82 cm), number of rows cob 4 (4.82), number of grains cob -1 (385.04), grain weight plant -1 (107.07 g) and 100-grain weight (22.45 g) of maize were observed in P 4 (55,500 plants ha -1 ) treatment. However, the highest grain yield of maize (8.27 t ha -1 ) was observed in P 2 2 -1 (83,000 plants ha ) treatment. Growth and yield of maize were significantly influenced due to the effect of leaf clipping at reproductive stage on maize. The highest cob length (15.02 cm), cob diameter (4.69 cm), number of rows cob -1 (14.33), number of grains cob (352.65), grain weight plant -1 (98.00 g), 100-grain weight (21.97 g) and grain yield (7.44 t ha ) were observed in C 1 (no clipping) treatment. In case of combination, the P combination treatment gave the highest grain yield of maize (8.53 t ha -1 ). Based on the above findings, experimental results revealed that different population densities and leaf clipping on maize at reproductive stage significantly influenced the growth and yield of maize. Therefore, it was suggested that planting 83,000 plants ha -1 along with no leaf clipping (P 2 C ) combination treatment would help to influence plant growth and increase its ability to give better yield production of maize

    EFFECT OF RICE HUSK ON ARSENIC ACCUMULATION IN POTATO PLANT UNDER DIFFERENT LEVELS OF ARSENIC TREATED SOIL

    A pot experiment was conducted in the experimental field of Sher-e-Bangla Agricultural University, Dhaka during the period from November, 2020 to May, 2021 to find out the effect of rice husk as an bio-adsorbent to decontaminate As toxicity in potato. The experiment consisted of two factors. Factor A: Arsenic levels (4) viz., As0: control (0 mg As kg-1 soil), As1: 20 mg As kg-1 soil, As2: 40 mg As kg-1 soil, and As3: 60 mg As kg-1 soil. Factor B: Rice husk levels (4) viz., R0: control (0 g kg-1 soil), R1: 20 g kg-1 soil, R2: 40 g kg-1 soil and R3: 60 g kg-1 soil. The experiment was laid out in a factorial randomized complete block design with three replications. Results revealed that As and/or rice husk had significant effect on arsenic load in different plant parts of potato t. Arsenic content in potato tuber flesh, peel, haulm and root gradually increased with the increase of its levels. On the contrary, As content in plant parts decreased with increasing rice husk levels. The soil treated with As1R3 exhibited As accumulation in tuber flesh (0.1070 mg kg -1 fresh weight) and peel (0.443 mg kg -1 FW), respectively. As load in different plant parts was in the sequence: root > haulm > tuber peel > tuber flesh. Although, the least As loading in tuber flesh was observed in As1R1, As1R2, As1R3 (range 0.1258-0.1070 mg kg-1 FW) which also showed higher productivity (range 402.67 - 416.67 g plant-1), but the treatment combination of As1R1 may be suitable for safe potato cultivation in lower level As contaminated soil. Therefore, potato growers can grow potato up to 20 mg As kg-1 contaminated soil treated with 20 g rice husk kg1 soil, which contains safe As load than the critical one (0.157 mg As kg -1 FW) for human consumption. So, application of rice husk for potato cultivation may a good option to reduce the arsenic hazards in lower arsenic endemic areas

    EFFECTS OF POTASSIUM AND SULPHUR WITH VERMICOMPOST ON GROWTH AND YIELD OF BORO RICE (BRRI dhan81)

    A Thesis Submitted to the Faculty of Agriculture Sher-e-Bangla Agricultural University, Dhaka, in partial fulfillment of the requirements for the degree of MASTER OF SCIENCE IN SOIL SCIENCEThe experiment was conducted at the research field of Sher-e-Bangla Agricultural University, Dhaka from November 2020 to May 2021 to investigate the effects of potassium and sulphur with vermicompost and to find out the best combination for better performances of Boro rice (BRRI dhan81). The experiment consisted two factors, Factor A: four potassium and sulphur fertilizer treatments viz: F : Control (no fertilizer), F 1 : 70 kg K ha -1 + 10 kg S ha -1 , F 2 : 100 kg K ha -1 + 20 kg S ha : 130 kg K ha -1 + 30 kg S ha -1 and Factor B: three vermicompost treatments viz: V : Control (no vermicompost), V 1 : 2 t vermicompost ha -1 and V 2 0 -1 and F : 4 t vermicompost ha . The experiment was laid out in randomized complete block design with three replications. Data were recorded on growth parameters, yield and yield components. The treatments had significant effects on most of the parameters. Results indicated that F , among the potassium and sulphur treatments and V , among the vermicompost treatments performed best in case of most growth and yield related characteristics like plant height, total number of tillers hill -1 2 , number of panicles hill -1 , panicle length, number of spikelets panicle -1 , number of filled spikelets panicle -1 , number of unfilled spikelets panicle -1 , 1000 grain weight, grain yield and straw yield. In case of treatment combinations the total number of tillers hill -1 (17.9) and panicles hill (16.7) were received from F 2 V 2 . Again, F 2 V treatment combination also performed the best result in case of the panicle length (23.25 cm), number of filled spikelets panicle 2 (126.41), total number of spikelets panicle -1 -1 (147.50) and 1000-grain weight (20.94 g). F treatment combination produced the highest grain and straw yield (6.60 t ha and 8.74 t ha -1 respectively). On the contrary, control treatments performed the lowest in all the growth and yield related parameters. Finally, the treatment combination F which is 100 kg ha -1 potassium and 20 kg ha -1 sulphur with 4 t ha -1 vermicompost performed the best for increasing the yield of Boro rice and improved the nutrient status of postharvest soil of AEZ 28

