蛍光性ナノ粒子は各種センサーや発光ダイオード、バイオイメージングなど幅
広い応用が期待される機能性発光材料である。なかでも有機化合物を材料として
用いた蛍光性有機ナノ粒子は材料化合物の分子設計の自由度の高さと多様な機能
性から近年注目されている。ナノ粒子の蛍光特性は孤立分子やバルク固体とは異
なることが知られており、さらに比表面積が大きいことから粒子表面がナノ粒子
の蛍光に強く影響を及ぼすものと考えられる。分子パッキングや粒子サイズ、粒
子表面環境に依存したナノ粒子の蛍光特性の解明は、機能性材料としての応用の
ために重要となる。本論文では代表的な蛍光色素であるペリレンジイミド誘導体
およびテリレンジイミド誘導体のナノ粒子を作製し、時間分解分光と単一粒子分
光の手法を用いた実験により上述のナノ粒子特有の蛍光特性を詳細に調べた研究
成果をまとめたものである。  
第 1 章は緒言であり、本研究の背景と目的についてまとめた。  
第 2 章では再沈殿法と液中レーザーアブレーション法によるペリレンジイミド
誘導体（DBPI、DTPDI、PR、DPDI）とテリレンジイミド誘導体（TDI）のナノ
粒子コロイド作製についてまとめた。蛍光色素の種類、作製手法および作製条件
により粒子サイズは約 40 から数 100 nm の範囲で異なり、また、その形状も球形
やロッド状になることが分かった。コロイド分散安定性や吸収・蛍光スペクトル
も異なることを明らかにした。  
第 3 章では DBPI と PR について、ナノ粒子の励起状態緩和ダイナミクスを単
一粒子分光と時間分解分光を用いて調べ、分子構造とナノ粒子中での分子パッキ
ングの関係について考察した。ペリレン骨格サイドベイに置換基をもつ PR では固
体中での分子間の電子的相互作用が弱く、サイズの小さなナノ粒子では表面蛍光
消光が顕著であることが示唆された。一方、DBPI では固体中で分子間の電子状態
相互作用が強く、光励起エネルギーはエキシマー（励起二量体）に捕捉される。
分子のパッキングの違いに起因したナノ粒子とバルク結晶でのエキシマーの生成
緩和ダイナミクスが観測された。DBPI アモルファスナノ粒子に有機溶媒を接触さ
せることで結晶性をもつ粒子へと相転移することを見出した。  
第 4 章では再沈殿法と液中レーザーアブレーション法の 2 つの異なる方法によ
り DBPI、TDI、DTPDI ナノ粒子を作製し蛍光特性を比較した。DBPI と TDI で
はナノ粒子中での分子パッキングが作製手法によって違うことが示された。一方
長鎖アルキル基を持つ DTPDI では、どちらの作製手法でも DTPDI 分子が一次元
的に π-π スタッキングしたカラム構造をもつナノ粒子が生成した。しかし、ナノ粒
子の形状は作製手法で大きく異なり、再沈殿法ではナノファイバー凝集体、レー
ザーアブレーション法では溶媒に安定に分散したナノブロックが得られた。  
第 5 章ではナノ粒子表面が与える蛍光特性への効果について検討した。粒子表
面での励起エネルギー移動または電子移動によりナノ粒子全体の蛍光が強く消光
され、粒子内での効率の良い励起エネルギーマイグレーションが示された。特に
PRと DBPIナノ粒子では表面蛍光消光を定量的に調べることにより消光メカニズ
ムを詳細に検討した。実験から見積もった蛍光反応消光速度を Förster 機構による
分子間エネルギー移動速度の見積もりと比較し粒子内でのエネルギーマイグレー
ション機構を考察した。  
第 6 章ではベンゾチアジアゾールにジアリールエテンを添加したナノ粒子を作
製し、ジアリールエテンのフォトクロミック反応による蛍光の ON/OFF スイッチ
ングを調べた。蛍光分子に対してジアリールエテン着色体の生成量が 1％で 90％
以上の蛍光が消光する、非線形蛍光消光が可逆的に起こることを混合ナノ粒子系
において初めて示した。蛍光消光挙動はジアリールエテンの添加量には依存しな
いが添加するジアリールエテンの種類によって蛍光消光挙動が異なることが明ら
かにした。これらの機構を Förster 型励起エネルギー移動消光に基づいて考察した。 
本論文における研究の成果を第 7 章で総括し、展望について述べた。

【はじめに】 
アルツハイマー型認知症(以下 AD)は、老人斑や神経原線維変化などの病理学上の変化が起こ
ることが発症に関わる器質性精神障害であるが、AD の病態に炎症が関わることが最近、注目さ
れている。これまで、私たちは、AD 患者の末梢血白血球での炎症に関わる TREM2 や ABCA7 など
