Study on rheological properties and stability of oil-in-water 
emulsions containing native maltodextrin or esterified 
maltodextrin in aqueous phase

The aims of the research are; to investigate the inf]uence of the concentration and dextrose 
equivalent (DE) value of native maltodextrin on rheological properties and the stabi1ity of oil-in-water 
emulsions; to investigate the optirnum condition for esterified maltodextrin production by enzymatic 
esterification; to study the physicochemical properties and emulsifying properties of esterified 
maltodextrin; and to investigate the influence of the concentration and type of estrified maltodextrin 00 
rheological properties and the stabiJity of oil-in-water emulsions. 
The effect of concentration of native maltodextrin with three different DE values 00 the viscosity， 
depletion attraction potential (Wdep)， rate of coalescence (Kc) and creaming rate of oil-in-water emulsion 
prepared with Tween80 have been investigated. The relative viscosity and Wd~p increased. with 
increasing maltodextrin concentration. Critical flocculation concentration (CFC) of emulsions containing 
maltodextrin with DE of 16 (DEI6)， 12 (DEI2) and 9 (DE9) were 11， 7 and 5.5% (w/w) respectively， as 
determined by viscosity measurement. A t native maltodextrin concentrations below CFC， there was no 
change in Kc and no creaming was observed. At native maltodextrin concentrations above CFC， an 
increase in the concentration of DE9 and DEl2 resulted in an increase in Kc until it reached a constant 
value. Kc values remained to be constant in the concentration range between 30 and 40% (w/w) for DE9 
and that between 35 and 45% (w/w) for DEI2. Further increasing in concentration of DE9 and DE12 
decreased Kc. Kc of DE16 monotonically increased with increasing concentration from CFC to 50% 
(w/w). A maltodextrin with a lower DE inhibited creaming more efficiently than maltodextrin with a 
higher DE because of higher viscosities. The Kc tended to increase with decreasing DE because the 
strength of interaction between oiI droplets increased 
The next research focuses on the relationship between rheological behavior and stability of 
oil-in-water emulsions containing native maltodextrin prepared with Tween80. The CFC's of oil-in-water 
emulsions containing native maltodextrin with DE16， DE12， and DE9 were 1， 8 and 6% (w/w) 
respectively， as revealed by microscopic method. The differences in the resulting measures are minimal， 
compared to viscosity measurement. Coalescence was observed as native maltodextrin concentration 
increased above the CF

depleting flocculation. The consistency index (kemuls;on) and yield stress (τ。)of emulsions containing 
native maltodextrin increased with increasing concentration or decreasing DE. The emulsions containing 
native maltodextrin showed Newtonian behavior as the concentration below the CFC. At native 
maltodextrin concentrations above the CFC， emulsions containing native maltodextrin exhibited shear 
thinning behavior. An increase in the concentration resulted in a decrease in the nemlllsion until 
concentration reached 20% (w/w) for DE9， DE12 and 25% (w/w) for DEI6. Further increase in 
concentration resulted in an increased the nemlllsion because of predominant influence of the continuous 
phase 
The new types of hydrophobically modified maltodextrin of DEI6 and DE9 were prepared by 
enzyme-catalyed reaction of maltodextrin and three fatty acids: decanoic acid (C.I 0)， lauric acid (C-12) 
and palmitic acid (C-16). Lipase obtained from T lan!lginosus was found to be a useful biocatalyst in 
the maltodextrin esterification. Esterified maltodextrin with a degree of substitution (DS) 0.015-0.084 
was prepared at the optimum conditions of 60'C for 4 hours. The DS was found to be at its highest 
when maltodextrin and fatty acids were taken in the ratio 1 :0.5. The functional properties of these 
esterified maltodextrin were investigated. Al1 esterified maltodextrin did oot completely dissolve in 
water. All esterified maltodextrin of DEI6 at a concentration of 25% (w/w) exhibited Newtonian 
behavior similar to that of native maltodextrin， whereas esterified maltodextrin of DE9 with long chain 
fatty acid showed shear thinning behavior. Esterified maltodextrin had a higher viscosity compare to 
native maltodextrin. X-ray diffraction pattern of esterified maltodextrin indicated crystallization of the 
fatty acid side chains. The thermal stability of esterified maltodextrin DE16 was checked by differential 
scanning calorimetry (DSC). The stabilization mechanism of esterified maltodextrin in emulsions was 
investigated. Esterified maltodextrin was then used as an emulsifier to make n-hexadecane O/W 
emulsions. The emulsifying properties of esterified maltodextrin depend on the degree of substitution， 
type of fatty acid and chain length of maltodextrin. 
Lastly， 1 studied the effect of the concentration and type of esterified maltodextrin in the aqueous 
phase on rheological behavior and stability of oil-in-water emulsions prepared with Tween80. Esterified 
maltodextrin DE16 solution exhibited Newtonian behavior regardless of an increase in concentration. 
Moreover， shear thinning behavior of esterified maltodextrin DE9 solution was induced with increase in 
concentration. The CFC's of emulsions containing esterified maltodextrin of DE16 and DE9 were 19 
and 14% (w/w) respectively， as revealed by microscopic method. There was no flocculation observed at 
esterified maltodextrin concentration below CFC and the emulsions exhibited Newtonian flow behavior 
At concentration above CFC， the emulsions were extensively flocculated hence share thinning behavior 
was observed. Further in increase in the concentration of al esterified maltodextrin DE 16 resulted in 
less pronounced shear thinning behavior due to the influence of colloidal nature of continuous phase 
However， flow behavior of emulsion containing maltodextrin. DE9 decanoate and laurate exhibited 
enhanced shear thinning behavior with increasing concentration because of pseudoplasticity of 
continuous phase. On the other hand， the less enhanced shear thinning behavior of emulsion containing 
maltodextrin DE9 palmitate was observed because of flocculation inhibition. Esterifed maltodextrin 
with lower DE and longer fatty acid molecules more efficient!y inhibited creaming than estrified 
maltodextrin with higher DE and shorter fatty acid molecules. The esterified maltodextrin extended 
CFC and was more effective in preventing creaming， compared to native maltodextrin.

