Digitale Transformation in der Hochschullehre unter Berücksichtigung des Blended Learning-Ansatzes
Die digitale Transformation gilt als „zentrale Herausforderung unserer Zeit“ (Barton, Muller und Seel, 2018, S. 88) und findet zunehmend auch in der Hochschullehre im Rahmen des elektronischen Wandels Einzug. Insbesondere zu Zeiten der Corona-Pandemie hat die Anzahl digitaler Lehrangebote an Hochschulen weltweit stark zugenommen. Dabei ist ein höherer Digitalisierungsgrad jedoch nicht gleichzusetzen mit einer höheren Lehrqualität. Positive Effekte auf den Lernerfolg konnte nachweislich eine Kombination aus klassischer Lehre in räumlicher und zeitlicher Synchronität und e-Learning- Elementen bieten, wie es im Blended Learning der Fall ist. Um eine solche Blended Learning- Veranstaltung zu entwickeln und erfolgreich zu implementieren, sind neben dem didaktischen Vorgehen auch die Zusammenarbeit und Akzeptanz verschiedener Stakeholder von zentraler Bedeutung.
In dieser Arbeit werden daher verschiedene Fragestellungen der digitalen Transformationen in der Hochschullehre nach dem Blended Learning- Ansatzes untersucht, die sich von didaktischen Vorgehensweisen über Entwicklungsmethoden bis zur Akzeptanz und Zusammenarbeit beteiligter Stakeholder strecken. Zum Einsatz kommen sowohl qualitative als auch quantitative Forschungsmethoden, die verhaltens- und kostruktwissenschaftliche Paradigmen in unterschiedlichen Forschungstiefen behandeln
Finding landmarks - An investigation of viewing behavior during spatial navigation in VR using a graph-theoretical analysis approach
Vision provides the most important sensory information for spatial navigation. Recent technical advances allow new options to conduct more naturalistic experiments in virtual reality (VR) while additionally gathering data of the viewing behavior with eye tracking investigations. Here, we propose a method that allows one to quantify characteristics of visual behavior by using graph-theoretical measures to abstract eye tracking data recorded in a 3D virtual urban environment. The analysis is based on eye tracking data of 20 participants, who freely explored the virtual city Seahaven for 90 minutes with an immersive VR headset with an inbuild eye tracker. To extract what participants looked at, we defined “gaze” events, from which we created gaze graphs. On these, we applied graph-theoretical measures to reveal the underlying structure of visual attention. Applying graph partitioning, we found that our virtual environment could be treated as one coherent city. To investigate the importance of houses in the city, we applied the node degree centrality measure. Our results revealed that 10 houses had a node degree that exceeded consistently two-sigma distance from the mean node degree of all other houses. The importance of these houses was supported by the hierarchy index, which showed a clear hierarchical structure of the gaze graphs. As these high node degree houses fulfilled several characteristics of landmarks, we named them “gaze-graph-defined landmarks”. Applying the rich club coefficient, we found that these gaze-graph-defined landmarks were preferentially connected to each other and that participants spend the majority of their experiment time in areas where at least two of those houses were visible. Our findings do not only provide new experimental evidence for the development of spatial knowledge, but also establish a new methodology to identify and assess the function of landmarks in spatial navigation based on eye tracking data
Allium pallasii and A. caricifolium - Surprisingly Diverse Old Steppe Species, Showing a Clear Geographical Barrier in the Area of Lake Zaysan
Polymorph Allium pallasii s.l. from monotypic A. sect. Pallasia was studied using a wide spectrum of methods and divided into two clearly morphologically, geographically, cytologically and genetically isolated species: A. pallasii s. str.—North-East Kazakhstan, Western Siberia, and the Altai Mountains; A. caricifolium—Kyrgyzstan, Northwest China, South-East Kazakhstan until Zaysan Lake in the east. Despite serious genetic differences, both species are sisters and are related to species of the A. sect. Codonoprasum (Subg. Allium). Allium caricifolium differs from A. pallasii s. str. by taller stems, dense inflorescence, and with filaments longer than perianth. The possible phylogenetic reasons for the separation of these species are discussed. A nomenclature analysis of synonyms was carried out
Effect of Environmental Temperatures on Proteome Composition of Salmonella enterica Serovar Typhimurium
