    Development of a Fluorescent Protein-based Laboratory Course for Life Science Students

    application/pdfDNA からタンパク質への遺伝情報の流れは生命系の学生にとって重要で基礎的な学修内容であるが、深い 学びのためには学生実験でDNA とその産物のタンパク質をともに細胞から抽出・精製し、それらの性質を調 べるといった実習が望まれる。しかし、学生実験では1 つのテーマに割ける時間は限られているので、短期間 でそれらの関連分野をまとめて学修できる内容を検討した。その結果、緑、赤、藍、橙の4 色の蛍光タンパク 質(それぞれ、GFP、RFP、CFP、OFP)の各遺伝子を大腸菌に導入し、少量の培養液から各蛍光タンパク質 を精製できる系を開発した。構築したプラスミドは、高コピーの複製起点とアンピシリン耐性遺伝子をもち、 ポリヒスチジンタグを融合させた各蛍光タンパク質の高発現を可能とする。各蛍光タンパク質を発現した大腸 菌はいずれも明瞭な色を呈し、Ni アフィニティカラムにより容易に発現タンパク質を精製できる。departmental bulletin pape

    A demonstration of visualizing sound waves by utilizing Lissajous curves

    application/pdfAn attractive demonstration, observation of chaotic Lissajous curves originating from sound waves, was presented for the general public. A simple experimental setup used in this demonstration consists of an optical lever, a rotating mirror, and a laser beam source. This system can detect a variety of Lissajous patterns without an oscilloscope. Thus, this system can be regarded as a budget oscilloscope.     Image feature extraction of comic artist by CNN

    application/pdfThis paper reports the extraction of image features of comic artist by convolutional neural network. In recent years, illegal uploading of comics has become a social issue. The comic artist wants to find his own illegally uploaded comic. A convolutional neural network extract image features representing a class from an image. Comics has the artist's style. The artist's style can be used as an image features that represents a class in the comic image classification problem. Therefore, convolutional neural networks can solve the artist classification problem for comic images. If the classification problem can be solved, it can be applied to image retrieval. Visualize the area of interest when convolutional neural networks predict artists. There is an element that can predict the artist in the attention area. The experimental results suggest that the comic characters face is important for convolutional neural network to predict the artist from the comic images.departmental bulletin pape

    Descartes and Turing

    application/pdfRené Descartes does not think that machines can speak like human beings because they do not have “reason.” About three hundred years later, Alan Turing insists that if machines can speak like human beings, they can think. In other words, Turing stands Descartes’s insistence on its head, as Roy Harris and Noam Chomsky point out. But Descartes insists that only human beings have “reason” so he never assumes that machines can speak. On the other hand, Turing insists that it is a matter of words whether machines can think or not. This means, as Turing claims, that the question, “Can machines think?” is not worth discussing.     Polyamine analyses in capsicum plants by a combined method of thin-layer chromatography and image analysis

    application/pdfPolyamines are widely distributed in living organisms and essential for their cell proliferation and cell differentiation. Although it has shown in recent decdes that polyamines have specific funtions in plants, such as environmental stress response and organ development, the molecular basis still remains unclear. A previous study has reported that polyamines are highly abundant in green peppers. In this study, to examine whether polyamines are abundant in the every cultivars of capsicum plants, we analyzed the amount of polyamines, which are putrescine, spermidine, and spermine, by a combined method of thin-layer chromatography and image analysis. Taking an organ-specific of polyamine accumulation into consideration, our results may suggest that high accumulation of putrescine in green peppers is caused by a competitive inhibition due to ethylene biosynthesis.departmental bulletin pape

    Learning Analytics Obtained from the Large-scale Online Testing

    application/pdfThe FP (follow-up program) system, a large-scale online testing system, is principally an assistance system for learning students, where the FP system was aimed at helping students who need basic learning and aimed at assisting teachers who have to engage in teaching a variety of educational students. However, all the teachers and stuffs do not always know such a system deeply. The author had a chance to explain the system in public at the research conference held in June 22, 2018, and thus he used this opportunity to do this. This paper describes the details of the lecture in June 22, 2018. Firstly, he mentions the item response theory comparing to the classical testing theory.     Inhomogeneous superconducting state in hydrogen storage alloy PdDx

    application/pdfDisorder affects the coherent state of a superconductor in characteristic ways. In conventional superconductors with s-wave pairing symmetry, magnetic defects act as strong pair breakers, whereas nonmagnetic defects have negligible effect. In this review, we will show that nonmagnetic defects promote the superconductivity mediated by optical phonon in palladium-hydride system. Temperature dependence of the critical current is well fitted by the Ambegaokar-Baratoff relation, suggesting the existence of Josephson junction between the superconducting(SC) grains. Appearance of the coherent SC grains through the Josephson coupling may be associated with the interaction between deuterium and lattice defects such as vacancies and dislocations. In addition, we theoretically investigated whether or not optical phonon modes formed by the collective motion of a large number of atoms exist in nonstoichiometric PdHx, possessing random or inhomogeneous lattices. We found that individual excitations compete with collective excitations in controlling the system’s randomness, and that optical phonons develop from localized phonon modes as the stoichiometry x increases.departmental bulletin pape

In previous studies research was conducted experiments of R-brace and numerical analysis of one-story-one-bay frame. In this paper numerical analysis with multi-story-multi-bay is carried out and performance of R-brace is discuss. As a result. The performance of R-brace is restrained residual deformation when the burden is large.     application/pdfThe education of the lower secondary school in France which corresponds to the Period of Integrated Study has been developed as a project study with the keyword “interdisciplinarité” from the end of last century. Its successor EPI (Interdisciplinary Practical Learning) starting from 2016 -2017 school year was planned as an effective learning approach for the French situation like declining academic performance. Japan, which showed the high performance in the PISA survey, seems to be oriented the more advanced one, with expecting that the school plan and the curriculum management is required to be more strictly executed. But there are uneasy factors to deepen the learning. Although the effectiveness of interdisciplinarité in France is still controversial, the French style that the specialty of the subject teacher in charge is utilized seems firm from the viewpoint of how to develop the class to deepen the learning.departmental bulletin pape
