    Exploring the Potential of Microalgae Cocultivation on Bacterial Community Dynamics in a Nile Tilapia and Garlic Aquaponic System

    The cocultivation of microalgae in aquaponic systems can improve nutrient removal and enhance water quality. Moreover, it can promote the biological and physicochemical balance in aquaponic ecosystems. However, the bacterial communities in aquaponic systems have not yet been comprehensively characterized and the effects of microalgae cocultivation remain unclear. Using 16S rRNA gene sequencing, we sought to elucidate the effects of a cocultivation of Chlorella vulgaris, Scenedesmus species, and Spirulina platensis cocultivation on the bacterial communities in the rearing tanks and the biofilter of a garlic (Allium sativum L.) and Nile tilapia (Oreochromis niloticus L.) aquaponic system during a 56-days experiment [...] also had a statistically significant effect on the bacterial composition: nonmetric multidimensional scaling analysis of a Bray–Curtis dissimilarity matrix demonstrated unique clustering patterns, indicating distinct bacterial communities in the biofilter and rearing tank water cocultivated with microalgal strains. We found significantly decreased abundances of the potentially pathogenic bacterial genera Janthinobacterium, Aeromonas, Pseudomonas, and Flavobacterium in aquaponic systems cocultivated with microalgae when compared to the control. Furthermore, the beneficial bacterial genera Fusibacter, Geothrix, Thiobaca, and Treponema were significantly enriched in the microalgae cocultivated systems. The results clearly demonstrate that integrating microalgae into an aquaponic recirculating aquaculture system (RAS), not only improves weight gain and survival rates of Nile tilapia, but also enriches beneficial bacterial genera and reduces potentially pathogenic microorganisms, offering a promising strategy to reduce antibiotic use and enhance water quality in aquaponics, without growth impairments of cocultivated plants

    CAU-52: An Iron Metal-Organic Framework Containing Furandicarboxylate Linker Molecules

    The V-shaped linker molecule 2,5-furandicarboxylic acid (H2FDC), which can be derived from lignocellulosic biomass, was used in a systematic screening with various iron salts and led to the discovery of a new iron-based metal-organic framework (Fe-MOF) with the composition [Fe3(μ3-O)(FDC)3(OH)(H2O)2]·5H2O·H2FDC, designated as CAU-52 (CAU = Christian-Albrechts-Universität zu Kiel). The crystal structure of CAU-52 was determined using 3D electron diffraction (3D ED) and further refined by Rietveld refinement against powder X-ray diffraction (PXRD) data. CAU-52 contains the well-known trinuclear [Fe3(μ3-O)]7+ cluster as the inorganic building unit (IBU) that is six-connected by FDC2- ions to form the pcu net. The connectivity leads to two types of cubic cages, similar to the ones observed in soc-MOFs. Comprehensive characterization of the title compound, including N2 and water vapor sorption measurements, confirmed its chemical composition. CAU-52 exhibits microporosity toward nitrogen with a type-I isotherm (77 K), yielding a specific surface area of as,BET = 1077 m2/g. The H2O sorption measurement at 298 K leads to an isotherm that exhibits three steps. The water sorption capacity was determined to be 390 mg/g, and it decreases slightly in subsequent sorption cycles. The MOF is stable up to 250 °C in air and chemically resistant in various solvents

    Toward an integrated pantropical ocean observing system

    Global climate is regulated by the ocean, which stores, releases, and transports large amounts of mass, heat, carbon, and oxygen. Understanding, monitoring, and predicting the exchanges of these quantities across the ocean’s surface, their interactions with the atmosphere, and their horizontal and vertical pathways through the global oceans, are key for advancing fundamental knowledge and improving forecasts and longer-term projections of climate, weather, and ocean ecosystems. The existing global observing system provides immense value for science and society in this regard by supplying the data essential for these advancements. The tropical ocean observing system in particular has been developed over decades, motivated in large part by the far-reaching and complex global impacts of tropical climate variability and change. However, changes in observing needs and priorities, new challenges associated with climate change, and advances in observing technologies demand periodic evaluations to ensure that stakeholders’ needs are met. Previous reviews and assessments of the tropical observing system have focused separately on individual basins and their associated observing needs. Here we provide a broader perspective covering the tropical observing system as a whole. Common gaps, needs, and recommendations are identified, and interbasin differences driven by socioeconomic disparities are discussed, building on the concept of an integrated pantropical observing system. Finally, recommendations for improved observations of tropical basin interactions, through oceanic and atmospheric pathways, are presented, emphasizing the benefits that can be achieved through closer interbasin coordination and international partnerships

