Approaches to observational study designs and analytical options to evaluate the safety of multi-dose vaccines: A systematic review
No full textObservational study designs are instrumental in evaluating the safety of vaccines post-licensure, particularly when randomized controlled trials lack statistical power to detect rate adverse events. Self-controlled case series and target trial emulation have attractive features to overcome biases from traditional observational designs such as cohort and case-control leading to their widespread use in pharmacoepidemiological studies. However, when evaluating the safety of multiple doses, bias can arise if key assumptions underpinning these designs are violated. Such violations occur when adverse events influence subsequent vaccination by either delaying or contraindicating them. Although remedies to these violations exist, available literature suggest less consideration of these condition, Moreover, despite the importance of employing multiple study designs to strengthen confidence and highlight sources of bias, literature suggest scarcity of such comprehensive studies. Consequently, we aim to systematically review current observational study designs and analytical options in evaluating the safety of multidose vaccines, critically highlighting gaps in these studies and provide recommendations for future research
