Expansion of myeloid suppressor cells and suppression of Lassa virus-specific T cells during fatal Lassa fever
International audienceLassa fever is a highly lethal hemorrhagic fever endemic to West Africa. In the absence of efficient prophylactic or therapeutic countermeasures, it poses a substantial threat to public health in this region. The pathophysiological mechanisms underlying the severity of the disease are poorly known because Lassa virus (LASV), its causative agent, has to be handled in BSL-4 laboratories and access to clinical samples is difficult. The control of Lassa fever is associated with a rapid and well-balanced immune response and viral clearance. However, severe disease is characterized by uncontrolled innate immune activation and symptoms reminiscent of sepsis and a cytokine storm. In a model of cynomolgus monkeys infected with two different strains of the virus, one causing moderate disease and the other a lethal outcome, we show that the control of LASV infection is characterized by the induction of a LASV-specific T-cell response, whereas severity is associated with the expansion of suppressive myeloid cells, alterations of the stromal network of secondary lymphoid organs, and the anergy of specific T cells. These results suggest that T cells are crucial for the control of LASV and that immunomodulatory therapeutics, such as checkpoint inhibitors, could contribute to new therapeutic strategies to treat Lassa fever. They also highlight how immunosuppressive mechanisms described in sepsis and cancer patients may play a role in the pathogenicity of Lassa fever, as well as in other similar hemorrhagic fevers
Epidemiological and clinical insights into enterovirus circulation in Europe, 2018 - 2023: a multi-center retrospective surveillance study
International audienceAbstract Background Enteroviruses (EV) cause yearly outbreaks with severe infections, particularly in young children. This study investigates EV circulation, age-distribution, and clinical presentations in Europe from 2018-2023. Methods Aggregated data were requested from ECDC National Focal Points for Surveillance and European Non-Polio Enterovirus Network. Data included detection month, specimen type, age-group, and clinical presentation for the ten most commonly reported EV types per year. Findings Twenty-eight institutions from 16 countries reported 563,654 EV-tests during the study-period with 33,265 (5.9%) EV-positive. Forty-two types were identified (n=11,605 cases) with echovirus (E)30, coxsackievirus (CV)A6, EV-D68, E9, E11, CVB5, E18, CVB4, EV-A71, and E6 most frequently reported. E30 detection declined after 2018/2019, while CVA6, CVB5, E9, E11, and EV-D68 were prevalent both before and after the COVID-19 pandemic, and CVB4 and E18 were prevalent after the pandemic. Over the study period, a shift in seasons (summer to fall) and specimen positivity (feces to respiratory) was observed. Neurological signs predominated among EV-A71, CVB4, CVB5, E6, E9, E11, E18, and E30 (30-72%). CVB4, CVB5, E9, E11, and E18 were also frequently reported among neonates (18-32%). CVA6 was frequently associated with HFMD, and EV-D68 with respiratory infections. Paralysis was reported among 22 infections, associated with ten non-polio types. Conclusion This study emphasizes the widespread circulation and severe nature of EV infections in Europe, particularly among neonates, as well as the (re-)emergence of specific types post-pandemic. Our findings highlight the need for continuous EV-surveillance to monitor variation in circulation, age, and clinical presentations, including paralysis among non-polio EV infections
QT Interval, Antipsychotics and Correlates Among Patients with Schizophrenia: Cross-Sectional Data from the Multicentric Real-World FACE-SZ
