Rheological parameters are an important characteristic of semisolid dosage forms. Structural and mechanical characteristics have a noticeable effect on the processes of release and absorption of drugs from ointments, as well as on their consumer properties: spreadability, adhesion, the ability to squeeze out of the tubes. The aim: to study the rheological parameters of aqueous solutions of sodium alginate in various concentrations to create medicinal hydrogel compositions. Materials and methods: sodium alginate (Shandong Topsea Seaweed Industrial Co., Ltd., China) was used as a gelling agent. The physicochemical properties of the samples were studied according to generally accepted methods of the State Pharmacopoeia of Ukraine. Rheological studies of experimental samples were carried out using a rotational viscometer «Rheolab QC», by Anton Paar (Austria) with coaxial cylinders C-CC27/SS at a temperature 20–25 °С. Results and discussion: the rheological profiles of sodium alginate gels have a non-Newtonian type of flow, which makes it possible to characterize them as systems with plastic-viscous properties. Gels with a sodium alginate concentration of 1.5–2 % have the best indicators of yield stress, hysteresis loop area, mechanical stability and dynamic flow coefficients. This indicates a low degree of destruction of the structural grid in the process of mechanical action and the presence of thixotropic bonds. Conclusions: sodium alginate gel base at 0.5 % and 1 % concentration is less stable under stress compared to sodium alginate bases at 1.5 % and 2 % concentration. The gel base of sodium alginate at a concentration of 2 % has the optimal mechanical stability value

    Get PDF
Non-alcoholic fatty liver disease (NAFLD) affects about a quarter of the world's population and it is closely linked to hypertension (HT). Pro-inflammatory and anti-inflammatory cytokines play a key role in the pathology progression, and the search for non-invasive biomarkers for the diagnosis of NAFLD remains an important issue. The aim of the study was to determine the diagnostic and prognostic value of IL-1β and IL-10 in assessing the progression of liver parenchyma changes in patients with NAFLD and HT comorbidity. Materials and methods. A study of 115 patients with non-alcoholic steatohepatitis (NASH) was performed. The main group consisted of 63 patients with NASH and HT, 52 patients with isolated NAFLD represented the comparison group. Clinical and laboratory parameters were evaluated, IL-10 and IL-1β levels were measured by ELISA method, ultrasound steatometry and elastography were performed in all patients. Results. The attenuation coefficient and median liver stiffness in NAFLD and HT group significantly exceeded the results in the isolated NAFLD group and in the control group. The IL-1β level in NAFLD and HT group was 17.55 pg/ml, and in isolated NAFLD group the indicator averaged 15.72 pg/ml, which exceeded the control values (8.26 pg/ml). IL-10 level was 12.69 pg/ml and 14.34 pg/ml in patients with comorbid and isolated NAFLD, respectively, while control results averaged 16.19 pg/ml. It were found strong relationship between IL-1β, IL-10 and CRP levels in patients with NAFLD and HT (r=0.61, p=0.024, and r=-0.69, p=0.036, respectively). Inverse correlations were also found between the cytokines IL-1β and IL-10 in NAFLD patients with and without HT (r=-0.61, p<0.001, and r=-0.57, p<0.001, respectively). Changes in the cytokine status of patients with NAFLD at different stages of steatosis and liver fibrosis had been identified. Conclusions. The presence of concomitant HT in patients with NAFLD is associated with greater severity of liver parenchyma changes. NAFLD manifestation is accompanied by increase of IL-1β and decrease of IL-10 levels, and deepening of these deviations were found in patients with comorbidity of NAFLD and HT. Interleukins IL-1β and IL-10 can be defined as biomarkers of NAFLD progression both in its isolated course and in its comorbidity with HT. The possibility of using biomarkers as an independent non-invasive test of diagnosing NAFLD requires further study

