PRELIMINARY STUDIES ON ISOLATION AND SCREENING OF ANTIBIOTIC PRODUCING SYMBIOTIC BACTERIA FROM STARFISH (Protoreaster nodosus) COLLECTED FROM COASTAL AREA TAKALAR REGENCY, INDONESIA
Marine organisms are well known for the availability of bioactive compounds which have various biological activities including antibacterial activity. Likewise, their symbiotic bacteria can also produce compounds that have similar activities. The purpose of this study was to isolate and screen the symbiotic bacteria from starfish (Protoreaster nodosus) collected from coastal area Takalar Regency, South Sulawesi, Indonesia. Isolation was carried out by the pour plate method using nutrient agar medium dissolved in sterile seawater. The isolated symbiotic bacteria were purified by using the quadrant method. The pure isolate was culture through submerged fermentation using nutrient broth media enriched with 1% yeast extract and sterile seawater for 7 days. The selected symbiont bacterial isolates were tested for their antibacterial activity against gram-positive and gram-negative bacteria using disc diffusion assays. The results of fermentation were separated from the biomasses and tested for antibacterial activity against Staphylococcus aureus (S. aureus, ATCC 25923), Bacillus subtilis (B. subtilis, ATCC 6633), Salmonella typhi (S. typhi, NCTC 786), and Escherichia coli (E. coli, ATCC 25923). The results of study revealed that four symbiotic bacteria (SB 1T, SB 2T, SB 3T, and SB 4T) were successfully isolated. All the SB isolates have good antibacterial activity against all tested bacterial strains with an average diameter of inhibition zone larger than 11 mm. Among all isolates, isolate SB 4T showed a remarkable size of zones growth inhibition (> 15 mm) against all tested bacterial strains. Thus, the symbiotic bacteria isolated from P. nodosus in this study have a promising broad-spectrum antibacterial activity
DECIPHERING THE PHARMACOLOGICAL INSIGHTS OF FRACTIONATED ELATOSTEMA PAPILLOSUM WED. AND HOLIGARNA LONGIFOLIA ROXB. THROUGH IN VITRO AND IN VIVO STUDIES
The present research intended to explore the biological activities, namely acute toxicity test and hypoglycemic as well as in vitro anti-arthritic along with the antibacterial activity of crude methanol extracts with its different soluble fractions like petroleum ether (PESF), carbon tetrachloride (CTCSF), chloroform (CSF) and aqueous soluble fraction (AQSF) of Holigarna longifolia and Elatostema papillosum. Phytochemical screening was performed by established protocols. Acute toxicity and hypoglycemic effects were performed in experimental and alloxan-induced diabetic rats. In vitro anti-arthritic and antibacterial activity were conducted by protein denaturation inhibitory and disc diffusion methods. It was observed that no rats exhibit any lethality types, which reveal the safety of plant fractionates. It was also seen that both plants' fractionates showed significant (p < 0.01) activity on hyperglycemia compared to standard. Upon investigation, it was observed that crude methanol and its CS fraction of E. papillosum and only CS fraction of H. longifolia significantly (p < 0.05) inhibited denaturation of bovine serum albumin protein compared to standard diclofenac sodium. Moreover, it was observed that crude methanol extract and its CS fraction of E. papillosum showed significant inhibitory action on all Gram-positive bacteria's growth. In contrast, the PES fraction highlighted an inhibitory zone of 26.7 and 24.7 mm, respectively, towards B. subtilis and S. aureus. This study provides some support to explain the traditional uses of H. longifolia and E. papillosum. 
