Caregivers' knowledge of local foods as a tool improving QOL among the elderly
No full textP(論文)The purpose of the present study was to survey the local foods of northern Miyazaki Prefecture using self-administered questionnaires and interviews for the caregivers of the dependent elderly persons. Many community-dwelling elderly individuals were familiar with local foods. Also, cooking and eating local foods contributed to improving communication among the elderly. Furthermore, we examined detailed information related to local foods. The caregivers recognized that local food was a very important factor for improving the QOL of the elderly. However, the caregivers had insufficient knowledge of local foods. These findings suggest that effective education about local foods for caregivers is necessary in order to raise the level of care.departmental bulletin pape
End of life care in Duchenne muscular dystrophy: grobal trends of researches and issues
No full textP(論文)In the present study, we examined the concept of end-life care for Duchenne muscular dystrophy and trends in related research with the objective of systematizing such care. We obtained the following findings: 1) A search on the PubMed database of literature published before December 31, 2007 containing the term "end-of-life" in the title or abstract yielded 2087 results, and this literature tended to become more common in recent years. However, when the search was narrowed down to include the term "muscular dystrophy", only one study (Hilton et al, 1993) was found. 2) A study on the components of end-of-life care led to the following. multidisciplinary support, the perspective of patients, and relatively long-term support for end-stage patients. 3) Although research had been conducted on support during the terminal stage for Duchenne muscular dystrophy both in Japan and abroad, no studies were based on the concept of end-of-life care.departmental bulletin pape
Preparation of recombinant lysosomal cysteine protease, cathepsin K in insect cells
No full textP(論文)軟骨細胞に局在するシステインプロテアーゼであるカテプシンKは,骨の差異九州に関わる酵素である。カテプシンKの阻害剤は骨粗鬆症の治療薬の候補になり得る。スクリーニングや酵素の機能を調べるためには組み換えカテプシンKを大量に作成することが必要である。我々はカイコ幼虫を用いた発現系が分泌型酵素であるカテプシンKの発現に適していると考えた。C末端にmycタグを付けたカテプシンK遺伝子をカイコ用の発現ベクターに挿入し,限界希釈法で組み換えウイルスを得た。このウイルスをカイコ培養細胞に感染させてカテプシンKの発現場所を調べたところ,細胞内で発現していた。細胞外への分泌が観察されなかったのでカイコ幼虫での発現をあきらめ,培養細胞からの精製を試みた。DEAE-セファロースカラム,Mono-Sカラム,抗myc抗体アフィニティーカラムで順次精製した結果,非活性を細胞可溶性画分の約300倍に高めることに成功した。Cathepsin K, which is localized in osteoclasts, plays an important role of bone resorption. Selective inhibitors of cathepsin K therefore can be promising therapeutic candidates for the treatment of diseases such as osteoporosis. Recombinant cathepsin K protein is necessary for screening the therapeutic candidates and elucidating the enzymatic functions because the amount of native cathepsin K is very low. Silkworm is one of the most attractive hosts for large-scale production of eukaryotic secretory proteins. The gene for fusion cathepsin K protein with myc-tag at the C-terminus was inserted into expression vector for silkworm and recombinant viruses were isolated by end-point dilution assay. Cathepsin K was expressed in the cell lysate, but not in the medium, and consequently we gave up the idea of cathepsin K expression in the silkworm. Thus, we tried to isolate cathepsin K protein from the cultured cells, and purified it by sequential operations with DEAE-Sepharose column, Mono-S column, and anti-myc antibody column. The purification fold of cathepsin K activity increased approximately 300 times in comparison with that in soluble fraction of cell lysate.departmental bulletin pape

The shift in family policy from combining work with having a family to supporting the family work-life balance : The problem with child caring support policy.
Get PDFP(論文)The Japanese total fertility rate (TFR) continues to decline. After the "1.57 shock" of 1990, the national government now gives support for child care in order to stop the decline in TFR, but with little impact. This child care support targets working mothers, but also can result in producing a double bind. Females often have to choose marriage with children to gain support, or they are forced to work and remain unmarried. The current system of support excludes persons which do not fit a particular family or gender role. This paper argues that government child care support should be a true family support that includes fathers, single parents and a variety of family combinations. Furthermore, the focus should be on a balance between work and private life.departmental bulletin pape
