【背景・目的】 
 原発性免疫不全症(PID)はヒトの生体防御機構に破綻をきたした病態であり、易感染性を示す
ほか、自己免疫疾患、悪性腫瘍、自己炎症疾患、アレルギー性疾患を合併することが知られてい
る。その多くは単一遺伝子の異常により発生し、これまでに 300 以上の疾患が報告されている。
PID 患者の予後の改善には早期診断および早期治療が不可欠であるが、比較的頻度の低い疾患で
あることに加え、臨床的な多様性も存在するため、日常診療で見逃されることも少なくない。 
 本研究では、マルチカラーフローサイトメトリーを用いて網羅的に免疫担当細胞のサブセット
解析ができるパネルを構築すること、構築したパネルを用いて健常人における年齢層別の正常値
を検討すること、および様々な PID における免疫担当細胞のフェノタイプを同定することを目的
とした。 
【対象・方法】 
 PID 患者 75 例、および健常コントロール 59 例の末梢血を用いて免疫担当細胞のサブセット解
析を行った。PID 患者の内訳は X 連鎖重症複合免疫不全症(X-SCID)7 例、うち maternal T 細胞を
持つものが 5 例、X 連鎖無ガンマグロブリン血症(XLA)18 例、X 連鎖高 IgM 症候群(X-HIGM)9 例、
毛細血管拡張性運動失調症(AT)14 例、Wiskott-Aldrich 症候群(WAS)10 例、STAT3 遺伝子機能喪
失型変異による高 IgE 症候群(HIES)9 例、STAT1 遺伝子機能獲得型変異による慢性皮膚粘膜カン
ジダ症(CMCD)8 例であった。健常コントロールは 4 群(0-1 歳、2-6 歳、7-19 歳、20 歳以上)に
分けて検討を行った。 
 末梢血より分離した末梢血単核細胞（PBMC）に対して、FITC、PE、ECD、PerCP/Cy5.5 または 
PC5、PE-Vio770 または PC7、APC、AF700、APC-Vio770、Vio Blue、および Vio Green で標識した 
氏名 髙 島 健 浩  
モノクローナル抗体を組み合わせて 10 カラー染色を行い、BD LSRFortessa™(3 レーザー 405mm/
488nm/640nm, Becton Dickinson)を用いて解析した。 
【結果】 
1)健常コントロールにおける検討 
 T 細胞、B 細胞は年齢を問わず一定の比率であったが、NK 細胞は加齢に伴い増加を認めた。 
T 細胞分画では、helper T(Th)細胞の比率が 0-1 歳群で有意に高く、加齢に伴い減少する傾向
を認めた。それに対し cytotoxic T(Tc)細胞は加齢に伴い増加する傾向を認めた。CD45RA 陽性
naïve T 細胞は加齢に伴い有意な減少を認めた。αβ T 細胞、γδ T 細胞、および Th1 細胞、Th2
細胞、Th17 細胞は年齢を問わず一定であった。iNKT 細胞、活性化 T 細胞、double negative T
(DNT)細胞は末梢血中にほとんど認めなかった。recent thymic emigrant(RTE)の比率は 20 歳
以上の群で有意に低く、制御性 T(Treg)細胞の比率は同年齢群で高値であった。濾胞性 helper
T(TFH)細胞は加齢により増加する傾向を認めた。B 細胞分画の中では、memory B 細胞の比率が
0-1 歳群で有意に低く、加齢に伴い増加する傾向を認めた。IgM memory、switched memory B
細胞は加齢により増加する傾向を認めた。transitional B 細胞は全年齢で概ね低値であった
が、0-1 歳群の一部で高値のものが存在した。樹状細胞は年齢を問わず一定の比率であった。 
2)PID 患者における検討 
 X-SCID 患者では T 細胞の著明な減少および B 細胞の増加を認めた。maternal T 細胞を持つ 
X-SCID 患者では Th 細胞の減少、Tc 細胞の増加、naïve T 細胞の減少、活性化 T 細胞の増加を
認めた。XLA 患者では B 細胞の欠損を認めたが、そのほかのサブセットでは異常を認めなかっ
た。X-HIGM 患者では Tc 細胞の増加、switched memory B 細胞および IgG 陽性 memory B 細胞の
減少を認めた。AT 患者では T 細胞の減少、NK 細胞の増加を認めたほか、T 細胞分画では Th 細
胞、naïve T 細胞、RTE の減少、γδ T 細胞、Treg 細胞の増加を認めた。B 細胞分画では memory 
B 細胞、特に IgM memory B 細胞および switched memory B 細胞の増加を認めた。WAS 患者では
健常コントロールと比較して全てのサブセットで有意差を認めなかった。HIES 患者では Th1 細
胞の増加および Th2 細胞、Th17 細胞の減少を認めたが、そのほかのサブセットでは異常を認め
なかった。CMCD 患者では HIES 患者と同様に Th17 細胞の減少を認めたほか、Th 細胞および naï
ve T 細胞の減少、DNT 細胞および TFH細胞の増加を認めた。 
【結論】 
 本法を利用することで末梢血中の免疫担当細胞を網羅的に検出することが可能となり、その
フェノタイプと臨床情報とを併せて評価することが、PID 患者を診断するうえで有用である。

