    A monoclonal antibody, mNI-58A, against CD11a induces homotypic cell aggregation and promotes interferon-γ production by the human NK-like cell line, KHYG-1

    P(論文)我々が開発したCD11a(LFA-1α)モノクローナル抗体(mNI-58A)のヒトNK様細胞株(KHYG-1)に与える影響を検討したところ、mNI-58A抗体はKHYG-1細胞に対して細胞間凝集を特異的に誘導した。この細胞間凝集反応は、conventional protein kinase C (cPKC)抑制剤であるGo6976とnovel protein kinase C (nPKC)抑制剤であるRottlerinでブロックされ、その効果はRottlerinの方がGo6976より強かった。また、mNI-58A抗体はKHYG-1細胞にインターフェロンγの産生能も増強させた。以上の結果は、mNI-58A抗体がヒト免疫応答におけるNK細胞の機能を解析する上で非常に有益な抗体であることを示唆している。A monoclonal antibody (mAb), designated as mNI-58A, against CD11a (leukocyte function-associated antigen-1 α; LFA-1α) was established in our laboratories by immunizing mice with the lipopolysaccharide (LPS)-stimulated human monocyte-like cell line, U937. This mAb specifically induced homotypic cell aggregation of the human NH-like cell line, KHYG-1.This mNI58A-induced homotypic cell aggregation was markedly blocked by the addition of an optional concentration of a conventional protein kinase C (cPKC) inhibitor, Go6976, and was completely blocked by the addition of an optimal concentration of a novel protein kinase C (nPKC); a PKC delta isoenzyme) inhibitor, Rottlerin. Interestingly, KHYG-1 cells effectively promoted interferon-γ (IFN-γ) production in the culture supernatants of the homotypic cell aggregations of KHYG-1 cells induced by mNI-58A. These findings strongly indicate that CD11a mAb (mNI-58A) has some unique properties and many be useful for analyzing the cell-to-cell interactions of NK cells in several human immune responses.departmental bulletin pape

    Neuropsychological characteristics of probable Alzheimer type mild cognitive impairment and vascular mild cognitive impairment: the analysis of the profile of cognitive impairment in mild cognitive impairment using Japanese version COGNISTAT

    P(論文)Objective: The goal of this research is to investigate the neuropsychological characteristics of probable Alzheimer type mild impairment (MCI) and vascular MCI (VaMCI). Participants: Post acute stroke patients (n=22) were recruited from the post stroke rehabilitation unit; and community dwelling elderly people without cardiovascular disease, stroke and dementia (n=11) were also recruited. Methods: Cognitive performance was assessed using the Neurobehavioral Cognitive Status Examination (COGNISTAT). Participants were classified into five groups by their COGNISTAT scores and MCI criteria: elderly people with normal cognitive functions (controls) (n=7), MCI (n=4), VaMCI (n=18), vascular cognitive impairment (VCI) (n=4) and post stroke with non cognitive impairment (NCI) (n=2). Results: Deficits of construction and judgment were seen in MCI and VaMCI compared to controls. However, MCI were significantly more impaired on tests of repetition and Memory compared to the controls and VaMCI. Construction and judgment scores correctly classified participants (96.9%) into two groups (controls and MCI groups), and repetition and memory score correctly discriminated between MCI and VaMCI (100.0%) by the discriminant analysis. Conclusions: The results suggested that impairments of construction and judgment were characteristics of both MCI and VaMCI, though deficits of repetition and memory were specific features of MCI.departmental bulletin pape

    Acoustic phonetic features of Japanese geminate consonant and elongated vowel produced by non-native Japanese learners: case study of temporal coordination of English/Chinese native speaker

