Downregulation of MGLL and microRNAs (miR-302b-5p, miR-190a-3p, miR-450a-2-3p) in non-small cell lung cancer: potential roles in pathogenesis
Genes involved in lipid metabolism have been considered potential therapeutic targets in lung cancer because lipid metabolism is severely disrupted in this cancer. Monoglyceride lipase (MGLL) is a lipolytic enzyme that converts monoacylglycerides to fatty acids and glycerol. MicroRNAs (miRNA), one of the most important epigenetic regulators of gene expression, are also considered potential biomarkers in diagnosing, treating, and prognosis lung cancer. This study aimed to investigate the potential effects of MGLL and related miRNAs (miR-302b-5p, miR-190a-3p, miR-450a-2-3p) in the pathogenesis of non-small cell lung cancer (NSCLC) by examining their expression levels and regulatory mechanisms. We analysed the expression levels of MGLL and miRNAs in 30 NSCLC and 20 non-cancerous tissues by qPCR. We performed in silico analyses to determine the biological functions of MGLL and miRNAs in NSCLC. A protein-protein interaction (PPI) network was constructed for MGLL, and gene ontology (GO) analysis, and the interacting genes were analysed using the TCGAnalyzer tool. Our study showed that the expression levels of MGLL, miR-302b-5p, miR-190a-3p and miR-450a-2-3p were significantly decreased in NSCLC tissues (p < 0.05). Also, according to TCGAnalyzer, MSRB3, HTR4, and FCER1G genes were downregulated genes for NSCLC. We showed that miR-302b-5p, miR-190a-3p, and miR-450a-2-3p significantly regulate the TGF-β signalling pathway. In conclusion, this study provides evidence for the potential role of MGLL and microRNAs (miR-302b-5p, miR-190a-3p, miR-450a-2-3p) in NSCLC. In subsequent studies, it was determined that MSRB3, FCER1G and LTB4R2 genes, especially the HTR4 gene, could be potential target genes for lung cancer
The relationship between attitude toward physical activity and weight gain in children and young adolescence
IntroductionThe purpose of this study was to investigate the association between attitudes toward physical activity and weight gain among children and young adolescents with an additional focus on the impact of gender on these attitudes.MethodsEmploying a descriptive survey method, data were systematically gathered via purposive sampling from 11 specific cities in Türkiye, ensuring representation from all seven regions. A total of 3,138 students, aged between 9 and 14 years, participated in this study, with a distribution of 46% girls and 54% boys. To assess the attitudes of children and young adolescents toward physical activity, the Youth Physical Activity Attitude Scale was utilized. Height and body weight measurements were taken to determine the body mass index of participants. SPSS 26.0 software facilitated the statistical analyses, including Pearson correlation analysis to explore relationships between variables. Multivariate Analysis of Variance was employed to evaluate the impact of age, BMI, and gender on attitudes toward physical activity.ResultsParticipants classified as normal weight exhibited a more positive attitude towards physical activity compared to their obese and overweight counterparts. Moreover, a significant gender difference emerged, with boys demonstrating significantly higher positive attitudes toward physical activity than girls. However, no significant difference was observed in negative attitudes based on gender. The study also revealed that an escalation in negative attitudes towards physical activity correlated with students being categorized as underweight, overweight, or obese, as opposed to having a normal weight status. Additionally, a statistically significant divergence in both positive and negative attitudes towards physical activity was found based on age. Specifically, the results indicated that students aged 9 and 14 exhibited lower levels of positive attitude when contrasted with their counterparts of different age groups. Conversely, in the domain of negative attitudes, students at the age of 9 scored higher than their peers in other age categories.DiscussionAttitudes towards physical activity can serve as a convenient indicator and guide for assessing the effectiveness of various practices or interventions aimed at promoting physical activity, with recognition of the significant gender difference in positive attitudes among children and young adolescents.</jats:sec
The effect of python programming language teaching on 7th grade students programming self-efficacy skills
