Dissertation Abstract 
Diabetes mellitus is now becoming a common metabolic disorder resuJting from 
the inability of our body's response to high blood glucose level. Type 2 diabetes mellitus 
is reported to be 90-95% of al diabetic cases， characterized by hyperglycemia. There is 
considerable evidence that hyperglycemia results in the generation of reactive oxygen 
species， ultimately leading increased oxidative stress in a variety of tissues. Inhibition of 
a-glucosidase is one approach to suppress postprandial hyperglycemia， thereby delaying 
glucose absorption and controlling the hyperglycemia in diabetic patients and reducing 
chronic vascular complications. Therefore， the ideal antidiabetic compound should 
possess both hypoglycemic and antioxidant properties 
Aspergi/lus terreus is a filamentous， cosmopolitan and ubiquitous fungus whicb is 
primarily isola!ed from soil. Tbe compounds isolated from A. terreus mostly possess 
pbarmacological and commercial values， sucb as lovastatin as ant卜cbolesterol.However， 
tbere are only a few reports on production of α-glucosidase inhibitor and antioxidant by 
species of Aspergi/lus. Tberefore， the aims of the study are to isolate and characterize tbe 
active compounds of A. terreus guided by inbibitory on yeast α-glucosidase and 
antioxidant assays. In tbis study we selected four isolates from A. terreus namely LSO 1 
and RCC 1 which cuJtured on potato maJt peptone (PMP) medium， and MC751 and LS07 
which cultured on Czapek-dox (Cz) medium 
Tbe isolation of antioxidant compounds from ethyl acetate (EtOAc) extract of 
LSOl using column chromatography on silica gel was guided by antioxidant activity 
yielded tbe crystalline compounds 1 and 2 wbich were identified as terreic acid and 
terremutin， respectively， on tbe basis of spectrometric analyses. Terreic acid (1) and 
terremutin (2) exbibited tbe highest level of DPPH free radical scavenging activity witb

ICso values of 115.0 and 114.0μM， respectively. This is the first report on antioxidative 
activity of terreic acid (1) and terremutin (2) from A. terreus. 
The bioassay-guided isolation and purification of an EtOAc extract of A. terreus 
MC751 led to the characterization of butyrolactone 1 (3) as an antidiabetic and 
antioxidant. The compound 3 demonstrated significant concentration-dependent， 
mixed-type inhibitory activity against yeast a-glucosidase with an ICso of 52.2 J.lM. The 
antioxidative activity of compound 3 was evaluated based on the scavenging effects on 
DPPH with ICso value of 51.4μM. This is the first report on α-glucosidase inhibitory 
activity of butyrolactone 1 (3). Further studies revealed that the production of compound 
3 and activities of EtOAc extract of MC751 increased when Czapek-dox medium was 
added with 0.5%. yeast extract (CzY) and incubated under static condition for 15 days. 
The EtOAc extract was successively subjected to silica gel column chromatography to 
afford butyrolactone 1 (3) and I (4) as major constituents. In order to verify this 
structure and to study the structure activity re1ationship (SAR)， compound 3 was 
converted to aspernolides A (3a) and acetylated to give butyrolactone 1-triacetate (3b) 
and -diacetate (3c)ー TheSAR of butyrolactone derivatives revealed that the prenyl side 
chain and hydroxyl group at lactone group in butyrolactone 1 has contribution for 
inhibitoryα-glucosidase activity， however not for antioxidant activity. The compound 3 
was found to be the most active compound as inhibitor of α-glucosidase (ICso= 52.2μ恥1)，
while compound 4 was the most potential for antioxidant (lCso= 17.6μM). 
A. terreus LS07 was cultured in CzY medium under static condition for fifteen 
days (i) and shaking condition for seven days (i) in order to increase the yieJd of extract 
Chromatographic separation of the EtOAc extract (i) led to the isoJated of compounds 3 
and 4. The isolation of the EtOAc extract (i) gave compound oleic acid (5)， ergosterol 
(6) beside compounds 3 and 4 as m吋ormetabolites. Ergosterol (6) exhibited poor 
inhibitory activities on α-glucosidase and scavenging DPPH radicals. 1n particular oleic 
acid (5) showed the highestα-glucosidase inhibitory activity (ICso=8.5μM). By 
comparing the activity of stearic， linoleic， and linolenic acid， itrevealed that a double 
bond in compound 5 was essential for a-glucosidase inhibition. However， the increasing