International audienceBackground: The life expectancy of patients with schizophrenia is reduced, partly due to cardiovascular diseases. Antipsychotics are associated with QT interval prolongation, which is a risk factor for arrhythmia and cardiac arrest. The differences between antipsychotic with regard to QT interval prolongation are not well understood.Objective: The aim was to compare the QT values associated with different antipsychotics within a real-world population of subjects with clinically stable forms of schizophrenia.Methods: The FACE-SZ cohort comprises subjects with psychotic disorders, referred to schizophrenia expert cents. QT interval was measured, as well as all treatments (psychotropic and others). The following maintenance treatment for schizophrenia was analysed cross-sectionally: aripiprazole, clozapine, haloperidol, amisulpride, olanzapine, quetiapine, risperidone. Age, sex, smoking status, body mass index, blood potassium levels, and the co-prescription of another QT-prolonging treatment were used as adjustment factors in multivariable linear regression analyses.Results: Among 792 patients, the mean corrected QT (QTc) interval in the sample of patients under monotherapy was 407 ms. The mean age was 31.7 years, and the majority were male (73.3 %). In comparison to the rest of the sample, clozapine was associated with a longer QTc interval (β = 0.012, 95% CI [0.006-0.018]), while aripiprazole was significantly associated with a shorter QTc interval (β = - 0.010, 95% CI [- 0.016 to - 0.005]). Other antipsychotics were not associated with significant variations of the QTc.Conclusions: The prescription of antipsychotics should always be accompanied by close monitoring of the QTc interval to prevent the risk of severe cardiac arrhythmia, particularly concerning clozapine

International audienceWhile contemporary architectural issues focus on urban and terrestrial housing, living on or in water is a special experience, at least for humans. The ocean may at first seem like a place that cannot be lived in, but environmental humanities make it an ontological one. Living in/on the Ocean has become mandatory for the inhabitants of Lower Manhattan in Kim Stanley Robinson’s New York 2140 (2017), following a double rise of water level during the 21st century that submerged the southern part of the city, and an escape for the Noahs, victims of the deadly military imperialism of the Earthlings in Hugo Verlomme’s Mermere (2020), whose plot is set in an undated future. In fact, we’ll be examining the ways in which the ocean can be “inhabited” in the Anthropocene era, whether it is the ecotone interweaving land and ocean that New York has become, or the oceanic ecotopia of Mermere.Si les problématiques contemporaines de l’architecture insistent sur l’habitation urbaine et terrestre, habiter sur ou dans l’eau est une expérience particulière en tout cas pour les humains. L’océan apparaît comme un lieu a priori inhabitable, mais les humanités environnementales en font un lieu ontologique. Habiter l’Océan est devenu une nécessité pour les habitants de Lower Manhattan dans New York 2140 (2017) de Kim Stanley Robinson à la suite d’une double montée des eaux au cours du XXIe siècle qui a submergé la partie sud de la ville, une échappatoire pour les Noés victimes de l’impérialisme militaire et mortifère des Terriens dans Mermere (2020) d’Hugo Verlomme dont l’intrigue est située dans un avenir non daté. PD-L1<sup>+</sup> plasma cells suppress T lymphocyte responses in patients with sepsis and mouse sepsis models
International audienceSepsis, a leading cause of death in intensive care units, is associated with immune alterations that increase the patients’ risk of secondary infections and mortality, so better understandings of the pathophysiology of sepsis-induced immunosuppression is essential for the development of therapeutic strategies. In a murine model of sepsis that recapitulates immune alterations observed in patients, here we demonstrate that PD-L1+ CD44+B220LowCD138+IgM+ regulatory plasma cells are induced in spleen and regulate ex vivo proliferation and IFNɣ secretion induced by stimulation of T splenocytes. This effect is mediated both by cell-cell contact through increased PD-L1 expression on plasma cells and by production of a soluble factor. These observations are recapitulated in three cohorts of critically ill patients with bacterial and viral sepsis in association with increased mortality. Our findings thus reveal the function of regulatory plasma cells in the pathophysiology of sepsis-induced immune alterations, and present a potential therapeutic target for improving immune cell function impaired by sepsis
Saturation dans les Réseaux de Neurones Récurrents: Expressivité, Apprenabilité et Généralisation.