Salmonella enterica serovar Typhimurium (STM) is a major cause of gastroenteritis and transmitted by consumption of contaminated food. STM is associated to food originating from animals (pork, chicken, eggs) or plants (vegetables, fruits, nuts, and herbs). Infection of warm-blooded mammalian hosts by STM and the underlying complex regulatory network of virulence gene expression depend on various environmental conditions encountered in hosts. However, less is known about the proteome and possible regulatory networks for gene expression of STM outside the preferred host. Nutritional limitations and changes in temperature are the most obvious stresses outside the native host. Thus, we analyzed the proteome profile of STM grown in rich medium (LB medium) or minimal medium (PCN medium) at temperatures ranging from 8 °C to 37 °C. LB medium mimics the nutritional rich environment inside the host, whereas minimal PCN medium represents nutritional limitations outside the host, found during growth of fresh produce (field conditions). Further, the range of temperatures analyzed reflects conditions within natural hosts (37 °C), room temperature (20 °C), during growth under agricultural conditions (16 °C and 12 °C), and during food storage (8 °C). Implications of altered nutrient availability and growth temperature on STM proteomes were analyzed by HPLC/MS-MS and label-free quantification. Our study provides first insights into the complex adaptation of STM to various environmental temperatures, which allows STM not only to infect mammalian hosts but also to enter new infection routes that have been poorly studied so far. With the present dataset, global virulence factors, their impact on infection routes, and potential anti-infective strategies can now be investigated in detail. Especially, we were able to demonstrate functional flagella at 12 °C growth temperature for STM with an altered motility behavior
Performance Evaluation of GNSS Position Augmentation Methods for Autonomous Vehicles in Urban Environments
Global Navigation Satellite Systems provide autonomous vehicles with precise position information through the process of position augmentation. This paper presents a series of performance tests aimed to compare the position accuracy of augmentation techniques such as classical Differential Global Navigation Satellite System, Real-time Kinematic and Real-time eXtended. The aim is to understand the limitations and choose the best position augmentation technique in order to obtain accurate, trustworthy position estimates of a vehicle in urban environments. The tests are performed in and around the German cities of Wuppertal and Duesseldorf, using a vehicle fitted with the navigation system POS-LV 220, developed by Applanix Corporation. In order to evaluate the real-time performance of position augmentation techniques in a highly challenging environment, a total of four test regions are selected. The four test regions are characterized mainly by uneven terrain with tall buildings around the University of Wuppertal, flat terrain with roads of varying width in the city centre of Wuppertal and Duesseldorf and flat terrain in a tunnel section located in the city of Wuppertal. The performances of the different position augmentation are compared using a Root Mean Square (RMS) error estimate obtained as an output from the Applanix system. Furthermore, a High-Definition map of the environment is used for the purpose of model validation, which justifies the use of RMS error estimate as an evaluation metric for the performance analysis tests. According to the performance tests carried out as per the conditions specified in this paper, the Real-time eXtended (RTX) position augmentation method enables to obtain a more robust position information of the vehicle than Real-time Kinematic (RTK) method, with a typical accuracy of a few centimeter in an urban environment
ORCID coverage in research institutions - Readiness for partially automated research reporting
Reporting and presentation of research activities and outcome for research institutions in official, normative standards are more and more important and are the basis to comply with reporting duties. Institutional Current Research Information Systems (CRIS) serve as important databases or data sources for external and internal reporting, which should ideally be connected with interfaces to the operational systems for automated loading routines to extract relevant research information. This investigation evaluates whether (semi-) automated reporting using open, public research information collected via persistent identifiers (PIDs) for organizations (ROR), persons (ORCID), and research outputs (DOI) can reduce effort of reporting. For this purpose, internally maintained lists of persons to whom an ORCID record could be assigned (internal ORCID person lists) of two different German research institutions—Osnabrück University (UOS) and the non-university research institution TIB—Leibniz Information Center for Science and Technology Hannover—are used to investigate ORCID coverage in external open data sources like FREYA PID Graph (developed by DataCite), OpenAlex and ORCID itself. Additionally, for UOS a detailed analysis of discipline specific ORCID coverage is conducted. Substantial differences can be found for ORCID coverage between both institutions and for each institution regarding the various external data sources. A more detailed analysis of ORCID distribution by discipline for UOS reveals disparities by research area—internally and in external data sources. Recommendations for future actions can be derived from our results: Although the current level of coverage of researcher IDs which could automatically be mapped is still not sufficient to use persistent identifier-based extraction for standard (automated) reporting, it can already be a valuable input for institutional CRIS
Enzyme assays with supramolecular chemosensors – the label-free approach
