Geopolitics of Food
The paper analyzes the food security with the four dimensions physical availability, economic and physical access, food use/utilization and stability, the economy and ecology of food production (plants, meat, fish), the risk factors for food security and the geopolitical activities. The food production increased almost 2.2 times in the past 50 years and was able to compensate the population growth, but the UN Food and Agriculture Organization (FAO) estimates that the food production must increase from 2015 to 2050 by 60%. The growth was achieved by a combination of increased irrigation, fertilizers, pesticides, genetically modified organisms (GMOs) to protect plants from harmful insects and pesticides and a general trend to larger farms and monocultures. Issues are nitrate overload, pesticide contamination, reduction of insects, pollination and birds, loss of biodiversity and land loss and degradation. Smart or precision farming based on the Internet of Things, 5 G networks, drones and Artificial Intelligence may lead to a more effective and ecologic resource use. Over the past 50 years, meat production has more than tripled. Meat production requires land for pastures and animal feed production. Meat replacements and alternatives like mock food, meat from cell cultures and insects have a small, but growing market. Another issue are livestock emissions like carbon dioxide and methane. In the past 50 years, fish production has increased by almost 4 times. Approximately one-third of the fish resources are overfished and some species are in a critical condition. The USA were the largest exporter of food in 2020 while Asia was the largest net importer. Europe was a net importer of food for many decades, but became a net exporter since 2013. While Africa’s imports and exports were balanced until the mid 1970ies, the gap between imports and exports is widening. The global food market is increasingly dominated by a few multinational companies from seeds to supermarkets. The most critical risk factors for food security are climate change with soil loss and water scarcity due to higher temperatures, extreme weather with storms, droughts (and floods with continued soil loss due to wind and water erosion. Water waste and low water prices are drivers of water scarcity, e.g., by overuse of groundwater resources like aquifers. A new threat to freshwater is the global contamination with microplastic. After the food price crisis from 2007/2008, food was recognized as strategic resource leading to land lease and buying, also known as Foreign Direct Investments (FDIs) or more critically as ‘land grabbing’ to prevent bottlenecks in the long term. The Black Sea Grain Initiative demonstrates the vulnerability of global food supply chains and the possibility to weaponize food. Two international water conflicts are ongoing with the Grand Ethiopian Renaissance Dam GERD in East Africa and the India-China Transboundary Water Dispute. In summary, the food, meat, and fish production could be successfully increased to compensate the population growth and the increasing meat consumption, but hunger and undernourishment still exist. Main risk factors for future food security are climate change, land loss and water scarcity. In response, food is seen as strategic resource resulting in geopolitical activities and food and water conflicts
Russlanddeutsch(e)

Obligate mutualistic cooperation limits evolvability
Cooperative mutualisms are widespread and play fundamental roles in many ecosystems. Given that these interactions are often obligate, the Darwinian fitness of the participating individuals is not only determined by the information encoded in their own genomes, but also the traits and capabilities of their corresponding interaction partners. Thus, a major outstanding question is how obligate cooperative mutualisms affect the ability of organisms to adapt evolutionarily to changing environmental conditions. Here we address this issue using a mutualistic cooperation between two auxotrophic genotypes of Escherichia coli that reciprocally exchanged costly amino acids. Amino acid-supplemented monocultures and unsupplemented cocultures were exposed to stepwise increasing concentrations of different antibiotics. This selection experiment reveals that metabolically interdependent bacteria are generally less able to adapt to environmental stress than autonomously growing strains. Moreover, obligate cooperative mutualists frequently regain metabolic autonomy, resulting in a collapse of the mutualistic interaction. Together, our results identify a limited evolvability as a significant evolutionary cost that individuals have to pay when entering into an obligate mutualistic cooperation
Repairing the Breach: Uncovering ER-dependent Membrane Repair during Mycobacterial Infection
