    ゴマ毛状根において生産されているデヒドロアントラセサモンBと関連するアントラキノン系化合物の生合成経路

    In order to investigate the biosynthesis of dehydroanthrasesamone B and its related anthraquinone derivatives in hairy roots of sesame (Sesamum indicum L.), 2H‐labeled MPAQ was prepared and fed to the sesame hairy root cultures. Efficient conversion of fed MPAQ to (Z)‐MPDEAQ was observed in this feeding experiment. However, the conversion of fed MPAQ to dehydroanthrasesamone B, anthrasesamone B and 2,3‐epoxyanthrasesamone B was not observed. These results demonstrated that MPAQ is the actual intermediate for the production of (Z)‐MPDEAQ and that (Z)‐MPDEAQ is produced via dehydrogenation of the monoene side chain in MPAQ. Moreover, dehydroanthrasesamone B may be produced via dehydrogenation of the monoene side chain in anthrasesamone B, but not via hydroxylation of the anthraquinone ring in (Z)‐MPDEAQ.ゴマは、その根や毛状根(培養根)において共通の炭素骨格を有するアントラキノン系化合物(アントラセサモン類)を生産していることが知られている。これらのアントラキノン系化合物のうち、デヒドロアントラセサモンBは、(Z)‐MPDEAQと同じジエン側鎖とアントラセサモンBと同じヒドロキシ化されたアントラキノン環を有することから、その生合成経路に興味が持たれた。本研究では、デヒドロアントラセサモンBと他のアントラキノン系化合物との生合成的関連を明らかにするために、重水素標識したMPAQのゴマ毛状根培養への投与実験を行った。ゴマ毛状根による投与したMPAQからの(Z)‐MPDEAQへの変換は確認されたが、アントラセサモンBとそのエポキシ体への変換は確認されなかった。また、デヒドロアントラセサモンBへの変換も確認されなかった。以上の結果から、デヒドロアントラセサモンBは、(Z)‐MPDEAQのアントラキノン環がヒドロキシ化されることにより生合成されているのではなく、アントラセサモンBのモノエン側鎖が脱水素化されることにより生合成されていることが示唆された。departmental bulletin pape

    Production of monoclonal antibodies against histamine

    食中毒の原因物質であるヒスタミン(His)のモノクローナル抗体(mAb)の作製した。Hisと5種類の架橋剤で結合させた Keyhole Limpet Hemocyanin (KLH)を作製し、これらでマウスの免疫を行った。これらの抗血清と5種類の架橋剤で結合させた Bovine Serum Albumin (BSA)との反応性を調べた。その結果、His‐BS3‐KLH、His‐DTSSP‐BSAまたはHis‐GA‐KLHで免疫したマウスの抗血清の反応性が高かった。次に、こられの抗血清で競合ELISAを行った結果、His‐GA‐KLHで免疫し、固相抗原にHis‐GA‐BSAを用いた場合に効率よくHis特異抗体を検出できた。そこで、His‐GA‐KLHで4回免疫し、His特異抗体を検出できたマウスの脾臓細胞の細胞融合を行った。限界希釈法でクローニングを2回以上行い、9クローンの安定な抗His mAb産生ハイブリドーマ(His.1〜9)を樹立した。この中で最もHisに反応性の高かったHis.6抗体を用いた至適化した競合ELISAで0.04μg/mLまでのHisの検出が可能であった。今後、これらの抗体を用いたHisの高感度な免疫化学的分析法の確立が期待される。Monoclonal antibodies (mAb) against histamine (His), a causative agent of food poisoning, was prepared. We prepared Keyhole Limpet Hemocyanin (KLH) bound with His and five kinds of cross‐linking agents, and immunized mice with these. The reactivities of these antisera with bovine serum albumin (BSA) conjugated with five different crosslinkers were examined. Antisera from mice immunized with His‐BS3‐KLH, His‐DTSSP‐BSA or His‐GA‐KLH were highly reactive. Competitive ELISA was performed with these antisera, and His‐specific antibodies were efficiently detected when His‐GA‐KLH was immunized and His‐GA‐BSA was used as the solid‐phase antigen. Therefore, we immunized mice four times with His‐GA‐KLH and performed cell fusion of spleen cells of mice in which His‐specific antibodies could be detected. Cloning was performed more than twice by the limiting dilution method, and 9 clones of stable anti‐His mAb‐producing hybridomas (His.1–9) were established. Optimized competitive ELISA using His.6 mAb, which had the highest reactivity to His, was able to detect 0.04 μg/mL of His. n the future, it is expected that these antibodies will be used to establish a highly sensitive immunochemical analysis method for His.departmental bulletin pape

