Instruments to assess gait in individuals with multiple sclerosis: a systematic review
Background: Gait impairment is considered a significant cause of disability and is a clinical feature of Multiple Sclerosis. For this reason, their regular assessment is essential for better management and treatment and to evaluate the effectiveness of rehabilitation. Objective: This study aims to know what instruments are available in the literature to assess gait in patients diagnosed with MS. Data Sources and Searches: A complete literature search was conducted in MEDLINE, Web of Science, Embase, and Scopus, following Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA). The study protocol was registered at the International Prospective Register of Systematic Reviews (PROSPERO). Study Selection: The authors selected studies written in English and representing validation of gait assessment tools exclusively for adults with a diagnosis of MS. Data Extraction and Quality Assessment: Two authors independently extracted the data for reliability, measurement error, validity, and assessed methodological quality using the COnsensus-based standards to select health Measurement Instruments (COSMIN). Tocar, doer, pensar: os efeitos da técnica Alexander em estudantes de música com dores crónicas relacionadas com a performance
Relatório Final de Estágio do Ensino Especializado, apresentado à Escola Superior de
Música de Lisboa, do Instituto Politécnico de Lisboa, para cumprimento dos requisitos à
obtenção do grau de Mestre em Ensino de Música, conforme Decreto-Lei nº79/2014,
de 14 de maio.Resumo I - A parte inicial deste trabalho consiste no relatório de estágio realizado no âmbito do
programa de Mestrado em Ensino de Música.
Este estágio foi dividido em duas etapas. A primeira ocorreu na Escola Artística do
Conservatório Nacional, em regime de observação, com a professora cooperante
Cândida Matos, no período de setembro a janeiro de 2022. A segunda etapa
correspondeu ao período entre janeiro e junho de 2023, em regime docente como
professora substituta no Conservatório de Música de Loulé, tendo a coordenadora
pedagógica Ana Figueiras como professora cooperante.
No primeiro período, foram observadas três aulas por semana, sendo as turmas
compostas por alunas de iniciação, 5º e 12º anos, respetivamente. Por questões
logísticas, ministrei cinco aulas neste período (duas para a aluna de iniciação, duas para
a aluna do 5º ano e uma para a aluna do 12º ano), as quais foram relatadas nos planos
de aula, enquanto as outras foram registadas em fichas de observação.
Nos segundo e terceiro períodos, exerci atividade docente no Conservatório de Música
de Loulé, como professora substituta, lecionando para sete alunos de cravo. Para este
relatório de estágio, selecionei três alunos: uma aluna do 1º grau, um aluno do 4º grau
e um aluno do 7º grau. Foi especialmente interessante trabalhar numa escola pública e
aprender a lidar com a carga burocrática que esta envolve, em comparação com escolas
privadas que já conhecia. Nessa escola, consegui integrar-me na comunidade educativa
com a ajuda dos outros professores. Ao mesmo tempo, pude exercer a minha
autonomia, sempre respeitando o programa aprovado pela comissão pedagógica. A
professora que substituí deixou instruções detalhadas de repertório para cada aluno, as
quais segui fielmente. Também adicionei nas aulas alguns exercícios criados
especialmente para a preparação de determinadas obras, bem como momentos de
improvisação.Resumo II - Numerosos estudos, como o de Gembris et al. (2020), demonstram que pelo menos 60%
dos músicos profissionais sofrem de dor ou distúrbios musculoesqueléticos ao tocar. Em
Portugal, foram realizadas pesquisas sobre a incidência dos problemas de dor
relacionada com o desempenho, ou DRD (também conhecidos como distúrbios
musculoesqueléticos relacionados com a performance), em músicos profissionais (Silva
et al., 2015; Sousa et al., 2017). Contudo, sabe-se ainda muito pouco sobre terapias
eficazes e apropriadas, bem como sobre as formas pelas quais os músicos podem
prevenir tais problemas.
De acordo com Williamon e Thompson (2006), os estudantes de música do ensino médio
apresentam alta incidência de dor relacionada ao desempenho, portanto, há uma
necessidade urgente dos alunos desenvolverem habilidades em autogestão e
autoconsciência para evitar o acumular de tensão muscular e stresse. O estudo de J.