    SURVEILLANCE OF SEED BORNE PATHOGENS AND EVALUATION OF HEALTH STATUS OF HYV SEEDS OF RICE GROWN IN BADC FARMS OF BANGLADESH

Experiments were performed during the period July 2021 to June 2022 at Department of Plant Pathology, Sher-e-Bangla Agricultural University, Dhaka, Bangladesh. The rice variety were BRRI dhan48, BRRI dhan82, BRRI dhan83, BRRI dhan85, BRRI dhan98, BRRI dhan49, BRRI dhan75, BRRI dhan87, BRRI dhan28, BRRI dhan29, BRRI dhan89. Seed health study was conducted following dry inspection and blotter method of ISTA. In inspection of dry seed method the highest purity percentage was found in BRRI dhan49 (99.98%) whereas the highest inert matter percentage (0.37%) was found in BRRI dhan28.10 fungal pathogen, bacterial ooze and unidentified mycelial growth observed on seeds of HYV rice in blotter seed health test method. The identified fungi were Bipolaris oryzae, Aspergillus flavus, Aspergillus niger, Aspergillus sp, Curvularia lunata, Fusarium oxysporum, Fusarium moniliforme, Nigrospora oryzae, Chaetomium sp, Pyricularia oryzae and Rhizopus stolonifer. Pathogen incidence of identified fungi, Bacterial ooze and unidentified mycelial growth of fungi were recorded. Among Aus varieties, BRRI dhan48 and BRRI dhan98 showed the highest (49.25%) and the lowest (28.8%) of seed borne pathogenic infection, respectively. In case of Aman varieties, the highest (58.8%) pathogenic infection was found in BRRI dhan75. However, BRRI dhan49 and BRRI dhan87 showed the same (50.25%) seed borne pathogenic infection. On the other hand, among Boro varieties, BRRI dhan29 and BRRI dhan89 showed the highest (35.5%) and the lowest (17.75%) seed borne pathogenic infection respectively. Considering the frequency and occurrence of seed borne pathogen, Fusarium sp., Aspergillus flavus, Bipolaris oryzae and unidentified white mycelial growth were found predominant seed borne pathogens

    GROWTH AND YIELD OF BRINJAL AS INFLUENCED BY DIFFERENT LEVELS OF POTASSIUM

Sher-e-Bangla Agricultural University, Dhaka- In partial fulfillment of the requirements for the degree of MASTERS OF SCIENCE (MS) IN DEPARTMENT OF HORTICULTUREA field experiment was conducted at horticulture farm of Sher-e- bangla Agricultural University, Dhaka during the period from March to July 2022 to investigate the growth and yield of brinjal as influenced by varieties and potassium. The experiment consisted of two factors and followed by randomized complete block design (RCBD) with three replications. Factor A: three brinjal varieties viz. V 1 (BARI hybrid begun-4), V 2 (BARI begun-8) and V 3 (BARI begun-10) and factor B: four levels of potassium (K) viz. K 0 (0 kg K ha- 1 ), K 1 (95 kg K ha -1 ), K 2 (115 kg K ha -1 ) and K 3 (135 kg K ha ). In case of variety, significant variation was found for different parameters. The tallest plant (83.19 cm), maximum number of leaves per plant (35.18), highest number of fruits per plant (31.88), single fruit weight (58.18 g) and the highest yield (45.13 t ha ) was achieved from the variety V 1 . On the other hand, the different levels of potassium showed significant differences among them. The treatment K 3 provided the highest plant height (84.66 cm), number of leaves per plant (36.87), number of fruits per plant (29.93), single fruit weight (55.46 g) and the highest yield (39.96 t ha -1 ). Combined effect of variety and potassium also showed significant difference among the treatments. The treatment combination of V 1 K 3 performed the highest plant height (93.72 cm), number of leaves per plant (38.12) whereas V 1 K 2 gave the highest number of fruits per plant (37.30), single fruit weight (66.05 g) and the highest yield (54.67 t ha -1 ). Regarding economic analysis, V 1 K 2 also gave the highest gross return (Tk. 1093400.00), net return (Tk. 773330.00) and BCR (3.42). So the treatment combination of V 1 K 2 may be considered as a suitable combination for growth, yield and economic return of brinjal production and this treatment combination might be recommended at farmers level for better achievement

    PROFITABILITY AND RESOURCE USE EFFICIENCY OF TOBACCO CULTIVATION IN SOME SELECTED AREAS OF LALMONIRHAT DISTRICT OF BANGLADESH