Since their humble beginning in the 40s, passing by the dashed hopes of researchers in the 70s and their comeback in the 90s, neural networks have never ceased to surprise us and have taken on an increasingly important role in everyday life. The explosion in the computing capacity of our machines and the abundance of data has propelled neural based algorithms to the forefront of the scene. But their undeniable success is coupled with a lack of understanding of their inner functioning. The more complex the algorithms become, the less we can understand what drives their decisions. In this vast class of algorithms that make up Neural Networks, Recurrent Neural Networks (RNNs) have a singular place. On the one hand, their main function is to process sequential data for large-scale applications involving text, audio, video and so on. On the other hand, because of the ambivalence of their inner workings, resembling at once non-recurrent neural networks in their structure, but at the same time exhibiting behaviors incomparable with the latter, the problem of establishing theoretical results and algorithms enabling a better understanding of their operation is a major one.In this thesis we present our theoretical contributions on RNNs. Our contributions span across several fields such as: \textbf{expressivity}, \textbf{learnibility} and \textbf{generalisation}. Expressivity is a global property, in the sense that we talk about the expressivity of a set of functions. Determining the expressivity of a set of RNNs is the act of finding the most complex class of functions that the RNNs in can simulate. Learnability is, in a way, expressivity but in a more constrained way, where the aim is to determine what is possible (or not possible) to learn for a given RNN architecture. Guarantees of generalisation have been a topical issue since the introduction of learning algorithms. In very simple terms these guarantees are used to ensure that the model, in our case a RNN, has learned what it is supposed to learn. In the field of expressivity, we started from a recently introduced framework and extended it in order to study a larger portion of RNN architectures. We propose a reformulation of this framework allowing us to import some of the handy properties to a real world scenario. On the one hand, this reformulation enables us to fill a theoretical gap, but it also paves the way for the other contributions. In the field of learnability our contribution is a theoretical demonstration of the limits of what can be learned by gradient descent for a class of RNNs the Simple Recurent Networks (SRNs). We build on the phenomenon of the vanishing gradient and combine this fact with the finite precision of computers, in order to delimit areas of the parameter space inaccessible for SRNs with current gradient descent algorithms. This demonstration is accompanied by experiments and a proof that the inaccessible areas of parameter space contain functions of interest such as finite-state machines. Finally in the field of generalisation guarantees, we derive PAC-Bayes bounds for SRN that is \emph{independent of data length}. We would like to emphasize the originality of this result in relation to comparable works in the literature, whose results are either more limited or systematically dependent on data length. The singular features of our work that set us apart are: 1) all our main results are independent of data length, 2) we have always considered the case where RNNs are defined with non-linear activation functions. Contrary to results where RNNs are assumed to be defined with linear or ReLU functions to simplify the analysis, we have instead incorporated the particularities of bounded functions such as the hyperbolic tangent function and the sigmoidal function. 3) we have incorporated the computer finite precision into our theoretical work.Depuis leurs modestes débuts dans les années 40, en passant par les espoirs déçus des chercheurs dans les années 70 et leur retour dans les années 90, les Réseaux Neuronaux n’ont cessé de nous surprendre et ont pris une place de plus en plus importante dans la vie de tous les jours. L’explosion de la capacité de calcul de nos machines et l’abondance des données a propulsé les algorithmes à base de neurones sur le devant de la scène. Mais leur succès indéniable s’accompagne d’une incompréhension de leur fonctionnement interne. Plus les algorithmes deviennent complexes, moins nous pouvons comprendre ce qui induit précisément leurs décisions. Dans cette vaste classe d’algorithmes que composent les Réseaux Neuronaux, les