This study aimed to investigate the impact of teaching the Python programming language on the programming self-efficacy of 7th-grade students. A quantitative research design was employed, utilizing a one-group pretest-posttest model. The study sample consisted of 10 students enrolled in the 7th grade at a public school located in a rural district of northern Turkey during the spring semester of the 2023-2024 academic year. These students attended a school affiliated with the Ministry of National Education. The study group was selected using the convenience sampling method, a type of purposive sampling. Data for this research was collected using the Programming Self-Efficacy Scale for Secondary School Students, developed by Kukul, Gökçearslan, and Günbatar (2017). The collected data were analyzed using the Jamovi software. To assess the suitability of the data for normal distribution, skewness and kurtosis coefficients, as well as Shapiro-Wilk test values (a normality test), were examined, alongside an inspection of histogram plots. The results indicated that the data were normally distributed with respect to both research questions. For the first research question, a paired sample t-test was conducted to determine whether there was a statistically significant difference between the pre-test and post-test scores. The analysis revealed that Python instruction did not have a significant effect on the programming self-efficacy of the 7th-grade students. Regarding the second research question, the homogeneity of the data was assessed. Levene's test for equality of variances was performed, confirming that the assumption of homogeneity was met. Consequently, independent sample t-tests were conducted to examine the significance of the difference in pre-test and post-test scores based on gender. The results indicated no statistically significant difference in programming self-efficacy scores between male and female students.</jats:p
A Comparison of Kawasaki Disease during the SARS-CoV-2 Pandemic with Multisystem Inflammatory Syndrome in Children
Objectives: The purpose of this study was to compare and contrast Kawasaki disease (KD) with multisystem inflammatory syndrome in children (MIS-C) during the SARS-CoV-2 pandemic. Methods: A retrospective analysis of the medical records of patients diagnosed with KD and MIS-C at a single institution from July 2020 to November 2021 was performed. Results: The study included 39 MIS-C patients (84.6% male) with a median age of 138 months and 17 KD patients (58.8% male) with a median age of 36 months. The MIS-C patients were older (p < 0.001) and had prolonged hospitalizations (p = 0.023), elevated neutrophil counts (p < 0.001), C-reactive protein (p < 0.001), procalcitonin (p < 0.001), interleukin-6 (p < 0.014), ferritin (p < 0.001), fibrinogen (p < 0.001), troponin I (p = 0.001), NT-proBNP (p < 0.001), and D-dimer levels (p < 0.001). There were more cases of hypotension (p = 0.024), decreased left ventricular function (p = 0.023), and a greater need for corticosteroids (p < 0.001), enoxaparin (p = 0.045), and therapeutic plasma exchange (p < 0.001). Kawasaki disease patients had a greater incidence of rash (p < 0.001), changes in oral mucosa (p < 0.001), conjunctival injection (p < 0.001), extremity changes (p < 0.001), and cervical lymphadenopathy (p < 0.001). They had a longer duration of fever (p < 0.001), elevated white blood cell count (p < 0.001), platelet count (p < 0.001), and alanine aminotransferase level (p < 0.001). The two groups were similar regarding the hemoglobin levels, erythrocyte sedimentation rates, albumin levels, and the frequency of coronary aneurysm, myocarditis, pericarditis, invasive mechanical ventilatory support, and intravenous immunoglobulin treatment. Conclusions: Advanced patient age, a greater presence of gastrointestinal and cardiac findings associated with hypotension, increased NT-proBNP levels, decreased left ventricular function, the use of various treatment modalities, and longer hospital stays suggest MIS-C, whereas prolonged fever and classical clinical features of KD favor KD.</jats:p
High-temperature sensitivity complex dielectric/electric modulus, loss tangent, and AC conductivity in Au/(S:DLC)/p-Si (MIS) structures
AbstractComplex dielectric (ε* = ε′ − jε″)/electric modulus (M* = M′ + jM″), loss tangent (tanδ), and ac conductivity (σac) properties of Au/(S-DLC)/p-Si structures were investigated by utilizing admittance/impedance measurements between 80 and 440 K at 0.1 and 0.5 MHz. Sulfur-doped diamond-like carbon (S:DLC) was used an interlayer at Au/p-Si interface utilizing electrodeposition method. The capacitance/conductance (C/G) or (ε' ~ C) and (ε″ ~ G) values found to be highly dependent on both frequency and temperature. The increase of them with temperatures was attributed to the thermal-activated electronic charges localized at interface states (Nss) and decrease in bandgap energy of semiconductor. The observed high ε′ and ε″ values at 0.1 MHz is the result of the space/dipole polarization and Nss. Because the charges are at low frequencies, dipoles have sufficient time to rotation yourself in the direction of electric field and Nss can easily follow the ac signal. Arrhenius plot (ln(σac) vs 1/T) shows two distinctive linear parts and activation energy (Ea) value was found as 5.78 and 189.41 from the slope; this plot at 0.5 MHz is corresponding to low temperature (80–230 K) and high temperature (260–440 K), respectively. The observed higher Ea and ε′ (~ 14 even at 100 kHz) show that hopping of electronic charges from traps to others is predominant charge transport mechanism and the prepared Au/(S:DLC)/p-Si structure can be used to store more energy.</jats:p