         審査結果の要旨 
 
本研究は、東日本の淡水魚類についてその遺伝的集団構造の解析により明らかにし、淡水魚類の
保全に関わるデータをえようとしたものである。今回は、東日本に広く生息し、商業的価値もない
ために放流の影響がほとんどなく、在来種の遺伝的集団構造が現在も維持されていると考えられる
コイ科の淡水魚アブラハヤ（Rhynchocypris lagowskii）を用いている。本論文では、ミトコンドリア
DNA シトクロム b 領域の部分配列を遺伝標識として用い、種内各標本群の遺伝的多様性および地域
間の遺伝的関連性についての解析を行なったものである。研究成果は 2 つの章に分けられている。
1 つ目は東日本におけるアブラハヤの遺伝的集団構造を明らかにすること、2 つ目は、国内外来種
として侵入の可能性のある河川を明らかにすることである。これらの情報をえることは、アブラハ
ヤの資源管理のみならず、その他淡水魚類の遺伝的集団構造と比較することで、日本列島の淡水魚
全般の遺伝資源の管理にも活用できる重要な研究であると考えられる。 
 
（１）東日本におけるアブラハヤの遺伝的集団構造の解析 
東北から琵琶湖にかけて東日本 25 河川 29 地点より採集したアブラハヤ、221 個体についてミト
コンドリア DNA シトクロム b 領域・部分配列 460bp の解析を行なった。基本 1 河川 1 地点である
が、最上川については支流 3 地点、阿武隈川と利根川についてはそれぞれ支流 2 地点の解析を行な
った。えられたハプロタイプは全体で 67 タイプであった。それらの出現ハプロタイプは、大きく 3
つのグループに分けられた。グループ 1 にはハプロタイプ 1～36 が、グループ 2 にはハプロタイプ
37～40 が、グループ 3 にはハプロタイプ 41～67 が含まれた。これら 3 つのグループは近縁種のタ
カハヤ（Rhynchocypris oxycephalus）明らかに異なっていた。各標本群においてグループ 1～3 に属
するハプロタイプの出現頻度には違いがみられた。天野川標本群(琵琶湖)はグループ 3 のみの集団
であり、また、太平洋側に流入する天竜川、狩野川、酒匂川についてもグループ 3 のみで形成され
ていた。さらに、同じく太平洋側に流入する利根川水系烏川(群馬県)、荒川、鮫川(福島県)につい
ても高頻度でグループ 3 のハプロタイプが出現した。グループ 2 のハプロタイプはほぼ北陸地方の
 