Enzyme activity measurements are essential for many research areas, e.g., for the identification of inhibitors in drug discovery, in bioengineering of enzyme mutants for biotechnological applications, or in bioanalytical chemistry as parts of biosensors. In particular in high-throughput screening (HTS), sensitive optical detection is most preferred and numerous absorption and fluorescence spectroscopy-based enzyme assays have been developed, which most frequently require time-consuming fluorescent labelling that may interfere with biological recognition. The use of supramolecular chemosensors, which can specifically signal analytes with fluorescence-based read-out methods, affords an attractive and label-free alternative to more established enzyme assays. We provide herein a comprehensive review that summarizes the current state-of-the-art of supramolecular enzyme assays ranging from early examples with covalent chemosensors to the most recent applications of supramolecular tandem enzyme assays, which utilize common and often commercially available combinations of macrocyclic host molecules (e.g. cyclodextrins, calixarenes, and cucurbiturils) and fluorescent dyes as self-assembled reporter pairs for assaying enzyme activity
Optimizing the retrieval effort in stack-based storage systems
In this thesis we investigate three different aspects of stacking problems. In the context of stacking problems, a certain amount of items are usually stored in a limited number of stacks with limited stack height. At first we focus on an elementary loading problem, the parallel stack loading problem (PSLP). We consider the PSLP with the objective to minimize the number of reshuffles in the unloading phase. Since in the PSLP the incoming items have to be stored according to a fixed arrival sequence, some relocations cannot be avoided later on. In this context, we give a strengthened NP-completeness proof and study the two surrogate objective functions minimizing the total number of unordered stackings of adjacent items and the total number of badly placed items. These objective functions are established in the literature to estimate the number of reshuffles and we compare them theoretically as well as in a computational study with the desired objective function to minimize the number of reshuffles. For this purpose, mixed-integer programming (MIP) formulations and a simulated annealing (SA) algorithm are proposed. Altogether, it turned out that minimizing the number of reshuffles directly with a two-stage SA approach is superior to the application of exact MIP approaches to the surrogate objective function mentioned above. Furthermore, we consider the premarshalling problem, where items in a storage area have to be sorted for convenient retrieval. A new model for uncertainty is introduced, where the priority values induced by the retrieval sequence of the items are uncertain. We propose a robust optimization approach for this setting, study complexity issues and provide different MIP formulations. Moreover, we investigate helpful properties which can speed up the solution process in several cases. In a two-stage approach we use the results from the theoretical analysis and MIP formulations to calculate optimal objective values of optimally robust storage configurations in a first step and compute premarshalling solutions with a minimal number of reshuffles to reach optimal configurations in a second step. In a computational study using a wide range of benchmark instances from the literature, we investigate both the efficiency of the approach as well as the benefit and cost of robust solutions. We find that it is possible to achieve a considerably improved level of robustness by using just a few additional relocations in comparison to solutions which do not take uncertainty into account. Finally, we consider the process of unloading items from a storage in the case of the blocks relocation problem with item families. For a given sequence of families, it has to be decided which item of each demanded family is unloaded from the storage with the objective to minimize the total number of reshuffles. Besides new complexity results, we propose IP formulations and a two-stage SA algorithm. Computational results are presented for real-world data from a company, benchmark instances from the literature, and randomly generated instances with different characteristics
(Student) Biology Teachers and Sustainable Nutrition - Eating, Teaching, Indoctrinating?
Das derzeitige Nahrungsmittelsystem und die individuellen Ernährungsgewohnheiten der Menschen sind mitverantwortlich für eine Vielzahl der globalen Umweltprobleme, wie dem Klimawandel, dem Biodiversitätsverlust und einem erhöhten Risiko für nicht übertragbare Krankheiten, wie Herz-Kreislauf-Erkrankungen und Diabetes. Um die negativen Auswirkungen des Nahrungsmittelsystems zu verringern, ist eine Transformation hin zu nachhaltigeren Ernährungsweisen, inklusive einer Veränderung der individuellen Ernährungsgewohnheiten unabdingbar. Nachhaltige Ernährung hat somit eine Schlüsselfunktion, um die Sustainable Development Goals (SDGs) zu erreichen. Im Sinne der SDGs kann die notwendige Transformation durch eine Bildung für nachhaltige Entwicklung (BNE) in Schulen beschleunigt werden. Dazu gehört auch die Ausbildung von Biologielehrkräften zur Umsetzung von BNE in der Schule. Dabei eignet sich besonders das Thema nachhaltige Ernährung, um BNE im Biologieunterricht zu integrieren.