の mRNA 発現や DNA メチル化率が変化していることを報告し、これらの末梢血遺伝子変化がバイ
オマーカーとなる可能性を示してきていた。Monocyte enhancer factor 2C (MEF2C)は、抗原が
B 細胞レセプターに結合したおりに起こる B 細胞分化に関与する炎症時の機能性タンパク質であ
る。MEF2C は、全ゲノム関連解析（GWAS）からも AD の発症に関連することが報告されている。
加えて、MEF2C は前頭葉皮質、海馬傍回、歯状回、偏桃体といった学習や記憶に関連した脳部位
に多く発現している。以上から、脳での MEF2C 遺伝子発現変化は AD の病理に関係していると予
想される。私は、脳での MEF2C の変化が AD の病理に関係しているのであれば、末梢血白血球の
変化も起こっていると予想され、AD の診断マーカーとして有用となる可能性があると考え、今
回の研究を発想し、実行した。 
【方法】 
 まず、AD 群では、Aging/Alzheimer's Association criteria を用い診断した後に、認知機能
検査の mini mental state examination (MMSE)や Alzheimer’s disease assessment scale (AD
AS)、精神機能を Neuropsychiatric inventory (NPI)、重症度を Clinical dementia rating (CD
R)にて評価した。対象者の末梢血を採取し、白血球分画から gDNA を抽出し、cDNA を作成した
（年齢、性別を一致させた AD 患者群と健常対照群各 50 例）。AD 患者群は塩酸ドネペジル投与
群と非投与群に分け、それぞれ 25 例とした。cDNA を用いて白血球 MEF2C mRNA 発現量を Taqman 
probe 法にて計測し、gDNA を用いて、その遺伝子発現に影響を与える MEF2C プロモーター領域に
ある 3 つの CpG サイトにおけるメチル化率をパイロシークエンス法にて計測し解析した。また、
AD の発症に影響する APOE の 2SNPs（rs429358 および rs7412cDNA）の関与を調べた。統計解析に
は SPSS を用いた。これらの研究は全て当院の倫理委員会の承認を得ている。 
【結果】 
 MEF2C mRNA 発現量は健常対照群と比較し AD 患者群で有意に低下していた（p = 0.007, Z=-
2.709, effect size=-0.271）。しかし、プロモーター領域の 3 つの CpG サイトの DNA メチル
化率は AD 患者群と健常対照群との間で有意差を認めなかった。

MEF2C mRNA 発現量は、MMSE 総得点、MMSE 想起得点、ADAS 総得点との間に有意な相関を認め、ME
F2C の末梢血遺伝子発現量とこれらの認知機能に有意な相関があることが示唆された（いずれも
p<0.05）。しかし、MEF2C mRNA 発現量は、年齢、性別、発症からの年数、塩酸ドネペジル投薬
の有無、NPI および CDR 得点には相関を認めなかった。APOE のε遺伝子型において、AD 患者群
は健常対照群と比べ有意にε4 アリルを多く持っており、既報と一致した(OR=7.2, 95%CI: 2.71
-19.09, Cramer’s V=0.172, p < 0.001)が、MEF2C mRNA 発現量との間に関連は認めなかった。
以上より、MEF2C mRNA 発現量は、独立して AD の発症や認知機能低下に関連していることが示唆
された。 
【考察及びまとめ】 
MEF2C mRNA 発現量は健常対照群と比較し AD 患者群で有意に低下していた。そして、MEF2C mR
NA 発現量と認知機能との間にも相関が認められた。一方、プロモーター領域の CpG サイトにお
ける DNA メチル化率は、AD 患者群と健常対照群との間で有意差を認めず、この発現量変化に影
響を与えていることは確認できなかった。AD 患者群と健常対照群との間で DNA メチル化率に有
意差を認めなかった理由として、プロモーター領域だけでなく他の部位、たとえば、イントロン
領域の DNA メチル化率が mRNA 発現量に影響しているという報告もあり、MEF2C でもプロモータ
ー領域以外のＤＮＡメチル化率が遺伝子発現に関与していることや、別の発現調節機構が働いて