 
2 河川に限定して出現した。一方、日本海側への流入河川ではグループ 3 に属するハプロタイプが
混在する河川がいくつかあったものの主要なハプロタイプはグループ 1 であった。また、太平洋側
に流入する河川のうち那珂川(茨城県)より北に位置する久慈川、七北田川(宮城県)はグループ 1 の
みであった。ただし、例外的に、那珂川よりも南に河口が形成されている利根川において、同水系
鬼怒川(栃木県)についてはグループ 1 のみとなった。このようにグループ 1～3 のハプロタイプの
分布は地理的な位置関係とおおよそ一致しており、東日本のアブラハヤ集団は大きく 3 つのグルー
プに分けられることが示唆された。ハプロタイプ多様度、塩基多様度の平均値は太平洋側 15 標本
群でそれぞれ 0.9295±0.0100、0.020183±0.010360 であったのに対し、日本海側 14 標本群では
0.7838±0.0339、0.008505±0.004747 となり、太平洋側で高い値を示した。それぞれのグループのハ
プロタイプ多様度、塩基多様度の平均値は、グループ 3 でそれぞれ 0.8720±0.0282、0.012231±
0.006596 となりグループ 1、グループ 2 に比べて高い値を示した。 
なお、29 標本群のうち太平洋側に流入する青森県・岩手県の 2 河川(馬渕川、北上川)はグループ
1 のハプロタイプのみが出現し、那珂川以北はグループ 3 が出現するという地理的な分布の予想か
らは外れていた。これらの 2 河川の標本群については在来集団ではない可能性が示唆された。岩手
県三陸沿岸にはもともとはアブラハヤが生息していないとする記録がある。今回の結果は、岩手県
以北の 2 河川については、アブラハヤの空白地帯に侵入・定着した国内外来種の可能性のあること
を示唆していた。なお、北上川標本群については天野川標本群(琵琶湖)と共通のハプロタイプも検
出されており、このことから判断すれば、琵琶湖よりアユの種苗放流等を介して侵入・定着した非
在来集団である可能性が高いと考えられた。 
 
（２）東北地方における非在来ハプロタイプの出現 
  地理的な位置関係から判断すればグループ 1 に属するハプロタイプが主となると考えられる地域
の標本群について、ここでは新たに日本海側の最上川の 9 支流、赤川 3 支流の計 12 標本群につい
てミトコンドリア DNA シトクロム b 領域・部分配列 429bp の解析を行なった。最上川水系におい
ては 9 標本群中 2 標本群、赤川水系において 3 標本群中 1 標本群においてグループ 3 が高頻度で出
現し、グループ 3 の混合率は最上川水系の押切川、丹生川でそれぞれ 50％、40％、赤川水系の八沢
川で 42.8％であった。これらのハプロタイプについても天野川標本群(琵琶湖)と共通のハプロタイ
プが検出されており、琵琶湖よりアユの種苗放流などを介して侵入した個体である可能性が高いと
考えられた。 
 
  以上のように、本論文は東日本におけるアブラハヤの遺伝的集団構造を明らかにし、今後の遺伝
資源としての活用において新しくかつ重要な知見を見出している。学術的にも実用的にもきわめて
質の高い研究で、博士論文として評価できるものと考える。 
 
本論文における公開審査会は、平成 29 年 8 月 5 日に高知大学農林海洋科学部において開催され、
学位論文の口頭発表と質疑応答が行われた。引き続いて行われた学位論文審査委員会で本論文の内
容を慎重に審査した結果、審査委員全員一致して博士（農学）の学位を授与するに値するものと判
定した。

