(Dissertation Title) 
The formation and evolution of supermassive black holes (SMBHs) are important topics in the 
modern astrophysics for understanding the nuclear activity of galaxies and the total picture of th巴
galaxy formation， b巴causeit is now widely recognized that the 5MBH evolution is closely coupled 
with the galaxy evolution (so-called galaxy-SMBH coevolution). Especially， the quasar luminosity 
function (QLF) provides us a clue to understand the evolution of 5MBHs from a statistical 
viewpoint. Recent quasar surveys in optical and X-ray have derived QLFs at various redshifts over 
a wide luminosity range. They have suggested that the redshift evolution of the quasar number 
density is luminosity-dependent. However， the number density of low-Iuminosity quasars at z > 4 is 
unclear due to the following two reasons. One reason is that the observationally-inferred number 
density includes possible large systematic errors due to drawbacks in the widely-adopted method 
for evaluating the survey completeness and the resultant quasar number densities. The other reason 
is the large statistical error in the quasar number density due to the lack of a sufficient number of 
low-Iuminosity qllasars at z > 4. Therefcire in this doctoral dissertation， we revisit the survey 
completeness and QLF at high redshift with a new method and derive the QLF with a large sample 
of low-Iuminosity quasars at high redshift. 
In order to derive th巴numberdensity of low-Iuminosity qllasars accurately at high redshift， we first 
improve the evaluation method of the number density of 10w-llIminosity quasars. Generally high 
redshift quasars are selected by using their color properties. Therefore it is important to make 
realistic model spectra， which well reproduce actual quasar spectra， for simulations to estimate the 
survey completeness. The sllrvey completeness is lIsually estimated by adopting a typical template 
of the quasar spectrllm， constructed empirically or based on simple models. However the 
equivalent width (EW) ofbroad emission lines in the quasar spectrum strongly depends on the 
quasar luminosity， inthe sense that lower-Iuminosity qllasars show emission lines with a larger EW 
(the Baldwin effect). If the quasar luminosity investigated in a survey is mllch lower than that of a 
template used to estimate the survey completeness， the quasar colors calculated from the template 
may be systematically different from the actual quasar colors due to the unexpected contribution of 
emission-line fluxes into broad-band magnitudes. Th巴reforeit is essential to know how the 
Baldwin effect affects the estimate ofthe completeness in qllasar sllrveys and the resultant QLFs. 
We made composite spectra for each luminosity based on the Baryon OsciIlation Spectroscopic 
Survey (BOSS) quasars and quantifi巴dthe Baldwin effect. As th巴result，we found that 
lower-luminosity quasars at z ~ 4 -5 are more easily selected by color selection criteria than 
higher-luminosity quasars when the effects of the Baldwin effect are properly considered. 
Furthermore the correction of the effect of the inter-galactic medium (lGM) absorption is important 
for preparing realistic model spectra ofhigh redshift quasars. Recently， a new 1GM absorption 
model， which weIl reproduces recent observational data ofthe 1GM column density， was publish巴d.
We evaluate the effect ofthe difference in 1GM models for the estimate ofthe survey completeness 
by using COSMOS images observed with Subaru / Suprime-Cam. We find that the latest 1GM 
model predicts a smaIler Lyman-break feature at z = 4 -5 than older 1GM model， which makes the 
color selection of quasars more chaIlenging. 
The results indicate that the completeness estimates are sensitive to the luminosity dependence of 
the quasar spectrum and also to 1GM attenuation models. At z ~ 4， the number density of quasars 
derived with the newly estimated completeness is (3.49土1.62)x10-7ルlpc-3mag-1 for・24.09く M1450
く ー23.09and (5.24士2.13)x10-7 Mpc-3 mag-1 for・23.09く M1450く ー22.09，which are ~24% lower tban 
those estimated by the conventional method. On the other hand， atz -5， the 1σconfidence upper 
limit ofthe number density at -24.5く M1450くー22.5in the new estimates is -43% higher than that 
estimated pr巴viously.These results suggest that the luminosity dependence of the quasar spectrum 
and the 1GM model are important for deriving the accurate number density ofhigh redshift quasars. 
Secondly we construct a large sample of low-luminosity quasars at z -5 to study the statistical 
evaluation of the number density of quasars， by taking the above results into account. Because of 
the insufficient area and depth of previous surveys， the low-luminosity quasar survey was difficult. 
However the strategic survey program of the Subaru telescop巴withthe Hyp巴rSuprime-Cam 
(HSC-SSP) proyides a key solution to search for faint quasars at high redshift. The HSC-SSP 
wide-field optical imaging survey started in March 2014 and plans to spend 300 nights for 5 years. 