いることが想定される。まだ、試料数が十分ではないため、今後の課題として、サンプル数を増
やした追加研究が望まれる。健常対照群においては、MMSE と ADAS を施行しておらず、健常対照
群でも認知機能検査を加味した追加研究が望まれる。 
以上より、白血球における MEF2C mRNA 発現量は、AD 発症や認知機能低下のバイオマーカーに
なる可能性が示唆された。なお、この学位論文の内容は、以下の原著論文に既に公表済である。

学位論文要旨 
            
High Mobility Group Box 1（以下 HMGB1）は、核内で DNA に結合する非ヒストンタンパクで、
核内のホメオステーシスに関与している。しかし、細胞壊死などにより細胞外へ排出されると、
他のサイトカインや DNA などと結合し、Toll 様受容体（Toll like receptor : TLR）や RAGE な
ど自然免疫系の受容体を介して直接または間接的に炎症性サイトカイン産生に関与することが知
られている。皮膚の炎症性疾患やリウマチなどでは HMGB1 の血中濃度が高くなることが報告され
ており、また敗血症における Late mediator としても注目されている。一方、HMGB1 は細胞遊走
能を持ち、創傷治癒において重要な役割を果たしていることが分かってきた。我々は、表皮角化
細胞に対する細胞外 HMGB1 の働きについて研究してきたので報告する。 
方法）HMGB1 は redox status により炎症性サイトカインの性格をもつ disulfide-HMGB1 と chemo
tactic に働く reduced-HMGB1 の状態がある。我々は、ヒト培養角化細胞にそれぞれの HMGB1 を
作用させ、その効果を調べた。 
結果）Disulfide-HMGB1 は角化細胞に対しても炎症サイトカインの産生を促すが、Reduced-HMGB
1 は炎症性サイトカインを産生せず、さらに poly(I:C)により誘導した炎症性サイトカインの産
生を抑制することがわかった。 
結論）現在までに HMGB1 によるヒト角化細胞直接的な抗炎症作用については報告されておらず、
今回我々が初めて見出した。この研究結果は皮膚の慢性炎症性疾患のメカニズム解明と新たな治
療薬の開発につながる可能性がある。

Several methods of gas phase CVD such as Hot Filament， DC arc，. Jet DC Plasma， Laser Plasma 
Oxygen-acetylene Flame， and Microwave Plasma are dificult to synthesize on a substrate which has low heat 
resistance， la叩edevices， and considered very costly in large scale production. So， the synthesizing diamond 
thin film using In-liquid plasma technique was our也市etin this study. In-liquid plasma is generated plasma in a 
bubble in liquid and synthesizing diamond in liquid alcohol is available. A Remarkable efect of liquid plasma 
process is high growth rate because the molecular density of a liquid is much higher than that of a gas. And the 
substrate which has low heat resistance can be deposited because of the cooling e仔'ectof the liquid. 
Furthermore， the experimental device becomes simple and easy. Therefore， synthesis of diamond using 
in-liquid plasma CVD is expected as altematives besides the conventional gas phase plasma CVD. 