    Collaborative research in healthcare: uncovering the impact of industry collaboration on the service innovativeness of university hospitals

    In the face of the challenges of rising demand and increasing economic pressure, medical service innovations and university hospitals are becoming increasingly important as cornerstones for the continuous development of medical care. Simultaneously, increasing technological complexity, scarce funding, and resource-intensive technology development are forcing university hospitals to open up their internal innovation processes. Numerous studies have highlighted the beneficial effects of cross-sectoral collaboration on the innovation performance of industrial entities, while neglecting the perspective of academic institutions. The impact of industry collaborations on the service innovativeness of university hospitals in particular has not been addressed in previous research. Although the potential for nonlinear effects of university-industry collaborations has been discussed in research, adequate evidence, particularly in the healthcare context, has not been provided. Therefore, this study addresses the questions: (1) What is the impact of university-hospital-industry collaboration (UHIC) on the service innovativeness of university hospitals? (2) Can a nonlinear relationship between UHIC and university hospitals' service innovativeness be empirically verified? The hypotheses are tested based on an unbalanced panel data set, which combines three distinct data sources and comprises annual observations from 2011 until 2019 of all German university hospitals. The findings indicate that the UHIC intensity has a positive effect on university hospitals’ service innovativeness with a one-year time lag. Furthermore, a negative quadratic UHIC effect was found. In doing so, this study complements existing research and opens the black box by uncovering the downside of cross-sectoral collaboration for a more complete understanding of the underlying dynamics of this growing area of research, with a particular focus on university hospitals

    Genetics impact risk of Alzheimer's disease through mechanisms modulating structural brain morphology in late life

    Background Alzheimer's disease (AD)-related neuropathological changes can occur decades before clinical symptoms. We aimed to investigate whether neurodevelopment and/or neurodegeneration affects the risk of AD, through reducing structural brain reserve and/or increasing brain atrophy, respectively. Methods We used bidirectional two-sample Mendelian randomisation to estimate the effects between genetic liability to AD and global and regional cortical thickness, estimated total intracranial volume, volume of subcortical structures and total white matter in 37 680 participants aged 8-81 years across 5 independent cohorts (Adolescent Brain Cognitive Development, Generation R, IMAGEN, Avon Longitudinal Study of Parents and Children and UK Biobank). We also examined the effects of global and regional cortical thickness and subcortical volumes from the Enhancing NeuroImaging Genetics through Meta-Analysis (ENIGMA) Consortium on AD risk in up to 37 741 participants. Results Our findings show that AD risk alleles have an age-dependent effect on a range of cortical and subcortical brain measures that starts in mid-life, in non-clinical populations. Evidence for such effects across childhood and young adulthood is weak. Some of the identified structures are not typically implicated in AD, such as those in the striatum (eg, thalamus), with consistent effects from childhood to late adulthood. There was little evidence to suggest brain morphology alters AD risk. Conclusions Genetic liability to AD is likely to affect risk of AD primarily through mechanisms affecting indicators of brain morphology in later life, rather than structural brain reserve. Future studies with repeated measures are required for a better understanding and certainty of the mechanisms at play

    Charakterisierung der Protein-, Kohlenhydrat- und Lipidverdauung in Larven des Seeigels Strongylocentrotus purpuratus

    The dissertation investigates the digestive physiology of the larvae of the sea urchin Strongylocentrotus purpuratus, in particular the degradation of proteins, carbohydrates and lipids. Extracellular digestion is considered a crucial step in the evolution of multicellularity and heterotrophy. While digestion is well described in adult sea urchins, it has been little studied in planktonic larvae. Molecular, biochemical and physiological methods were used to investigate the activity and gene expression of digestive enzymes under different feeding conditions (high vs. low food supply). The results show that larvae already have a fully developed digestive machinery at an early stage (from day 7). Gene expression was age-dependent and responded to food availability. The expression of protease genes was particularly high, followed by lipases and significantly lower levels of glycosidases. Enzymatic analyses showed that proteases responded rapidly to feeding - even with a low diet - while lipases were only activated with a high diet. Glycosidases, on the other hand, showed a slower, continuous activation. The pH optima of the enzymes correspond to the conditions in the larval intestine: Proteases and lipases worked best under alkaline conditions (pH 9.0-9.5), whereas glycosidases worked best at neutral to slightly acidic pH (pH 5.0-6.0), indicating a possible localisation outside the midgut. Overall, the work provides new insights into the functional development and adaptability of the digestive system of sea urchin larvae. It forms a basis for further studies on dietary changes during metamorphosis and the reaction of these organisms to environmental changes