Investigation of the 5-hydroxymethylyfurfural value and antioxidant properties of Kazdaği Fir cone syrup
Background and aims Cone syrup, unlike cone molasses, is a food product prepared without adding beet sugar. The use of high temperatures in the production of syrups and molasses causes the formation of 5-Hydroxymethylfurfural (HMF), which causes serious toxic effects on the human body, from glucose, fructose aldose and ketose monosaccharides. Although pine cone syrup is just becoming widespread in our country, it is a new food that is in high demand during the winter months. Therefore, in the study, HMF levels, sugar content, phenolic content and antioxidant levels in Kastamonu local cone syrup prepared without adding beet sugar were experimentally determined. Methods The color intensity resulting from the reaction of hydroxymethylfurfural, paratoluidine and barbituric acid was determined by reading the absorbance at a wavelength of 550 nm with a spectrophotometer. HMF measured spectrophotometrically was detected at very low levels in the syrup. Brix value, phenolic content and antioxidant values of fir syrup were also evaluated. Results Spectrophotometric analysis, the HMF content of traditional fir syrup was found to be very low, 8.1 mg/L. In addition, the total phenolic content value of locally produced fir cone syrup was found to be 6,050 mg GAE/g and the % inhibition value calculated by the DPPH method was 48,553, thus shedding light on its potential nutritional content through antioxidant activity analyses. Conclusions This study is the first to investigate fir syrup. As a result of the study, antioxidant activity analyzes shed light on its potential nutritional content. More importantly, this study was the first to show that fir syrup has a low glycemic index with a % Brix value (2.370±0.058) and that diabetic patients can safely use it instead of molasses.</jats:p
Analysing seafarer selection criteria in the context of talent management: Implications for Turkish seafarer market
It is predicted that the seafarer shortage in the global seafarer market will continue in the next years. In particular, the expected shortage of officer-licensed seafarers in the coming years should be considered as an opportunity for the seafarer supplier countries. For this purpose, this study aims to make implications about how to improve the skills of seafarers by analysing seafarer selection criteria in Turkish maritime companies. In this context, the criteria related to the selection of seafarers were defined, these criteria were evaluated by the selected experts, and managerial implications in the context of talent management were made. The results showed that 'Knowledge of the English Language' was seen as the most significant criterion, and it was followed by respectively 'Having Sufficient Knowledge of the Safety Management System' and 'Validity Period of Seafarer Documents (passport, certificates, qualification, etc.)'. According to the results obtained from the study, suggestions have been made that are thought to enable Turkish seafarers to get a larger share of the international seafarer market

Testing a social cognitive model of career self‐management with Turkish undergraduate students
AbstractThis study investigated the direct and indirect associations between social–emotional learning (SEL) skills, career decision self‐efficacy (CDSE), vocational outcome expectations (VOE), and career engagement (CE) within social cognitive career theory (SCCT) in a sample of Turkish university students. Junior and senior students (N = 460) completed the Social–Emotional Learning Scale‐Young Adult Form, the Career Decision Self‐Efficacy Scale‐Short Form, the Vocational Outcome Expectations Scale, the Career Engagement Scale, and the Demographic Information Form. Data was collected during COVID‐19 lockdowns in 2021. The results of a multiple serial mediation model indicated that higher SEL skills were related to higher CDSE and VOE, and, in turn, to CE. Overall, the results show that SCCT provides a promising framework for identifying predictors of CE, self‐directed career management behavior, in young adults during the transition from school to work. These findings have implications for theory, research, and practice by identifying potential influences on CE during COVID‐19.</jats:p