The aim of the study. To analyze the effect of probiotics on the microflora of the gastrointestinal tract in calves and biochemical parameters of blood in calves up to one month. Materials and methods. The research was conducted during 2020 in the conditions of Ukrainian farms for cattle breeding. Five experimental groups of five one-week-old calves were formed in each and one control group. Calves were kept separately in the same conditions on the same diet, but with feeding together with colostrum substitute probiotics of five grams per animal: Bacillus amyloliquefaciense, Bacillus mucilaginosus, Bacillus coagulans, Bacillus megaterium, Bacillus pumilus. The strains are deposited and produced by “Kronos Agro” Ukraine. Results. It was found that as a result of studies when feeding calves B. coagulans, B. pumilus and B. mucilaginosus the number of Lactobacillus sp. was 80 % higher than in the control group. The level of opportunistic pathogens in the experimental group with B. coagulans had minimal values. Animals in the group where B. mucilaginosus was given had a higher amount of Candida - up to 300 CFU/g and Enterobacteriaceae – 200 CFU/g; which is 50 % less compared to control groups, but more than in the experiment with B. coagulans. According to the results of biochemical examination of blood serum in calves, the absence of toxic effects of probiotic strains: Bacillus amyloliquefaciense, Bacillus mucilaginosus, Bacillus coagulans, Bacillus megaterium, Bacillus pumilus on the internal organs of animals was established. Conclusions. It was found that the maximum positive effect on the microflora of the gastrointestinal tract of calves up to 30 days of age had B. coagulans (1×109) when fed at a dose of 5 g per animal. The amount of Lactobacillus sp. was the maximum and reached 800 CFU/g, which is 80 % more than in the control group. At the same time, the level of opportunistic pathogens in the experimental group with B. coagulans had minimal indicators and was: Clostridium by 20 %, Escherichia coli – by 70 %, Enterobacteriaceae, Staphylococcus and Candida – 100 % less than the control. In the study of biochemical parameters, it was found that the activity of enzymes, protein and glucose levels in the serum of experimental animals fed with B. coagulans were within the physiological norm, indicating a normal metabolic process and no toxic effects

The aim. Study of hemostasis, antiphospholipid antibody levels and immunological parameters in pregnant women with antiphospholipid syndrome (APS), in particular with retrochorial hematoma (RCH). Materials and methods. 90 women were selected and divided into two groups: the control group – 30 pregnant women with a normal pregnancy (without APS) and the main group – 60 pregnant women with APS. Women in the main group were diagnosed with APS before pregnancy. In turn, the main group was divided into two subgroups: 1 subgroup – 41 pregnant women without RCH and 2 subgroup – 19 pregnant women with RCH. The main indicators of hemostasis were determined in all pregnant women, namely: the degree and rate of platelet aggregation, Willebrand factor, D-dimers. In addition, the level of antiphospholipid antibodies (APLA), antibodies to β2-glycoprotein, to annexin V, to prothrombin was examined, and the level of annexin V was also determined. The absolute and relative content of Treg, CD3+, T-lymphocytes, CD4+ (T-helpers), CD8+ (cytotoxic T-lymphocytes), CD19+ (B-lymphocytes), CD16+CD56+ (NK cells), CD16+CD56+CD107a+ (activated NK cells). Results. The obtained results showed that in pregnant women with APS compared to pregnant women without APS there are statistically significantly higher values of the degree and rate of platelet aggregation (90.6±6.3% and 106.3±6.7% vs. 65.3±5.3 % and 73.4±5.6%, respectively). There were also higher values of Willebrand factor and D-dimers (2.5±0.3 IU/ml and 378.1±34.3 ng/ml against 1.7±0.2 IU/ml and 268.1±27, 3 ng/ml, respectively). APLAs were significantly higher in pregnant women with APS compared with pregnant women in the control group, namely: 16.1±1.5 vs. 3.8±0.4 U/ml. With regard to antibodies to β2-glycoprotein, to annexin V, to prothrombin and to the level of annexin V, their values were also statistically significantly higher in the group of pregnant women with APS. In addition, the results of the study showed that pregnant women with APS showed changes in subpopulations of immunocompetent cells. However, examining the difference in hemostasis, antibody content, and level of lymphocyte subpopulations between pregnant women with APS with and without RCH, it was found that their shifts in pregnant women with RCH were more pronounced than in women without RCH. Conclusions. Pregnant women with APS are characterized by significantly more significant changes in hemostasis, manifested by activation of intravascular thrombosis. In addition, such pregnant women had a significantly higher concentration of autoantibodies. There are also changes in the immune system, in particular, a decrease in Treg-cells, which have the ability to reduce the specific proliferation and effector functions of lymphocytes, thereby participating in the pathogenesis of APS