The theory of IしMPCVDis relatively new. 8ut in general， the theory is almost the same as gas phase CVD. 
80th use the 8achmann diagram as a reference. This submerged plasma is a proprietary technology of Ehime 
Unive陀ity，it uses a microwave or high frequency as a plasma energy source and generated in liquid. 
The mechanism of diamond synthesis using an in-liquid plasma chemical vapor deposition (CVD) method 
has been investigated. Chemical reactions from a liquid mixture of methanol and ethanol (in-liquid plasma CVD) 
and a gas mixture of methane and hydrogen (gas-phase CVD) are used for analyzing. The procedure was， 
initialy， synthesizing CO and then induces a chemical reaction using the remaining C and H to synthesize a 
carbon substance. From the experimental results of carbon deposits using various liquid and gas mixtures as 
the source materials， found that the region of the remaining H and C after CO synthesis satisfying H/C注20is 
necessary to synthesize diamonds using in-liquid plasma CVD method. The IしMPCVDmethod has succeeded 
to synthesize single c叩staldiamond (100) and (111) plane. Deposited diamond film on (100) has film growth 
gradually and smooth surface. Whereas (111) face has a polyc叩st剖linefilm with iregularity growth and rough 
suげace.The surface suitable for homoepitaxial growth was found to be (10). 
Growing diamond on ferrous base materials is challenging because of its dificulties due to several factors. 
First， carbon can difuse into the substrate with relatively high difusion rate， so diamond nucleation time can be 
long. Second， the catalytic efect of iron on the growth of Sp2 dominated amo巾housand nanoc叩stalinecarbon. 
Thi吋， poor adhesion and high residual st陪ssappear because the thermal expansion coeficient between 
diamond and ferrous metal are not compatible. 
Diamond nucleation on non-diamond substrate is generally proposed to occur mostly on an intermediate 
layer of amorphous carbon， metal carbides and 9問phite.In the current study， silicon as substrate and Titanium 
as interlayer were used. 80th of them were successful for growing diamond. 
The application of the IL-MPCVD method for the synthesis of diamond film on stainles stel using double 
interlayer Ti/Si was investigated. The thickness of the interlayer for appro阿梅 diamondfilm formation has 
achieved， despite the fact that the result should be improved. 
For the future experiment， nickel is one of the materials that should be used for growing diamond. Nickel is 
one of the few materials that have a close lattice-parameter match with diamond (a = 3.52 A for Ni and 3.56 A for 
diamond). However， itshigh solubility for carbon and its strong catalytic efect on hydrocarbon decomposition 