    学位論文の要約 
目的： 
プラチナ製剤等細胞障害性抗癌剤及び分子標的薬の開発により抗癌剤治療が劇的に進化しつ
つあるが、治療過程で常に薬剤耐性が誘導されることが治療上の限界となる。ところで、臨床
においてはこれら異なる二つのタイプの薬剤を連続して使用するケースがあるものの、一方の
薬剤により誘導された耐性が他方の薬剤に対する感受性・耐性に与える影響に関しては殆ど解
明されていない。本研究では、肺癌細胞株を用いた cisplatin、分子標的薬連続治療モデル系
を構築し、プラチナ製剤治療が分子標的薬治療に与える影響を検討した。また、プラチナ製剤、
分子標的薬双方に共通する耐性機構である上皮間葉移行が、本連続治療モデルに関与する可能
性を併せて検討した。 
方法： 
EGFR 阻害薬 gefitinib に感受性の肺腺癌細胞株 HCC4006 及び HCC827、HER2 阻害薬 lapatinib
に感受性の肺扁平上皮癌細胞株 NCI-H2170 を cisplatin 暴露下で継代培養し cisplatin 耐性細
胞を樹立した（HCC4006-CR、HCC827-CR 及び NCI-H2170-CR 細胞株）。更に親株、cisplatin 耐
性細胞に存在する分子標的薬耐性ポテンシャル細胞を二週間の gefitinib または lapatinib 暴
露 に よ り 選 択 し （ HCC4006-GR2w 、  HCC4006-CR-GR2w 、 HCC827-GR2w 、 HCC827-CR-GR2w 、
NCI-H2170-LR2w 及び NCI-H2170-CR-LR2w 細胞株）、得られた二つの耐性細胞を比較解析するこ
とにより、シスプラチンに対する耐性獲得が分子標的薬に対する耐性獲得に与える影響につい
て検討した。各分子標的薬への感受性は MTS assay により、上皮間葉移行マーカーの発現はウ
エスタンブロット及び qRT-PCR 法により確認した。上皮間葉移行に伴う運動能の亢進を
transwell migration assay により評価し、この運動能の制御に関わる因子を、siRNA ノックダ
ウン法により検討した［Kurokawa, Ise, Omi, Goishi and Higashiyama, 2013：主論文］。 
結果： 
cisplatin 耐性細胞株 HCC4006-CR は親株に比して gefitinib 感受性を部分的に失った。また
本耐性細胞を gefitinib に僅か 2 週間暴露することにより選択された細胞株 HCC4006-CR-GR2w
は、HCC4006-GR2w と比して gefitinib 高耐性を示した。これらの結果は、cisplatin 耐性獲得
が gefitinib 耐性化に影響を与えたことを示す。さらに耐性細胞では上皮間葉移行様の変化を
認め、cisplatin によって誘導された上皮間葉移行様の変化が gefitinib 耐性獲得に影響を与
えることを示す結果を得た。 
HCC827 細胞株から同様に樹立された耐性細胞 HCC827-CR-GR2w もまた、上皮間葉移行様の変
化を示すマーカー変動を認めた。ところで、HCC827 の gefitinib 耐性機構として MET 遺伝子増
幅が報告されているが、HCC827-CR-GR2w では MET 遺伝子増幅は見られなかった。一方、親株
HCC827 から選択された gefitinib 耐性細胞 HCC827-GR2w は上皮間葉移行様の変化を示さなかっ
た。以上の結果は、HCC827 では cisplatin 暴露が gefitinib に対する耐性獲得機構に影響を与
え、上皮間葉移行様の変化が優勢的に gefitinib 耐性獲得に関与したことを示す。 
また、cisplatin 耐性細胞株 NCI-H2170-CR から lapatinib 二週間暴露により選択された
NCI-H2170-CR-LR2w でもまた上皮間葉移行様の変化を示すマーカー変動を認めたが、親株から
選択された NCI-H2170-LR2w では上皮間葉移行様の変化は見られなかった。以上の結果は、
NCI-H2170 においても cisplatin 暴露が lapatinib に対する耐性獲得機構に影響を与え、上皮
間葉移行様の変化が優勢的に lapatinib 耐性獲得に関与したことを示す。 
樹立した耐性細胞の運動能を評価したところ、cisplatin 耐性細胞から分子標的薬で選択さ
れた、上皮間葉移行様の変化を伴った細胞株で顕著な運動能の亢進が認められた。また、HCC4006
及び HCC827 由来耐性細胞では、この運動能の亢進が受容体チロシンキナーゼ AXL の発現亢進と
相関し、これらの亢進した運動能は AXL ノックダウンにより有意に抑制された。 
結論： 
cisplatin、分子標的薬による連続治療においては、cisplatin によって誘導された上皮間葉
移行様の変化が、続く分子標的薬に対する耐性獲得にも影響を与えうる可能性が示された。ま
た、上皮間葉移行様の変化を伴ってこれら二剤に耐性を獲得した癌細胞は運動能の亢進など高
転移能を獲得する可能性が示され、それら耐性癌に対しては AXL 等の上皮間葉移行関連因子を
標的とした抗転移治療が有効ではないかと考えられた。

