The effect of the combination therapy with chlorophyllin, a glutathione transferase P1-1 inhibitor, and docetaxel on triple-negative breast cancer invasion and metastasis in vivo/in vitro
Abstract The expression of glutathione S-transferase P1 (GSTP1) enzyme increases in cancer cells, leading to anticancer drug resistance. The antioxidant chlorophyllin has an inhibitory effect on GSTP1. In this study, we investigated the effect of chlorophyllin and its combined administration with the chemotherapeutic agent docetaxel on metastatic processes. For this purpose, both the 4T1 triple-negative breast cancer cell line and metastatic animal model were used. The MTT, flow cytometry, and wound healing assays were used to investigate cell viability, cell cycle, and cell migration, respectively. Total gelatinase activity, GST activity, and glutathione levels in cell and liver tissue lysates measured by colorimetric methods. Micrometastases were evaluated histochemically in liver tissue sections. As a result, the coadministration of chlorophyllin and docetaxel significantly inhibited cell migration in vitro. There was a significant decrease in the total gelatinase activity in vivo. We found that only combined treatment reduced the micrometastatic lesions in the liver tissues, though this reduction was not statistically significant. In conclusion, the coadministration of chlorophyllin and docetaxel may have a potential role in controlling metastatic processes by suppressing cell migration, gelatinase activity, and micrometastasis formation in triple-negative breast cancers. TTürk kültürünün yapı taşları içerisinde son derece önemli bir yer teşkil eden âşık edebiyatı, kolektif hafızaya ait verileri ihtiva eden müstesna bir değerdir. Bu değerin icracısı konumunda olan âşıklar, kadim geleneklere bağlı duygu biçimleri ve geleceği kurgulayan düşünüş tarzları ile toplumun sözcülüğünü yapan bilge kişilerdir. Âşıkların, sosyal hayat içerisinde ortaya çıkan aksaklıkları şiirlerinde eleştiri konusu yaptığı ancak çoğu zaman verdikleri öğütlerle bir farkındalık ortaya koymayı amaçladıkları söylenebilir. Bu amaç etrafında pek çok şiir kaleme alan Âşık Ruhsatî, sahip olduğu irfanla bilinç dışına ait verileri gün yüzüne çıkaran değerli bir şahsiyettir. Âşık Ruhsatî, 19. yüzyılda Sivas’ın Deliktaş beldesinde dünyaya gelmiştir. Asıl adı Mustafa olan âşığın yaygın kanaate göre saz çalamadığı düşünülmektedir. Bununla birlikte doğaçlaması oldukça güçlü bir kalem şairidir. Şiirlerinde köy hayatı, aşk, tabiat, tasavvuf gibi konuları işlemiş ayrıca yergi şiirleri kaleme almıştır. Ruhsatî’nin üslubunda topluma yol gösterme düşüncesi ön plana çıkmıştır. Dolayısıyla şiirlerinde didaktik tarz önemli bir yere sahiptir. Bu açıdan destan türü etrafında insanlara öğüt veren eserler ortaya koymuştur. Yaşadığı dönemden günümüze zihinleri aydınlatan Ruhsatî; tütün bağımlılığı hakkında da bir destan kaleme almış, bu destanda tütünün sosyal ve ekonomik tahribatları ile özellikle insan sağlığı üzerindeki olumsuz etkilerinden bahsetmiştir. Usta âşık, hayatta olduğu çağda tütünün zararları konusunda yeterince bilimsel çalışma bulunmamasına rağmen gözlem yeteneğinin ve toplumsal duyarlılığının sonucunda, insanlığa akla uygun bir mesaj vermiştir. Tütün bağımlılığına karşı mücadele kapsamında ülkemizde kararlılıkla gerçekleştirilen çalışmalar düşünüldüğünde, tiryaki destanı olarak isimlendirilebilecek eserin bağımlılıktan kurtulmak için yol gösterici olduğu ifade edilebilir. Çünkü Ruhsatî, bahsedilen eserinde tütün müptelası olan bireylere karşı olumlu bir tutumla yaklaşılması gerektiğini, onları incitebilecek söylemlerden kaçınılmasının önemini öğütlemektedir. Âşık Ruhsatî’nin tiryaki destanının, tütün bağımlılığının zararlarına yönelen söylemi etrafında çözümlendiği bu makalede ayrıca bağımlılıkla mücadele çalışmalarına âşık edebiyatı penceresinden dikkat çekme ve olumlu bir katkı sunma hedefi güdülmüştür. Bu amaç etrafında, ilgili alt başlıkta değerlendirmelerde bulunulmuştur. Makalenin sonucunda, yaşadığı toplumun sorunlarına duyarlı bir sanatçı olan Ruhsatî’nin, tütünün insan sağlığı üzerindeki olumsuz etkileri ile ekonomik ve sosyal zararlarına karşı güçlü bir farkındalık ortaya koyduğu anlaşılmıştır.</jats:p
Effects of Health Education Based on the Roy Adaptation Model on the Healthy Lifestyle Behaviors and Self‐Esteem of Working Adolescents: A Randomized Controlled Trial