and subsequent 9問phiteformation at low pressures have prevented CVD diamond nucleation on the Ni surface 
without the deposition of an intermediate graphitic layer.

本学位論文の内容は、 2つの幾何学的最適化問題の数値的解法の計算効率の向上についてであ
る。
最初の一つは、スタイナ一極小木問題 (Steinerminimum tree problem)である。 これは、 d
次元ユークリッド空間内に与えられた点の集合をつなぐ最小の長さの木(サイクルが無いグラ
フ)を見つけよというものである。その際、余分な点(スタイナー点 (Steinerpoint)と呼ばれ
る)を付け加えても良いという条件が付いている。付け加える点の最適な数と位相(他の点とど
のようにつなげるかという情報)が未知であるため「組み合わせの数の爆発Jが起こり、そのた
めこの問題はNP困難な問題である ことが知られている。可能性がある全ての場合を検証するた
め、巨大な探索木を探索する際になるべく効率的に「枝刈り」をおこなう必要がある。本論文で
は、探索木の探索と「枝刈りJについて、以下に述べる新しい手法を提案した。
スタイナ一点の数と位相を決めた際には、その位相において木全体の長さが最小になるスタイ
ナ一点の位置は一意に決まることが知られている。その位置を求めるために緩和法を用いるが、
その際スタイナ一点同土が衝突することがある。 Chapeau-Blondeauらは、衝突した2つのスタ
イナ一点で位相を交換する (heuristicな)アルゴリズムを提案した。本論文では、彼らの手法を
用いて巨大な探索木を効率よく探索し、さらに枝刈りするアルコリズムを提案している。
Chapeau-Blondeauらの手法では局所的な最適解しか見つけられないが、本手法では探索木全体
を探索するために大域的な最適解を確実に発見することができる。数値実験の結果、提案手法に
より計算効率が従来手法より 20%以上向上することを確かめることができた。提案手法の優れ
ているところは、スタイナ一極小木を求めるさまざまな既存の手法と組み合わせることができる
ということで、さらなる発展が期待できる。
2つ自の問題は、 d次元ユークリッド空間内に与えられたジヨルダ ン閉曲線内に張る極小曲面
を求めよというプラトー問題 (Plateauproblem)に対する数値解法である。極小曲面とは、全て
の点において平均曲率がOとなる曲面である。ジヨ ルダン曲線が平面に埋め込まれている場合は、
極小曲面は等角写像になる。 閉曲線内の曲面が極小曲面であるための必要十分条件は、 それがあ
るエネルギー汎関数の極値を与える曲面になっていることであることが知られている。
プラトー問題に対しては、 Tsuchiyaにより区分的 l次有限要素法を用いた数値解法が提案さ
れていた。まず円板を三角形分割し(三角形の集合で円板を覆うこと)、三角形分割上の区分的
1次写像で極小曲面の近似を試みるスキームである。しかし、エネルギー最小の写像を求める際
に、円板の境界上にある三角形分割の節点の像がジヨルダン曲線上でかなり自由に動けるため、
その数値スキームはしばしば不安定になることが知られていた。特に、曲線が大きく変化する部
分に対して対応する円板境界上の節点の数が少ない場合、一部の三角形が"潰れて"しまい、数値
解の精度が大きく損なわれる現象がしばしば観察された。
本論文では、その対策として円板の三角形分割の動的再分割を導入した。そのために 「三角形
の像が defectiveであるJという指標を導入し、円板の境界付近で defectiveな三角形が検出さ
れた場合に、動的にその三角形(と必要ならば周辺の三角形)を分割するアルゴリズムを開発し
た。その結果、極小曲面および、等角写像に対する有限要素法の精度、効率が、大幅に向上した。
この方法は、さらに次の 2つの場合について、数値解の精度と計算効率の向上に寄与した。
一つは、ジヨルダン閉曲線が角を持つ場合である。ジヨルダン閉曲線が角を持つ場合、区分的
l次写像でその角を近似しなければならないので、必然的にそこでは精度が大きく損なわれる。
しかし円板の境界上に動的に節点を導入することにより、角の周辺の近似の精度がかなり回復す
ることが確かめられた。
もう一つの場合は、自由境界を持つ極小曲面を求める問題である。例えば、針金の両端を平板
にくっつけた物体において、針金と平板の間に張る石鹸膜の形状を求めよという問題が考えられ
る。このとき、曲面と平板の間には角が生じるので、やはり近似精度が損なわれる。本論文では、
提案手法がこの場合も有効であることを確かめた。シャボン液に浸したジヨルダン曲線を引き上
げる途中にも自由境界が生じるが、本論文の手法を適応することにより、その様子をアニメーシ
ョンにすることができた。さらに、自由境界を持つ極小曲面を求める問題に対する有限要素解の
精度について、数学的に厳密な収束証明を与えることにも成功した。
以上、 2つの幾何学的最適化に関する本論文は、この分野の研究に大きく貢献したと考えられ
る。本論文の内容は 2編の英文論文として投稿中であり、また本論文の内容を数々の国際会議で
発表し、高い評価を得てきた。
よって、本論文は博士(理学)の学位を受けるに十分な価値があるものと認める。
本論文の公聴会は、 2月 1日に理学部2号館201号室(大演習室)で開催され、論文内容に
ついての口頭発表と質疑応答が行われた。

environmental contarninants. The aim of my studies is to investigate the effects of 
environment chemicals at the transcriptome and proteome levels in laboratory rodents. The 
results from these studies may help us to understand the mode of actions of these 
chemicals with specificity in strains， genders， growth stages， and doses of exposure. 