Davies (2020) sugere que aprender estratégias que melhorem os fatores de
desempenho juntamente com a saúde relacionada ao tocar, pode ser mais adequado
para estudantes universitários, interessados em desenvolver as suas habilidades
musicais. A Técnica Alexander (TA), desenvolvida por F.M. Alexander (1867-1955) como
um conjunto de princípios que visam identificar e mudar hábitos de uso
contraproducentes, já é popular entre músicos e artistas do espetáculo em países anglo-
saxónicos, e tem sido associada à melhoria dos sintomas de DRD e qualidade de
desempenho como mostraram os estudos de J. Davies em 2019 e Fun Ying et al em
2015. Porém, até 2014, quando foi realizada a revisão sistemática de U. Klein et al., a
maioria das evidências empíricas dos efeitos da TA em músicos estão relacionadas a
resultados associados à ansiedade de desempenho.
Este estudo de caso visa explorar os efeitos das aulas da Técnica Alexander em vários
alunos da Escola Superior de Música de Lisboa (ESML) e da Escola Artística do
Conservatório Nacional (EAMCN), que sofrem de DRD crónica. Este estudo foi realizado
ao longo de 2 anos, durante os quais cada aluno recebeu de 5 a 10 aulas individuais de
50 minutos dadas pelo autor do estudo, professor da Técnica Alexander desde 2002. O
questionário musculoesquelético nórdico (QMN) desenvolvido por Kuorinka et al. em 1987, com uma escala modificada de Borg (0 = sem dor a 10 = dor máxima) desenvolvida
por Borg em 1982, foi utilizado antes e após a intervenção. Apesar do número reduzido
de aulas, uma melhoria progressiva foi observada em todos os alunos. Alguns deles
chegando ao 1 ou 0 na escala de Borg, após as aulas de TA. Os resultados apoiam os
estudos anteriores mencionados, indicando que aulas de TA projetadas com propósito
podem contribuir para o gerenciamento de DRT e beneficiar habilidades relacionadas
ao desempenho.Abstract I - The initial part of this work consists of the internship report carried out within the
framework of the Master's program in Music Education.
This internship was divided into two stages. The first stage took place at the Escola
Artistica do Conservatório Nacional, in an observation capacity, with cooperating
teacher Cândida Matos, from September to January 2022. The second stage
corresponded to the period between January and June 2023, during which I worked as
a substitute teacher at the Conservatório de Música de Loulé -Francisco Rosado, with
pedagogical coordinator Ana Figueiras as the cooperating teacher.
In the first period, three classes per week were observed, with the classes composed of
beginner, 5th grade, and 12th grade students, respectively. Due to logistical reasons, I
taught five classes during this period (two for the beginner student, two for the 5th
grade student, and one for the 12th grade student), which were reported in lesson plans,
while the others were recorded in observation sheets.
In the second and third periods, I carried out teaching activities at the Loulé Music
Conservatory as a substitute teacher, teaching seven harpsichord students. For this
internship report, I selected three students: one from the 1st grade, one from the 4th
grade, and one from the 7th grade. It was particularly interesting to learn how to work
in a public school, dealing with all the bureaucratic workload that it entails. In this school,
with the help of the other teachers, I was able to integrate into the educational
community. At the same time, I was able to exercise my autonomy, always respecting
the program approved by the pedagogical committee. The teacher I replaced left
detailed instructions regarding repertoire for each student, which I followed faithfully. I
also added some exercises during the classes, specifically created for the preparation of
certain works, as well as moments of improvisation.Abstract II - Performing arts medicine has grown substantially since 1980, and numerous studies
demonstrate that at least 60% of professional musicians suffer from pain or
musculoskeletal disorders when playing according to the study of Gembris, et al. (2020).
In Portugal, research has been conducted on the incidence of performance-related pain
problems, or PRP (or Playing related Musculoskeletal disorders), in professional musicians (Silva
et al., 2015; Sousa et al., 2015). However, very little is known about effective and appropriate
therapies and the ways in which musicians can prevent such problems.
According to Williamon & Thompson (2006), High School music students show a high
incidence of playing related pain (PRP), therefore, there is an urgent need for students
to develop skills in playing self-management to avoid the build-up of dangerous levels of
muscle tension and stress. To learn strategies that enhance performance factors
alongside playing-related health might be better suited to university students,
concerned in developing their musical skills (Davies, 2020).
