Melatonin and vitamin C were tested on 155 reproductive tracts (310 ovaries) of local Iraqi sheep in aimed to investigate their effects on blastocyst generation. Oocytes were counted and cultured for 24 hours in a CO2 incubator with melatonin and vitamin C as antioxidants. Fresh semen was collected from two fertile rams by electroejaculator with the addition of heparin for capacitation at a rate of 10pg/ml. Matured oocytes grade good and fair were incubated in a CO2 incubator for 24 hours. Fertilised oocytes were cultivated in TCM-199 media with melatonin and vit c. Daily media replacement was 50%. The divided zygote developed every 24 hours until it formed a blastocyst after 168 hours and hatched after 216 hours after fertilisation. Addition of melatonin increased maturation rate to 60% (27/45) from 37.7% (20/53) with vitamin C and 22.2% (4/18) without antioxidants. Melatonin significantly differed (p<0.05) from vitamin C or control groups in the maturation group. The melatonin group had 59.2% (16/27) fertilisation, while the vitamin C 65% (13/20). There was no significant difference between groups. Maturing and fertilisation rates differed between the treated and control groups 22.2% (4/18) and 25% (1/4), respectively (p<0.5). With melatonin, blastocyst production was 62.5% (10/16). It was 53.8% (7/13) in vitamin C. Blastocysts were absent in the control group. Blastocyst production differed significantly (p<0.5) between the melatonin, vitamin C, and control groups. It was concluded from this study that the addition of an antioxidant (melatonin or vitamin C) could improve IVM, IVF, and IVEP in sheep

    No full text
Salmonella spp. are negative gram bacteria, rod-shaped; it belong to Enterobacteriaceae family. Salmonella is a wide variety of species that causes many diseases in humans and animals. This investigation was carried out to study the molecular investigation of Salmonella spp. from broilers. It was determined that approximately one hundred broilers were suffering from diarrhea, so feces and cloacal swabs were collected from them. These swabs were cultured on various media before sub-culturing on SS. agar and XLD. Following this, biochemical tests were performed to identify the bacteria. DNA extraction was carried out per the company's guidelines, and PCR was carried out to look for the invA gene, which is unique to Salmonella species. According to the findings, the percentage of individuals infected with Salmonella species was 17.39%. During this investigation, PCR was utilized to locate the in vA gene, and out of the eight tested samples, Salmonella species were identified in 8of them. Despite the severe and dangerous effects of the Salmonella pandemic on the agricultural and economic sectors as well as on human health, the findings showed that the PCR method had a high specificity in detecting Salmonella spp. when compared to other traditional methods of spotting the bacteria

Statistical Analysis of Compressive Strength of Concrete

    No full text
Concrete is used in the construction practice as it is easily available, cheap, has flexibility of handling and can be shaped in any form desired. The use of concrete in the construction industry is now being done for a very long time. It is the most used material in the parts of structure where compressive strength is required. So in the construction process evaluation of the compressive strength of concrete plays a vital role. There is no universal method for the evaluation of the compressive strength. In India, the method prescribed by IS 456:2000 is used for checking whether the concrete made is fulfilling the requirement. An attempt is made to analyses this method statistically. The characteristic strength of the concrete is represented by test results of concrete cubes at 28 days. The compressive strength of concrete in India is defined based on this characteristic compressive strength. The sampling process of the concrete which is based on the amount of work of concrete to be done is studied and a trend between the number of samples and the failure percentage of samples in terms of compressive strength is studied. The concept of population and sample in terms of the concrete mix design is studied. Hypothesis testing is performed on various groups of samples and their deviation from the population is calculated. An attempt to calculate optimal number of samples for testing and an improvised method using statistics as a tool for checking the compressive strength of concrete is made