Réseaux Neuronaux Récurrents (RNN selon l’acronyme anglais) occupent une place singulière. D’une part, leur fonction principale est de traiter des données séquentielles pour des applications importantes sur du texte, de l’audio, de la vidéo, etc. D’autre part, en raison de l’ambivalence de leurfonctionnement interne, ressemblant à la fois aux réseaux neuronaux non récurrents, de par leur structure, mais à la fois présentant des comportements incomparables avec ces derniers, le problème de l’établissement de résultats théoriques et d’algorithmes permettant de mieuxcomprendre leur fonctionnement constitue le problème majeur. Dans cette thèse, nous présentons nos contributions théoriques sur les RNN. Ces contributions couvrent plusieurs problématiques scientifiques de ces modèles : \textbf{expressivité}, \textbf{apprenabilité} et \textbf{généralisation}. L’expressivité est une propriété globale, dans le sens où nous parlons de l’expressivité d’un ensemble de fonctions. Déterminer l’expressivité d’un ensemble de RNN consiste à trouver la classe de fonctions la plus complexe que les RNN de peuvent simuler, témoignant en quelque sort de l’adaptabilité de . La capacité d’apprentissage est, en quelque sorte, l’expressivité, mais d’une manière plus contraignante, l’objectif étant de déterminer ce qu’il est possible d’apprendre pour une architecture de RNN. Les garanties de généralisation est une question d’actualité depuis l’introduction des algorithmes d’apprentissage. D’une manière très simplifiée ces garanties sont utilisées pour s’assurer que le modèle, dans notre cas un RNN, a appris ce qu’il est supposé apprendre. Dans le domaine de l’expressivité, nous sommes partis d’un cadre récemment introduit et l’avonsétendu ce qui nous a permis d’étudier l’expressivité d’architectures de RNN ne pouvant pas être étudiées dans le cadre initial. Nous proposons une reformulation de ce cadre qui nous permet d’importer certaines des propriétés pratiques dans un scénario du monde réel. D’une part, cettereformulation nous a permis de combler une lacune théorique, mais elle a également ouvert la voie aux autres contributions de cette thèse. Dans le domaine de l’apprenabilité, notre contribution est une démonstration théorique des limites de ce qui peut être appris par descente de gradient pour une classe de RNN, les Réseaux Récurrents Simples (SRN acronyme anglais). Nous nous appuyons sur le phénomène de disparition du gradient et combinons ce fait avec la précision finie des ordinateurs, afin de délimiter des zones de l’espace des paramètres inaccessibles pour les SRNs avec les algorithmes usuels de descente de gradient. Cette démonstration est accompagnée d’expériences et d’une preuve que les zones inaccessibles de l’espace des paramètres contiennent des fonctions d’intérêt telles que les machines à états finis. Enfin, dans le domaine des garanties de généralisation, nous dérivons des bornes PAC-Bayésiennes pour les SRN qui sont indépendantes de la longueur des données. Nous souhaitons insister sur le caractère original de ce résultat par rapport aux travaux comparables de la littérature qui des résultats soit plus limités soit dépendants systématiquement de la longueur des données. Les caractéristiques singulières de notre travail qui nous distinguent sont les suivantes : 1) tous nos résultats principaux sont indépendants de la longueur des données, 2) nous avons toujours considéré le cas où les RNN sont définis avec des fonctions d’activation non linéaires. Contrairement a une grande proportion des résultats où les RNN sont supposés être définis avec des fonctions linéaires ou ReLU pour simplifier l’analyse, nous avons au contraire incorporé les particularités des fonctions d’activation bornées telles que la fonction tangente hyperbolique et la fonction sigmoïdale dans nos résultats. 3) nous avons intégré dans notre travail théorique la précision finie utilisée dans les calculs des ordinateurs
Caractérisation diélectrique des hydroxyapatites carbonatées à hautes températures