         審査結果の要旨 
 肥満および高脂血症は、高血圧や糖尿病等との因果関係も指摘され、ともに様々な健康問題の重要な
危険因子と認識されている。重篤な場合には医療処置に頼らざるを得ないが、運動や健康的な食事によ
る予防が望まれている。食事成分によるこれらの疾病予防の研究も多数なされており、食物繊維や植物
ステロール、ポリフェノールが有効である可能性が報告されている。こうした中、大豆の摂取が肥満お
よび高脂血症の予防に有効である可能性が多くの研究により指摘されている。大豆の多くの成分の中で、
これらの疾病予防に寄与する可能性のあるものとして大豆タンパク質とともに大豆イソフラボンが挙げ
られている。大豆イソフラボンはポリフェノールの一種で、主にダイゼインおよびゲニステインからな
る。抗酸化性、エストロゲン性および抗エストロゲン性を持つことが知られている。抗肥満については
ほとんど知見が無く、抗高脂血症については大豆タンパク質との相互作用による効果が検討されていた
が、先に大豆イソフラボンを高含有する発酵脱脂大豆エタノール抽出物（FSBE）が、雌ラット特異的に
抗肥満効果、コレステロール低下効果を持つことが見出された。FSBE は大豆タンパク質を含まないが、
大豆イソフラボン以外の成分も多数含んでおり、大豆イソフラボンも主にダイゼインとゲニステインの
混合物である。これについて純粋な大豆イソフラボン・ダイゼインおよびゲニステインを用い、抗肥満
効果およびコレステロール低下効果を検証し、その作用機構を探った。FSBE 摂取時には血中にダイゼ
イン及びゲニステインは高濃度に存在せず、ダイゼインの腸内細菌代謝物エコールのみが高濃度で存在
したため、特にダイゼインに着目して検討を行った。 
 まず抗肥満効果について、コレシストキニン（CCK）感受性欠損またはレプチン感受性欠損により過
食性の肥満を引き起こす OLETF ラットおよび Zucker fa/fa ラットの雌を用いた。両ラット種とも卵巣摘
出術または偽手術を施したものそれぞれについて、ダイゼインの混餌投与の影響を調べた。また同様の
実験をそれぞれのラット種の原生種である LETO ラットおよび Zucker +/+ラットにおいても行った。
OLETF ラットについては、LETO と同様にダイゼインの摂取による有意な飼料摂取量低下および体重増
 
 
加抑制が認められた。卵巣摘出するとその効果は減弱したが、同様の傾向は見られた。Zucker fa/fa ラッ
トにおいては、卵巣摘出を行った場合のみダイゼインの効果が認められた。血清エコール濃度は OLETF
および LETO ラットでは、多くの実験で用いられており我々も FSBE の実験で用いた SD ラットに比べ
て低いレベルであり、Zucker fa/fa および+/+ラットではさらに低いレベルとなった。また、卵巣摘出
を施すことによりさらに低いレベルとなった。以上よりダイゼインが雌ラットの飼料摂取量を低下させ
体重増加を抑制していることが明らかになり、さらにこの効果に CCK およびレプチンによる食欲抑制
シグナルは関与していないことが明らかになった。 
 飼料摂取量低下の作用機構を検討するために、ダイゼイン摂取により雌ラットの飼料摂取量が低下し
ている際の食欲制御因子の変動をプロファイリングした。摂取エネルギーの過不足を感知した消化管や
脂肪組織がホルモンや神経系を介して視床下部にその情報を伝え、食行動が抑制又は促進されることに
よりエネルギー恒常性が保たれている。これについて視床下部、消化管の食欲制御因子の遺伝子発現を
調べた。ラットは通常自由摂取下で飼育され、その摂食タイミングには大きな個体差がある一方先のエ
ネルギー恒常性を維持する機構は飼料摂取に対し刹那的に応答しており、自由摂取下でプロファイリン
グすることは難しい。そこで摂食タイミングの同期を図り 1 日 3 回、一定時刻にそれぞれ 1 時間だけ自
由摂取させる 3 食制を導入し、食事前後の各種遺伝子発現をプロファイリングした。ラットは SD ラッ
トを用いた。ダイゼイン摂取による飼料摂取量の低下は 2 食目においてのみ見られ、この際視床下部に
おいては食欲促進因子であるニューロペプチド Y 遺伝子発現の食後の上昇をダイゼインが抑制し、同じ
く食欲促進因子であるガラニンの遺伝子発現にも同様の傾向が見られた。食欲抑制因子であるコーチコ
トロピン放出ホルモンの遺伝子発現はダイゼイン摂取により食後に有意に上昇した。小腸粘膜の CCK
遺伝子発現は食前に有意に上昇し、食後に上昇傾向を示した。どのタイミングにおいても血清エコール
濃度は非常に高かったが、ダイゼインはほとんど検出されなかった。以上よりダイゼインがエコールに
代謝されて視床下部の食欲制御因子を変更することにより飼料摂取量を低下させることが明らかになっ
た。小腸粘膜の CCK については先の OLETF ラットの実験で CCK シグナルの関与が否定されているた
め、作用に関与している可能性は低く、ダイゼインもしくはエコールがいかにして視床下部遺伝子発現
を変更しているかはまだわからない。 
 コレステロール低下効果については雌の SD ラットをそのまま、または卵巣摘出術を施して用いた。
このラットにダイゼイン、ゲニステイン又はこの両方を混餌投与し 4 週間飼育し血清コレステロール濃
度低下を観察した。ダイゼインはゲニステインとの併投与とも、卵巣摘出術の施術の有無とも独立して
血清コレステロール濃度を有意に低下させ、肝臓の遊離コレステロール含量も同様に低下させた。また
糞中胆汁酸排泄も有意に増加させた。糞中胆汁酸はコレステロールの最大の排泄経路でありその排泄増
加はコレステロール低下作用と密接に関与していると考えられており、作用機構を説明しうる。しかし
肝臓の遺伝子発現解析ではコレステロールより胆汁酸を合成する経路のいずれの酵素の遺伝子発現も変
化しておらず、実際にコレステロールが消費されているかは断定できなかった。この他肝臓のアポリポ
プロテイン B（ApoB）遺伝子発現がダイゼインの摂取により有意に増加した。ApoB は超低密度リポタ
ンパク質（VLDL）の構成要素であり、VLDL の分泌は血中コレステロール濃度を制御する重要な要素
である。しかし、肝臓のコレステロール含量の低下を反映した結果である可能性も否定できない。以上
よりコレステロール低下作用をもたらしているのはダイゼインであり、ゲニステインにはその効果もダ
イゼインに対する相互作用も無いことが明らかになった。作用機構は解明に至っていないものの、胆汁
酸排泄促進による機構や、VLDL 分泌の低下による機構が示唆された。 
 以上一連の結果から大豆イソフラボン・ダイゼインが腸内細菌によりエコールに代謝され、抗肥満効
果およびコレステロール低下作用をもたらすことが明らかになり、両効果の作用機構の一端も明らかに
された。さらなる検討により作用機構が明示されればヒトの健康栄養への利用も期待される。 
学位論文の公開審査会は 2017 年 8 月 5 日高知大学農林海洋科学部で実施され、口頭発表と質疑応答
が行なわれた。続いて学位論文審査委員会を開催して本論文の内容を審査した。その結果、全員一致し
て本論文が博士（農学）の学位を授与するに値するものと判定した。

