Molecular changes in hippocampal energy metabolism in mice selectively bred for extremes in stress reactivity: Relevance of mitochondrial dysfunction for affective disorders

    Get PDF
Affective disorders, such as major depression, are frequently associated with metabolic disturbances involving mitochondria. Although dysregulation of the hypothalamic–pituitary–adrenal (HPA) axis is known to alter energy metabolism, the precise mechanisms linking stress and metabolic disturbances are not sufficiently understood. We used a mouse model of affective disorders to investigate the impact of a genetic predisposition for extremes in stress reactivity on behavioural and metabolic phenotypes as well as energy metabolism. Adult males of three independent mouse lines selectively bred for high, intermediate or low HPA axis reactivity were tested for exploratory and locomotor activity as well as stress-coping behaviour. Additionally, basal and stress-induced plasma corticosterone levels, body weight, food intake and body composition were measured. At the molecular level, the hippocampal transcriptome was analysed using microarray, serial analysis of gene expression and qRT-PCR. Finally, mitochondrial DNA copy number, damages and mitochondrial respiration were assessed. We found clear effects of the differential stress reactivity on the behavioural, morphometric and metabolic measures. Remarkably, the hyperactive behavioural and neuroendocrine stress-coping style of high-reactivity mice was associated with significant changes in the expression of an extended list of genes involved in energy metabolism and several mitochondrial functions. Yet, only minor changes were found in mitochondrial DNA copy number, damages and respiration. Thus, our findings support a prominent role of glucocorticoids in shaping the major endophenotypes of the stress reactivity mouse model and contribute towards understanding the important role of HPA axis dysregulation and changes in energy metabolism in the pathophysiology of affective disorders

The Role of Non-Governmental Organisations in Politicising European Security

    Get PDF
The role of non-governmental organisations (NGOs) in politicisation and European security is highly undertheorised and suffers from a lack of research. The realisation that little is known about those organisations engaging in the EU policy field (empirical interest) and their involvement in politicisation (conceptual interest) is the main driver of the thesis and was transferred in the following research question: “What role do NGOs play in politicising European security?” The thesis responds to this question with taking a) Brussels-based and national NGOs and b) the subfield of EU counter-terrorism into account. In concrete, the dissertation project analyses the involvement and engagement of these organisations in regard to three counter-terrorism legislations: The EU data retention directive, the EU PNR directive and the EU terrorist content online regulation. With recourse to the prominent literature, politicisation is understood as a process of drawing an issue discussed behind closed doors in the public sphere and making it part of public deliberation. To study the role of NGOs, three strategies present in interest group literature voice (outside lobbying), access (inside lobbying) and litigation (as legal means) are scrutinised in detail and linked to the conception of politicisation. A final comparison of the introduced cases demonstrates that politicisation processes are distinct with regard to the three legislative acts examined. While NGOs succeeded in drawing the EU data retention directive into the public sphere, the politicisation connected to terrorist content online was characterised by a debate between experts, who work with the affected technology, while the issue of passengers’ flight data was only hardly publicly deliberated. The main finding of the project is, that privacy and data protection NGOs play a role in politicisation, but that role is highly context-dependent: It depends on whether a favourable political-security culture is in place, whether the issue is conducive (“intrusive”) and provides an anchor for framing as well as whether NGOs have sufficient (financial and human) resources to become active as politicisers. The innovative theoretical framework to study NGO-driven politicisation processes can be regarded as a basis for future research focusing on NGOs working in EU security (e.g. EU migration and border management), different oriented NGOs (e.g. with a focus on environmental, trade, LGBTQ policy) or on other types of non-state actors (e.g. interest groups, social movements)

The Politics of Presence in Asylum. Negotiating the Inclusion:Exclusion of Refugees in Osnabrück

    Get PDF
In her cumulative dissertation, Sophie Hinger examines local responses to and consequences of asylum migration. She starts from the observation that the way asylum migration and refugees are dealt with locally is not only conditioned by (inter-)national policies and their implementation, but by a multitude of actors and factors and their interaction. By interrogating different ‘zones of negotiation’ (reception, accommodation, deportations, integration policy) and drawing on numerous original interviews, observations, and documents, she analyses discourses and practices of inclusion and exclusion of refugees. The medium-sized German city of Osnabrück serves as an entry point for the investigation of these multi-scalar and multi-local zones of negotiation. Sophie Hinger argues that at the heart of local negotiations over asylum lies the contested presence of protection-seekers and the opportunities for participation that result from this presence or are claimed based on it. She advances the notion of a ‘politics of presence’, which denotes those practices that challenge attempts to control migration and existing boundaries of belonging and opportunities for participation. The politics of presence comprise the implicit politics of refugees' coming to and being in the city, explicit demands for opportunities to participate, as well as institutional measures of inclusion (even or especially if these oppose restrictive national migration and asylum policies). Such practices are not necessarily local or urban, but as is argued in the dissertation, it is often in and through cities that conflicts over the presence of asylum seekers are played out and that the politics of presence come to the fore. With the concept of the politics of presence, Sophie Hinger contributes to debates on local migration politics at the intersection of migration, urban, and social movement studies