    糖質制限食がストレプトゾトシン誘発性糖尿病ラットの耐糖能に及ぼす影響

    Diabetes is a chronic disease that occurs when insulin production by the pancreas is reduced or when the body cannot effectively use the produced insulin. Type 2 diabetes is a serious problem among middle‐aged and elderly individuals. Therefore, a healthy and balanced diet, moderate exercise, and proper weight control are effective in preventing and alleviating type 2 diabetes. In recent years, a carbohydrate‐restricted diet (a very low‐carbohydrate diet) has been recognized as effective for the prevention and alleviation of diabetes. In this study, we investigated the effects of moderate and severe carbohydrate‐restricted diets on glucose tolerance in streptozotocin (STZ)‐induced diabetic rats, an insulin‐deficient diabetes model. Forty‐eight male Wistar rats (3‐week‐old) were obtained, half of which were administered STZ to induce diabetes. Both groups of rats were fed moderate and severe carbohydrate‐restricted diets for four weeks. We confirmed that both moderate and severe carbohydrate‐restricted diets increased fasting serum glucose levels and did not improve glucose tolerance in STZ‐induced diabetic rats. However, firm conclusions regarding carbohydrate restriction have not yet been reached, and further research is needed to investigate the long‐term effects and metabolic mechanisms of carbohydrate restriction.糖尿病は、インスリン分泌不全およびインスリン抵抗性により高血糖状態が持続する慢性疾患である。2型糖尿病は中高年者にとって深刻な問題であり、その予防・改善のためには、健康的でバランスの取れた食事、適度な運動、適切な体重管理が必要となる。近年、糖尿病の予防・改善に糖質制限食(超低糖質食)が有効であることが報告されている。本研究では、ストレプトゾトシン(STZ)誘発性糖尿病ラットの耐糖能に及ぼす中程度および厳しい糖質制限食の影響を検討した。48匹の3週齢Wistar系雄ラットの半数にSTZを投与して糖尿病を誘発させ、両群のラットに中程度および厳しい糖質制限食を4週間摂取させた。その結果、STZ誘発性糖尿病ラットにおいて、中程度および厳しい糖質制限食はいずれも空腹時血清グルコース濃度を増加させ、耐糖能を改善しなかった。糖質制限食に関する明確な結論はまだ得られていないため、糖質制限食の長期的な効果と代謝メカニズムを明らかにするためには、さらなる研究が必要である。departmental bulletin pape

    Serum carbohydrate antigen 19-9 and metabolite hypotaurine are predictive markers for early recurrence of pancreatic ductal adenocarcinoma

    香川大学Kagawa University博士(医学)Objective: A significant number of patients experience early recurrence after surgical resection for pancreatic ductal adenocarcinoma (PDAC), negating the benefit of surgery. The present study conducted clinicopathologic and metabolomic analyses to explore the factors associated with the early recurrence of PDAC.Materials and methods: Patients who underwent pancreatectomy for PDAC at Kagawa University Hospital between 2011 and 2020 were enrolled. Tissue samples of PDAC and nonneoplastic pancreas were collected and frozen immediately after resection. Charged metabolites were quantified by capillary electrophoresis-mass spectrometry. Patients who relapsed within 1 year were defined as the early recurrence group.Results: Frozen tumor tissue and nonneoplastic pancreas were collected from 79 patients. The clinicopathologic analysis identified 11 predictive factors, including preoperative carbohydrate antigen 19-9 levels. The metabolomic analysis revealed that only hypotaurine was a significant risk factor for early recurrence. A multivariate analysis, including clinical and metabolic factors, showed that carbohydrate antigen 19-9 and hypotaurine were independent risk factors for early recurrence ( P = 0.045 and P = 0.049, respectively). The recurrence-free survival rate 1 year after surgery with both risk factors was only 25%.Conclusions: Our results suggested that tumor hypotaurine is a potential metabolite associated with early recurrence. Carbohydrate antigen 19-9 and hypotaurine showed a vital utility for predicting early recurrence.doctoral thesi

    心理臨床家が傷つくこととお金という2つの観点から、心理療法や心理臨床家のあり方、教育などについて考察した。心理臨床家には、心理療法をとおしての傷つき、実人生での傷つき、教育・訓練の中での傷つきがある。だが、傷つきを手がかりにクライエント理解が深まり、傷つきから回復する過程が心理臨床家を成長させる。そのため、傷つきに対する耐性やレジリエンス、反脆弱性が心理臨床家には求められる。また面接料金などのお金は、心理療法に現実検討をもたらす。クライエントが限られた人生の中の時間とお金と労力を費やして自分自身へと向き合う姿に触れることが、心理臨床家の傷つきに意味を付与し、回復を促すものとなる。departmental bulletin pape

    Reduction rate of anti-acetylcholine receptor antibody titer levels is an early prognostic indicator for myasthenia gravis

    香川大学Kagawa University博士(医学)Background: A realistic treatment goal for myasthenia gravis (MG) is achieving minimal manifestations or better status with prednisolone at ≤5 mg/day (MM-or-better-5 mg), considering a patient's health-related quality of life. Prognosis prediction during the early phases of immunotherapies might be critical for determining subsequent treatment strategies; however, the appropriate biomarkers remain unknown.Aim: This study aimed to clarify whether the reduction rate of anti-acetylcholine receptor antibody (RR-AChR Ab) titer levels is a useful biomarker for predicting MM-or-better-5 mg achievement.Methods: We retrospectively investigated patients with MG and AChR Abs who received immunotherapy for the first time. The RR-AChR Ab titer levels were calculated in the early (within 30 days), middle (31–60 days), and late (61–100 days) periods after starting immunotherapies. A receiver operating characteristic (ROC) curve was generated to determine an appropriate cutoff value for RR-AChR Abs to achieve an MM-or-better-5 mg.Results: Of 53 patients, 24 (45%) achieved MM-or-better-5 mg after 1 year. For the early period, the RR-AChR Ab cutoff value to predict MM-or-better-5 mg was 1.68%/day with an area under the curve (AUC) of 0.75 (sensitivity, 85%; specificity, 70%). However, the middle and late posttreatment AUC values did not predict MM-or-better-5 mg achievement.Conclusion: The RR-AChR Ab might be an appropriate prognostic biomarker during the early period of MM-or-better-5 mg achievement. In the era of early fast-acting treatment strategies, the RR-AChR Ab trend after starting immunotherapies may guide the subsequent treatment choices.doctoral thesi