International audienceLa maîtrise de la chimie et de la microstructure des hydroxyapatites carbonatées (CHA) est un enjeu crucial pour contrôler leur comportement biologique et favoriser leur emploi en tant que matériaux d’implants osseux. Leur biodégradabilité étant régie par la localisation et la quantité de carbonates, la taille des grains et la porosité [1], il est nécessaire de maîtriser les phénomènes se déroulant durant l’étape de densification/carbonatation. Ces matériaux peuvent être obtenus par frittage réactif d’hydroxyapatite (HA) sous une atmosphère contenant du CO2. Par ses lentes vitesses de chauffe et son importante inertie thermique, le chauffage résistif « conventionnel » a pour conséquence de favoriser le grossissement granulaire et réduire la surface disponible pour interagir avec CO2. Permettant un chauffage rapide, le frittage micro-ondes (MO) peut fournir une solution versatile pour obtenir des CHA présentant des propriétés physico-chimiques et biologiques nouvelles.Alors que le frittage MO de HA stœchiométrique est bien documenté [2,3], sa carbonatation sous CO2 pour produire CHA reste à étudier [4]. L’absorption et la dissipation d’énergie d’un matériau sous champ MO dépend de ses propriétés diélectriques. La connaissance de l’évolution de ces caractéristiques au cours du frittage réactif MO constitue une étape nécessaire pour une meilleure compréhension de certains phénomènes tel que la formation de points chauds et la gestion des gradients de température.Cette présentation vise à étudier les propriétés diélectriques de HA lors du frittage réactif MO en fonction des évolutions de sa composition et son état de densification. Un procédé capable de caractériser la réponse diélectrique de l'échantillon pendant le processus de frittage a été développé. Il comprend une cavité MO résonante, un générateur, une caméra thermique haute résolution et un programme pour automatiser l'acquisition des courbes de résonance. Une analyse de données rigoureuse a posteriori permet l’obtention des propriétés diélectriques. Transition vers l'approche par compétences : la perception des enseignants dans la mise en place du BUT
International audienceIn the IUTs, the introduction of the BUT competence-based approach in 2021 represents a paradigmshift, the impact of which has yet to be assessed. The aim of this paper is to contribute to this processthrough a quantitative and qualitative analysis of teachers' perceptions of the objectives andimplementation of the reform. The results show mixed opinions on the implementation of this reformin terms of the dimensions measured (overall coherence, collegiality, contextualisation of teaching,situational assessment). Three profiles of adaptation to the reform emerge: teachers with little impact,as they were already aligned to the paradigm before the reform; others who perceive benefits of thereform but encounter obstacles in its implementation, related to interdisciplinary coherence and collaboration; and finally, teachers who express difficulties in implementing the reform. The obstaclesmentioned and the levers for overcoming these difficulties should lead us to pay particular attention toinvolving the various actors in a process of co-construction of such a reference system.Dans les IUT, la mise en place du BUT en approche par compétences en 2021 constitue un changement de paradigme dont l'impact n'a pas encore été évalué. Cette communication vise à y contribuer par une analyse quantitative et qualitative du ressenti d'enseignants sur les objectifs visés et la mise en œuvre de la réforme. Les résultats montrent des avis mitigés sur la mise en œuvre de cette réforme, sur les dimensions mesurées (cohérence globale, collégialité, contextualisation des enseignements, évaluation en situation). Trois profils d'adaptation à la réforme émergent : des enseignants peu impactés car déjà alignés avec le paradigme avant la réforme ; d'autres percevant des bénéfices à cette réforme, mais rencontrant des obstacles dans sa mise en œuvre, relatives à la cohérence interdisciplinaire et la collaboration ; et enfin, des enseignants exprimant des difficultés dans la mise en œuvre de la réforme.Les freins et leviers évoqués pour surmonter ces difficultés devraient conduire à une attention particulière à impliquer les différents acteurs dans une démarche de co-construction d'un tel référentiel
Influenza vaccine effectiveness against detected infection in the community, France, October 2024 to February 2025
International audienceInfluenza circulates at high levels in Europe since November 2024. Using a test-negative study based on data from French community laboratories between October 2024 and February 2025, we estimated vaccine effectiveness (VE) against PCR-detected influenza infection (44,420/15,052; positive/negative individuals). For all age groups, the overall VE was 42% (95% CI: 37–46%), with 26% (95% CI: 18–34%) against influenza A and 75% (95% CI: 66–82%) against influenza B. Among individuals ≥ 65-year-olds VE was 22% (95% CI: 13–30%) and among 0–64-year-olds, 60% (95% CI: 56–65%)
Leopardi à l'heure de l'anthropocène: actuel, trop actuel ?