IN VIVO AND IN VITRO EVALUATION OF PHARMACOLOGICAL ACTIVITIES OF Hedychium coccineum RHIZOMES EXTRACT
The study reports the in vivo antidiarrheal and in vitro anthelmintic, cytotoxic, and thrombolytic activity of methanol extract of Hedychium coccineum rhizomes (MEHC). The antidiarrheal activity was determined using Castor oil-induced diarrhea and Gastrointestinal motility test in mice at the doses of 200 and 400 mg/kg body weight, whereas an aquarium worm, Tubifex tubifex, was used to determine the anthelmintic activity. The cytotoxic and thrombolytic activity of MEHC was performed by Brine shrimp lethality bioassay and clot lysis method respectively. In antidiarrheal, castor oil-induced diarrhea and gastrointestinal motility exhibited a significant reduction in diarrhea and defecation and an extremely significant inhibition in intestinal motility and peristalsis index by 200 and 400 mg/kg of MEHC. The MEHC (5, 10, and 20 mg/mL) showed a significant dose-dependent manner paralysis time and times to death in multiple comparisons to the different levamisole concentrations (0.5, 0.8, and 1 mg/mL) at in vitro anthelmintic activity. The brine shrimp lethality bioassay exhibited a weak LC50 (681.95 µg/mL; R² = 0.951) while in thrombolytic a significant percentage of clot lysis (32.70%, P < 0.05) demonstrated. The findings demonstrate that H. coccineum rhizomes could be potential sources for biological activity
EFFECT OF INDIVIDUAL AND COMBINED APPLICATION OF Trichoderma sp. AND VERMICOMPOST ON THE MANAGEMENT OF Sclerotium rolfsii AND GROWTH OF CHILLI UNDER PEATLANDS AGRO-CLIMATIC CONDITIONS
This study was conducted to evaluate the biocontrol potential of Trichoderma sp., and vermicompost on the management of Sclerotium rolfsii and chilli plant growth under peatlands agro-climatic conditions. The research was conducted under the laboratory as well field condition of Palangka Raya University in a completely randomized factorial design. As a treatment, four doses of Trichoderma sp. i.e. 0, 5, 10, and 15 g/box, and three doses of Vermicompost i.e. 0, 100, and 200g/ box was used individually and in combination and each treatment have three replications. Three kilograms of sterilized peat soil planting media were placed in the 36 plastic boxes and Trichoderma sp., and vermicompost was applied 1 week before the chilli seeds are planted. After 10 weeks of plantations, observations related to the disease intensity and chilli growth and yield were recorded. The results of the study revealed a synergistic effect of Trichoderma sp. and vermicompost combination in controlling the stem rot disease caused by S. rolfsii. Individual and combined application of Trichoderma and vermicompost had an effective range of 73.33-100% over the control. Further, among the various tested treatments, a combination of Trichoderma sp @ 5g/box with vermicompost @ 200g/box and individual application of Trichoderma sp @ 10g/box were found superior in the management of stem rot disease. In the case of chilli growth parameters, the results of the study suggested that the application of vermicompost had a significant effect on the chilli plant's growth and yield, while the application of Trichoderma sp. significantly affects the chilli plants number of leaves
VIRTUAL SCREENING AND IDENTIFICATION OF PLAUSIBLE NOVEL THERAPEUTIC EGFR INHIBITORS AGAINST BREAST CANCER
Present days increasing concern about the identification of potential non-toxic drug candidates against several cancers is very important. The current study was carried out to discover the novel phytochemicals as effective anticancer agents against the selected protein (i.e., EGFR), which is a promising target for moderating triple-negative breast cancer (TNBC). Various studies showed that the natural constituents have a strong anti-tumor capacity and inhibiting tumor growth. Here structure-based virtual screening and molecular docking studies have been recognized as rational tactics for the recognition of novel drug candidates against the binding domain of EGFR (PDB code: 3GKW & 5FEE). Furthermore, the drug-likeness, adverse effects, and toxicogenomics effects were assessed with the help of various computational tools. Virtual screening was reported that 4 drug candidates i.e., CID: 65064; CID: 5280443; CID: 440735, and CID: 5280343 showed reliable consequences with fewer side effects and more efficient for the selected proteins. The overall effects indicated that renowned hits could be developed as reference skeletons for novel inhibitors envisaging EGFR to ameliorate TNBC