    Analyse der Effekte von Doxycyclin auf den Ischämie-/Reperfusionsschaden am Rattendarm

    Akute Ischämien des Darms machen nur einen geringen Anteil der akuten Organischämien aus, sind aber mit einer hohen Mortalität von 60-90% assoziiert. Für die hohe Mortalität ist die Vulnerabilität des Darms gegenüber dem Phänomen des Ischämie-/Reperfusionsschadens (I/R-Schaden) mitverantwortlich. Erste experimentelle Untersuchungen deuten darauf hin, dass Doxycyclin durch seine antioxidativen, zytoprotektiven und antiinflammatorischen Eigenschaften den I/R-Schaden am Darm reduzieren kann. In dieser Forschungsarbeit sollten die Effekte von Doxycyclin auf den I/R-Schaden am Rattendarm genauer analysiert werden. Dafür wurden die Auswirkungen von intestinaler I/R, Doxycyclin und der Kombination von intestinaler I/R und Doxycyclin auf die Integrität des Darmgewebes, die Entstehung von oxidativem Stress und auf inflammatorische Prozesse untersucht. Die gewonnenen Ergebnisse deuten darauf hin, dass die Applikation von Doxycyclin vor intestinaler I/R den intestinalen Mukosaschaden verringerte, nicht aber das Ausmaß der stattfindenden apoptotischen Prozesse. Da keine Anhaltspunkte dafür gefunden werden konnten, dass die Induktion von intestinaler I/R in dieser Forschungsarbeit die Entstehung von oxidativem Stress im Darmgewebe beeinflusste, können keine Aussagen über mögliche protektive Effekte von Doxycyclin auf die Entstehung von oxidativem Stress bei intestinaler I/R getroffen werden. Weiterhin ergab sich kein eindeutiger Anhalt dafür, dass die Induktion von intestinaler I/R im Rahmen dieser Forschungsarbeit einen Einfluss auf inflammatorische Prozesse hatte, sodass auch die Wirkung von Doxycyclin auf inflammatorische Prozesse bei intestinaler I/R nicht beurteilt werden kann

    Charakterisierung Cisplatin-induzierter extrazellulärer Vesikel des Ovarialkarzinoms unter besonderer Berücksichtigung der Metalloprotease ADAM17

    Das Ovarialkarzinom ist der gynäkologische Tumor mit der zweithöchsten Mortalität. Unter Therapie entwickelt der Großteil der Patientinnen ein Rezidiv oder eine Chemotherapieresistenz. Extrazelluläre Vesikel (EV) und die Metalloprotease ADAM17 könnten eine bedeutende Rolle in der Chemotherapie-Resistenzentwicklung einnehmen. In dieser Arbeit wurde ein Protokoll verwendet, mit welchem mehrheitlich kleine EV aus den Überständen PMA- und Cisplatin-stimulierter Ovarialkarzinomzellen (Igrov-1) angereichert und mittels der Mindeststandards der International Society of Extracellular Vesicles (2018) charakterisiert werden konnten. Die EV-Charakterisierung wurde durch eine massenspektrometrische Lipidprofilanalyse ergänzt. Es zeigt sich, dass Cisplatin in Abhängigkeit und PMA unabhängig vom Zelltod eine EV-Freisetzung induziert. Die freigesetzten Cisplatin-EV zeigten die höchste Abundanz an maturer ADAM17 nach Stimulation mit geringen Cisplatin-Konzentrationen (1 µM, 2,5 µM). Die Aufnahme von PMA- bzw. Cisplatin-EV in Igrov-1-Zellen wurde qualitativ und quantitativ nachgewiesen. Der Aufnahmeprozess ist zeitabhängig und wird zumindest teilweise durch Aktin-abhängige Endozytose vermittelt. In einem initialen Experiment zeigte sich, dass auch aus dem malignen Aszites einer Ovarialkarzinom-Patientin mit großer Wahrscheinlichkeit ADAM17-tragende EV angereichert werden konnten. Die Ergebnisse dieser Arbeit haben die Grundlage dafür geschaffen, die Bedeutung von EV-assoziierter ADAM17 bei der Entstehung von Chemotherapieresistenzen im Ovarialkarzinom weiter aufzuklären. Dies ist die Grundlage für eine diagnostische oder auch therapeutische Perspektive, um dadurch die Prognose des Ovarialkarzinoms zu verbessern

