2011年東北地方太平洋沖地震では，震源から遠く離れた海岸埋立地でも液状化
による被害が確認され，復興遅延につながった．類似の被害は発生確率の高い南
海トラフ地震や首都圏直下型地震などでも予測されており，被害を最小化するた
めには事前対策が急務となっている．一方，地球温暖化は地球規模の課題であり，
土木・建築工事でも低炭素型社会の構築が求められ，工事中に排出される温室効
果ガス削減に向けた工法開発，普及が必要とされている．近年では，限りある資
源の有効利用の確保が求められるようになり，建設工事で副次的に得られる建設
発生木材の再資源化が課題となっている．  
以上の課題解決に向けた対応策の一つとして，丸太による液状化対策工法（LP-
LiC工法）が開発，実用化に至っている．今までに実施工を行い，地盤改良効果を
確認しているが，液状化強度を定量的に検証した事例はないことから経済的な設
計法の提案には至っていない．施工面では，供用期間中の生物劣化が懸念される
が，地中での木材利用を目的とした対策は見当たらない．加えて，持続可能な社
会形成のためには限りある資源の有効利用が重要とされ，解体工事で発生する建
設発生木材の再利用が課題となっている．それらの性状は解体工事前に把握でき
ない場合が多く，再利用が可能な材かどうかを現地で判断するのは困難である．  
本研究では，工法の発展および普及を図り，更なる木材の地中利用拡大を目指
すため，以下の項目に対する検討を行った．  
 
1.丸太による液状化対策工法の地盤改良効果の検証  
大型土槽を用いた大型振動実験を行い，丸太打設による地盤改良効果を検証し
た．振動実験は無対策，丸太打設，密度増大のケースを作製し4Hz，20波の正弦波
で同時加振させ，液状化による被害の程度を比較した．その結果，無対策地盤は

100Gal程度の入力加速度で沈下が生じたが，丸太を密に打設するほど改良効果が
顕著になり，丸太打設地盤は良く締固めた地盤に匹敵する小さな沈下量に収まる
ことが明らかになった．模型土槽のサウンディング試験結果では，丸太打設によ
り打設後のN値がいずれも増加し，加振時の応力 -ひずみ関係，有効応力経路では，
丸太間地盤は密な地盤の特性を示していた．これらから，丸太打設は地盤を密実
化する効果に加え，丸太間地盤が密になることにより剛性低下が生じず，沈下が
抑制されるとの結論が得られた．相対密度と液状化抵抗の関係で整理し，既存の
密度増大工法との地盤改良効果の違いを比較した結果，丸太打設した地盤の繰返
し抵抗は，密度増加させた地盤に比べ大きくなることが明らかになった．  
 
2.設置環境の異なる地中利用木材の健全性評価  
実際に土木構造物として長期間使用された木材を対象に，材種や設置期間，施
工環境，水位などの観点から地中利用木材の健全性を評価した．本研究では目視
判定とピロディン試験により劣化度の調査を行い，縦圧縮試験と縦振動法により
ヤング係数を求めた．約60年間水面下に設置された牛枠工による結果では，目視
判定結果から被害度は全体で2以下と判定され，ピロディン試験結果は貫入量の多
くが20～30mm程度の範囲内であった．また，回収丸太のヤング係数は生木とほぼ
同程度の性能を保持しており，常時水面下に設置された地中利用木材は長期間経
過後も設置環境や使用用途，樹種の違いによらず健全であることを明らかにした． 
次に，約20年前に施工された杭出し水制工を対象に，より詳細なピロディン試
験を行い，水位変動域における健全性を評価した．ピロディン試験結果から，透
水性が高い砂礫地盤の場合，地中においても生物劣化が生じることを確認した．
一方，土質の毛管現象によって地盤が高含水率状態を保持できる範囲では，常時
水面下に類似の環境条件となりうることから，木材を地中利用する際には水位と
土質の毛管現象を考慮して打設深度が検討できることを示した．  
 
3.建設発生木材の地中利用への再利用と判別法の提案  
土木・建築利用事例に基づき，末口径0.12～0.18m，矢高が末口径の 40％以下，
長さ2.0～6.0mの丸太を任意に選別し，健全性と強度性能を評価した．その結果，
目視判定の被害度が2以下，ピロディン貫入量が30mm以下では基準弾性係数と同
等のヤング係数を有し，強度面も問題がないことが確認された．縦振動法の結果
では，回収直後の高含水率丸太であっても，縦圧縮試験および縦振動法より求め
たヤング係数は概ね線形関係を示し，簡易な縦振動法により現場で回収丸太の基
本的性質を判断できる可能性が示唆された．健全性が確認され生木と同等のヤン
グ係数を示す建設発生木材は，一般的な木杭打設機の圧入力よりも大きな座屈荷
重を有することから，地中への打設が可能と判断した．建設発生木材の健全性や
強度を評価することで，再利用が可能な建設発生木材を現場で判定できる判別法
を提案した．