COMEDICATION OF RABEPRAZOLE SODIUM CAUSES POTENTIAL DRUG-DRUG INTERACTION WITH DIABETIC DRUG LINAGLIPTIN: In-vitro AND In-silico APPROACHES
Drug-drug interaction is a notable concern among physicians when prescribing multi-therapy to the patients as concomitant administration of multi-drugs might cause unexpected adverse drug reactions. The main objective of this research is to predict a potential drug-drug interaction between two frequently used drugs by diabetic patients, an antidiabetic drug (linagliptin) and a proton pump inhibitor (rabeprazole sodium). Here, several in vitro techniques, including thermal (melting point, thermogravimetric analysis [TGA]), morphological (scanning electron microscopy [SEM] and X-ray powder diffraction [XRPD] analysis), highly sophisticated synchronous fluorescence, and in silico methods were applied to anticipate the potential drug-drug interaction between these stated drugs quickly. The melting point and TGA study revealed thermochemical properties, thermal stability profiles, and degradation patterns upon temperature rising of the formed complex and these precursor drugs. The SEM and XRPD have provided the morphological changes like particle shape and size distribution of the desired molecule that might be caused due to the potential drug-drug interactions. Besides, the drastic reduction of the quenching rate constant of linagliptin during interaction with bovine serum albumin in synchronous fluorescence also endorsed the potential drug-drug interaction. Furthermore, the drug-receptor docking analysis demonstrated that the binding affinity of the precursor ligands might be reduced due to the predicted drug-drug interaction. However, the current evidence warrants extensive investigation to confirm the above-stated potential drug-drug interaction in the larger animal model. Finally, clinical data need to be closely monitored during the treatment of diabetic patients prescribed with linagliptin and rabeprazole sodium
BIPLOT ANALYSIS FOR IDENTIFICATION OF SUPERIOR GENOTYPES IN A RECOMBINANT INBRED POPULATION OF WHEAT UNDER RAINFED CONDITIONS
Physiological traits of wheat genotypes and their trait relation to drought conditions are important to identify the genotype in target environments. Thus, genotype selection should be based on multiple physiological traits in variable environments within the target region. This study was conducted at Punjab Agricultural University during rabi crop seasons 2012-13 and 2013-14 to study the recombinant inbred lines (RILs) of wheat genotypes derived from traditional landraces and modern cultivars (C518/2*PBW343) based on various morpho-physiological traits. A total of 175 RILs were selected for this study based on various tolerance indices. The genotype by trait (GT) biplot analysis was applied to data from seven high-yielding RILs grown under irrigated (E1) and rainfed environments (E2). The GGE biplot explained 100% of the total variation for chlorophyll content, grain filling period, peduncle length, water-soluble carbohydrates, grain number, grain yield, and 95.1% for canopy temperature, 94.9% for thousand-grain weight. GT-biplots indicated that the relationships among the studied traits were not consistent across environments, but they facilitated visual genotype comparisons and selection in each environment. RIL 84 and RIL108 were close to the average environment (ideal genotype) for all traits studied except chlorophyll content. A well-performing genotype with great environmental stability is called an "ideal genotype. Among all entries, these genotypes performed well. Therefore, among the traits studied, grain filling period, peduncle length, canopy temperature, water soluble carbohydrates, and 1000 grain weight contributed to grain yield under a stress environment. Furthermore, it may be used as a donor material in breeding programs and QTLs mapping
IMPACT OF MUSIC AS MOTIVATION TO PERFORM EXERCISES AMONG UNIVERSITY STUDENTS – A SURVEY