緒 言
alanine : glyoxylate aminotransferase2（AGXT2）は，
D体アミノイソ酪酸（D-aminoisobutyrate，D-AIB）と
ピルビン酸から2-メチル-3-オキソプロパネートとア
ラニンを，生成するアミノ基転移酵素である．AGXT
2は，腎臓，肝臓に多く存在するため，これまでに我々
はラット肝臓より精製し特徴を決定し報告1）している
が，脳でなぜAGXT2が発現しているかについては，
わかっていなかった．最近，AGXT2が一酸化窒素合
成酵素（nitric oxide synthase，NOS）の競合阻害物質
である非対称性ジメチルアルギニン（asymmetric di-
methylarginine，ADMA）を分解することが報告され
た2）．今回私は，AGXT2の脳内での機能を推測するた
め，ブタおよびラットでのAGXT2の脳内分布を調べ
ることとした．
対象と方法
対象：屠殺後の新鮮なブタの脳，腎臓，肝臓を用意
し，脳は大脳灰白質，大脳白質，小脳灰白質，小脳皮
質，尾状核，視床，海馬，中脳，橋，延髄，脳梁の11
部位に分け－80度で保存した．6－8週齢のオス
Wistar系ラットは，ジエチルエーテル麻酔下で断頭し，
脳を摘出し，前頭葉，側頭葉，小脳，線条体，視床，
海馬，中脳，橋，嗅脳の9か所に分け－80度で保存し
た．すべての実験は愛媛大学動物実験倫理委員会の承
認を得て行った．
ブタAGXT2 cDNA配列決定：ブタcDNA配列を決定
するため，常法により抽出したtotal RNA分画を用い，
逆転写反応によりcDNAを作成し，他の種のAGXT2遺
伝子cDNA配列を参考にプライマーを作成し，5’-およ
び3’-rapid amplification of cDNA ends（RACE）法に
よって増幅後，アミノ酸配列を決定した．

Summary
Alanine : glyoxylate aminotransferase 2（AGXT2 ; EC 2.6.1.44）is known to be an important amino
enzyme that transfers not only D-3-aminoisobutyrate, an intermediate of thymine degradation, to 2-methyl-
3-oxopropanoate but also an asymmetric dimethylarginine, a competitive inhibitor of nitric oxide synthase,
to α- keto -δ-（NG, NG- dimethyl guanidino）valeric acid with pyruvate as an amino acceptor. Large
amounts of AGXT2 are distributed in the kidney and the liver but they are also found in the brain. In order
to clarify the role of AGXT2 in the brain, AGXT2 enzyme assay was performed in a pig brain tissue.
Following this experiment, the pig AGXT2 cDNA sequence was determined by 5’- and 3’-race methods and
the mRNA distribution of the AGXT2 gene in the pig and the rat brains was studied with quantitative real-
time PCR methods. In both brains, the AGXT2 mRNA was distributed throughout the brain tissues, with
relatively high expression found in the frontal cortex, striatum and hippocampus. It is suggested that the
AGXT2 gene may play an important role in the central nervous system.

