bonisierung – stellt neue Anforderungen an Arbeitskräfte in diversen Bran-
Sustainability at the Watt En Schlick Festival - A sustainability analysis using the example of the Watt En Schlick music festival taking into account regional and national characteristics
Between Input and Output: The Importance of Modelling Transients in Meal Preparation Tasks
We are moving closer to autonomous robots preparing meals. While restaurant robots in static environments already are successfully performing single actions like making pizza, the goal is to enablerobots to perform changing actions, in various environments and with any available object. Towards this goal, a methodology for creating actionable knowledge graphs that can be used to parameterise general action plans has been proposed. However, for extended failure handling towards fully automated action execution, we argue that transients need to be considered. A transient can be described as a transitory object in a task that is not the same as the input object anymore but not yet the output object of the task. For example, when pouring ingredients into a bowl to make the dough, the added ingredients form a mass of ingredients (here: a transient) that only becomes dough through mixing them. This work shows how transients can be modelled and how robots can integrate and possibly benefit from this modelling

The ecology and evolution of the extracellular vesicles of halophilic Archaea
Extracellular vesicles (EV) are small, spherical structures that bud from the cell envelope. These particles are produced by cells across all three domains of life, marking them as a ubiquitous feature of cell biology. EVs transport proteins, lipids, nucleic acids, and other bioactive compounds in membrane-enclosed packages, which allows them to mediate a multitude of different functions. While much has been uncovered for bacterial and eukaryotic EVs, little is known about the functions and production of EVs from the archaeal domain.
The work presented in this dissertation aims to explore the mechanisms of production, biochemical composition, and function of EVs produced by halophilic Archaea. In Chapter II, I present a standardized method for the isolation and purification of EVs from archaeal cultures. This method details the isolation of EVs from liquid cultures, the purification from other extracellular contaminants, and protocols related to the subsequent downstream analysis. By providing a standardized protocol, I aim to promote additional research into archaeal EV production that is comparable to each other. Using these methods in Chapter III, I characterize the biochemical composition of EVs from Haloferax volcanii and other halophilic Archaea. I uncover that halophilic Archaea produce EVs enriched with small-sized RNA with regulatory potential, which has not been demonstrated for archaeal EVs previously. Proteomic analysis reveals a conserved small GTPase that is crucial to EV production, which we have named archaeal vesiculating GTPase (ArcV). Structurally and functionally, ArcV bears similarities to the Arf-family of GTPases, which are responsible for regulating vesicle formation in the eukaryotic endomembrane system. We propose that this provides evidence for previously established hypotheses regarding the archaeal origins for the eukaryotic endomembrane system, and that small GTPase-dependent vesicle formation could have emerged earlier than previously considered. In Chapter IV, I further characterize EV-associated RNA from Halorubrum lacusprofundi, with and without virus infection. I observe that the RNA population exported in EVs changes upon viral stress, leading to the enrichment of specific RNAs. I also explore the effects on the cell transcriptome after incubation with EVs, suggesting that EVs are able to affect gene expression in the receiving cell.
The characterization of archaeal EVs has implications on both microbial community dynamics and prokaryotic evolutionary history. The ability for EVs to induce changes in gene expression in the receiving cell suggests a regulatory mechanism that acts on a population-wide scale. Additionally, the presence of a small GTPase-dependent mechanism for EV production in Archaea requires us to reevaluate our current understanding of prokaryotic membrane remodeling mechanisms and the evolutionary history of eukaryotic-like features
Urban resilience - development of a touristic climate change adaptation concept for the city of Bremen
Past interaction of ocean circulation and West Antarctic Ice Sheet dynamics in the Amundsen Sea Sector
The West Antarctic Ice Sheet is highly sensitive to ocean forcing and is currently experiencing grounding line retreat and ice shelf thinning. Since the response of the ice sheet to a warming world is unknown, it is important to analyze past warmer-than-present periods, e.g. the Pliocene, for comparison with possible future changes. Two drill sites, Site U1532 and Site U1533 of IODP Expedition 379, were linked to a large network of 2D high-resolution seismic reflection data to analyze changes of the West Antarctic Ice Sheet from the latest Miocene to the Pleistocene.