Mangroves are one of the most productive forests located at the upper intertidal coastline in 
tropical and subtropical latitudes. Mangroves provide essential ecosystem services such as 
sequestering large quantities of carbon, exporting organic matter to coastal waters, nursery 
habitats for coastal species, protection of coastal areas from erosion, flooding, catastrophic 
events and forest resources for coastal communities. The organic matter exported from 
mangrove forests to nearby ecosystems both inland and offshore are important food sources. In 
recent decades, mangrove forests have been suffered from land conversion for aquaculture, 
agriculture, and urbanization, causing a massive degradation of mangrove forests and loss of 
their ecosystem services. Therefore, mangrove restoration is considered as a solution to reverse 
the declining trend in mangrove forest areas. However, the ecological function of restored 
mangrove forests, particularly in ecosystem carbon (C) storage and organic carbon (OC) 
accumulation, and food sources for benthic invertebrates are still not well working. Addressing 
these gaps will be allowed for more detailed assessment of the ecological function of restored 
mangrove ecosystems and strengthened understanding of OC fluxes and budgets in the coastal 
area. It provides the scientific information on conservation and restoration of mangrove forests 
in the coastal developing countries, such as Vietnam. Thus, the study has aimed at clarifying 
the ecological function of restored mangrove ecosystems by the analysis of ecosystem OC 
storage and OC dynamics in different mangrove species (Kandelia obovata and Sonneratia 
caseolaris), different forest ages (from young to older forests), and different tidal range (from 
low to high tidal elevation) in Hau Loc mangrove forests (HLMF), Thanh Hoa Province, North-
Central Vietnam. The investigation of ecosystem C storage, OC content, sediment properties, 
and stable isotope signatures of carbon (δ13C) and nitrogen (δ15N) have been combined to solve 
the questions: (1) How do restoration activities influence on the sediment OC and ecosystem C 
storage in the restored mangrove ecosystems? (2) What is the roles of mangrove restoration in

contributing a significant proportion of OC to the sediments, and which factors control the 
accumulation of mangrove-derived organic matter in the mangrove sediments? (3) How will 
benthic invertebrate communities and their diet composition change in restored mangrove 
forests? 
The present study is composed of five chapters, of which the main findings are outlined as 
follows: 
(1) Restored mangrove forests enhance ecosystem carbon storage and sedimentary 
organic carbon: The ecosystem C storage in five different-aged restored mangrove forests and 
a mudflat in Hau Loc was estimated by measuring the above-ground biomass of trees, 
understory vegetation, downed woody debris, and the below-ground biomass of roots, OC in 
the sediment cores and overall depth. The mean above- and below-ground C stocks (mean ± 
SE) of these forests ranged from 12.7 ± 2.4 to 107.5 ± 10.7 MgC ha-1, and 103.8 ± 2.9 to 412.4 
± 6.5 MgC ha-1, respectively. The ecosystem C storage of mangrove forests tended to increase 
with forest age, varying between 201.3 ± 4.3 and 519.9 ± 12.5 MgC ha-1, with an overall mean 
of 310.3 ± 21.3 MgC ha-1 and was relatively lower than those of mangrove forests in the Mekong 
Delta, Vietnam and other mangroves worldwide. These results indicated that the oldest 
mangrove forest was potentially successful in recruitment and confirmed the efficiency of the 
restoration programs. The increase of the OC content and ecosystem C storage from the recent 
planted to the maturity stage of restored mangroves forests was a result of higher biomass 
production following forest development, including root in the young forests or both leaf litter 
and root, and OM derived from previous mangrove vegetations in the old forests. Also, the 
position of stand in the intertidal zone well influenced on litter accumulation or deposition of 
materials on the forest floor under tidal inundation and flushing, resulting in a low OC content 
in the mudflat and the young mangrove forests, and a higher OC content in the older mangrove 
forests. Therefore, mangrove restoration programs should be paid attention to as a potential 
solution for increasing ecosystem C storage and sediment OC, and as a major component of 
coastal ecosystems management and mitigation strategies in climate change. 
(2) Restored mangrove forests are predominant organic carbon source for the sediments: 
The stable carbon isotope signatures (δ13C) of core and surface sediments and organic carbon 
sources such as mangrove litter (termed as autochthonous sources) and marine phytoplankton 
(termed as allochthonous sources) were analyzed for examining sources and contribution of 
mangrove-derived organic matter to sedimentary organic carbon (SOC). The sediment δ13C 
values considerably decreased with forest age, was from −24.7 ± 0.1 to 27.4 ± 0.1‰ and from 
−25.1 ± 0.1 to −26.2 ± 0.1‰ for core and surface sediments, respectively. The sediment cores 
δ13C values decreased progressively with depth in the mudflat and the S. caseolaris forests, 
while a reverse trend was observed in the K. obovata forests. The δ13C values of POM increased 
from the upstream to the mouth of the river, which was considered as the δ13C values of marine 
phytoplankton, while δ13C values of mangrove litter were lower than previously reported for 
mangrove leaves in the Northern Vietnam. The outputs from mixing model indicated that the 
mangrove-derived organic matter contributed from 45.8 to 64.1%, and from 36.3 to 82.4% to 
the source of SOC in the core and surface sediments, respectively, and significantly increased 
following forest age. The maturity of mangroves forest supplied more production of mangrove 
roots with a rapid decomposition rate of the mangrove-derived OM at the high intertidal zone, 
and it led to more depleted in δ13C signatures in the old mangrove forests. The inundated 
conditions depending on the position of mangrove forests influenced on the accumulation of 
mangrove litter with low δ13C signatures and deposition of organic matter from marine 
phytoplankton with higher δ13C signatures on the forest floor, causing more enriched δ13C 
values in the young mangrove plantations and the mudflat. These results pointed out the 
importance of restored mangrove forests as C sources for the sediments and proved the role of 
mangrove restoration programs in C cycling in the coastal regions.