    Get PDF
The aim: to improve the early diagnosis of cardiac arrhythmias and conduction disorders in children with gastroesophageal reflux, by studying the prognostic significance of the life history in this group of patients. Materials and methods. 56 children aged 8 to 18 years, mean age 14,93±2,62 years were examined. All examined children were divided into two alternative groups: Group I (main) was represented by 28 children with gastroesophageal reflux in combination with cardiac arrhythmias and conduction disorders, and Group II (control) - 28 children with only gastroesophageal reflux without cardiac arrhythmias and conduction disorders. Patients underwent clinical, anamnestic (with a detailed study of life history) and instrumental studies (electrocardiography, Holter daily ECG monitoring, esophagogastroduodenoscopy). Results. When analyzing the prognostic significance of the patient's life history, it was found that its very high level was observed for the number of foci of chronic infection (I=6.0) and the frequent incidence of a child up to year old of life (I=3.05). High prognostic value was registered relative to the number of diseases suffered by the child per year on average (I=2.35), and moderate predictor properties are characteristic of chickenpox (I=0.89) and the number of chronic diseases at present (I=0.71). The duration of breastfeeding showed low (I=0.30) prognostic significance. Suffered previously bacterial infections did not have predictor properties (I=0.12). In general, the prognostic significance of the patient's life history was high (I̅=2.20). Conclusions. It was found that in general the prognostic significance of the patient's life history was high (I̅=2.20) It is determined that in favour of the development of cardiac arrhythmias and conduction disorders evidence: the number of foci of chronic infection ≥1-2; lack of frequent morbidity in a child up to a year of life; the number of diseases suffered per year ≤5; the presence in past medical history of chickenpox; the number of chronic diseases in a child at present ≤2 and short-term (≤9 months) breastfeeding. It has been established what denies probability of development of the researched pathology: absence of the centers of a chronic infection; the presence frequent incidence of a child up to year old of life; the number of diseases suffered per year≥6, and chronic diseases at present ≥3; longer (≥9 months) breastfeeding and no history of chickenpo

The results of the study of the effect of cord blood serum and “Actovegin” (Takeda, Ukraine) on the reproductive function of cows in a comparative aspect are presented. The aim of the study was to analyze the effect of cord blood serum and Actovegin on the reproductive function of uterine cows in a comparative aspect. Materials and methods. The research was conducted during 2017-2020 in four dairy farms of Sumy region with tethered and untied maintenance. In 128 cows of different breeds and productivity, the timing of sexual cycling was studied and analyzed in comparison with the spontaneous sexual cycle with the use of cord blood serum and the drug "Actovegin" (Takeda, Ukraine). Results. It was found that in cows with tethered content, the number of days from the introduction of 15 ml of cord blood serum to the manifestation of the excitation stage was significantly less compared to the spontaneous manifestation of sexual desire (5.63±0.36 and 14.38±2.1, respectively). When cows with loose content were administered 10 ml of cord blood serum in combination with the drug "Actovegin", the number of days before the manifestation of sexual cycling was lower by 18.3 % compared with the rate of spontaneous sexual desire (4.31±0.38 and 18.25±1.89, respectively). Conclusions. As a result of using 10 ml of cord blood serum in combination with the drug "Actovegin", in cows of different productivity and under different housing conditions, the number of days before the onset of the stage of arousal is significantly reduced compared to the spontaneous manifestation of sexual desire. Cows of different productivity and with different housing conditions come to the sexual desire significantly faster with the use of 15 ml of cord blood serum compared to the spontaneous manifestation of sexual desire

20 % of ischemic stroke appear to originate from carotid artery atherosclerotic disease. Serum biomarkers reflecting the activity of atherosclerotic process and may help for estimate risk of acute cerebrovascular events. Several serum inflammatory markers have been proposed for risk assessment, but their prognostic role less known. The aim of this study is to clarify the prognostic value of biomarkers of atherosclerosis lipoprotein-associated phospholipase A2 (Lp-PLA2) and E-selectin in patients with symptomatic and asymptomatic carotid stenosis. Materials and methods. The study involved 106 patients with atherosclerotic carotid stenosis >50 % (74 men and 32 women, mean age 62.6±0.9) from which 76 symptomatic (35 with acute ipsilateral atherothrombotic stroke and 41 after carotid endarterectomy) and 30 asymptomatic patients. The control group consisted of age- and sex-matched 20 healthy subjects. The level of serum Lp-PLA2 and E-selectin was determined using a commercially available enzyme-linked immunosorbent assay kit. Results. The level of Lp-PLA 2 was in general significantly higher (p<0.05) in patients groups than in the control group and most high Lp-PLA2 concentration was in groups of symptomatic patients who underwent carotid endarterectomy. The level of E-selectin in the study patients was significantly higher than in the control group (p<0.05). The correlation of Lp-PLA 2 with E-selectin was significant for total patients (R=0.365664, p=0.00085) and group after carotid endarterectomy (R=0.429143, p=0.01796), but not for asymptomatic group (p>0.05). Receiver Operating Characteristics curves of logistic regression models which takes into joint both indicators was specificity and sensitive for predicting the occurrence of ischemic stroke. Conclusion. Conducted study show that the levels of Lp-PLA 2 and E-selectin have a significant impact on the development of stroke in patients with atherosclerotic carotid stenosis and can be used to predict it. A multidimensional model of the dependence of the probability of stroke on a linear combination of Lp-PLA 2 and E-selectin allows to obtaining significantly higher characteristics of the accuracy of stroke prediction than models with each factor alone