The Alexander Technique (AT), developed by F.M. Alexander (1867-1955) as a set of
principles aiming to identify and change counterproductive habits of use, is already
popular among musicians and performing artists in Anglo-Saxon countries, and has been
associated with the improvement of PRP symptoms and performance quality by J.
Davies (2019) and Fun Ying et al. (2015), although most empirical evidence of its effects
in musicians concerns outcomes associated with performance anxiety, as demonstrated
by U. Klein et al. systematic review in 2014.
This case study aims to explore the effects of Alexander Technique lessons on various
students at the Escola Superior de Música de Lisboa (ESML) and Escola Artistica de
Música do Conservatório Nacional, suffering from chronic PRP. This study was carried out
along 2 years, in which each student received from 5 to ten individual 50-minute lessons
given by the author of the study, teacher of the Alexander Technique since 2002. The
Nordic musculoskeletal questionnaire (NMQ) developed by Kuorinka et al., in 1987, with
a Borg modified scale (0= no pain to 10= maximum pain) developed by Borg in 1982,
were used before and after the intervention. Despite the reduced number of lessons, a
progressive improvement was noticed in all the students. Some of them arriving at a 1 or 0 in the Borg scale after the AT lessons. The results support mentioned previous
studies indicating that purpose designed AT classes may contribute to the management
of PRP and benefit performance related skills.N/
Utilização de animais em espetáculos: perceção do bem-estar animal em alunos do 5.º ano de escolaridade
Relatório de Prática de Ensino Supervisionada apresentado à Escola Superior de Educação de Lisboa para
obtenção de grau de mestre em Ensino do 1.º Ciclo do Ensino Básico e de Matemática e Ciências Naturais
no 2.º Ciclo do Ensino BásicoO presente relatório foi desenvolvido no âmbito da Unidade Curricular de Prática de Ensino Supervisionada II do 2.º ano do Mestrado em Ensino do 1.º Ciclo do Ensino Básico e de Matemática e Ciências Naturais no 2.º Ciclo do Ensino Básico.
Na primeira parte apresenta se uma descrição sintética das práticas de intervenção pedagógica que ocorreram no 1.º e no 2.º Ciclo do Ensino Básico, numa turma de 2.º e em duas turmas de 5.º ano, respetivamente, e uma análise crítica e reflexiva sobre as mesmas, devidamente fundamentada. Na segunda parte, apresenta se um estudo empírico intitulado Utilização de animais em espetáculos: Perceção do bem
estar animal em alunos do 5.º ano de escolaridade. Esta investigação teve como propósito identificar a perceção dos alunos acerca do bem estar dos animais utilizados para entretenimento humano. Neste sentido foram definidos os seguintes objetivos gerais: i) desenvolver a consciencialização para o bem estar animal; ii) identificar os principais locais onde são utilizados animais para o entretenimento humano; iii) identificar as necessidades básicas dos animais e se as mesmas se encontram ou não asseguradas.
O estudo é de natureza quantitativa e tem características de uma investigação
ação. Participaram na investigação cinquenta e nove alunos, distribuídos por três turmas distintas (dois grupos experimentais diferenciados pela sua origem portuguesa e de várias nacionalidades e um grupo de controlo). A recolha de dados foi efetuada através da administração de um questionário (antes e depois do processo de ensino
aprendizagem formal acerca de alguns espetáculos com animais) e da análise das respostas dadas pelos discentes. De forma a conduzir a intervenção foi delineado um plano de intervenção para os grupos experimentais que possibilitasse averiguar a eficácia das estratégias/ atividades da prática pedagógica para o cumprimento dos objetivos gerais formulados.
Os resultados do estudo indicam que, em determinadas situações, os alunos dos grupos experimentais foram capazes de identificar situações em que o bem-estar dos animais não se encontrava assegurado, melhorando após a intervenção embora não para todos os espetáculos trabalhados. Desta forma, conclui-se que os recursos construídos
foram ainda assim eficazes para uma maior consciencialização acerca do bem-estar animal e promoveram o confronto de ideias, o que contribuiu para o desenvolvimento de capacidades argumentativas de pensamento critico.This report is part of the Curricular Unit of Supervised Teaching Practice II of the 2nd year of the Master's Degree in Teaching in the 1st Cycle of Basic Education and Mathematics and Natural Sciences in the 2nd Cycle in Primary Education.