(3). Restored mangrove forests supply the food sources for macro-invertebrates: Stable 
carbon (δ13C) and nitrogen (δ15N) isotope ratios of potential food sources and benthic 
invertebrates including gastropods, crabs, bivalves, polychaetes, shrimps and mantis shrimps 
were used to identify the change in the diets of consumers in mudflat and various aged restored 
mangrove forests in the HLMF. The potential food sources for benthic invertebrates varied 
among differently aged forests, including sediment organic matter (SOM), particulate organic 
matter (POM), benthic microalgae (BMA) and marine phytoplankton (Phy); and SOM, POM, 
BMA and mangrove leaves (Man) in the mangrove forests. The isotope signature of food 
sources tended to increase from mangroves forests to mudflat, reflecting the influence of forest 
maturity and the position of forest in the intertidal zone, which related to tidal conditions. The 
crabs and gastropods communities had a broad range in δ13C and δ15N signatures among species 
and forest ages, whereas the bivalves, polychaetes, and shrimps and mantis shrimps 
communities showed a narrow range of them. The difference of actual isotope composition 
between food sources and consumers and the mixing model analysis indicated that the benthic 
invertebrates of the mudflat mainly consumed on a mixing of BMA and Phy. However, a 
combination of BMA and SOM was the primary organic source for the benthic invertebrates of 
the mangrove forests, with a higher contribution of organic sources in the old forests. The dense 
tree canopy in the old planted mangroves created more dark shades for the development of 
BMA on the forest floor, and thus led to the alternative of the diet composition of benthic 
invertebrates among the mudflat and the mangrove forests. Moreover, the difference in 
assimilated food sources of benthic invertebrate between mangroves and non-vegetated mudflat 
was driven by tidal import and export, and accumulation of SOM deposition on the forest floor, 
which associated with the location in the intertidal zone. The assimilated food sources of crabs 
in the present study were not consistent with the results of the earlier studies, and it could be 
clarified by the difference in the application of trophic discrimination values, the feeding 
practices, and the assimilation efficiency of mangrove crabs. This study is vital for assessing 
potential effects of mangrove restoration activities on local benthic invertebrate communities, 
as well as for evaluating the success of mangrove restoration programs. 
(4). Recommendation for future studies on the ecological functioning of restored 
mangrove ecosystems: This study showed the functions of restored mangrove forests in 
enhancing ecosystem C storage and sediment OC, was the predominant contribution of 
mangrove-derived organic matter to the sources of SOC and supporting food sources for benthic 
invertebrate communities. However, the decomposition of mangrove leaf litter was a major 
component in the OC dynamics, and roles of restored mangrove forests in the provision of food 
sources for fishes and nearby habitats, have not yet been addressed in the present study. 
Although the sea-level rise has positive effects on death and degradation, and changes in habitat 
conditions of coastal ecosystems, the functional role of restored mangrove forests in adaptation 
to changes in sea level has not been studied. Hence, the future studies should measure rates of 
elevation change and sediment accumulation rates in restored mangrove ecosystems for 
understanding the benefits of mangrove restoration programs in controlling sediment elevation 
to mitigation and adaptation to climate change and sea-level rise.

【背景】 
小児期の低酸素性虚血性脳症（Hypoxic-ischemic encephalopathy: HIS）に対する治療は現
在でも対症療法に限られる。HIS は全年齢で発生し、原因として心筋梗塞、心停止、窒息など、
小児期では新生児仮死、溺水、心停止などの頻度が高い。HIS の治療は、従来の呼吸循環管理
や薬物療法に加え、近年では脳低温療法が行なわれているが効果は限定的である。また自己臍
帯血幹細胞療法も試みられているが研究段階であり新たな治療が望まれる。 
脳浮腫は HIS に伴い認められ、頭蓋内圧亢進や脳ヘルニアにより障害部位以外の組織障害を
引き起こす。近年、アクアポリン 4（AQP4）が脳浮腫形成に関与するとの報告がある。しかし
小児期 HIS の病態と AQP4 の関わりや治療法についての報告は少ない。一方、五苓散が脳浮腫
に有効との基礎研究が散見され、成人の慢性硬膜下血腫に対し有効であったとする臨床研究も
あるが、HIS への効果は不明である。 
既存の HIS 動物モデルは成人と新生児期が対象で、小児期のモデルはない。そこで本研究で
は小児期 HIS 動物モデルを作成し、五苓散の有効性及び AQP4 発現に及ぼす影響を検討した。 
【方法】 
１：低酸素性虚血性脳症における五苓散の有効性 
 ヒトの小児期に相当する日齢 21 の幼弱 Wister 雄性ラット 26 匹を、五苓散群（13 匹）と対
照群（13 匹）に分け、各々に五苓散（2g/kg）と生理食塩水を投与した。薬剤投与 1 時間後に
麻酔鎮静下に頸部正中切開を行い、右側外頸動脈から右側中大脳動脈にスレッドを挿入し 60
分間の塞栓後にスレッドを抜去し右側総頸動脈永久結紮を加えた。術後 24 と 48 時間後に動物
用 MRI を用いて拡散強調画像（DWI）及び T2 強調画像（T2w）で病変面積を測定し、病変面積
比率＝（患側病巣面積－健側高信号面積）/全面積（％）を算出した。また処置後 14 日間の生
存率を比較した。