The first study focused on the strain di首位encesin the effects of 2ム7，8・tetrabromo・
dibenzo・p-dioxin(TBDD). TBDD is an analogue of 2ム7，8・tetrachloro-dibenzo・p-dioxin
(TCDD)， which is one of the most potent dioxins and dioxin-like-compounds. Studies on 
the toxicology of TBDD indicated白at也iscompound influences a spectr山nof effects 
likely to that ofTCDD in laboratory animals. It has been demonstrated that there are sむam-
and species-specific di舵 rencesin the aryl hydrocarbon receptor (AHR)-mediated 
responses to dioxin exposure. Here， we used the two inbred mouse strains C3H1HeJ-Ipr/ 
lpr (C3H/lpr) and 1¥忽L/MpJヤrllpr(MRL/lpr) which have different AHR genotypes at 
the 375血 position.C3H1争rAHR presents an Ala residue at the 375也 position，which has a 
high a伍nityto TCDD， while MRL/伊rAHR encodes a Val which has lower affinity to 
TCDD. Thus， C3H1lpr and MRL/lpr are considered as sensitive-and resistant-strain， 
respectively. Here， we adopted a proteomic approach using two-dimensional electro-
phoresis (2-DE) and matrix-assisted laser desorption/ionization time offlightltirne offlight 
(MALDI-TOF/TOF) mass spectrometry小IfS)to identifシthedifferences in the effects of 
TBDD exposure on the hepatic proteome between the two mouse strains. To confirm the 
strain-difference in response to TBDD treatment， cytochrome P450 (CYP) lAl and lA2 
protein levels were measured in both strains. A dose of 10μglkg body weight of TBDD 
induced hepatic CYPIAl and CYPIA2 expression in both strains， but the expression 
levels of both CYPIA proteins were higher in C3H1争rmice than in MRL!.伊rmice. The 
result supports that C3HJlpr mice are more sensitive to dioxins也anMRL/lpr mice. We 
have successfuly identified proteins白atwere more induced or suppressed by TBDD 
treatment in C3HJ炉rmice. These include proteins responsible for AHR activation through 
production of endogenous ligands such as aspartate aminotransferase， indolethylamine 
N-methyltransferase， and aldehyde dehydrogenases， as well as reducing oxidative stress， 
such as superoxide dismutase and peroxiredoxins. Taken together， our results provide 
insights into the molecular mechanism underlying the high dioxin-susceptibility of the 
C3H/伊rstrain， inwhich AHR activation by TBDD is more prompted by the production of 
endogenous lig姐 ds，but the adaptation to oxidative stress is also acquired. 
The second study focused on an emerging environmental contaminant， bisphenol A 
(BPA). BPA has been known as a m司orendocrine disrupting chemical which is widely 
detected in the environment and human. BPA has been detected in body fluids and tissues 
including umbilical cord blood， human breast milk， anmiotic fluid， fetal serum， and 
placental tissue， suggesting that BPA Can be transfen吋企ommother to fetus during 
pregnancy and lactation. Therefore， there is a growing concem about the potential 
developmental toxicity of BPA. Studies in developmental exposure demonstrated that BPA 
has adverse effects on male and female reproduce systems， metabolic homeostasis， and 
may associate with liver dysfunction，旬pe2 diabetes， and cardiovascular disorders. These 
effec臼 maydepend on sexes， ages， and dose expos山 es.The aim of this study is to unveil 
the mechanism of the transgenerational actions of BPA based on the changes in hepatic 
transcriptome and proteome in rat 0億 pring.We exposed pregnant Wistar rats to low (50 
μg/kg bw/day) and high (5000μg/kg bw/day) doses ofBPA or 17s・estradiol(E2， 50μg/kg 
bw/day) from embryonic day 3 to 18. Firstly， the offspring were observed for any changes 
in their body and organ weights and anogenital distance (AGD)拭 PNDl・2(newbom)， 
PND21・22(weaning)， and PND59・60(young). We observed significantly increased body 
weight and longer AGD出 bothfemales and males at some periods. There were also 
several changes in organ weights.百lesechanges were found differently between sexes， 
ages， and doses of exposure. 