Study of the role of lipid peroxidation and antioxidant system in patients with infectious diseases is of great interest to researchers. Numerous studies have led to a common understanding of their contribution to the pathogenesis of infectious diseases, as well as to the complications development. However, the state of lipid peroxidation and antioxidant protection in patients with chickenpox (varicella) has not been sufficiently studied which is important for the development of new targeted treatments. The aim of the research - to assess the state of lipid peroxidation (LPO) and the antioxidant system (AOS) in the dynamics in patients with chickenpox, depending on the severity of the disease. Materials and methods. We selected for participating in the study 240 chickenpox patients (142 women and 98 men) aged 18-40 years. Chickenpox was diagnosed using clinical, serological, and molecular genetic methods. The state assessment of LPO in chickenpox patients was based on the determination of serum diene conjugates (DC), malondialdehyde (MDA) levels and the total oxidative activity (TOA) of blood plasma. Results. Study showed that the process of lipid peroxidation increases and the activity of the antioxidant system decreases in patients with chickenpox during the acute period of the disease. The extent of these processes becomes greater as the severity of chickenpox increases. A decrease in the activity of antioxidant enzymes and a disruption in the functioning of the glutathione system in patients with moderate to severe hypertension leads to disruption of protective mechanisms and, as a result, to an increase in the free radical chain reactions, the uncontrolled growth of which causes irreversible damage to the membranes of various cells, which underlies visceropathy in chickenpox patients. Conclusions. Antioxidant system is activated in the acute period in patients with mild chickenpox. It is evidenced by an increase in the overall antioxidant activity of blood plasma (p=0.045) and red blood cells (p=0.00087), in the activity of catalase (p=0.001), superoxide dismutase (p=0.0093), glutathione peroxidase (p=0.036), glutathione reductase of plasma and red blood cells, and an increase in the concentration of oxidized and reduced glutathione in blood. Lipid peroxidation is hyperactivated in the acute period in patients with a moderate and severe course of the disease and an excess amount of primary and secondary hydroperoxides of fatty acids accumulates in the blood. This is accompanied by indicates a developing imbalance between the oxidative and antioxidant systems in group of patients with moderate and severe chickenpox. Our findings confirm the usefulness of antioxidants in the treatment of chickenpox

Peripheral artery diseases are a worldwide medical and social problem. Approximately 30 % of patients with critical limb ischemia will undergo amputations and 25 % will die after 1 year. These patients require reconstructive and angioplastic interventions to preserve the limb. The aim of the study is to compare the effectiveness of different endovascular surgical technologies in patients with multilevel steno-occlusive lesions of the infrarenal aortic arteries. Materials and methods. Among 243 endovascular interventions, 51 patients were presented with multilevel steno-occlusive lesions of the infrarenal aortic arteries as a result of atherosclerosis obliterans. 42 patients (82.4 %) had two-level lesions and 9 (17.6 %) had three-level lesions. Results. Among 42 patients with a two-level lesion, 29 (56.9 %) ones had femoral arterial segment affection in combination with tibial artery affection. 13 (25.5 %) patients suffered from the lesion of the iliac segment in combination with the affection of the femoral segment arteries. In 9 (17.6 %) patients the lesions of three or more levels were determined: an iliac segment in combination with femoral and popliteal arteries – in 4 patients, and in 2 patients there were lesions of the femoral, popliteal and tibial segments, in 3 patients there was a lesion of the iliac, femoral, popliteal and tibial-foot segments in different degrees of severity. 7 balloon angioplasties and 13 stenting procedures were performed in two and three-level lesions in which the iliac arterial segment was affected. Only balloon angioplasty was used to revascularize the infraingvinal arterial segments. In the early postoperative period, the complications included thrombosis appeared in 6 patients. It was possible to restore the blood flow and save the limb only in 3 persons. For the other 3 ones, the attempts to restore the patency of the arteries were unsuccessful and resulted in the amputation of the lower extremity. Mortality in the early postoperative period was 3.9 % (2 death due to myocardial infarction). Conclusions. The persons with multilevel steno-obstructive lesions of the infrarenal aortic arteries are the most difficult category of patients with CLI. The surgical method of choice for patients with multilevel steno-occlusive lesions is the endovascular angioplasty with or without stenting