The current study aimed to examine the association between music and motivation to do exercise among university students in Nilai, Negeri Sembilan, Malaysia. The demographics data, use of music during exercise, preferred type of music, and criteria to select music were assessed by a validated self-administered questionnaire. The motivation towards exercises was evaluated using the Behavioural Regulations in Exercise Questionnaire (BREQ-3) while the level of physical activity was assessed by Godin-Leisure Time Exercise Questionnaire (LTEQ). The demographics information, use of music during exercise, preferred type of music, and criteria to choose music were tabulated using descriptive statistics. A point-biserial correlation was used to analyze the relationship between music and motivation to do exercise while chi-square was utilized to determine the association between the use of music during exercise and physical activity level. The results of the current study showed that 80.3% of the participants listen to music during exercise. Energetic and rhythmic was the preferred type of music for exercise. Tempo/ speed/ bpm was the most popular factor to be considered during the exercise. Most of the participants prefer to listen to an individual music player during exercise rather than an open audio system.. Listening to music during exercise shown significant correlation with a motivation (p=0.006), external regulation (p=0.014), identified regulation (p=0.006), integrated regulation (p=0.002) and intrinsic regulation (p=0.015). There was a significant association between the use of music during exercise and physical activity level (p=0.003) in this study. Future research that involves the type of exercise performed with the music is encouraged to explore the significance of music as a motivational tool in exercise
STUDY OF ACUTE AND SUB-ACUTE TOXICITY OF Boehmeria virgata (FORST) GUILL LEAF EXTRACT IN WISTAR RATS
This study was carried out to evaluate the acute and sub-acute toxicity of the standardized Boehmeria virgata leaf extract (BLVE) in Wistar rats. B. virgata is a traditional herb utilized by the people of Makassar, Indonesia to cure inflammation and cancer. In the current study, thirty (30) Wistar rats were divided into 6 groups (3 groups of males and 3 groups of females) for acute treatments. Similarly, for investigating sub-acute treatment forty (40) Wistar rats were split into 8 groups (4 groups of males and 4 groups of females). For acute toxicity treatment, selected rats received 2000 mg and 5000 mg/kg of BVLE by oral administration for 14 days while for the estimation of sub-acute toxicity, experimental rats were treated with 250, 500, and 1000 mg/kg BVLE for 28 days. After 14 days of treatment, the rats were monitored for any form of changes in behavior, weight, food, water intake, and histopathology. The treated animals underwent hematological, biochemical, histopathological, and organ weight analysis after 28 days. Results of the study revealed no significant differences in body and organ weight, intake of food, and water after acute BVLE treatment in rats compared with those in the control group. The histopathological study suggested a general hydropic degeneration of the liver after acute treatment with 5000 mg/kg; such degeneration did not occur in the kidneys and kidney glomerulus of BVLE treated rats. Further, no significant toxicity was shown in hematological, biochemical, organ weight, and histopathological data in the sub-acute BVLE group after comparing treated rats with the control group. Results of the study can be concluded that BVLE is not toxic at doses of up to 1000 mg/kg. Therefore B. virgate fulfilled a preclinical criterion that is necessary for its further establishment as a clinically useful extract
EFFECT OF HONEY CONSUMPTION AFTER PHYSICAL EXERCISES ON ELECTROLYTES AND BLOOD GLUCOSE LEVELS
Honey has been used as food and medicine for thousands of years. The purpose of this study was to determine the effect of honey in restoring the levels of electrolytes and blood glucose after performing physical exercises. The effect of honey consumption on electrolyte and blood glucose levels was examined on12 healthy male subjects, which were divided into 4 groups with three members in each group. Group I was treated as control and provided only 250 ml of mineral water after exercise, while group II was provided 250 ml commercial isotonic beverage, group III provided honey solution 1 (15 ml honey/250 ml water), and group IV provided honey solution 2 (45 ml honey/250 ml water). The physical exercise performed by the subjects is running on a treadmill at a speed of 5.6 km/h for 40 minutes. Measurements of electrolyte and blood glucose levels were performed 24 hrs before as initial baseline, and these were also measured after the physical exercise as well as after the treatment. Results of the study revealed that honey played a significant role in the restore electrolyte and blood glucose levels in people who have performed physical activities such as exercising and the effect of honey is similar to the commercial isotonic beverage. However, no significant difference (p-value > 0.05) was reported between the honey solution 1 and 2 and other treatment groups in elevating sodium and chloride level