The first part presents a brief description of the pedagogical practices that took place in the 1st and 2nd Cycles of Primary Education, in a 2nd grade and two 5th grade classes, respectively, and a critical and reflective analysis of them. In the second part, an empirical study entitled Use of animals in shows: Perception of animal welfare in 5th grade students is presented. This investigation aimed to identify the students' perception of animals welfare used for human entertainment. In this sense, the following general objectives were defined: i) to develop awareness of animal welfare; ii) to identify the main places where animals are used for human entertainment; iii) to identify the basic needs of the animals and whether they are met.
The study is quantitative in nature and has characteristics of an action research. Fifty-nine students participated in the research, divided in three groups (two experimental groups differentiated by their nationality – Portuguese and other nationalities – and a control group). The data were collected through the administration of a questionnaire (before and after the formal teaching-learning process about animal shows) and the analysis of the answers given by the students. To conduct the intervention, an intervention plan was designed for the experimental groups that would make it possible to verify the effectiveness of the strategies/activities of the pedagogical practice for the fulfillment of the formulated general objectives.
The results of the study indicate that, in certain situations, the students in the experimental groups were able to identify situations in which the animal´s welfare was not assured, improving after the intervention, although not for all the shows approached in class. In this way, it is concluded that the designed resources were still effective for a greater awareness of animal welfare and promoted the confrontation of ideas, which help to the develop argumentative capacities of critical thinking.N/
Dissection of the IFN-γ versus IL-17-specific transcriptomes of γδ T cells: a new role for signaling adaptor Themis
The crucial role of murine γδ T cells in several (patho)physiological contexts stems from a complex process of ‘developmental pre-programming’ in the thymus, after which a significant fraction of γδ T cells populate peripheral sites already endowed with the capacity to secrete either IL-17 or IFN-γ. However, despite the relevance of these γδ T cell effector subsets, we still lack knowledge on the transcriptomes that specifically associate with IL-17 or IFN-γ production. To address this, we established a double reporter IL-17-GFP:IFN-γ-YFP mouse strain, which allowed us to isolate pure peripheral IL-17-producing (γδ17) or IFN-γ-producing (γδIFN) γδ T cells to perform RNA-sequencing. This led to the identification of the distinct transcriptomes of γδ17 and γδIFN cells, which surprisingly diverged in 6337 differentially (over 1.5-fold) expressed genes. Pathway and gene ontology analyses indicated that γδ17 cells differ from γδIFN cells in their selective ability to sense and integrate external cues, whereas γδIFN stands out in replication, transcription, and translation processes. A detailed analysis of the top differentially expressed genes between γδ17 and γδIFN cells revealed that most of the signature genes of each subset increased their expression levels in the periphery (compared to the thymus), suggesting that γδ17 and γδIFN cells only terminate their differentiation process at peripheral sites. Among the top differentially expressed genes, we found Themis, a T cell-specific gene involved in the regulation of TCR signal strength, to be enriched in γδIFN cells. Importantly, we found that Themis deficiency leads to a dysregulated effector γδ T cell peripheral compartment at steady state, which upon infection with Plasmodium berguei ANKA sporozoites confers Themis-deficient mice full protection from experimental cerebral malaria, a γδIFN-dependent pathology. Accordingly, we observed a less activated and less proliferative γδIFN population in the peripheral lymph nodes of infected Themis-deficient mice compared to Themis-sufficient controls. This work demonstrates the relevance of the characterization of the γδIFN and γδ17 transcriptomes to uncover new players in the regulation of γδ T cell effector functions, which may open new avenues for their manipulation in disease settings.info:eu-repo/semantics/publishedVersio
Produção de zeólitos hierárquicos por ação de surfatantes para reações de friedel-crafts em condições ambientalmente sustentáveis