２：低酸素性虚血性脳症における五苓散がアクアポリン 4 に与える影響 
 日齢 21 の Wister 雄性ラット 18 匹を、五苓散群（6 匹）、対照群（6 匹）、sham 対照群（6
匹）に分け、五苓散群には五苓散（2g/kg）、対照群と sham 対照群には生理食塩水を投与した。
五苓散群および対照群には、薬剤投与 1 時間後に麻酔鎮静下に前述の方法で右側中大脳動脈に
60 分間の一過性虚血を負荷し、右側総頸動脈に永久結紮を加えた。術後 24 時間で動物用 MRI
にて病変形成を確認し、術後 36 時間で全身麻酔下に脳を取り出し、病変側の虚血部位と非病
変側組織における AQP4 mRNA 発現量および AQP4 蛋白質量に対する五苓散の効果を査定した。 
【結果】 
１：五苓散は低酸素性虚血性脳症の病変を縮小させ生存率を改善した 
 成熟ラットで虚血性病変を作成可能な中大脳動脈の一過性虚血負荷のみでは幼弱ラットでは
病変を全く認めず、中大脳動脈一過性虚血に同側総頸動脈に永久結紮を加えることで高率に虚
血性病変を作成することができた。 
 五苓散の低酸素性虚血性脳症への効果は、DWI では五苓散群で術後 24 時間（p=0.0042）及
び 48 時間（p=0.0072）ともに有意に対照群に比し病変面積比率が低下した。T2w でも五苓散
群で術後 24 時間（p=0.0023）及び 48 時間（p=0.038）で対照群に比し有意な病変面積比率の
低下が認められた。また術後 14 日間の生存率は、五苓散群は対照群に比して有意に改善した
（p=0.03）。 
２：五苓散は低酸素性虚血性脳症の亜急性期における AQP4 mRNA の上昇を抑制した 
 対照群病変部位の AQP4 mRNA が sham 対照群に比し有意に増加した（p<0.0001）のに対して、
五苓散群では有意な抑制が認められた（p=0.0008）が、この時期における蛋白質量に対しては
有意な効果は示さなかった。 
【考察・結論】 
 ヒト小児期相当の幼弱ラットを用いた本研究で、五苓散は AQP4 mRNA 発現抑制という新規な
効果を介して HIS に伴う脳浮腫を抑制し生存率を改善することが明らかとなった。成熟ラット
における実験で、AQP4 mRNA は、実験的 HIS 誘導後は漸進的に増加するのに対し、AQP4 蛋白質
量は誘導後急性期(術後 6～12 時間)には上昇するが、亜急性期(24～48 時間)には一旦減少し、
のち慢性期(72 時間以降)に再び発現亢進するという異なる経過を辿ることが報告されている。
本研究では五苓散は HIS に伴う脳浮腫を抑制し生存率を高めたが、これには亜急性期の AQP4 
mRNA 発現上昇抑制が伴っており、このことが慢性期の AQP4 発現亢進を阻害したことで脳浮腫
を抑制したと考えられる。AQP4 遺伝子欠失モデルでは脳虚血に伴う脳浮腫が抑制されるとの既
報はこの考察を支持する。今回の結果は、ヒト小児のみならず全年齢の HIS に対する新たな治
療法開発への発展が期待される。

The succulent Mesembryanthemum crystallinum, a facultative CAM species, exhibits 
extreme tolerance to salinity level as much as 800 mM NaCl, which is 1.5 times higher 
than that within the seawater (Adam et al. 1998). Epidermal bladder cells (EBC), which 
are single-celled trichomes on the leaf and shoot surfaces of the common ice plant, are an 
interesting component of salinity tolerance in this species. Upon the onset of salt stress, 
EBCs would swell up and could accumulate the Na+ in excess of 1 M (Adams et al. 1992, 
1998). Recently, several physiological, biochemical, or molecular investigations of the 
contributions of EBCs to plant’s salt response and adaptation to salt stress have been 
published. These include a mutational study of contribution of EBCs to salinity tolerance 
(Agarie et al. 2007), a proteomic study of protein expressed in response to salt stress 
(Cosentino et al. 2013), and an investigation of cell-type specific responses to salinity by 
EBCs (Oh et al. 2015) have implicated the important roles of EBCs in conferring the 
salinity tolerance to the plant. However, not much is known about molecular mechanism 
of EBC development in the common ice plant. 
 
