Head and Neck Squamous Cell Carcinoma: Cuproptosis-related Long Non-coding RNAs Predict Prognosis and Immunotherapeutic Response
Objective Head and neck squamous cell carcinoma (HNSCC) has a poor prognosis because of high recurrence and metastasis rates and failure of immunotherapy caused by escape of immune cells. The aim of this study was to explore potential biomarkers and precise drug therapies for HNSCC.
Methods We extracted the RNA transcriptome dataset and related clinical data of patients with HNSCC from The Cancer Genome Atlas (TCGA) database to analyze differentially expressed long non-coding RNAs (lncRNAs) in HNSCC. We screened differentially expressed lncRNAs related to cuproptosis among regulatory genes and identified those that best indicated the prognosis of HNSCC univariate Cox (uni-Cox) regression analyses and the least absolute shrinkage and selection operator. We constructed models and evaluated their accuracy in determining high- and low-risk groups using the Kaplan–Meier, receiver operating characteristic curve, and uni-Cox regression and multivariate Cox (multi-Cox) regression analyses. Subsequently, we subjected the high-risk group to gene set variation analysis, principal component analysis, immunoassay, and half-maximal inhibitory concentration prediction. To further improve tumor immunotherapy, we divided the entire sample into cold and hot groups based on cuproptosis-related lncRNAs and analyzed the therapeutic differences. Results Combined with cuproptosis-related genes and by analyzing the HNSCC data from TCGA, we obtained five cuproptosis-related prognostic lncRNAs. The model showed certain reliability. Patients in the high- and low-risk groups had different tumor microenvironments and drug sensitivities. Cuproptosis-related lncRNA expression differed between cold and hot tumors. Further, hot tumors were more sensitive to immunotherapy drugs compared to cold tumors.
Conclusion This study established and verified the lncRNAs that stratified the prognostic risk related to cuproptosis in HNSCC. The model also predicted immunotherapeutic responses of patients with HNSCC and provided a reference for the individualized treatment of cold and hot tumors
From Lelia’s and Mrs. Albrecht’s Classrooms to Explore Who is the Native Speake
Chang-Rae Lee’s novel, Native Speaker, narrates the story of the protagonist from a first-person perspective. Through observing his wife’s teaching instructions, Henry recalls his own experiences in an English classroom during his youth, where Mrs. Albrecht’s harsh methods and classmates' mockery eroded his confidence and even caused serious anxiety. The descriptions of Mrs. Albrecht’s classroom demonstrate the traumatic experience of marginalized immigrant children. Henry Park, a 1.5 generation of Korean-American immigrant, imitates a polished English accent to be assimilated into American mainstream society. However, his excessive carefulness to details prevents him from becoming a true “native speaker”, and from the inner world, he is simultaneously influenced by both Korean culture and American culture. Henry’s “dual identity” leads him to be employed by an immigration intelligence agency, and he is assigned to infiltrate the camp of Korean-American politician--John Kwang, a candidate for the city mayor. In his interactions with John Kwang, Henry is confronted with an identity crisis as an Asian-American. Consequently, the issues of language, identity, and the political rights of minority ethnic groups arise.
Cultivation of Chinese-to-Foreign Translation Talents in the Context of New Liberal Arts
Under the background of new liberal arts, the cultivation of Chinese-to-foreign translation talents faces new opportunities and challenges. This study systematically analyzes the current status and problems of the cultivation of Chinese-to-foreign translation talents through methods such as questionnaire surveys and in-depth interviews, and proposes corresponding improvement strategies. The research finds that the current cultivation of Chinese-to-foreign translation talents has problems such as monotonous curriculum settings, conservative teaching methods, and insufficient practical teaching, which are difficult to meet the diverse needs for translation talents under the new liberal arts background. In response to these problems, this study suggests starting with optimizing the curriculum system, innovating teaching methods, and strengthening practical teaching to construct a cross-disciplinary integration, practice and innovation-oriented Chinese-to-foreign translation talent cultivation model. At the same time, the study also emphasizes the key role of teachers in the cultivation process, proposing to promote the overall improvement of the teacher team through enhancing teachers' professional quality and strengthening teacher training. In addition, this study also pays attention to the latest trends and market demands of the translation industry, and proposes a cultivation strategy that combines the concept of new liberal arts with market demands, aiming to cultivate more Chinese-to-foreign translation talents with innovative spirit, practical ability, and cross-cultural communication skills