    Etablierung einer digitalen Vermessungsmethode von Gingivarezessionen

    Ziel der Arbeit war es, die Genauigkeit und Reproduzierbarkeit der Messung von gingivalen Rezessionen zu optimieren sowie eine valide, präzise, reproduzierbare und dokumentierbare digitale Vermessungsmethode zu etablieren und die longitudinale Entwicklung dieser Rezessionen zu beobachten. Eine kritische Analyse etablierter Klassifikationssysteme und der Messung mittels Parodontalsonde verdeutlichten die methodischen Limitationen herkömmlicher Verfahren. In der vorliegenden Studie wird ein Vergleich von drei Messverfahren vorgenommen: der klinischen Messung mittels Parodontalsonde, der analogen Vermessung unter Einsatz derselben Sonde an Gipsmodellen sowie der digitalen Messung an digitalisierten Gipsmodellen unter Verwendung eines Skala-Werkzeugs. Die Ergebnisse bestätigen, dass die digitale Vermessungsmethode valide Ergebnisse generiert und eine signifikant höhere Präzision aufweist. Abschließend wird das Potenzial der digitalen Methode für den klinischen Alltag diskutiert, da sie eine präzisere Diagnostik und damit eine optimierte Therapieplanung in der Parodontologie ermöglicht. Die Arbeit leistet somit einen wichtigen Beitrag zur Weiterentwicklung der parodontalen Diagnostik und bietet einen fundierten Ausblick auf zukünftige Forschungsansätze im Bereich digitaler Messverfahren

    Raumerleben von muslimischen Frauen mit Kopftuch in Deutschland im Hinblick auf den strategischen Umgang mit räumlichen Unsicherheiten in Alltagsräumen

    Die vorliegende Dissertation befasst sich mit dem Raumerleben kopftuchtragender muslimischer Frauen in Deutschland und fokussiert sich insbesondere auf ihren strategischen Umgang mit den Unsicherheiten, die sie im alltäglichen Leben erfahren. Ziel der Arbeit ist es, die vielfältigen Herausforderungen aufzuzeigen, denen kopftuchtragende Muslimas in unterschiedlichen Alltagsräumen begegnen, und zu analysieren, wie sie diesen strategisch begegnen. Die qualitative Forschung basiert auf Interviews mit 14 kopftuchtragenden Muslimas, die ihr Raumerleben in einer Vielzahl von Umgebungen beschreiben. Diese Räume werden von den Frauen je nach persönlicher Erfahrung unterschiedlich wahrgenommen und lassen sich in drei Kategorien einteilen: topophobe Räume, topoambivalente Räume und topophile Räume. Die Ergebnisse der Studie verdeutlichen, dass kopftuchtragende Muslimas in Deutschland im täglichen Leben mit spezifischen sozialen und strukturellen Herausforderungen konfrontiert sind. Gesellschaftliche Vorurteile, Diskriminierung und mangelnde Akzeptanz führen dazu, dass die Frauen viele Räume als unsicher wahrnehmen. Um diesen Schwierigkeiten zu begegnen, entwickeln sie individuell angepasste Handlungs- und Denkstrategien, die je nach Situation entweder konfrontativ oder vermeidend eingesetzt werden. Dabei zeigt sich, dass die Wahl der Bewältigungsstrategien eng mit den Selbst- und Fremdbildern der Frauen verknüpft ist, wobei sowohl ihre religiöse Identität als Muslima als auch ihre intersektionalen Erfahrungen von Diskriminierung eine zentrale Rolle spielen. Die vorliegende Arbeit verdeutlicht, dass Räume für kopftuchtragende Muslimas in Deutschland nicht lediglich physische Orte darstellen, sondern als emotional aufgeladene und subjektiv erlebte Umgebungen zu verstehen sind
