    MMP-7 expression is associated with a higher rate of tumor spread through air spaces in resected lung adenocarcinomas

    香川大学Kagawa University博士(医学)Objectives: The spread through air spaces (STAS) of adenocarcinoma (ADC) is a unique pattern for local invasion, which comprises the spread of tumor cells within air spaces beyond the tumor edge without a direct connection with the primary tumor. Matrix metalloproteinase-7 (MMP-7), a secreted proteolytic enzyme that degrades various extracellular matrix components and other substrates, regulates several pathophysiological processes as well as the occurrence and development of cancers in humans. Here, we retrospectively analyzed a cohort of Japanese patients with treatment-naive, surgically-resected lung ADC to assess whether MMP-7 is associated with STAS development and if it could be used as a predictor of STAS.Materials and methods: We performed histological evaluation using hematoxylin and eosin staining and immunohistochemical analysis using microarrays. Thereafter, we scored the examined tissues for immune markers to identify significant tumor STAS predictors.Results: We identified that high MMP-7 expression is an independent predictor of a high STAS incidence. Multivariate analysis revealed that MMP-7 expression was correlated with tumor behavior and poor prognosis. Furthermore, STAS remained significantly associated with a higher risk of ADC recurrence.Conclusion: The development of tumor STAS could be promoted by the functioning of MMP-7. This study could be a crucial basis for future investigations on the detection of tumor STAS.doctoral thesi

    Evolution and function of galectins in Xenopus laevis: Comparison with mammals and new perspectives

    Galectins are metal-independent sugar-binding proteins that recognize galactose (the β-galactoside structure) and regulate the cross-linking of sugar chains between cells and the extracellular matrix. Their specificity for galactose is attributed to their highly conserved carbohydrate recognition domain. Galectins participate in biological processes across species, including development, differentiation, morphogenesis, tumor progression, metastasis, immunity, and apoptosis. However, the relationship between the binding of galectin to sugar chains (glycans) and their biological functions remains unclear. Thus, a comprehensive functional analysis of galectins is required to better characterize their evolutionarily conserved and unique functions. We have previously identified and characterized 12 Xenopus laevis galectins (xgalectins), the only non-mammalian vertebrate species in which galectins have been comprehensively characterized to date. In this review, we present the latest findings on the types and functions of xgalectins and discuss prospects for elucidating their diverse functions from an evolutionary perspective through comparisons with mammalian galectins.journal articl

    Efficient production of D-sedoheptulose using transketolase from Thermus thermophilus HB8 and epimerization reaction using D-tagatose 3-epimerase from Pseudomonas cichorii ST24

    Heptose is a rare monosaccharide with seven carbon atoms. In this study, we demonstrated that two heptoses can be produced cost-effectively and efficiently using transketolase from Thermus thermophilus HB8 (TtTK). First, alternative substrates to a conventional donor substrate, lithium hydroxypyruvate, were investigated, suggesting that 5-keto-D-fructose (5-KF) was the best donor substrate among the 14 donor substrates evaluated. Using 5-KF and aldopentoses as acceptors, we confirmed that four heptoses could be produced, and that the optimal reaction temperature and donor and acceptor substrate concentration are 50°C and 500 mM, respectively. Furthermore, the D-sedoheptulose produced by the partially purified TtTK was converted to D-allopeptulose through the epimerization by D-tagatose 3-epimerase (D-TE) from Pseudomonas. cichorii ST24. The equilibrium ratio of the epimerization reaction between D-sedoheptulose and D-alloheptulose was 45:55. This is the first report that D-TEcould catalyze epimerization reaction between D-sedoheptulose and D-alloheptulose.journal articl

    Efficacy of photodynamic diagnosis for non-muscle invasive bladder cancer: Exploratory subject-based analysis in a prospective, single-arm, multicenter phase III trial

    Background This analysis utilized a subject-based analysis of data from the prospective single-arm phase III trial (jRCT2061210055) to evaluate the efficacy of photodynamic diagnosis (PDD) when 5-aminolevulinic acid hydrochloride (5-ALA) was administered 4–8 h before transurethral resection of bladder tumors (TURBT).Methods This study evaluated 144 patients suspected of having non-muscle invasive bladder cancer (NMIBC), including 127 with NMIBC (38 patients had normal-to-flat-appearing cancer) and 17 without cancer. The diagnostic accuracy and clinical utility of blue light (BL) during TURBT were assessed for each patient and compared with those of white light (WL).Results Subject-based analysis revealed that WL alone missed tumor lesions in 33.1 % of patients, whereas BL alone missed 3.1 % in a biopsy-based analysis (P < 0.001). WL alone failed to diagnose NMIBC in 4.7 % of patients, while BL missed 1.6 %. Adding BL to WL led to an upgrade of NMIBC risk classification in 12 of 127 patients (9.4 %), including 5 upgraded to high-risk and 4 to highest-risk categories.Conclusion This study is the first to demonstrate that PDD's efficacy is maintained up to 8 h after oral 5-ALA administration and indicates that PDD improves the detection rate of bladder cancer, potentially optimizing risk stratification and offering optimal additional treatment.journal articl