During the Pliocene, between 4.2 to 3.2 Ma, a warm period was recognized with a highly dynamic West Antarctic Ice Sheet, correlating with reduced ocean bottom current activity. This period can be described as an overarching warm period with several advance and retreat phases of the West Antarctic Ice Sheet in the Amundsen Sea Sector
Tidal dynamics and ice cavities in AWI-ESM2: modelling and implications
Earth system models (ESMs) serve as effective numerical tools for studying our planet. Advances in computational capability have provided scientists with the opportunity to explore the Earth’s complex system. While many processes have been considered within individual components of the Earth’s system or in multi-sphere interactions, there remains a significant demand for establishing more integrated ESMs. In this thesis, the author primarily implements a global tide module and an ice-shelf cavity module into AWI-ESM2.
Ocean tides play a crucial role in the global ocean system due to tidal mixing. Tidal mixing is an important process in the deep ocean because it can lift dense water from the depths, thereby maintaining global stratification and the Thermohaline Circulation. Previous studies have examined tidal effects using either an explicit tide model or a parameterisation of tidal mixing. However, comparing these two approaches could yield further insights into the tidal impact on the global ocean and climate. By evaluating both methods within the same model, the author discusses their advantages and disadvantages, contributing to a more comprehensive understanding of the tidal effect on global ocean circulation and climate.
Previous climate modelling simulations often ignore Antarctic ice-shelf cavities due to their relatively small area. However, ice-sheet modelling studies have highlighted the potential for accelerated melting of the Antarctic Ice Sheet in a warming climate. Inspired by this, the author incorporates Antarctic ice-shelf cavities into AWI-ESM2. By comparing simulations with and without these cavities, the results reveal a significant impact on both pre-industrial and future-warming climates. This underscores the crucial role of hydrosphere−cryosphere interactions in the Earth’s system.
Marine geological studies have proposed that during past glacial periods, a kilometer-thick ice shelf covered the Arctic Ocean and Nordic Seas. Additionally, freshwater is hypothesised to have been stored in this vast basin. To explore this further, the author conducts simulations using an assumed pan-Arctic ice shelf in AWI-ESM2. Through two-dimensional idealised experiments, the author identifies two distinct time scales for freshening beneath such an ice shelf. The upper ocean experiences a rapid freshening process, while the lower ocean undergoes a slower freshening process. Theoretical estimates provide insight into the time scale required for freshening the Arctic−Nordic Seas basin in the presence of such an ice shelf.
These implementations in AWI-ESM2 not only validate the studies presented in this thesis but also pave the way for potential research on Earth’s past, present and future climate
Synthese und temperaturabhänige Struktur-Eigenschafts-Beziehungen von Mullit-typ Zinn(II) Verbindungen
Within the scope of this dissertation, precursors for O8 mullite-type phases and the phases themselves were synthesized and studied. Due to unforeseen results, an investigation of dolomite-type compounds is included in this work.
As the investigation of newly synthesized mullite-type compounds was the main part of this dissertation, the remaining part concerns this structure type. To investigate the influence of the lone electron pair (LEP) of Sn2+ on the structure and the difference to the influence of the LEP of Pb2+, different SnMBO4 (M = Al, Ga, Cr, and V) are synthesized by a conventional solid-state synthesis in sealed quartz tubes (chapter 2 - 5). Rietveld refinement of X-ray powder diffraction data indicates that all compounds crystallize in the orthorhombic space group Pnam, where the edge-sharing MO6 octahedra form an infinite chain along the crystallographic c axis and are interconnected by trigonal planar BO3 groups as well as square pyramidal SnO4 groups. To measure the strength of the stereochemical activity of the LEP, Wang-Liebau eccentricity (WLE) parameter was calculated from the structural data which led to a highly influenced structure. Further investigations were done to consolidate the structural features with Sn Mössbauer, solid state NMR, Raman, IR, and UV/Vis spectroscopic measurements