By taking advantage ofthe wide field-of-view (FoV) ofHSC (a circular FoV with 1.5 deg in 
diameter)， the Wide-layer component of the HSC-SSP survey wiIl cover ~ 1，400 deg2 when the 
survey is completed. The 5σsurvey depth of the Wide-layer component reaches ~3 mag deeper 
than that of the SDSS surv巴y.Thanks to the deep and wide HSC-SSP survey data， we can search 
faint quasars at high-redshift. 
By the new method of the newly estab!ished high redshift quasar selection， we construct a large 
sample of z -5 low-luminosity quasars based on the wide survey data in a part of the HSC-SSP. 1n 
81.8 deg2 area within the Wide layer ofthe HSC-SSP survey， 224 candidates of low-luminosity 
quasars at z -5 are selected by adopting the Lyman-break method in the magnitude range of 19.1く
iく 24.1.We derive the QLF at z ~ 5 by removing the sample of M1450 > ・23.32，where the sample is 
largely affected by the contamination of compact galaxies. The resultant binned QLF at z -5 is
consistent with that of previous studies at M1 450 く ~24 . 32 . We fit a double power-law model to our 
HSC and Sloan Digital Sky Survey (SDSS) sample that is distributed in a wide luminosity range 
(-28.76く M1450く ・23.32)witb the maximum likelihood method. 1n this process， the bright-end 
slope is fixed to -2.90 based on the number densities of some previous studies for bright quasar 
+0.03 
samples. As a result， it turn out to be tbat the best-fit faint-end slope，α= -1.2rv.v>_O.10， isflatter 
than that reported by previous studies at z -5. The break of the double power-law is fainter and 
higher number densities than the r巴sultsofz -5 pr巴viousstudi巴s.This is because our sample 
covers a wider luminosity range extending enough fainter range than the break luminosity of the 
double power-law function. Also we investigate the QLF parameters at 3三z三6with those in the 
literature. The HSC resuIts suggest that the faint and bright-end slope and the break luminosity are 
roughly consistent at 4三z三6，although other previous studies suggested that the faint-end slope 
becomes steeper at higher redshifts. Furth巴rmorewe explore the redshift evolution of the quasar 
number density for each luminosity. Our results show that the number density of faint quasars 
decreases toward high redshift at z > 3， similar to bright quasars. This evolution is consistent with 
th巴AGNdown-sizing scenario. 1n addition， as indicated by previous studies for luminous quasar丸
the number density of low-luminosity quasars also d巴creasemore rapidly from z -5 to z -6 than 
from z -4 to z -5. 