Some genetic determinants of EBC development in the common ice plant were elucidated 
in our laboratory. Soejima et al. (2009) investigated the steady-state mRNA abundance of 
19 fiber development-related homologs, and eight trichome development-related 
homologs in wild-type and EBC-less mutant ice plants and found that that a 
GLABRA2-like gene (McC4HDZ) and a MYB transcription factor homolog (McMYB2) 
were preferentially expressed in wild-type plants, whereas a putative  TRIPTYCHON 
(McTRY) and CAPRICE-like gene (McCPC) were preferentially expressed in the EBC 
mutant. Yet, it  is believed that there might be novel genes that might be involved in the 
EBC development of the ice plant. Using a cDNA based-suppression subtractive 
hybridization (SSH) approach, Hoshino et al. (2004) identified genes preferentially 
expressed between the wild type and the EBC-less mutant. RT-PCR analysis of the 12 
SSH-derived clones revealed that 5 clones were differentially expressed between 
wild-type and the EBC mutant, of which two clones (WM10 and WM28) were

preferentially expressed in wild type plants, whereas the other three clones (MW3, 
MW21 and MW31) were preferentially expressed in the EBC mutants. 
 
The present study is aimed to provide the detailed analysis of the functional roles of 
those common ice plant homologs of trichome development-related genes and the 
SSH-derived clones. I have constructed binary vectors containing those genes, and 
introduced them into Arabidopsis to begin to dissect their functional roles in plant 
trichome and EBC- development.  
 
Sequence analysis revealed that McMYB2 encodes a R2R3 MYB protein and shares high 
similarity with Arabidopsis AtMYB5 and cotton GbMYB2, which are reported to be 
involved in plant trichome formation. The present study showed that the ectopic 
expression of McMYB2 in Arabidopsis thaliana (ecotype Columbia) resulted in increased 
trichome number by 20% to 40% compared to the wild type, through activation of the 
trichome development-related gene, GL2. The common ice plant McHDZ4 encodes a 
HD-ZIP IV protein and is closely related to Arabidopsis GLABRA2 (GL2), a main 
machinery in trichome formation. The results showed that the expression of McC4HDZ 
could partially complement the trichomeless phenotype of the gl2-1 mutant. Moreover, 
the overexpression of McC4HDZ in the wild-type Arabidopsis (ecotype Columbia) 
increased trichome number, in association with the upregulation of key trichome-positive 
regulators GL1 and TTG1. Furthermore, Sequence analysis indicated that a SSH-derived 
clone WM28 conserved G/AAVVY and Y/WLVAW amino acid sequences which are 
highly conserved in jasmonate-induced genes reported in several halophytes such as 
Suaeda glauca, B e t a  vu l g ar i s  su b s .  vu l ga r i s ,  A t r i p l ex  can es c en s .  Functional studies 
of the effects on trichome number of expressing a candidate gene WM28 in Arabidopsis 
were also performed. It is found that the overexpression of WM28 resulted in increased 
trichome density, acting as an activator of positive regulators of trichome development, 
GL1 and GL3, presumably through jasmonate signaling pathway. 
 
 
 
Taken together, these results showed that the common ice plant homologs of trichome 
development-related genes, McMYB2 and McC4HDZ and the SSH- derived clone WM28 
could function in trichome formation in Arabidopsis, implying that those genes are 
involved in the development of EBC in the common ice plant. Moreover, the present 
study confirmed that EBC development in the common ice plant has similarities to and 
shares molecular mechanisms with trichome development in Arabidopsis.