Chloroplasts harboring green coloured chlorophyll pigment are the vital cell organelles and are 
prevalent in photosynthetic eukaryotes. Apart from their characteristic function, in which it converts 
light energy into chemical energy through the process of photosynthesis, it is also a home for many 
other important metabolic processes. Most of the chloroplastic genes were transferred to the nuclear 
genome during the course of evolution; under these circumstances, where more than 95 % of the 
chloroplastic proteins were synthesized on the cytosolic ribosomes, a proper targeting mechanism to 
chloroplasts as well as translocation machinery has been evolved in the photosynthetic cells. Much 
knowledge regarding the entire process of protein import into chloroplasts is still unclear and need to 
be explored soon by considering the importance of chloroplast on which whole life on earth depends. 
Most of the nuclear encoded chloroplastic proteins are synthesized as precursors in the cytosol 
with their N-terminal transit peptide and are imported into the chloroplasts through protein 
translocation machinery (translocons) embedded in the outer and inner envelope membranes (Figure 1). 
Protein import process of these proteins is mainly divided into two stages, “docking” and 
“translocation”, depending on an energy requirement in vitro (Figure 2). In translocation, precursor 
proteins fully translocate through the double membrane to reach to the interior of chloroplasts, in the 
presence of a high level of ATP (> 1 mM) at a higher temperature (Figure 2). In contrast, in docking,

precursor proteins irreversibly bind to the chloroplastic surface to form early protein translocation 
intermediates (PTIs) under limited energy conditions (Figure 2). Precursor proteins are expected to 
interact sequentially with the translocon components, however regardless of how PTIs were formed 
during the early stage of protein import under different conditions, addition of a high level of ATP at a 
higher temperature freed precursors from early PTIs, which results in the completion of translocation. 
Our current knowledge of chloroplastic translocon is based mainly on the biochemical analyses of the 
early PTIs formed under docking conditions. However, once precursors depart from the early PTIs, it is 
impossible to pause the movement of precursors until translocation is completed. Therefore, molecular 
mechanisms of post-docking stage have not been well-understood. If we are able to manage the 
movement of translocating precursor by trapping precursors carrying steric hindrance at their carboxyl 
termini in the translocon to form PTIs under translocation conditions, then we may find a way to solve 
this subject (Figure 3).