   学位論文要旨   
背景：プロポフォール(2,6-diisopropylphenol)は麻酔の導入・維持に広く使われている静脈
麻酔薬である。プロポフォールの睡眠量以下での投与（subhypnotic dose）に鎮痛効果が
あるかどうかは議論されており、動物実験では鎮痛効果に一定の結果が得られておらず、
人においては、術後痛軽減作用があるとの報告がある。我々は末梢組織に残留したプロポ
フォールが抗侵害受容、抗痛覚過敏作用を有し、術後痛を軽減すると仮説を立てた。末梢
組織に残留したプロポフォールの効果を調べる目的で、プロポフォールをラットの皮膚への
塗布し、抗侵害受容作用および抗痛覚過敏作用を行動学的、電気生理学的に調べた。 
方法 
行動学的実験：SDラット（n=8,510-698g）を用い、後肢皮膚にプロポフォール(1,10,25%)を塗
布し後肢の侵害性熱刺激に対する逃避反応時間（Paw ｗithdrawal latency：PWL）および、
機械刺激に対する後肢の逃避反応閾値の経時的変化を調べた。また侵害熱刺激に対し
痛覚過敏をおこすアリルイソチオシアネート(AITC; mustard oil)塗布10分後に10％プロポフ
ォールを塗布し侵害性熱刺激に対するPWLの経時的変化を調べた。侵害性熱刺激に対
するPWLの測定にはHagreaves’ plantar test apparatus (Plantar Test 390; IITC Life 
Science)を使用し、機械反応に対する逃避反応閾値の測定にはelectronic von Frey 
filament (1601C; IITC Life Science)を使用した。コントロールとしてプロポフォールの基剤
（10％脂肪乳剤）を用いた。 
 電気生理学的実験：SDラット(n=38,410-510g)を用い、麻酔導入はペントバルビタール腹
腔投与（65mg/ｋｇ）で、維持はペントバルビタールの持続静脈投与(10–20 mg/kg/h)で行っ
た。腰椎椎弓切除後、脊髄を露出しタングステンマイクロ電極による細胞外記録法用いて、

脊髄後角の広作動域ニューロン(WDRニューロン)の単一ユニット記録を行った。WDRニュ
ーロンの末梢受容野にプロポフォール(1,10,25%)を塗布し、熱刺激（15秒間で34℃から
52℃）、冷刺激（15秒間で34度から0℃）に対するWDRニューロンの発火頻度を経時的に調
べた。また、綿、触、ピンチ刺激に対する反応も調べた。またAITC塗布5分後に10％プロポ
フォールを塗布し、熱刺激に対するWDRニューロン発火頻度を調べた。熱および冷刺激に
はPeltier thermode (0.5-in. diameter, NTE-2A; Physitemp) を使用し、コントロールとして
10％脂肪乳剤を用いた。 
結果 
行動学的実験：侵害性熱刺激に対するPWLは、25%プロポフォール群でコントロール群、1%
プロポフォール群と比較して有意に延長し、また25％プロポフォール群で塗布前と比較して
5分後有意に延長した。PWLはAITC塗布で有意に短縮したが、10％プロポフォール塗布
群ではAITC群、コントロール群と比較して有意に延長し、塗布前と比較して15分後のPWL
は有意に延長した。機械刺激に対する逃避反応閾値はプロポフォール塗布で変化しなか
った。 
電気生理学的実験：侵害性熱刺激に対するWDRニューロンの発火頻度は10%、25%プロポ
フォール群で、塗布前と比較して15分後に有意に減少した。コントロール群および1%プロポ
フォール群では変化しなかった。10％プロポフォール塗布による最大の発火抑制効果は塗
布後15分に認められ、30分後に基準値に戻った。AITC塗布後の10％プロポフォール塗布
はAITCによるWDRニューロン発火頻度増加作用を抑制した。冷刺激、綿、触、ピンチ刺激
に対するWDRニューロンの発火頻度はプロポフォール塗布の影響を受けなかった。 
考察：今回の研究により、プロポフォールの塗布は侵害性熱刺激に対する逃避反応を抑制
し、WDR ニューロンの侵害性熱刺激に対する発火を抑制し、AITC による侵害性熱刺激に
対する痛覚過敏を抑制することがわかった。これらの結果は末梢組織に残留したプロポフ
ォールが術後痛軽減に寄与する可能性を支持する。プロポフォール塗布の抗侵害受容、
抗痛覚過敏作用の機序は本研究では明らかではないが、末梢神経における TRPV1 もしく
は TRPA1 チャネルの脱感作や、カンナビノイド受容体や末梢のγ‐アミノ酪酸 A 受容体の
活性化の関与が考えられる。 
結論：本研究によりプロポフォール塗布は侵害性熱刺激に対する逃避反応を抑制し、WDR
ニューロンの侵害性熱刺激に対する発火を抑制し、AITC による痛覚過敏を抑制することが
明らかとなった。
