鹿児島大学博士(歯学)Doctor of Philosophy in Dental Science博士論文全文, 博士論文要旨, 最終試験結果の要旨, 論文審査の要旨Silver nanoparticles (AgNPs) have demonstrated antibacterial properties and are widely recognized as one of the most prominent types of nanoparticles. Recent studies have highlighted their effectiveness against coronaviruses. However, the detailed molecular mechanisms underlying the action of AgNPs on viruses and their impacts on the human body remain to be fully elucidated. Thus, we attempt to delineate the preventive effects of AgNPs against SARS-CoV-2 infection. Our findings indicate that treatment with AgNPs reduces ACE2 expression, a key receptor for SARS-CoV-2 particularly in normal oral and lung cells. Additionally, we observed a decrease in the binding affinity of the spike protein to the cell after the AgNP treatment. Through western blot analysis, we identified the involvement of the AKT and/or mTOR signaling pathway in this process. Because AKT and mTOR signaling affects mitochondrial function, we investigated the effects of the AgNP treatment on mitochondria. As a result, we observed the localization of AgNPs within mitochondria. Further, an increase in mitochondrial Fe2+ and reactive oxygen species levels was observed, ultimately resulting in mitochondrial dysfunction. Analysis of microRNA (miRNA) expression signatures and The Cancer Genome Atlas (TCGA) database revealed that that two passenger strand miRNAs, miR-30c-1-3p and miR-30c-2-3p, were downregulated in cancer tissues, and their low expression was closely associated with worse prognosis in patients with BrCa. Functional assays showed that miR-30c-1-3p and miR-30c-2-3p overexpression significantly inhibited cancer cell aggressiveness, suggesting these two miRNAs acted as tumor-suppressors in BrCa cells. Notably, involvement of passenger strands of miRNAs is a new concept of cancer research. Further analyses showed that seven genes (TRIP13, CCNB1, RAD51, PSPH, CENPN, KPNA2, and MXRA5) were putative targets of miR-30c-1-3p and miR-30c-2-3p in BrCa cells. Expression of seven genes were upregulated in BrCa tissues and predicted a worse prognosis of the patients. Among these genes, we focused on TRIP13 and investigated the functional significance of this gene in BrCa cells. Luciferase reporter assays showed that TRIP13 was directly regulated by these two miRNAs. TRIP13 knockdown using siRNA attenuated BrCa cell aggressiveness. Inactivation of TRIP13 using a specific inhibitor prevented the malignant transformation of BrCa cells. We aimed to examine which morphology of dendritic cells (DCs) and macrophages corresponds to “preconditioning” of the SN against cancer. Although macrophages are generally able to tolerate cancer metastasis, the CD169-positive subtype is believed to be a limited exception. Immunohistochemical and morphometric analyses were performed to examine DC-SIGN-, CD68-, and CD169-positive cells in SNs and non-SNs of 23 patients with gastric cancer with or without nodal metastasis. All patients survived for >5 years without recurrence. DCs were present in the subcapsular, paracortical, and medullary sinuses, the endothelia of which expressed DC-SIGN and smooth muscle actin (SMA). In the non-SNs of patients without metastasis, subcapsular DCs occupied a larger area than SNs, and this difference was statistically significant. Conversely, subcapsular DCs were likely to have migrated to the paracortical area of the SNs. DC clusters often overlapped with macrophage clusters; however, histiocytosis-like clusters of CD169-negative macrophages showed a smaller overlap. We found a significantly larger overlap between DC-SIGN and CD169-positive clusters in SNs than in non-SNs; the larger overlap seemed to correspond to a higher cross-presentation of cancer antigens between these cell populations. DC-SIGN–CD169-double positive cells might exist within this overlap. SNs in gastric cancers are usually preconditioned as a frontier of cancer immunity, but they may sometimes be suppressed earlier than non-SNs. 重症心身障がい入院患者における栄養状態予測のための多周波生体電気インピーダンス分析の臨床応用可能性の評価

The purpose of this study was to investigate the clinical utility of body composition, which has been suggested to be related to biochemical tests in persons with SMID
Methods: We calculated cutoff values of body composition predicting malnutrition and compared the usage of peripheral parenteral nutrition for the two groups divided by each body composition cutoff value. We also compared body composition at the baseline between non-survivors and survivors.
Results: Only group comparisons based on skeletal muscle ratio and extracellular water to total body water (ECW/TBW) cutoffs showed significant differences in the usage of peripheral parenteral nutrition. Non-survivors had significantly higher ECW/TBW than survivors at the baseline.
Conclusions: In persons with SMID, skeletal muscle ratio, and ECW/TBW measured using multifrequency bioelectrical impedance were associated with biochemical indices and ill health.