International audienceSituating Leopardi in the era of the Anthropocene means first of all questioning the relevance of rereading his work in the light of a debate and terminology that largely post-dates him. From a methodological, critical and hermeneutical point of view, this poses real difficulties. Will we see yet another updating of Leopardi's work, as the history of its reception and fortunes has constantly offered us over the last two centuries? In~this paper, we will first address the methodological pitfalls that an eco-criticism of Leopardi would inevitably encounter if it sought in him a precursor or a visionary of the ecological aporias of our century. We will then attempt to situate our questioning within a cultural, ideological and geological framework that integrates current questions about the Anthropocene into the long history of Western Europe, going back to the Enlightenment and the Industrial Revolution. Rather than updating Leopardi's thought, we propose a distant reading that historicises the discourse of the Anthropocene by tracing its origins, to see to what extent Leopardi had something to say, as early as the nineteenth century, about and against the founding myths of this Anthropocene that was not yet called that.Poser la question de Leopardi à l'heure de l'Anthropocène, interroger la pertinence d'une relecture de son øeuvre à l'aune d'un débat, d'événements et d'une terminologie qui lui sont largement postérieurs, entra\ⁱne, du point de vue critique, de réelles difficultés méthodologiques et herméneutiques. S'agit-il d'une énième actualisation de l'øeuvre de Leopardi, comme l'histoire de sa réception et de sa fortune n'a cessé de nous en offrir durant les deux derniers siècles~? Dans cette contribution, nous aborderons d'abord les écueils méthodologiques auxquels se confronte inéluctablement une éco-critique de Leopardi qui chercherait en lui un précurseur ou un visionnaire des apories écologiques de notre siècle. Nous tenterons ensuite d'inscrire notre interrogation à l'intérieur d'un cadre culturel, idéologique et géologique, qui intègre les questionnements actuels autour de l'Anthropocène dans les temps longs de l'histoire européenne occidentale, remontant jusqu'aux Lumières et à la révolution industrielle. Plutôt que d'actualiser la pensée de Leopardi, nous proposons ainsi une distant reading, diachronique et panoramique, susceptible d'historiciser le discours de l'Anthropocène en retraçant ses origines, pour voir dans quelle mesure Leopardi avait quelque chose à dire, dès le xixe~siècle, sur et contre les mythes fondateurs de cet Anthropocène qui ne portait pas encore son nom.Collocare Leopardi nell'era dell'Antropocene significa innanzitutto chiedersi se~sia opportuno, nonché legittimo dal punto di vista ermeneutico e metodologico, rileggere la sua opera alla luce di un dibattito e di una terminologia che gli sono di gran lunga posteriori. Significa quindi chiedersi se non si tratti di un'ennesima attualizzazione dell'opera di Leopardi, simile a quelle cui la burrascosa storia della sua fortuna critica ci ha abituati nel corso degli ultimi due secoli. In questo contributo ci confronteremo alle insidie metodologiche cui sembra dover andare inevitabilmente incontro un'ecocritica di Leopardi, qualora cercasse in lui un precursore o un visionario delle aporie ecologiche del nostro secolo. L'obiettivo sarà poi quello di impostare la questione all'interno di una cornice culturale, ideologica, nonché geologica, che integri le domande del presente sull'Antropocene nella storia lunga dell'Europa occidentale, risalendo all'Illuminismo e alla Rivoluzione industriale. Più che attualizzare il pensiero di Leopardi, proponiamo una distant reading che storicizzi il discorso dell'Antropocene rintracciandone le origini, per capire meglio ciò che Leopardi aveva da dire, sin dal primo Ottocento, contro i miti fondanti di quell'Antropocene che cos\`i ancora non si chiamava.