Trabalho final de Mestrado para obtenção do grau de Mestre em Engenharia Química e Biológica – Perfil de Processos QuímicosO presente trabalho teve por objetivo a produção de zeólitos hierárquicos por ação de surfatantes, em meio básico, para estudo posterior como catalisadores em reações de acilação de Friedel-Crafts em condições ambientalmente sustentáveis. Numa primeira fase do trabalho, no decorrer da Bolsa de Iniciação à Investigação, refª IPL/2021/ZeoGemini_ISEL/BII/5M, foi estudado a influência do tempo de tratamento alcalino na presença de CTAB no zeólito ZSM5 (estrutura MFI), tendo-se chegado a um tempo de tratamento ótimo de 6h. No material de partida e respetivas amostras modificadas foi estudado o efeito da temperatura na reação de acilação de Friedel-Crafts tendo-se usado os substratos furano e 1-metoxinaftaleno, obtendo-se uma temperatura de reação ótima de 80 ºC. Numa segunda fase, já em âmbito de TFM, estudou-se o efeito da quantidade de surfatante adicionado no zeólito ZSM5, tendo por referência múltiplos da concentração micelar crítica (CMC), previamente determinada. Assim, os múltiplos da CMC utilizados no estudo com o zeólito ZSM5 foram os seguintes: 5, 10 e 25. No caso do zeólito Y (estrutura FAU), os múltiplos da CMC utilizados foram: 5, 10, 25 e 50. Posteriormente, com a amostra que demonstrou melhores resultados, quer no caso do zeólito ZSM5 quer no caso do zeólito Y, efetuaram-se estudos de regeneração aos catalisadores sendo que, foram realizadas várias técnicas para o zeólito ZSM5 e no zeólito Y apenas as duas técnicas que demonstraram ser mais promissoras no zeólito ZSM5. Os materiais de partida, bem como as amostras modificadas, foram caracterizados através das seguintes técnicas: difração de raios-X pelo método dos pós, SEM e TEM,
permitindo a caracterização estrutural dos materiais e a adsorção de azoto a -196 ºC para o estudo das propriedades texturais. Na reação de acilação de Friedel-Crafts, as amostras modificadas preparadas a partir de ambos os zeólitos apresentaram valores de rendimento na mesma ordem de grandeza. Para a série do estudo do efeito do tempo, no zeólito ZSM5, a amostra que obteve melhor desempenho foi a submetida ao tratamento alcalino assistido pelo CTAB durante 6h, ou seja, a amostra ZSM5_6_14, e para a série do estudo da quantidade de surfatante a amostra que se demonstrou mais promissora foi a ZSM5_6_5, ou seja, aquela onde se adicionou uma quantidade de surfatante 5 vezes superior ao valor da CMC de referência. 8 Para as amostras à base do zeólito Y, no estudo do efeito da quantidade de surfatante, a amostra que demonstrou melhor desempenho catalítico foi a amostra Y_12_10, onde se utilizou uma concentração de surfatante 10 vezes superior à CMC de referência.The present work aimed to produce hierarchical zeolites through the action of surfactants for later study as catalysts in Friedel-Crafts reactions under environmentally sustainable conditions. In the first phase of the work, in the Research Initiation Grant, ref. IPL/2021/ZeoGemini_ISEL/BII/5M, the ZSM5 zeolite (MFI structure) was studied. The optimum reaction temperature was studied using the substrate 1-methoxynaphthalene, obtaining an optimum temperature of 80 ºC, and the influence of the treatment time with CTAB, using furan as substrate, reaching an optimum time of 6h. In a second phase, within the scope of TFM, the effect of multiples of the critical micellar concentration (CMC) for the ZSM5 zeolite (MFI structure) was studied, based on its reference value, which had previously been calculated for the two types of zeolites. Thus, the multiples used in the study with the ZSM5 zeolite were the following: 5, 10 and 25. In the case of the Y zeolite (FAU structure), the CMC multiples used were the following: 5, 10, 25 and 50. Subsequently, with the sample that demonstrated the best results, both in the case of the ZSM5 zeolite and in the case of the Y zeolite, regeneration studies were carried out on the catalysts, and several techniques were carried out for the ZSM5 zeolite and in the Y only the two techniques that demonstrated to be most promising in the ZSM5 zeolite. The starting materials, as well as the modified samples, were characterized using the following techniques: X-ray diffraction, SEM, TEM (structural characterization) and nitrogen adsorption at -196 ºC (textural characterization). In the Friedel-Crafts acylation reaction, samples prepared from both zeolites showed yield values in the same order of magnitude. For the study series of the effect of time, on the ZSM5 zeolite, the sample that obtained the best performance was the one subjected to alkaline treatment assisted by the CTAB for 6h, that is, the ZSM5_6_14 sample, and for the study series of the amount of surfactant the sample that proved to be most promising was ZSM5_6_5, that is, the one where an amount of surfactant 5 times higher than the reference CMC value was added. 10 For samples based on zeolite Y, in the study of the effect of the amount of surfactant, the sample that demonstrated the best catalytic performance was sample Y_12_10, where a surfactant concentration 10 times higher than the reference CMC was used.N/
Hepatitis A in Portugal: epidemiological overview of incidence in the last decade