審査結果の要旨 
 水稲における直播栽培は就農者人口の減少と高齢化に対処するための省力・低コスト技術として期待
されている．しかし，直播栽培における課題である種子（種籾）の出芽性，直播栽培による食味の変動，
播種時期と収量や食味との関係などについては未だ十分検討されていない．そこで本研究では，今後の
乾田直播栽培に資する知見を得るために，種籾の乾燥出芽性および深播出芽性の品種間差異，乾田直播
栽培が食味に及ぼす影響，乾田直播栽培における播種時期の問題などについて解析するとともに，実際
の農家水田で乾田直播栽培を行って，生育，収量，品質，食味を慣行の移植栽培と比較した． 
 日本各地の新旧 50 品種について調査した結果，乾燥出芽性には有意な品種間差が認められ，コシヒ
カリ，あきたこまち，キヌヒカリなどの良食味品種は乾燥出芽性が高いと判定された．乾燥出芽性と出
芽後の草丈，根長との関係は弱く，出芽後の形質によって乾燥出芽性の良否を推定することは難しいと
考えられた．同じ 50 品種の深播出芽性にも有意な品種間差が認められたが，深播出芽性が優れる品種
は宝満神社赤米，亀治，愛国などの古い品種が多かった．良食味品種は，乾燥出芽性とは逆に，深播出
芽性が劣る傾向がみられた．深播出芽率は播種後日数に対して指数関数的に増加し，その関係式から求
めた深播出芽開始期および深播出芽率増加指数には有意な品種間差が認められた．深播出芽開始期と深
播出芽率増加指数との間に有意な相関関係は認められず，出芽開始の早晩とその後の出芽率増加の緩急
は独立した遺伝形質であると推測された．そこで，これら両特性と中茎長，下位節間長との関係を検討
したところ，深播出芽率増加指数は第 2 節間長および第 1 節間長と，深播出芽開始期は中茎長および第
1 節間長との関係が強く，両特性を規制する要因は異なっていた．深播出芽率には第 2 節間長が最も強
く影響し，次が第 1節間長で，中茎長の影響力はそれほど強くないと推測された． 
 日本産の 10 品種について，種籾の水分状態 (吸水籾，乾燥籾) と土壌の水分状態 (灌水量多，中，
少，無) との組み合わせが乾燥出芽性に及ぼす影響を検討した．同じ土壌水分条件であっても，吸水籾
を使うと出芽率は大きく上昇し，平均出芽日数は短縮した．また，同じ吸水籾を使っても，土壌水分条
件が厳しくなるほど出芽率は低下したが，平均出芽日数はそれほど変化しなかった．したがって，播種
前に種籾を浸漬することで，乾田直播栽培での出芽率をある程度高めることができると考えられた． 
日本および中国産の各 10 品種を供試し，乾田直播栽培した米の食味官能試験を行うとともに食味に関
わる理化学的特性を調査した．その結果，直播栽培によって食味が向上する品種と低下する品種があっ
 
 
たが，穂重型の日本の古い品種と中国産品種の多くは，乾田直播栽培で食味が低下する傾向がみられた．
移植栽培における食味特性と乾田直播栽培における食味特性との間には有意な正の相関関係が存在し，
移植栽培で理化学的食味特性が優れ，官能試験の評価が高い品種は乾田直播栽培でも同様に評価された．
しかし，官能試験の総合評価に対する理化学的特性の寄与率は栽培法によって異なり，移植栽培では最
高粘度，直播栽培ではタンパク質含有率と最高粘度の貢献が大きかった． 
 香川県の基幹品種である中生のコガネマサリ，おいでまい，ヒノヒカリを供試し，不耕起乾田直播栽
培における播種期の違いが収量，食味，品質に及ぼす影響を調査した．播種期をⅠ区（4 月 23 日播種），
Ⅱ区（5 月 7 日播種），Ⅲ区（5 月 21 日播種）として検討した結果，収量はⅢ区，Ⅱ区，Ⅰ区の順に多
い傾向を示した．食味官能試験の総合評価は，収量とは逆にⅠ区，Ⅱ区，Ⅲ区の順に高い傾向がみられ
た．玄米の外観品質ではⅡ区とⅢ区の差は小さく，Ⅰ区で大きく低下した．すなわち，Ⅲ区は収量面で
は好ましいが食味が劣り，Ⅰ区は食味面では優れるが収量が低く，品質も劣る傾向にあることが判明し
た．この原因は，播種期の違いによる登熟気温の差にあった．これらより，収量，品質，食味を総合し
てみた場合，香川県における中生品種の不耕起直播栽培の播種適期は本研究におけるⅡ期の前後（5 月
2半旬頃）と考えられた． 
 香川県で育成されたおいでまいの乾田直播栽培適性を同熟期のヒノヒカリおよびコガネマサリと比較
した．おいでまいの出芽性と耐倒伏性は，他の 2 品種並と考えられた．しかし，乾田直播栽培した場合
のおいでまいは，収量面では不安定な傾向を示した．おいでまいの外観品質と理化学的食味特性および
食味官能試験評価項目は，他の 2 品種と同程度かやや優る傾向がみられた．これらを総合して，おいで
まいの乾田直播栽培適性はヒノヒカリやコガネマサリに劣ることはないと判断された． 
 2013 年と 2015 年に農家水田においてヒノヒカリの不耕起乾田直播栽培の実証試験を行った．直播栽培
の収量は，移植栽培に比べて 2015 年は 6％の減収にとどまったが 2013 年は 37％減収した．2013 年の減
収には播種期前後の降雨不足による穂数の減少が最も強く影響していた．外観品質に処理区間差はなか
った．直播栽培米の理化学的食味特性は移植栽培米よりも最高粘度とブレークダウンが大きく，官能検
査における味が良く，粘りが強くて軟らかいために総合評価が向上する傾向がみられた．この原因は，
直播栽培の水稲は登熟性の劣る 2 次枝梗着生籾が少ないので登熟歩合と千粒重が高くなり，粒が厚く充
実が良くなるためと考えられた．これらより，ヒノヒカリを乾田直播栽培した場合，品質と食味は移植
栽培に比べて遜色はなく，食味はむしろ向上する可能性が示された．一方，収量は気象条件による苗立
ちの良否に強く影響され，移植栽培よりも不安定であることが確認された． 
 