Mitochondria also has double-membrane system as well as chloroplasts, many mitochondrial 
proteins are also nuclear-encoded, so that these proteins should be imported into mitochondria. In case 
of mitochondrial protein import, nuclear encoded mitochondrial proteins are synthesized on the 
cytosolic ribosomes as precursors and are imported into mitochondria through multi subunit protein 
complexes located in the outer and the folded inner membrane by using the driving force of an 
electrochemical gradient present across the inner membrane. In mitochondria, the precursor protein 
fused to mouse dihydrofolate reductase (DHFR) has blocked import once DHFR structure was 
stabilized in the presence of methotrexate (MTX) (Eilers and Schatz, 1986). By analyzing the trapped 
precursor, progress of research on mitochondrial protein import was dramatically enhanced. To this end, 
two strategies which could be employed for blocking the process of protein import into chloroplasts 
mainly includes, developing a precursor protein which is fused to the tightly folded domain and post 
translational attachment of the folded structure to the precursor protein. 
Firstly, we have employed a precursor which is fused to tightly folded domain. The similar 
kind of strategy has already been applied on chloroplastic protein import. However, protein import into 
chloroplasts was not blocked by the tightly folded precursors fused with mouse cytosolic DHFR in the 
presence of MTX (America et al, 1994; Endo et al, 1994). In the current investigation, we made an 
attempt to reevaluate whether DHFR—MTX interaction did not significantly affect protein import into 
chloroplasts. Precursors fused to DHFR from mouse reported previously was cell-free synthesized 
incorporating radio-active amino acids without purification, indicating that the amount of precursors 
were limited and synthesized precursors were contaminated with various cellular factors and small 
molecules, which might affect blockage of protein import. By applying recombinant precursors 
expressed in E. coli cells, we had developed in vitro chloroplastic protein import assay system (Inoue et

al, 2008). Important features of this assay system are: it can handle the large amount of precursor, 
import reaction is analyzed without exogenous factors, thus manageable by strict energy conditions. 
But all of E. coli overexpressed precursors prepared so far, were recovered in the inclusion bodies and 
had to be solubilized in 8 M urea prior to the import reaction. However, denaturation step should be 
avoided, if the precursor with a tightly folded domain was applied for plugging translocation channel. 
Therefore, we designed a precursor protein which was assembled with soluble components. Since 
DHFR is tightly folded in the presence of MTX, in addition, to avoid the use of a gene from 
heterologous origin, DHFR from E. coli (EcDHFR) was selected to be a major component of the 
mature part of the precursor. For a detection and purification purpose, human influenza hemagglutinin 
protein (HA) epitope tag, biotin acceptor peptide (BAP), and histidine affinity tag (HAT) were chosen. 
HAT, instead of hexahistidine (H6) tag, was picked because it was considered to be more soluble than 
artificial H6 tag. In addition, to extend the distance between the transit peptide and EcDHFR, one unit 
of the random coil linker (r1) consisted with the quintuple penta-peptides of GGGGS ((G4S)5) was 
inserted to complete the precursor construction. Our newly prepared precursor protein was recovered 
in a soluble form in E. coli cells after overexpression and has retained chloroplast import competency 
(Pohare and Akita, 2016). DHFR portion of this precursor was tightly folded in the presence of MTX 
and import of MTX bound precursor was inhibited, though not completely. In addition, after 
fractionation of chloroplasts, processed precursor protein was recovered in the soluble fraction (Akita 
and Pohare, 2016). These results indicate the presence of unfolding activity within chloroplasts, which 
allow EcDHFR-fused precursor to be translocated through envelope membranes. 
Secondly, for the post translational attachment of the folded structure, biotin-streptavidin (SA) 
technology was utilized. Interaction between biotin and SA is known to be one of the strongest