Hepatitis A is an acute liver disease with fecal-oral transmission caused by a hepatotrophic picornavírus - Hepatitis A Virus (HAV). Food contamination with HAV can occur at any time: cultivation, harvesting, processing, handling, and even after cooking. Food and water contamination takes place more frequently in developing countries where the disease is common but can also occur in developed countries. Although uncommon, foodborne outbreaks have also occurred due to people consuming contaminated fresh and frozen imported food products. Therefore, monitoring the disease is an essential tool for the early implementation of preventive measures, applying the One Health approach to both the human and environmental dimensions to mitigate the impact that an outbreak may have on the population. As representações dos professores sobre o impacto de um projeto de Aprendizagem Socioemocional, no âmbito das Academias Gulbenkian do Conhecimento
Dissertação apresentada à Escola Superior de Educação de Lisboa para obtenção de grau de mestre em Educação Social e Intervenção ComunitáriaSegundo Cristóvão et al. (2017) já há inúmeros estudos que reconhecem a eficácia do impacto que os programas de Aprendizagem Socioemocional (ASE) têm no sucesso dos alunos, mas também nos professores. Este é um estudo que tem como problemática a reflexão crítica sobre a implementação de um desses programas, no âmbito das Academias Gulbenkian do Conhecimento (AGC). Desse modo, a partir das
representações dos professores, procura compreender o impacto do projeto no sucesso dos alunos, em si próprios e nas suas práticas pedagógicas. Através de uma abordagem de natureza qualitativa e de um paradigma exploratório, este estudo recorre à perspetiva dos professores através de entrevistas semiestruturadas com o objetivo de fornecer pistas para investigações futuras. Foram utilizados os métodos
dedutivo e indutivo, combinando sistemas de categorias previamente definidos com categorias que criámos indutivamente a partir dos dados, emergindo das respostas dos entrevistados às perguntas, em consonância com os objetivos específicos. Os professores reconhecem a importância do desenvolvimento das Competências Socioemocionais pelos alunos, assim como a importância que a ASE tem quando comparada com a aprendizagem de conteúdos. Compreendemos que o sucesso dos projetos ASE está relacionado com o conhecimento prévio dos professores sobre os mesmos, mas também com a sua abertura para estar em contacto com algo inovador. Os resultados também apontam para uma diferenciação entre as representações dos professores com mais e menos de 10 anos de serviço, revelando a necessidade de uma Formação Inicial de Professores (FIP) que contemple a ASE, assim como de um constante trabalho reflexivo e colaborativo entre escola, técnicos, comunidade escolar e família, podendo ter a Educação Social um papel relevante neste processo.According to Cristóvão et al. (2017), there are numerous studies that recognize the effectiveness of the impact that Social-Emotional Learning (SEL) programs have on students' success, as well as on teachers. This study aims to critically reflect on the implementation of one of these programs within the scope of the Gulbenkian Academies of Knowledge. Thus, based on teachers' representations, it seeks to understand the project's impact on students' success, on the teachers and on their teaching practices. Through a qualitative approach and an exploratory paradigm, this study resorts to teachers' perspectives through semi-structured interviews with the aim of providing insights for future investigations. Both deductive and inductive methods were used, combining predefined categories with categories created inductively from the data, emerging from the respondents' answers to the questions, in line with the specific objectives. Teachers acknowledge the importance of developing Socio-Emotional Skills in students, as well as the significance of SEL when compared to academic learning. It is understood that the success of SEL projects is related to teachers' prior knowledge about them, as
well as their openness to engage with something innovative. The results also point to a differentiation between the representations of teachers with more and less than 10 years of service, revealing the need for Initial Teacher Training (ITT) that includes SEL, as well as constant reflective and collaborative work among the school, professionals, the school community, and families, where Social Education can play a relevant role in this process.N/
Label-free discrimination of T and B lymphocyte activation based on vibrational spectroscopy – A machine learning approach
B and T-lymphocytes are major players of the specific immune system, responsible by an efficient response to
target antigens. Despite the high relevance of these cells’ activation in diverse human pathophysiological pro cesses, its analysis in clinical context presents diverse constraints. In the present work, MIR spectroscopy was used to acquire the cells molecular profile in a label-free, simple, rapid, economic, and high-throughput mode.