Durch den Austausch und die Bewertung unterschiedlicher Perspektiven auf nachhaltige Ernährung bietet Biologieunterricht zum Thema die Möglichkeit, die Bewertungskompetenz der Schüler:innen zu fördern. Insbesondere Biologielehrkräfte nehmen somit eine Schlüsselrolle ein, um Lernende als zukünftige Akteure des Wandels auszubilden. Da nachhaltige Ernährung nicht explizit in den schulischen Biologielehrplänen verankert ist, liegt es auch in der Verantwortung der Lehrkräfte zu entscheiden, ob und wie sie das Thema unterrichten. Zu diesem Zweck ist es wichtig, angehende Biologielehrkräfte als Individuen mit persönlichen Überzeugungen und Verhaltensweisen in Bezug auf nachhaltige Ernährung zu analysieren. Die Analyse wird umso wichtiger, wenn man bedenkt, dass viele Schüler:innen ihre Lehrkräfte als Vorbilder für nachhaltige Verhaltensweisen ansehen. Dies birgt die Gefahr, dass Schüler:innen dazu neigen, die Überzeugungen und Verhaltensweisen ihrer Lehrkräfte in Bezug auf nachhaltige Ernährung zu übernehmen, ohne deren Überzeugungen angemessen zu bewerten.
Wie der Titel "Eating, Teaching, Indoctrinating" bereits andeutet, zielt die vorliegende Dissertation vor dem Hintergrund des Indoktrinationsverbots in Deutschland darauf ab, einen Einblick in die persönlichen Ernährungsabsichten von angehenden Biologielehrkräften und ihrer Bereitschaft, nachhaltige Ernährung zu unterrichten zu geben. Daraus lassen sich Möglichkeiten für die Verankerung des Themas in der universitären Biologielehramtsausbildung ableiten, die möglicherweise auch die zukünftige Umsetzung von nachhaltiger Ernährung in der Schule fördern können. Jedes dieser drei Kernelemente der Dissertation verweist auf wesentliche Erkenntnisse aus den vier Studien.
Eating. Die ersten beiden Fragebogenstudien (Studie 1: N = 155; MAlter = 21.2; SD = 1.95 und Studie 2: N = 270; MAlter = 22.9; SD = 2.8) untersuchten, wodurch die Intention von angehenden Biologielehrkräften, sich nachhaltig zu ernähren beeinflusst wird. Beide Studien basierten auf einem erweiterten Modell der Theorie des geplanten Verhaltens (TPB). Zusätzlich zu den sozial-psychologischen Faktoren der TPB, wurde auch die Mensch-Natur-Beziehung durch die Hinzunahme mehrerer Variablen untersucht. Frühere Studien haben gezeigt, dass die Mensch-Natur-Beziehung ein Einflussfaktor für umweltfreundliches Verhalten ist, wozu auch eine nachhaltige Ernährung gezählt werden kann. Die Ergebnisse beider Studien zeigen, dass die Einstellungen über nachhaltige Ernährung, subjektive Normen und die wahrgenommene Verhaltenskontrolle die Intention, sich nachhaltig zu ernähren beeinflussen. Zudem konnten allgemeine Umweltbetroffenheit (Studie 1), altruistische Umweltbetroffenheit (Studie 2) und Naturverbundenheit (Studien 1 und 2) die Einstellungen gegenüber nachhaltiger Ernährung und die Intention, sich nachhaltig zu ernähren positiv voraussagen. Im Hinblick auf die Ausbildung von Biologielehrkräften regen beide Studien dazu an, die Bedeutung des eigenen nachhaltigen Ernährungsverhaltens von Biologielehramtsstudierenden und die zugrundeliegenden Determinanten für schulischen Biologieunterricht zum Thema zu untersuchen.
Teaching. Basierend auf den Ergebnissen der ersten beiden Studien wurde im Rahmen von Studie 3 untersucht, inwiefern die persönliche Intention, sich nachhaltig zu ernähren, die Intention, das Thema nachhaltige Ernährung zukünftig zu unterrichten beeinflusst. Neben der Intention, sich nachhaltig zu ernähren, wurde auch der Einfluss der traditionellen TPB-Variablen untersucht. Darüber hinaus wurde die TPB um Wissen über nachhaltige Ernährung sowie vorherige universitäre Erfahrungen mit (B)NE oder nachhaltiger Ernährung erweitert. Die Ergebnisse der Online-Fragebogenstudie mit 621 angehenden Biologielehrkräften (MAlter = 23.3; SD = 3.9) zeigten erstmalig, dass angehende Biologielehrkräfte mit einer höheren Intention, sich nachhaltig zu ernähren, positivere Einstellungen gegenüber dem Unterrichten von nachhaltiger Ernährung, eine höhere Selbstwirksamkeit und eine höhere Intention haben, um nachhaltige Ernährung zu unterrichten. Universitäre Vorerfahrungen zeigten ebenfalls einen Einfluss auf die Lehrintention. Höheres Wissen über nachhaltige Ernährung wurde lediglich in Verbindung zu einer höheren Selbstwirksamkeit gegenüber dem Unterrichten identifiziert, hatte aber keinen direkten Einfluss auf die Intention, nachhaltige Ernährung zu unterrichten.