non-covalent interactions in nature. SA is a homo-tetramer, thus binds up to four biotin molecules, that 
more than one biotinylated precursor may be bound to SA. Howarth et al (2006) developed 
monovalent SA consisted of one subunit of wild-type and three mutant subunits deficient in biotin 
binding. Unlike monomeric SA, monovalent SA retains the same level of biotin affinity and 
thermostability as the wild-type SA. Monovalent SA was associated with the precursor protein 
carrying BAP at its carboxyl terminal overexpressed in Escherichia coli. When this 
precursor-monovalent SA complex was applied for in vitro chloroplastic import assay, translocation of 
precursor was not blocked. Further, we developed a more stable form of monovalent SA; cross-linked 
monovalent traptavidin which has 10-fold slower biotin dissociation as compared to the monovalent 
SA and has enhanced stability due to insertion of a covalent cross-link across dimer-dimer interface. 
Although the import of biotinylated precursors was inhibited in the beginning of the reaction, but even 
cross-linked monovalent traptavidin does not prevent the biotinylated precursors from being imported 
into chloroplasts. In this case too, the processed precursor protein was recovered in the soluble fraction 
after fractionation of chloroplasts and remained protected from the thermolysin digestion. This 
suggested the presence of strong denaturing activity at the chloroplastic surface or the strong pulling 
force enough to dissociate monovalent SA or cross-linked monovalent traptavidin from biotinylated 
precursor. If in the latter case, apart from the interest in protein targeting mechanism, to investigate 
such strong motor, if presents, is valuable for innovating a biological nanomotor. 
During the course of precursor preparations, gene manipulation was essential. When we 
employed gene manipulation, we found a rapid and simple, recombination based cloning method in E. 
coli. Various types of enzyme kits serving the purpose of cloning have already been commercialized 
and are available from several sources. When cloning with such kits, control experiments in which the

enzyme mixture was omitted often also produced transformants that carried the desired recombinant 
plasmids. Such an undesirable outcome served as an impetus for designing a cloning technique. Here, 
we report a time-saving, cost-effective, and simple cloning technique that was serendipitously 
discovered. After optimizing the experimental conditions, such as the molar ratio of two fragments and 
the length of overlaps between the ends of two fragments, we found that the desired transformants may 
be obtained when a mixture of two DNA fragments containing at least 12 bp of identical nucleotide 
sequences at their terminal ends is introduced into E. coli cells (Pohare and Akita, 2017). To 
mechanistically explain our cloning procedure, double-stranded DNA specific exonucleases, such as 
RecE, are present in E. coli cells, once a vector and an insert fragment are introduced in cells after heat 
shock treatment, single-stranded DNA overhangs will be created at both ends of the fragments. Single 
stranded overhangs favor annealing of these fragments in vivo and the gap is filled in by a polymerase, 
followed by being sealed by a ligase. Thus, we propose a mechanism which operates in vivo and 
functions similarly to that of the SLiCE method (Zhang et al, 2012). Because this technique is based on 
homologous recombination in vivo, cloning is independent of restriction endonuclease recognition sites 
at the cloning site, and a gene of interest can thus be cloned into plasmid at the desired site without 
additional nucleotide sequences. Therefore, most of the time-consuming steps required in classical 
cloning protocols are avoided.