Recurring to machine learning algorithms MIR data was subsequently evaluated. Models were developed
based on specific spectral bands as selected by Gini index and the Fast Correlation Based Filter. To determine if it was, possible to predict from the spectra, if B and T lymphocyte were activated, and what was the molecular
fingerprint of T- or B- lymphocyte activation.
The molecular composition of activated lymphocytes was so different from naïve cells, that very good pre diction models were developed with whole spectra (with AUC=0.98). Activated B lymphocytes also present a very distinct molecular profile in relation to activated T lymphocytes, leading to excellent prediction models,
especially if based on target bands (AUC=0.99). The identification of critical target bands, according to the
metabolic differences between B and T lymphocytes and in association with the molecular mechanism of the
activation process highlighted bands associated to lipids and glycogen levels.
The method developed presents therefore, appealing characteristics to promote a new diagnostic tool to

Deteção e reconhecimento inteligente de embarcações
Este projeto foca-se na pesquisa e desenvolvimento de sistemas avançados para a deteção automática de embarcações. Nesta perspetiva, o trabalho está centrado na conceção e otimização de uma arquitetura denominada Single Shot MultiBox Detector (SSD), um algoritmo empregue na deteção de objetos em imagens. Este método é utilizado em visão computacional e sistemas de inteligência artificial para identificar e localizar objetos numa cena específica. Com o intuito de alcançar esse objetivo, foi proposta uma nova arquitetura baseada no SSD. O objetivo primordial dessas modificações centrou-se na redução do custo computacional, visando tornar o sistema apropriado para a deteção em tempo real, sem comprometer o nível de precisão exigido. Os resultados obtidos revelam que a nova arquitetura proposta do SSD demonstra uma redução no consumo de recursos computacionais, mantendo o seu desempenho na deteção. A arquitetura proposta consiste numa rede base mais otimizada, com um menor número de camadas de previsão e menos âncoras empregues por camada. Alcançou-se um mAP de 79.3% a uma taxa de 2 fps num sistema de computação com recursos limitados, como é o caso do Raspberry Pi 4. Esses 2 fps representam uma deteção em tempo real, considerando a natureza do problema, no qual as embarcações se movem a velocidades reduzidas. Este trabalho representa um avanço no campo da deteção e classificação de embarcações, detendo o potencial para ser utilizado em aplicações práticas, incluindo a monitorização do tráfego marítimo e a segurança costeira, além disso, possui a capacidade de armazenar imagens devidamente categorizadas para a criação autónoma de conjuntos de dados de embarcações. Os resultados alcançados abrem portas para futuros desenvolvimentos e aperfeiçoamentos nesta área de aprendizagem automática que se encontra numa constante evolução.This project focuses on the research and development of advanced systems for the automatic detection of vessels. From this perspective, the work is centered around the design and optimization of an architecture called Single Shot MultiBox Detector (SSD), an algorithm employed in detecting objects in images. This method is used in computer vision and artificial intelligence systems to identify and locate objects in a specific scene. In order to achieve this goal, a new architecture based on SSD was proposed. The primary objective of these modifications focused on reducing computational costs, aiming to make the system suitable for real-time detection without compromising the required level of accuracy. The results obtained reveal that the proposed new architecture of SSD demonstrates a reduction in computational resource consumption while maintaining its performance in detection. The proposed architecture consists of
a more optimized base network, with a smaller number of prediction layers and fewer anchors employed per layer. An mAP of 79.3% was achieved at a rate of 2 fps on a computing system with limited resources, such as the Raspberry Pi 4. These 2 fps represent real-time detection, considering the nature of the problem in which vessels move at reduced speeds. This work represents an advancement in the field of vessel detection and classification, holding the potential for practical applications, including maritime traffic monitoring and coastal security. Moreover, it has the capability to store properly categorized images for the autonomous creation of vessel datasets. Aspergillus fumigatus exposure through a qualitative microbial risk assessment in the poultry industry