AGXT2酵素活性の測定：既報3）に基づき測定し
た．まず抽出した各組織にPBSを加え，ホモジナイズ
し，遠心後，上清をとった．上清と14Cで標識した β-
アラニン，ピルビン酸を混合し，37度で反応させた．
反応後，2，4-dinitrophenylhydrazine（DNPH）を加え生
成物を処理した．トルエンを加え，よく混合し，有機
層を液体シンチレーションカウンターにて測定した．
AGXT2 mRNAの定量：RNA分画を常法に基づき抽
出した後に，AGXT2TaqMan primer-probe set（Applied
Biosystem，P101206-000H06，Rn00582928_m1）を 用
い，quantitative realr-time PCR法にて定量した．内部標
準としてGAPDHを用いた．
統計：統計解析はSPSS統計ソフトpackage11．5を用
いて行った．mRNA発現の組織ごとの差はBonferroni
post hoc testを行い，p値が0．05未満を有意とした．
結 果
ブタAGXT2 cDNA配列決定：図1に各種のAGXT2
アミノ酸配列を示す．ヒトを基準に各種のアミノ酸配
列の一致率を調べた．ラット83．7％，マウス88．0％，
ウシ86．8％，ブタ85．8％だった．
AGXT2酵素活性の測定：図2にブタ各臓器の酵素
活性分布を示す．腎臓，肝臓では高い活性がみられた
が，脳でも活性が確認できた．脳内では，尾状核，視
床，小脳での活性がやや高い傾向にあったが，脳部位
別の有意差は確認できなかった．
AGXT2 mRNAの定量：図3にラットとブタAGXT2
mRNAの全組織の比較，図4にラットとブタAGXT2
mRNAの脳内の比較を示す．ラット，ブタとも腎臓，
肝臓で発現は有意に高かった．しかし，脳内でも発現

が確認できた．各部位で比較したところ，ラット，ブ
タともに前頭葉，海馬で高い傾向にあったが有意差は
みられなかった．
考 察
今回私は，ブタAGXT2のアミノ配列を決定し，ブ
タおよびラットの組織別にAGXT2mRNAを定量した．
AGXT2アミノ酸配列は，哺乳類各種で一致率が高
く，重要なアミノ酸であると考えられた．ブタAGXT
2酵素活性は腎臓及び肝臓に高かったが，他の種同
様，脳にも活性が認められた．ラットおよびブタAGXT
2 mRNAを定量した結果，ともに脳に発現しているこ
とが確認された．脳部位別発現量では，有意差はなかっ
たものの皮質，海馬に高い傾向がみられた．
これまで，AGXT2は，チミンの代謝産物D-AIBの
分解に関わる唯一のアミノ基転移酵素であることか
ら，脳内でもD-AIBの分解に関わっていると考えてい
た4），5）．しかし，最近，AGXT2は，ADMAの分解に関
係し，生体内でADMAを実際に分解していること6），
AGXT2欠損マウスが高血圧を呈すること7）から，
AGXT2はADMAによって阻害されるNOS活性を増加
させ，機能していることが推測される．その場合，脳
内AGXT2は，NOに関係した作用を持っているかもし
れない．今後，AGXT2に対する抗体を用いて，AGXT
2の組織別分布，細胞別分布を調べ，より詳細な検討
を行う予定である．
結 論
ブタAGXT2cDNA配列を決定し，哺乳類を通して高
い保存性を有するアミノ基転移酵素であることを確認
した．ブタおよびラットにてAGXT2の分布を酵素活
性，mRNAで調べた結果，腎臓，肝臓に多く発現する
ものの，どちらの種においても，脳で発現しているこ
とが確認された．詳細な脳内分布を調べたが，広く発
現しており，なんらかの重要な機能を持っていること
が推測された．