    P(論文)Phonetically, Japanese is a mora-timed language which is very different from the majority of languages. Many learners of Japanese as a second language have difficulty producing the special mora phoneme. Our initial research task was to describe the acoustical phonetic features of special mora as produced by non-native Japanese learners, in the hope of establishing in the future a training method for special mora phoneme pronunciation for non-native learners of Japanese language. In particular, this paper reviews our data on the geminate consonant and elongated vowel produced by English and Chinese native speakers. We observed features of temporal coordination during native Japanese speakers word production taking measurements of the geminate consonants and elongated vowels and compared these measurements with data from the nonnative speakers. Subjects pronounced sets of words (minimal pairs differing in geminate and non-geminate consonant, and minimal pairing words with elongated vowels and non-elongated vowels) and compared the rate of change in the length of the stop gap and the vowel duration produced by speakers of differing native language backgrounds. Our results indicated that the stop gap produced by English native speakers was significantly extended in comparison to the stop produced by native Japanese speakers, while the duration of elongated vowels were less extended than the native speakers. The stop gap produced by Chinese native speakers was less than the stop gap of native Japanese speakers.departmental bulletin pape

    Survey regarding the use of contact lenses by university students

    P(論文)To investigate the adequate use of contact lenses (CL) and the necessity health guidance to prevent ocular disorders, we conducted a questionnaire survey regarding the use of CL and the presence or absence of ocular disorders in 127 students from whom informed consent regarding the survey contents was obtained. Seventy students (55.1%) used CL. The make-to-female ratio was 1:2. The mean interval from the start of CL application was 5.1±2.4 years. The types of CL consisted of hard CL in 12.9% of the 70 students, conventional soft CL (SCL) in 21.4%, frequent-replacement SCL (2 weeks) (FRSCL) in 50.0%, and daily disposable SCL in 15.7%. The proportion of students using FRSCL was the highest. The duration of wearing CL per day ranged from 12 to 14 hours; students wearing CL over many hours accounted for more than 50%. Concerning their experience regarding CL-related ocular disorders, 62.9% reported a history of oculopathy, including severe cases such as endophthalmitis. These results suggest that a method to inform CL users of the risk of CL-related ocular disorders, which develop despite appropriate care and periodic consultations, should be examined.departmental bulletin pape

    The possibility of using "Job Card" for career education

    P(論文)This paper described the advantage of using a "job card" for career education. "The job card system" is a new system that began this year. The original purpose of "the job card system" is to support a person looking for a job. When a job seeker writes a "job card", the job seeker can understand his own work experience and employment possibilities while getting the advice of the career counselor. If a teacher utilized a "job card" for career education in this way, the student can understand his career better. Therefore, I recommend that a teacher utilize the "job card" for career education. However, 2 problems may arise. First, the teacher may have insufficient training and second, "job card" are not yet widely known in Japan.     Current state and problems concerning out of home environment with mentally disabled persons: considerations focused on interactive relations

    P(論文)This paper reports on the state of out home environments for mentally disabled persons living in a city. I report the results of a questionnaire survey and discuss problems from the point of interactions between environment and other related factors. The aims of the study are to find suppressive factors preventing mentally disabled persons from going out of home and to improve their out of home experiences by addressing these factors. 65 mentally impaired individuals were given an interview. Results show the main factors obstructing these persons were a shortage of public transportation, difficulties in getting a drivers license, a luck of full-time employment, inadequate information about support services and few opportunities to meet with friends. Based on our findings the following areas were identified for improvement: (1) development of full-time employment for mentally disabled persons, (2) promotion of peer-support activities, (3) development more effective service information systems for mentally disabled persons, (4) on-going needs assessment for these persons.departmental bulletin pape

    Preschool memories of preschool teacher education students and images of preschool teachers that students hope to become

    P(論文)At the Department of Early Childhood Education, J Junior College and the Department of Child Welfare Services, K University of Health and Welfare, a questionnaire survey was administered to 90 students from 2007 to 2008 in order to examine what memories they had of preschool and what kind of preschool teacher they hoped to become after graduation. This paper discusses the educational methods applied at both departments on the basis of the obtained results. The results of this survey should be provided to the students at both departments, thus enabling them to once again experience childhood feelings in order to consider what qualities preschool teachers ought to have. 86% (77/90) of students replied that they wanted to become preschool teachers who have a positive relationship with children. Accordingly, students should be trained with the goal of fostering a positive connection between child and teacher. Conversely, only 22% (20/90) of students replied that they wanted to become preschool teachers who have the capacity to educate or guide children. Students should recognize the professional characteristics of preschool teachers-telling right from wrong, being in good health, and being skilled in arts and sports.departmental bulletin pape