The Control of the Lunar South Pole

    Get PDF
After the Apollo missions to the moon in 1969 and the 1970ies, the space research shifted to other goals. But since the 1990ies, the lunar research was restarted and intensified. Initially, the focus was on the development of science and technology for future moon missions. In addition to space agencies, commercial providers became increasingly active and were e.g., motivated by the Google Lunar X-Prize competition, which originally intended to award projects for a moon landing until 2018 the latest. But with the release of the US Commercial Space Launch Competitiveness Act in 2015, the focus began to shift from scientific to economic aspects, i.e., the potential exploitation of lunar resources. However, for a long-term presence, a habitable moon base would be needed with water, enough sunlight, and a stable landing place. The best combination of these factors is present on certain places of the Lunar South Pole which is now the key area of space geopolitics. Lunar settlements without access to lunar water are not possible and who controls the lunar water, has factual control over future lunar long-term activities. Indications for the existence of lunar water ice were found in the late 1990ies. Further research identified six craters on the Lunar South Pole as likely targets for water ice deposits and direct evidence for water ice resulted from lunar impactor studies. The United States with the Artemis program and China with the Chang’e program make steady progress to permanent moon bases (Artemis Base Camp vs. International Lunar Research Station ILRS) in overlapping lunar South Pole areas. Other competitors like Russia, India, Israel, and Japan were affected by landing accidents. Meanwhile, both East (China/Russia) and the West suspect each other that the true objective of their lunar research is the control of the Lunar South Pole and its water ice which would constitute a factual control over moon settlements and consequently of the lunar resources. There are growing tensions between United States and China in space policy. With respect to the moon, both sides have formed growing multi-national space alliances, the US Artemis Accord with over 20 countries including India and the Chinese International Lunar Research Station Cooperation Organization ILRSCO with 5 initial member states countries including Russia. Satellite hacking and laser weapons made substantial progress. Consequently, high altitude espionage balloons were re-activated as safer alternatives as they are low-cost, unmanned, can carry payloads for electronic warfare, surveillance, jamming and observation, are difficult to detect, and can be easily replaced. From a strategic perspective, the United States were leading for decades, but as there were substantial delays in the implementation of the Artemis program already, it is meanwhile possible that China’s moon base will come first. For this reason, every effort must be made by the United States to meet the future Artemis timelines, if they want to keep the leadership

Combinatorial Aspects of Horizontal Visibility Graphs, Symmetric Edge- and Laplacian Polytopes

    Get PDF
In this thesis we study three main objects; horizontal visibility graphs (HVGs for short), symmetric edge polytopes and Laplacian polytopes. Chapters 2 and 3 focus on HVGs. In particular, we show that HVGs corresponding to a time series of length N of pairwise distinct and arbitrary data points are counted by the (N-1)st Catalan number and the (N-2)nd Schröder number, respectively. Moreover, extending previous work by Lacasa and Luque, we prove that HVGs associated to data sequences without equal entries are completely determined by their ordered degree sequence. Additionally, we provide an algorithm, which extends the fast horizontal visibility algorithm of Zhu et al. (2012). The extended version remains in O(N) in the worst case, works efficiently on streamed data, and is parallelizable. This approach enables the computation of HVGs with millions of vertices in a short period, opening up new application areas of HVGs for time series generated batch-wise or resulting from measurements with a high sampling rate. In Chapter 4, we then focus on the symmetric edge polytope associated to a simple graph. We study their γ\gamma-vectors, which are associated with their hh^*-vectors, both from a deterministic and a probabilistic point of view. On the deterministic side, we prove non-negativity of γ1\gamma_1 and γ2\gamma_2 for any graph and completely characterize the case when γ2=0\gamma_2=0. On the probabilistic side, we show that the γ\gamma-vectors of symmetric edge polytopes of most Erdős-Rényi random graphs are asymptotically almost surely non-negative up to any fixed entry. This proves that Gal's conjecture holds asymptotically almost surely for arbitrary unimodular triangulations in this setting. In the last chapter of this thesis, we focus on Laplacian polytopes associated to simplicial complexes. Given a (finite) simplicial complex, we define its i-th Laplacian polytope as the convex hull of the columns of its i-th Laplacian matrix. This extends Laplacian simplices of finite simple graphs, as introduced by Braun and Meyer. After studying basic properties of these polytopes, we focus on the d-th Laplacian polytope of the boundary of a (d+1)-simplex (σd+1)\partial(\sigma_{d+1}). If d is odd, then as for graphs, the d-th Laplacian polytope turns out to be a (d+1)-simplex in this case. If d is even, we show that the d-th Laplacian polytope of (σd+1)\partial(\sigma_{d+1}) is combinatorially equivalent to a d-dimensional cyclic polytope on d+2 vertices. Moreover, we provide an explicit regular unimodular triangulation for the d-th Laplacian polytope of (σd+1)\partial(\sigma_{d+1}). This enables us to compute the normalized volume and to show that the hh^\ast-polynomial is real-rooted and unimodal, if dd is odd and even, respectively

    Geopolitik der Ernährung

    No full text