Furthermore， we conduct spectroscopic observations for a part of the HSC photometric sample of z 
-5 quasar candidates， and measure properties of their 5MBHs. Observed low-luminosity quasars 
show lower MBH than most SDSS quasars at z -5 and relatively high Eddington ratio that is close 
to the Eddington limit. lt suggests that the 5MBHs of the sample in this study are in a relatively 
young phase ofthe evolution.

【背景と目的】
マラリア原虫の生活環のうち、標的細胞に寄生できるのはメロゾイトとスポロゾイトの２つ 
の侵入型ステージである。そのなかでもスポロゾイトは蚊の唾液腺とヒトの肝細胞に侵入し、
生活環を確立する。その侵入にかかわる分子の同定とその機序の解析は、マラリアの感染、伝
播の機序、予防法の確立において重要な課題である。Bantuchai 氏は、メロゾイトとスポロゾ
イトの両方の侵入型に共通する分子の侵入機序を解明することを目的として、両ステージで発
現する侵入関連分泌型タンパク質の同定と、その機能解析を試みてきた。その過程において、
メロゾイトが赤血球に浸入する上で重要と想定されるロプトリー蛋白質に注目し、スポロゾイ
トの宿主細胞侵入における意義について検討した。特にロプトリー蛋白質の一つで膜貫通ドメ
インとカルシウム結合モチーフを有する rhoptry neck protein 11 (RON11)に着目して本研究
を行った。
【方法と結果】
マウスに感染するマラリア原虫である Plasmodium berghei を用いて、その RON11 (PbRON11) 
について、まず特異抗体を作成した。その抗体を用いて Western blot をしたところ、蚊の中腸
から採取したスポロゾイトで、予想される 100kDa 付近にバンドが確認できた。また PbRON11 の
マラリア原虫の生活環における発現プロファイルを調べ、赤血球ステージのシゾント期と、スポ
ロゾイトにおいて発現することを見いだした。また、両方のステージで先端部小器官のロプトリ 
ーに PbRON11 が局在することを確認した。スポロゾイトにおける mRNA 発現はオオシスト時期に
限定されており、スポロゾイトのロプトリー蛋白質群に特徴的な転写パターンを示した。
スポロゾイトにおける PbRON11 の役割を解析する為に、スポロゾイト特異的に PbRON11 発
現を抑制する遺伝子改変原虫（PbRON11-cKD）をプロモーター置換により作成した。この原虫
を媒介蚊に感染させると、中腸に形成されるオオシスト内でスポロゾイトが正常に形成され、
体腔液に放出されるものの、その後の唾液腺への侵入効率が極端に抑制された。これらのこと
から、PbRON11 はスポロゾイトが蚊の唾液腺に侵入することに必須の分子であることが示され
た。さらに、スポロゾイトを体腔から回収し、肝由来培養細胞株に添加したところ、侵入効率
が遺伝子改変をしていないコントロール原虫の 1/100 以下であることが明らかとなった。ま
た、体腔から回収したスポロゾイトの滑走運動能は、PbRON11-cKD でほとんど見られなかった
ことから、PbRON11 はスポロゾイトの接着あるいは運動能に必須であることが示唆された。以
上より、PbRON11 が唾液腺や肝細胞への侵入に重要な役割を担うことが示された。
【結語】
PbRON11 を低発現させたスポロゾイトにおいて、接着能、滑走運動能の低下が認められ、さ 
らにマラリアを媒介する蚊の唾液腺および哺乳類宿主の最初の感染標的である肝臓への侵入の
阻害が観察された。その結果、PbRON11 はマラリアの伝播に重要な役割を果たすことが明らか
となり、今後、感染予防および治療の標的となり得る可能性が示唆された。
本論文の公開審査会は平成 30 年 1 月 15 日に開催された。申請者は研究内容を明確に英語で
発表し、以下の内容を含む多くの質疑に対し的確に応答した。主な質疑の内容として、1) マ
ラリア媒介蚊の唾液腺および哺乳類の肝臓への感染において、RON11 の結合パートナーは何
か。スポロゾイトとメロゾイトにおけるロプトリー内の局在に差がある意義、RON11 の発現と
感染種特異性への影響。2) RON タンパクには RON11 と同じような機能をもつ RON2,4,5 が存在
するが、それらの役割の違いについて。Ca2+の結合活性は接着と侵入に必須か。N 末端の PEXEL 
モチーフの推測される機能。G-蛋白質の関与はあるのか。3) 遺伝子改変原虫に viability の
変化はあったのか。感染力の低下は RON11 の発現低下のみが影響しているのか。遺伝子発現と
蛋白質発現のプロファイリング解析はされているか。4) in vitro の実験においてなぜヒト肝
癌細胞を用いるのか。5) 実験動物であるマウスと原虫および遺伝子改変原虫について、ヒト
マラリアと同様の肝機能障害などの感染症状を来すのか。6) 今後の臨床応用のストラテジー
をどう考えるか。治療標的やワクチンの候補蛋白質になるのか、などであった。
マラリア原虫のスポロゾイトの侵入における key molecule として RON11 を世界で初めて同
定した本研究は評価される。今後の治療および予防の標的として利用される可能性があり、臨
床的にも有用な研究と考えられる。審査委員は，申請者が本論文関連領域に対して学位授与に