Tuberculosis is one of the top deadliest infectious diseases. Its causative agent, Mycobacterium tuberculosis, employs numerous strategies to survive and persist in macrophages. It induces membrane damage to reduce the endolysosomal toxicity and to exit the Mycobacterium-containing vacuole (MCV). Lesions in the MCV trigger host membrane repair machineries that restore its function and prevent the escape of bacteria. Recently, an ER-dependent repair of damaged lysosomes that is fostered by lipid transfer proteins of the oxysterol binding protein (OSBP) family was described. However, these membrane damage-repair events and especially the role of this pathway during M. tuberculosis infection are largely unexplored. To reveal the underlying molecular mechanisms, I employed the Dictyostelium discoideum/ M. marinum system. In the first part of this thesis, I established 3D-HPF/FS-CLEM together with the electron microscopy facility. This technique is robust, preserves membranes in their utmost native state and provides near-atomic axial resolution. Importantly, it allows envisioning organelle interactions and exit events of mycobacteria in D. discoideum. In the second part of this thesis, I uncovered the formation and function of ER-dependent repair pathway during mycobacterial infection. I found that the ER-Golgi protein OSBP8 re-localizes to the MCV as well as to cytosolic mycobacteria. Strikingly, the mobilisation of OSBP8 is dependent on the presence of the mycobacterial type VII secretion system ESX-1 and presumably on MCV damage. Using 3D-HPF/FS-CLEM and other advanced imaging approaches, I observed that ruptured MCVs recruit ER-tubules containing OSBP8 in its close proximity. Exploring the functional implications of ER-MCV contact sites revealed that depletion of OSBP8 leads to hyperaccumulation of phosphatidylinositol 4-phosphate PI4P on the MCV/lysosomes suggesting OSBP8 balances PI4P on damaged membranes. In addition to this, sterol accumulation in the MCV was slightly reduced. Next, by employing fluorescent probes I observed decreased lysosomal acidification and proteolytic activity in the OSBP8 knockout cells, which promoted mycobacterial growth. Conversely, I observed a growth restriction of mycobacteria in cells overexpressing OSBP8. Next, the conservation of the ER-dependent repair pathway in M. tuberculosis infected macrophages was tested. Remarkably, in agreement to OSBP8 mobilisation, the ER-Golgi protein OSBP re-localized to intracellular mycobacteria in human macrophages in an ESX-1-dependent manner. Taken together, these findings provide the evidence the ER-dependent membrane repair contributes to host resistance against intracellular pathogens including M. tuberculosis
Personalized Assessment and Network Model Feedback: An Empirically-Based Approach to Personalizing Psychotherapy
Background: Network modeling has become an increasingly popular method of data analysis within psychology. The appeal of visually displaying relationships between psychological variables (i.e., individual symptoms) may be explained by its potentials for use in clinical practice. Most network model studies suggest that future research could have relevant clinical implications. What that means, concretely, has not been explored in much depth. One potential avenue towards integrating network models into clinical practice constitutes providing treating psychotherapists with their patients’ networks, in the form of feedback. The research of feedback in psychotherapy shows that providing psychotherapists with more information about their patients most often leads to improved therapeutic outcomes (i.e., faster and greater reduction of symptomatology). In order to create these patient-level networks, intensive data collection schedules (e.g., Ecological Momentary Assessment; EMA), can be used. Methods: Providing therapists with their patients’ EMA-based network models, exploring how these models were utilized, and measuring to what extent this form of feedback beneficially impacts therapeutic outcomes are the foci of the TheraNet Project. This dissertation provides first insights into how the TheraNet Project was developed (Study 1), implemented (Study 2), and perceived by participating therapists (Studies 3 and 4). In Study 1, an overview of existing EMA methodologies were explored in an effort to provide guidance for the EMA design within TheraNet. Study 2 provided detailed information about the finalized design of TheraNet, and included two examples of patient feedback and their treating therapists’ reactions thereto. Study 3 focused on the therapists’ first reactions to their first TheraNet patients’ feedback in the context of an interpretation workshop. Lastly, Study 4 analyzed retrospective focus groups with therapists who had been participating for a longer amount of time, as a way to understand their use of the feedback over time/across patients and to gain more insights into potential obstacles to use. Results: Findings indicate that networks may hold value as clinical feedback tools for therapists, though several suggestions for the networks’ clinical optimization were made too. Other contextual factors also significantly contributed to the use or non-use of the network feedback, which should be considered in future research. Conclusions: Networks can be useful in clinical practice, though contextual/systemic barriers may cause problems in the implementation of these kinds of studies. Quantitative, session-to-session measurement of therapeutic progress and outcomes within the TheraNet Project are still ongoing and will need to be interpreted within the context of the results described here
Following the path of survey optimization - Effects of Survey Design on Various Quality Criteria of Surveys