都市雨水排水管渠網の空気圧縮を考慮、した雨水流出解析
モデルに関す る研究

近年， 各地の都市下水道流域で，気候の温暖化と も相 まって ゲリ ラ豪雨が頻発
し，浸水氾濫災害がたびたび発生するようになってきている.こうした都市域の
豪雨時の雨水流出(浸水氾濫)現象をシミュレートする雨水流出解析モデルにつ
いては，今日までに，開水路流れとサーチャージ流れが混在する複雑な雨水排水
管渠網内の流れと地表面上を複雑な様相を呈して流下する浸水氾濫流れを共に精
度高く追跡計算できる幾つかの流出解析モデノレが開発されてきている. しかしな
がら，マンホーノレ蓋飛散現象などが引き起こされる可能性のある，空気圧縮を伴
う雨水排水管渠網の雨水流出をもシミュレートしえる雨水流出解析モデルは未だ
開発されていない 本論文は，そうした空気圧縮を考慮した雨水流出解析モデル
の開発を目指したものである
第 1章では，都市雨水排水流域の雨水流出モデルに関する研究の歩みを明らか
にしてい る すなわち ，都市化現象が急速に進展 してきた 1960年代から ，地表面
および雨水排水管渠の雨水の流れをいずれも kinematicwave流れと し，雨水排水
管渠の流れは open-channelf10w として取り扱うところの都市雨水流出モデ、/レが
提案され始めた. 1980年代に入り ，都市化がさらに進展するようになると ，計画
を上回る大量の雨水が流出する よ うになり ，雨水排水管渠網では surchargedf10w 
(満管流れ)がしばしば発生するようになってきたため，雨水排水管渠の雨水の
流れを dyna皿 icwave流れとして取り扱い， open-channel f10wと surchargedf10w 
の両流れをシミュレートできる雨水流出モデルが提案されるようになってきた
さらに， 2000年代に入ると，世界の各地の都市域で，気候の温暖化の進行とも相
まって，豪雨が頻発するようになるとともに都市の雨水流出も激しさを増し?
ンホールからの氾滋 ・溢水による大規模な地表面浸水氾濫流が発生するようにな
ってきたため，この複雑な流れの様相目を呈する地表面浸水氾濫流を dynamicwave 
流れとして取り扱い，雨水排水管渠の流れと組み合わせてシミュレートできる雨
水流出モデルが提案されるようになってきた 近年では，豪雨時，雨水排水管渠
網の激しい流れが空気を圧縮してマンホール蓋を飛散させる雨水流出現象がしば
しば発生するようになっており，こうした豪雨時の空気圧縮を伴う激しい雨水の
流れをシミュレートできる都市域の雨水流出モデルが必要とされるようになって
きているが，現在までのところ，そうした雨水流出モデノレは未だ開発されていな
し、-
第 2章では s はじめに，雨水排水管渠網内に多数，存在し，それらを通じて空
気の排気 ・吸気が行われるところの，これまで全く考慮、されることのなかったマ
ンホール左取付管を組み入れ，空気の流れを等エントロビー流れと考えて，雨水
排水管渠網の空気・水二層流の流出における空気の流れの基礎式(運動方程式と
連続の式)を導出している.次いで，雨水排水管渠網の空気圧の時間的 ・場所的
変動を考慮して，空気・水二層流の水(雨水)の流れの基礎式(運動方程式と連
続の式)を導出している なお，これら基礎式は，雨水排水管渠網内の雨水と空
気の実際の流れを厳密に記述できているものとしては，都市雨水流出の分野では
初めて，導出されたものである.
第 3重量では，上述した雨水と空気の流れの基礎式を，厳密に数値解析するため，
特性曲線法を採用し，その数値解析のための数値計算式(差分式)を展開してい
る 雨水と空気の二層流を，特性曲線J去を用いて厳密にかつ精度高くまた安定的
に数値解析しようとする試みは，こうした分野の雨水流出解析法では，はじめて
の試みでもある
第 4章では，本論文で新たに導出・提案された雨水排水管渠網における空気・
水二層流の流出解析モデルの適合性 ・妥当性について，本解析モデノレを雨水排水
管渠水王里模型による二層流流出実験に適用し E 解析結果を実験結果と比較して調
べ，本流出解析モデル(流れの基礎式およびその数値解析手法)が高い適合性と
妥当性をもつものであることを明らかにしている
第 5章では，本研究で得られた主要な成果を要約し，結論としている.
以上のように，本論文は，未だ開発されていない，マンホール蓋飛散現象など
が引き起こされる可能性のある，空気圧縮を伴う雨水排水管渠網の雨水流出をも
シミュレートしえる都市域の雨水流出解析モデルの開発を目指したものであり
本研究成果が土木工学(水工学) ，特に都市流出分野の研究の進展に寄与すると
ころ大である このように，本論文は，博士(工学)の学位論文として価値ある
ものと認められる