値する十分な見識と能力を有することを全員一致で確認し，本論文が博士(医学)の学位論文に
値するものと結論した。

論  文  名  ABO 血液型と膵癌の長期予後の関連について 
 
      審査結果の要旨 
 本論文の要旨を以下に記す。 
【背景と目的】 
膵癌は最も予後不良な癌腫の一つであり、本邦での 5 年生存率は男女ともに 7− 8％である。
本邦における膵癌の罹患率と死亡率は増加しており、それぞれ全癌腫の中で第 7 位と第 4 位で
ある。発症リスク因子に関し、遺伝要因として遺伝性膵癌や家族性膵癌が明らかとなっている
が、環境要因として糖尿病や肥満、喫煙、飲酒などが指摘されている。 
これまでに遺伝的要因に関与している ABO 血液型について、膵癌や胃癌、腎細胞癌、非小細
胞肺癌などの癌腫では発症リスクや長期的な臨床経過との関連が報告されている。膵癌に関し
て、西欧からの研究では O 型が非 O 型に比べて切除後の生存率が有意に良好であるという報告
があるが、一方中国からは差がないという報告もあり、ABO 血液型と膵癌患者の長期予後との
関連についてまだ一定の見解が得られておらず、特に日本人においてはまだ不明である。 
愛媛大学医学部附属病院第三内科と関連病院では、Ehime Pancreato-Cholangiology (EPOCH)
 Study Group を組織し、愛媛県内多施設において膵癌と診断された患者の集計を行ってきた。
本研究では、同コホートへの後ろ向き調査を行い、ABO 血液型と膵癌患者の長期予後との関連
を明らかにすることを目的として検討および解析を行った。 
【方法】 
2011 年〜2013 年の間に EPOCH Study Group で診断された膵癌患者を対象として、年齢、性別、

ABO 血液型、UICC 病期、転帰などの情報を収集した。統計解析は、群間比較はカイ 2 乗検定ま
たは Student-t 検定、傾向検定では Cochran Armitage 検定、生存解析は Kaplan-Meier 法およ
び Cox 比例ハザード回帰を用いて行った。P<0.05 を有意差ありとした。 
【結果】 
 １）ABO 血液型が判明している膵癌患者は 409 例で、平均年齢は 71.8±10.0 歳、男性が 54.3%
であった。各血液型の頻度は A 型 182 例(44.5%)、B 型 86 例(21.0%)、O 型 91 例(22.2%)、AB
型 50 例(12.2%)であった。２）診断時の腫瘍径中央値(範囲)は A,B,O,AB 型それぞれで 3.0cm
(0.6-10.0), 3.0cm(1.0-7.0), 3.0cm(1.0-10.0), 3.4cm(0.7-7.0)で、進行病期 IIA 以下の割
合はそれぞれ 36.8%, 34.9%, 47.3%, 26.0%であった。３）治療法について外科切除、化学療
法、保存治療の割合は A 型 41.8%,38.5%,19.8%、B 型 45.4%,32.6%,22.1%、O 型 45.1%,40.7%,1
4.3%、AB 型 34.0%,48%,9%であった。４）生存期間中央値(95%信頼区間)は A,B,O,AB 型それぞ
れ 370 日(299-468),408 日(301-568),521 日(365-771),301 日(190-451)であった。５）A アレ
ル群（A 型+AB 型）と非 A アレル群（B 型+O 型）を比較すると、前者で診断時腫瘍径は有意に
小さく(P=0.044)、生存期間中央値(MST)は短い傾向にあり(P=0.051)、特に外科切除後の MST
は有意に短かった(P=0.025)。６）A アレル群と O 型のみの比較では、前者で診断時病期 IIA
以下の割合が有意に少なく(P=0.034)、全治療および外科切除後の MST は有意に短かった(P=0.
046, P=0.033)。７）年齢、性別、BMI、腫瘍径、リンパ節転移と遠隔転移の有無、治療法を調
整して行った多変量解析では、A アレル群は非アレル群に対して生存予後に関する独立した危
険因子であった(HR1.336, 95%CI 1.007-1.781)。 
【結論】 
 本研究により A アレル群(A 型+AB 型)は非 A アレル群(B 型+O 型)に比べて生存率が不良で、
病期や治療法を調整した多変量解析でも独立した予後不良因子であった。この結果より、ABO
血液型は日本人膵癌患者の長期予後に関与する可能性が示唆された。 
 
 本論文の公開審査は平成 31 年 2 月 4 日に実施された。申請者はまず、本論文の内容ならび
に研究意義について英語により明解な発表を行った。審査委員からは本研究に関する以下の質
問がなされた。１）日本人全体の ABO 血液型分布と比べて本コホートでは O 型の頻度がやや少
ないがその理由と研究に与える影響は、２）O 型の患者で BMI が高いが糖尿病など他の発症リ
スクとの関連はどうか、３）腫瘍径や病期診断の方法は CT などの画像なのか、切除例では術
後病理診断なのか、またその違いによる影響はどうか、４）データ欠損例の扱い方や統計的解
析方法について適切か、５）ABO の血液型による予後の違いの機序として血液型抗原はどのよ
うに膵癌悪性度に影響しているのか。申請者はこれらの質問やコメントすべてに対して明確に
応答した。特に ABO 血液型が膵癌予後に影響を与える機序について、血液型糖鎖抗原と細胞接
着の関連や、癌細胞の接着を阻害する血中の soluble intercellular adhesion molecule 1 
(sICAM-1)レベルが O 型患者では低下しているという報告などをもとに申請者の考えを詳細に
説明した。 
 
 以上から審査委員は，申請者が本論文関連領域に対して学位授与に値する十分な見識と能力
を有することを全員一致で確認し，本論文が学位授与に値すると判定した。