Indoctrinating. Die identifizierten Zusammenhänge der ersten beiden Forschungsschwerpunkte deuten darauf hin, dass die unterrichtliche Umsetzung von nachhaltiger Ernährung in einem potenziellen Spannungsfeld mit der Indoktrination von Schülern in Bezug auf nachhaltige Ernährung steht. Ausgehend von dieser Prämisse wurde in der vierten Studie untersucht, ob Biologielehrkräfte ein Indoktrinationsrisiko beim Unterrichten von nachhaltiger Ernährung wahrnehmen und welche Methden sie anwenden würden, um Indoktrination zu vermeiden. Mittels semi-strukturierter Einzelinterviews wurden Biologielehrkräfte (N = 7) aus Osnabrück und dem Münsterland befragt. Die Ergebnisse zeigen, dass die Befragten ein hohes Indoktrinationsrisiko beim Unterrichten von nachhaltiger Ernährung wahrnehmen, das sie vor allem auf das Handeln der Biologielehrkräfte selbst zurückführen. Eine der schwierigsten Entscheidungen für die Befragten war die Frage, ob sie ihre eigenen Ernährungsgewohnheiten offenlegen sollten, wenn sie nachhaltige Ernährung unterrichten, und ob die Weitergabe dieser Information an die Schüler als Indoktrination angesehen werden könnte–insbesondere wenn es sich um eine vegetarische oder vegane Ernährung handelt. Die Denkweise der Befragten reichte von der Überzeugung, dass Lehrkräfte sehr zurückhaltend sein müssen, wenn sie nachhaltige Ernährung unterrichten, bis hin zu der Ansicht, dass sie offen sein und ihren eigenen Ernährungsstil im Unterricht preisgeben sollten. In letzterem Fall sahen die Befragten nahezu keine Gefahr der Indoktrination, sondern eher Vorteile für die Schüler-Lehrer-Beziehung, die Authentizität der Lehrkräfte und für die Ermutigung der Schüler:innen, ihre eigene Position im Prozess der Nachhaltigkeitstransformation zu erkunden. Die Befragten diskutierten schülerzentriertes und multiperspektivisches Lernen als Beispiele für nicht-indoktrinierende Unterrichtsansätze. Der Einsatz dieser Unterrichtsansätze im Rahmen einer BNE und allgemein zur Förderung der Bewertungskompetenz von Schüler:innen im Biologieunterricht wurde als vorteilhaft angesehen.
Zusammenfassend geben die vier empirischen Studien einen ersten Einblick, ob und inwiefern (angehende) Biologielehrkräfte das Thema nachhaltige Ernährung im schulischen Biologieunterricht adressieren würden, von welchen Faktoren diese Intention abhängt und welche Bedenken sie beim Unterrichten von nachhaltiger Ernährung haben. Auf dieser Grundlage werden in der Dissertation erste Empfehlungen für zukünftige Forschung und die Ausgestaltung von Seminaren über nachhaltige Ernährung in der Biologielehramtsausbildung diskutiert. Sofern angehende Biologielehrkräfte im Rahmen ihres Studiums Möglichkeiten zur methodischen Umsetzung und unterrichtlichen Einbettung des Themas nachhaltige Ernährung erlernen, könnte auch die zukünftige Implementierung des Themas in den schulischen Biologieunterricht gefördert werden
NC-AFM high-resolution studies of the calcite(104) surface at low temperatures with and without submonolayer of CO and H2O
The central aim of this thesis is to give further insights into the structural properties of the calcite(104) surface and especially to unravel its surface reconstruction by clarifying the contradicting reports of the (2 × 1) and row-pairing reconstruction. Within this thesis, the pristine as well as the CO and H2O-covered calcite(104) surface is investigated for the first time with non-contact atomic force microscopy (NC-AFM) operated at 5 K. CO terminated
tips are used in the measurements to benefit from the improved contrast capabilities and detailed understanding of contrast formation with these tips