Tobacco is the narcotic crop. The purpose of the study was to describe the socio-economic characteristics of tobacco farmers in the study area; to measure the profitability and resource use efficiency of tobacco in selected areas and to identify problem faced by the farmers in tobacco production in the study area. Primary data were collected from randomly selected 61 farmers from two upazillas named Lalmonirhat sadar and Kaliganj of Lalmonirhat district. Both descriptive and econometric model were applied in this study. Total cost of tobacco production was Tk. 175601 per hectare. Gross return of tobacco was Tk. 209279 per hectare, gross margin of tobacco was Tk. 86340 per hectare and that of net return was Tk. 33677 per hectare. Benefit Cost Ratio (BCR) was found to be 1.19. Production function analysis suggested that, among the variables included in the model, cost of hired labor, cost of urea and cost of MOP had a positive and significant effect on the gross return of tobacco production. Resource use efficiency indicated that hired labor, seed, Urea, TSP and MOP were underutilized and irrigation and insecticide were overutilized by tobacco farmers. The study identified some problems and constraints faced by the tobacco farmers .Instituitional supports from government and Nongovernment organizations for credit, input and technology may be helpful to increase tobacco cultivation as well as income of tobacco farmers

    ASSESSMENT OF METAL POLLUTION IN THE CROP FIELDS ADJACENT TO INDUSTRIAL AREAS OF KUSHTIA AND JHENIAIDAH DISTRICTS OF BANGLADESH

    A Thesis Submitted to the Faculty of Agriculture, Sher-e-Bangla Agricultural University, Dhaka, In partial fulfillment of the requirements for the degree of MASTER OF SCIENCE IN AGRICULTURAL CHEMISTRYIn the present study, we investigated the heavy metals pollution in soil, water and crops collected from crop fields near the industrial area of the Kushtia and Jhenaidah districts. In case of water study low concentration of Pb(0.13-0.18 mg/kg) were present in all the studied location but Cd,Cu and Cr concentration were below detectable limits. In case of the soil sample, the mean Cr and Cu concentrations in the soil from the studied location were higher than the permissible limits set by FAO/WHO, indicating that the soil accumulate higher metals near industrial areas. In the case of the rice sample, the mean Pb and Cr concentration in rice grain from all of the studied locations was much higher than the permissible limits set by FAO/WHO, indicating the rice grain accumulate higher Pb and Cr from contaminated field soil. The results also indicate that the contamination factor (CF) values indicating only Cu(falling between 3-6) and Cr(greater than 6) pose significant to severe contamination in field soil, while all metals pose low contamination in water and rice grain samples collected from studied sampling sites(Below 1). The results of the study indicate that the contamination degree (CD) of the water and rice grain samples collected from different industrial areas in the Kushtia and Jhenaidah districts was found to be low(Below 1). However, the field soil samples collected from the same areas exhibited a moderate degree of contamination. A Pollution Load Index (PLI) greater than 1 indicates that the soil samples collected were significantly contaminated with metals.But the PLI values found in the water and rice grain samples were below 1, which means that their contamination levels are low

    SUSTAINABILITY OF AGRICULTURAL MECHANIZATION IN RANGPUR DISTRICT

Sher-e-Bangla Agricultural University, Dhaka- In partial fulfillment of the requirements for the degree of MASTERS OF SCIENCE (MS) IN AGRICULTURAL EXTENSION & INFORMATION SYSTEMThis study aims to assess the sustainability of agricultural mechanization in Rangpur district. Agricultural Mechanization covers all levels of farming and processing technologies, from simple and basic hand tools to more sophisticated and motorized equipment. Sustainable agricultural mechanization considers economic, social, and environmental dimensions that contribute to the sustainable development of the agricultural sector. The objectives of the study were to identify a set of indicators that represents social, economic and environmental issues of agricultural mechanization, to assess the sustainability of agricultural mechanization, and to determine contributory factors of the sustainability of agricultural mechanization. An essential set of indicators were developed by reviewing the literature. Data were collected from 120 farmers of nine villages of three upazilas namely Rangpur Sadar, Gangachara and Taraganj of Rangpur district. The sustainability of agricultural mechanization was assessed by developing a composite sustainability index consisting of three dimensions. Normalization, weighting and aggregation were used for constructing the sustainability index. The results indicate that: i) on average the agricultural mechanization of more than one-third (38.42%) of farmers was sustainable and ii) using multiple regression analysis (standardized beta), the major contributing factors were adequacy of extension services, access to financial institutions, knowledge on climate change, education, the extent of reducing pre-harvest, harvest, post-harvest losses and increasing productivity. Research findings indicate that these six factors played a vital role in achieving sustainability of agricultural mechanization in the study area. Several policy recommendations (e.g., higher adequacy of extension services, full access to financial institutions, higher education, increase in agricultural productivity, availability of agricultural machinery, frequent use of agricultural machinery etc.) were made based on the findings of the study to achieve sustainability of agricultural mechanization in Rangpur district