Secondly， we analyzed the hepatic transcriptome profiled of the offspring at newbom 
and weaning by a paired-end sequencing with Ilumina HiSeq2500. The genes which had 
significantly different FPKM (fragments per kilobase of transcripts per million mapped 
reads) in BPA-or E2・treatedgroups from those in the control (vehicle-treated) group (FDR 
< 0.05) were selected as differentially expressed genes (DEGs) and used for further 
analyses including biological function and transcription factor analyses. Transcriptome 
data revealed血atthere were more changes in newbom rats than in weaning rats. A 
principal component analysis using FPKMs of al DEGs demonstrated that the prenatal 
BPA exposure caused the masculinization of the hepatic transcriptome in females at 
newborn and weaning stages. It suggests antagonistic effects of BPA to endogenous 
es仕ogenson female offspring. Transcriptome profile of newbom offspring showed that 
BPA exposure up-regulated genes related to the cel cycle， but down-regulated genes 
related to the chemical carcinogenesis in both females and males. In newbom female pups， 
even the low dose of BPA induced the DNA replication and insulin resistance， and 
depressed the fa町 aciddegradation， steroid hormone synthesis， and PPAR signaling 
pathways. In newbom male pups， only the high dose of BPA altered gene expressions in 
the pe1'oxisome， PPAR signaling， fatty acid degradation， and othe1' metabolic pathways. 
T1'anscriptome profile of weaning 0百springshowed that even low dose ofBPA changed the 
expression levels of genes involved in several infectious and metabolic diseases in females， 
and in metabolic pathways in males. Interestingly， most of the effects found in newborn 
females were reduced or disappeared at weaning， whereas the effects in newbom males 
were increased or continued at weaning. These results indicate different modes of action of 
prenatal BPA exposure between male and female offspring as well as between early and 
later life stages. We aIso detected altered expression of epigenetics-related genes including 
DNA methyl-transferases (DNMTs)， histone acetyl仕ansferases(HATs)， methyltransferases 
(HMTs) ， deacetylases (HDACs)， and demethylases (HDMs). These changes we1'e mostly 
observed in newbom females， suggesting the role of epigenetics in susceptibility to BPA 
exposure. In addition， BPA was found to induce or inhibit n山nerousxenobiotic 
metabolizing enzymes including phase 1， I， and II enz戸nesin postnatal animals， 
suggesting BPA may have crosstalk with effects of othe1' contaminants and drugs. 
Thirdly， we examined the expression levels of live1' prote也sin newbom pups using a 
high-throughput approach， isobaric tags fo1' relative and absolute quantitation (iTRAQ) -
labelling followed by nano-liquid chromatography (LC) and MALDI MS/MS. Based on 
the ratios between iTRAQ-tags of treated group and control group C三1.5or ~ 0.7)， we 
decided the differentially exp1'essed proteins (DEPs) by each仕ea加lentand used them for 
biological function and network analyses. Proteornic data also showed differences between 
female and male pups. We also identified DEPs involved in lipid and ste1'oid hormone 
metabo1ism. Together叩thtranscriptomic data， these data suggest that p1'enatal exposure to 
BPA induces lipid accumulation and imbalance of sex hormones， consequently lead to an 
increase of body weight and AGD length in offsp1'ing. Network analysis of the DEPs 
indicated several hubs including CPSl， GADPH， TXNl， ALB， and ACTB， which may be 
considerable as genetic makers fo1' the developmental effects ofBPA. 
Overall， the studies in this dissertation demonstrated a valuable strategy to clarify the 
molecular mechanisms of genetic (strain)ー， sex-， age-， and dose-dependent effects of 
environmental contaminants.