小倉和幸氏が提出した学位論文“THEEVOLUTION OF GALAXIES AND THE LARGE-SCALE 
STRUCTURE IN THE EARLY UNIVERSE TRACED BY DAMPED LYαABSORPTION SYSTEMS"の公
聴会を、平成 29年 8月 21日午後 2時から午後 3時半まで、審査委員全員の出席の下で
理学部会議室において行った。また同日午後 3時半から午後 4時まで同室において審査
委員会を行ったので、その結果を報告する。
暗黒物質が支配する宇宙大規模構造の進化が詳細に理解されるようになった今なお、
この宇宙大規模構造の中で銀河がどのよ うに形成進化を遂げてきたかという問題は未
解明である。その大きな理由の一つは、バリオン物質がガスから星に転化していく過程
が高度に非線形な現象であるためであり、このため観測的研究により現象論的な理解を
積み上げていくことが求められている。銀河進化の初期段階を研究するには、進化の初
期段階にある天体種族に注目した観測が必要だが、この観点でとりわけ重要な天体種族
として DampedLyα吸収線系 (DLA)とライマンアルファ輝線天体(LAE)が挙げられる。 DLA
は明るい背景光源の手前に存在するガス塊が背景光源のスベクトノレ中に影のように吸
収線を示すことで認識される天体であり、その正体としてガスが豊富で進化の初期段階
にある銀河だという可能性が指摘されている。また LAEは強し、 Lyα輝線放射で特徴付
けられる銀河であり、星質量が小さく比星形成率が高いためやはり若い段階にある銀河
だと考えられている。ただしこれまでの観測的研究では、こうしたいくつかの天体種族
のそれぞれに着目して統計的性質の調査などが行われているため、銀河の形成進化の初
期に関する統一的措像の理解には至っていないのが現状である。
この状況を打破するため、小倉氏は本研究において DLAとLAEの相互関係に着目した。
これまでこの両者の相互関係が調査されてこなかった主な理由は、 DLAを系統的に調査
するために利用可能な背景光源が少なかったためである。また、 LAEは一般に極めて暗
く、十分な集光力を持つ望遠鏡を使わなければ調査できない。更に、両種族の天体とも
に数密度が低く、十分な体積に対する観測が必要な点も、系統的調査を阻んできた理由
として挙げられる。しかし最近になり、米国のバリオン音響振動分光サーベイの進展に
よって DLAの背景光源としてのクェーサーが多数発見され、 DLAの系統的探査が可能に
なってきた。小倉氏はこの好機を捉え、広視野かっ高感度を誇る国立天文台すばる望遠
鏡の主焦点撮像装置である Suprime-Camを用いることで DLAとLAEの相互関係を調査す
るとしづ着想に至った。小倉氏は自ら研究代表者として観測提案を行い、高い競争率の
審査を勝ち抜いですばる望遠鏡の観測時間を獲得した。ここで、 DLAが空間的に密集し
た領域にとりわけ注目して観測を行うというところに高い独自性が認められる。また、
LAEの空間分布や数密度・光度関数などの統計量をそういった領域において導出するこ
とにより DLAと LAEの相互関係を調査するという方法は、本研究において特に高い新規
性がある部分だと評価できる。
小倉氏自身によるデータ解析の結果、 LAEの空間分布や数密度・光度関数といった統
計量は、一般領域における統計量と無矛盾であることが分かり、大局的には必ずしも
DLAとLAEの聞に強い相関が見られる訳ではないことが今回の研究により初めて明らか
になった。更に、個々の DLAの周囲に注目すると、視野内で最も水素柱密度が大きな
DLAの周囲では LAEが局所的に密集している傾向が見られた。これらの事実から、 DLA
と LAEの聞には本来は空間的な相関があるが特に局所的に相関が強い領域以外では中
性水素による共鳴散乱の影響で LAEが観測されにくくなっている可能性など、具体的な
し、くつかのシナリオを提示した。更に提示したシナリオの検証方法についても示すこと
により、当該分野の今後の方向性に対しても示唆を与えた。
以上のように小倉氏は優れた研究結果を博士論文にまとめて査読付英文論文として
出版し、また多くの国際会議においても成果発表を行うことで当該分野の発展に資する
活動を行ってきたととは高く評価できる。公聴会では、研究の背景を分かりやすく説明
した上で、得られた研究成果およびそれに基づく議論と考察について論理的に説明した。
また、その後の質疑応答においても適切な回答を行い、小倉氏が研究内容に対して深く
理解していることが確認された。また、提出された論文が適切な形式・構成されていて、
研究倫理上の問題がないことも確認された。したがって、申請論文は博士(理学)の学
位論文にふさわしいと判断し、審査委員全員一致の意見で合格とした。