 本論文の公開審査会は，平成 29 年 2 月 4 日に愛媛大学農学部において開催され，論文発表と質疑応
答が行なわれた．引き続き開催した学位論文審査委員会において本論文の内容について慎重に審査した
結果，審査委員全員一致して博士（農学）の学位を授与するに値するものと判定した．

The retail landscape in Indonesia is transforming from a traditional to a modern retail format in 
response to supply factors (liberalized foreign investment and improved supply chain) and demand 
factors (income growth, urbanization, urban consumption, women in the labor force, changing lifestyle, 
and convenience). Given this, policy makers are concerned about the impact of this transformation on 
consumers’ choices and small-scale farmers’ livelihood. The first objective of this study is to analyze 
the determinant factors that affect consumer choices for retail formats when purchasing fresh fruit and 
vegetables. A structured questionnaire was constructed to gather shopping preferences from 887 
households in the Jabodetabek region in West Java. Descriptive analysis, factor analysis, and a 
multinomial logit model were used to analyze the data. The findings indicate that the traditional food 
retail markets remain as essential elements in the fruit and vegetable supply chain in Indonesia. We 
found that several consumers’ motive factors such as quality, safety, price, easiness & availability, brand 
& traceability, and store attributes have become important determinants for consumers in deciding fruit 
& vegetable retail choices. Modern retail shoppers are identified as consumers who are prefer to shop 
in convenient places (store environment) and purchase products with certain brand and traceability 
attributes. Meanwhile, we also verified that the main barrier to developing modern market outlets is 
continued dominance of the traditional markets in terms of price, ease and availability, and product 
quality and safety. Therefore, our results convey a message for food retail stakeholders regarding 
developing future strategies for modern and traditional retail formats. To attract additional buyers, 
modern food retailers may need to maintain and enhance their advantage in popularity by offering clean, 
convenient, secure, practical, product origin identified, branded, and certified products, as well as other 
main attributes such as quality, variety, ease, and service. To retain a dominant market share, traditional 
outlets may need to enhance their product quality by adopting the necessary storage technologies and 
improve the outlet environment. 
Alongside with the growing of modern food retail, the global market of organic vegetables has 
grown over the last few years. This is due to increased public concerns about modern agricultural 
practices and their effect on human health as well as on the environment. The excessive use of synthetic 
chemical inputs to protect crops against weeds, pests and diseases in order to improve agricultural 
productivity has caused consumers anxiety about their potential hazards such as residues in food that 
perceived to be associated with long-term and unknown effects on health. This has encouraging the 
increasing demand of sustainable foods such as organically food produced including in Indonesia. The 
second objective of this study is to explore the factors that drive Indonesian consumer preferences in the 
purchasing of organic vegetables, and determines the factors that help explain consumer preferences in 
the purchasing of organic vegetables. Through on consumer survey, our results show that consumers 
with fewer family members and have a higher income, and are price tolerant, are more likely to purchase 
organic vegetables. Meanwhile, female consumers are less likely to buy organic vegetables. Another 
important finding is that positive attitude towards organic products, safety and health, environmental

concerns, as well as degree of trust in organic attributes, are the determinants of organic vegetable 
purchasing among consumers. Therefore, based on the study results, the following recommendations 
are needed for organic vegetable development in Indonesia: (a) implementing an appropriate pricing 
strategy; (b) encouraging organic labeling and certification for vegetables; and (c) intensively promoting 
organic food with respect to consumers’ motives and concerns on health, safety, as well as environmental 
sustainability. 
The rise of supermarkets in Indonesia have been providing further market opportunities for 
small-scale farmers, in which most of Indonesia’s farmer falls into this category. The third objective of 
this study is to examine the supermarket participation and its effect on the well-being of small-scale 
farmers. Data were collected through a household survey with 137 vegetable farmers in Cipanas-Cianjur 
Regency and Leuwiliang-Bogor Regency, West Java Province, Indonesia. By applying a treatment 
effects model that allows capturing the possibility of selection bias, we examine the factors that 
determine farmers’ participation as well as the effect on their income. The results show that there were 
several constraints faced by small-scale farmers who participated in the supermarket channel that were 
related to their level of education, irrigated land, packaging equipment, and storage space ownership, 
rather than farm size, or other household and farm assets. These findings indicated that Indonesia’s 
modern market growth could also include small-scale farmer’s participation. In addition, we also found 
that access to asphalt roads is also an important factor for participating in modern channels since it will 
affect transaction costs. Therefore, our result conveys a message for the government to provide the 
training programs needed (in production methods, quality improvement, system traceability, and 
marketing access) and to improve rural infrastructure (irrigation, roads, packing houses). Furthermore, 
the impact analysis has shown that participation in the supermarket channels generates a higher per 
capita household income than traditional channels. It implies that linking small-scale farmers to the 
supermarket channels could be a useful strategy for improving small-scale farmers’ income. 
Implementing sustainable supply chain management (SSCM) practices can be considered an 
essential strategy for fruit and vegetable supply chain actors in response to the significant rise in 
consumer concerns about health, food security, and social and environmental issues. By implementing 
SSCM practices, profits can be achieved by reducing risks and environmental impacts and at the same 
time improving economic performance and providing social benefits. Thereafter, it should lead to 
increased competitive advantages and ensure the sustainability of the inclusion process for small-scale 
fruit and vegetable farmers in modern retail channels. Hence, the fourth objective of this study is to 
provide a framework and systematic review by analyzing enablers for implementing sustainable fruit 
and vegetable supply chain management based on small-scale farmer participation in modern retail 
channels. We have identified 15 influential enablers for implementing sustainable fruit and vegetable 
supply chain management, based on a review of the literature and opinions from experts and industry 
practitioners. Next, an interpretive structural modelling (ISM) methodology was used to establish a 
structural model of the enablers, which not only helps in understanding the contextual relationship 
among the enablers but also in determining their interdependence to be able to implement sustainability 
practices. A Matrix Cross-Reference Multiplication Applied to a Classification (MICMAC) analysis 
was then conducted to determine the importance of the enablers based on their driving and dependence 
power. Several enablers, such as physical and institutional infrastructure and collaboration among 
supply chain members and stakeholders, were found to have strong driving power and were 
fundamental to implementing SSCM practices. We also found enablers that depend strongly on other 
enablers, such as product safety and quality improvement and supply chain flexibility, responsiveness, 
and efficiency. These findings offer valuable insight for supply chain actors by helping them to evaluate 
the potential for successfully implementing SSCM practices.

薬剤耐性菌問題は，2015 年の G7 サミットで，最重要課題のひとつとして首脳
宣言にも盛り込まれた世界規模の問題である．環境中に放出される抗菌物質は，
選択圧として環境微生物群集に耐性菌を生み出し，人獣医療現場から放出される
薬剤耐性菌と耐性遺伝子（antibiotic resistance genes, ARGs）は，環境微生物群集に
伝播する．すなわち，耐性菌問題は，今や臨床医学・獣医学のみならず，環境問
題としても重大である．世界保健機構では，ワンヘルスのコンセプトでこの問題
に取り組んでいる． 
    ARGs は水平伝達で細菌間へ拡散するが，これはプラスミドなどの伝達因子で
行われる．これまでは，薬剤の選択圧がなくなると，耐性菌からプラスミドが脱
落することが知られてきた．しかし，近年，薬剤汚染のない海洋環境からも多く
の ARGs が検出されており，環境中でのプラスミドに担われる ARGs の動態を知
ることは耐性菌リスク対策の為に必要な情報である． 
   本研究では，マイクロコズム実験系で，まず多剤耐性プラスミドを保有する海
洋細菌と大腸菌を，選択圧のない貧栄養環境に暴露した時のプラスミド量変化を
経時的に定量した．その結果，30 日間でプラスミドの脱落は認められず，細菌が
viable but non-culturable (VNC)状態になっても保有されていた．このことから，プ
ラスミドは海水環境でも安定して細菌群集に保持されることが明らかになった． 
   細菌は海洋環境中では原生生物に捕食され，捕食後，プラスミドも消化分解さ
れて環境から除去されると考えられるが，それを証明した例はない．本研究では，
繊毛虫２種，鞭毛虫２種を使ったマイクロコズムでの耐性菌捕食実験において，
経時的に細菌細胞内外のプラスミドを定量した．その結果，いずれの原生生物種
の場合も，細菌が捕食されるのと同調してプラスミドが環境に放出され，その後，
環境中でプラスミドは減少するものの，一定量は残存することが分かった．加え
て，捕食圧のある環境中で，プラスミドは細菌細胞中にも保持され続けることが
分かった．これらの現象は，薬剤存在下・非存在下で観察された．これらのこと
から，捕食圧のある海洋環境でも，プラスミドは環境中および細菌群集において
 
 
除去される事なく，残存することが示唆された． 
   以上の研究から，これまで知られていなかった ARGs の環境動態において，水
圏とくに海水環境では，陸圏由来耐性菌および耐性プラスミドを保有する海洋細
菌は，薬剤選択圧がない自然環境でも細菌群集に存在し続け，また耐性菌が原生
生物に捕食された場合でも，ARGs は環境に残存し続けることが明らかになった． 
   本研究は，これまでに行われていない新規なアプローチを考案して ARGs の海
水環境中での動態を定量化したものであり，海水環境が，負荷された ARGs のリ
ザーバとなり続けることを実験的に明らかにした初めての成果である． 
 
   本論文は環境微生物学，公衆衛生学の見地からみてオリジナルな研究と評価さ
れる．さらに世界的に医学・獣医学・環境学分野等で問題となっている薬剤耐性
菌リスクの解決への提言を与えうる先駆け的研究として，幅広い分野で科学的貢
献をしており，博士論文としてふさわしい内容であると考えられる． 
 
   本論文に関する公開審査会は，平成 29 年 8 月 5 日に高知大学農林海洋科学部に
おいて開催され，学位申請者の適切な説明と応答があった．さらに，その後の学
位審査委員会において学位論文の内容と，既報の学術論文等が慎重に検討された
結果，審査委員全員一致して博士（学術）の学位を授与するに値するものと判定