ABSTRACTBackgroundWhile adolescents are an at‐risk group that is in a process of developmental crisis and requires guidance for adjustment. They may be traumatized by challenging conditions due to the negative experiences they are socially exposed to. Having to work is one of the most important social problems that increases the risk situation of this group. Roy's Adaptation Model is also quite suitable for use in interventions and research addressing developmental groups. It is believed that the adaptation of the individual to a new situation in their life contributes to their capacity to improve their health and increase their quality of life.ObjectiveThis study was conducted to determine the effects of a Health Education Program based on Roy's Adaptation Model on the healthy lifestyle behaviors and self‐esteem levels of working adolescents.DesignThis study was designed as a randomized controlled trial.MethodThe study was conducted at a vocational training center in Turkey between March 14 and June 13, 2022. The sample consisted of 66 adolescents aged 14–18 years (intervention: 30; control: 36) trained at the Vocational Apprenticeship and Training Center. The adolescents in the intervention group participated in an 8‐session program for 4 weeks. Data were collected using a Personal Information Form, the Healthy Lifestyle Behaviors Scale II, and the Rosenberg Self‐Esteem Scale (RSE). Data collection took place before the program started, immediately after its completion, and 3 months later. This document was prepared in accordance with the Preferred Reporting Items for Analyses of Randomized Controlled Trials (CONSORT) guidelines.ResultsA significant difference was found between the intervention and control groups in favor of the intervention group in terms of their Healthy Lifestyle Behaviors Scale II scores in the 2nd measurement and their RSE score in the 3rd measurement.ConclusionThe Health Education Program based on Roy's Adaptation Model is effective in increasing the healthy lifestyle behaviors and self‐esteem of working adolescents.Patient or Public ContributionNo patient or public contribution.ImpactRoy's Adaptation Model can provide a conceptual framework for holistic studies to promote health for developmental groups.</jats:sec
The effects of preheating on color values of polyacid-modified composite resins
Objectives: Color stability is crucial to the esthetic success of restorative materials. Internal and external factors affect the coloring of these materials. Research carried out on the development of restorative materials draws attention to the advantages of methods such as preheating resins, reducing the viscosity and light cure time, increasing the marginal adaptation, and increasing the degree of monomer conversion. The aim of this study was to examine the in vitro effects of preheating polyacid-modified composite resins (PMCRs) on their color values and evaluate the obtained findings comparatively. Materials and Methods: Four main groups were formed by preparing discs from A2-colored PMCR material, which were kept at four different temperatures (4°C, 23°C, 39°C, and 55°C) before polymerization. The colors of the discs were measured according to the Commission on Illumination Laboratory system before and after they were kept in distilled water for 24 hours. The data were statistically analyzed using IBM Statistical Package for the Social Sciences V3. Results: It was seen that the preheating treatment did not significantly affect the color sensation (∆E) and red-green (∆a) color values of the PMCRs. It was determined that this process was effective on the lightness (∆L) and blue-yellow (∆b) color values of the materials. Conclusion: It should be considered that preheating applied to PMCRs may be beneficial for the longevity of the color stability of restorations, but different oral hygiene and dietary habits may have different effects on PMCRs. </jats:sec
De-Escalation of Nodal Surgery in Clinically Node-Positive Breast Cancer
ImportanceIncreasing evidence supports the oncologic safety of de-escalating axillary surgery for patients with breast cancer after neoadjuvant chemotherapy (NAC).ObjectiveTo evaluate the oncologic outcomes of de-escalating axillary surgery among patients with clinically node (cN)–positive breast cancer and patients whose disease became cN negative after NAC (ycN negative).Design, Setting, and ParticipantsIn the NEOSENTITURK MF-1803 prospective cohort registry trial, patients from 37 centers with cT1-4N1-3M0 disease treated with sentinel lymph node biopsy (SLNB) or targeted axillary dissection (TAD) alone or with ypN-negative or ypN-positive disease after NAC were recruited between February 15, 2019, and January 1, 2023, and evaluated.ExposureTreatment with SLNB or TAD after NAC.Main Outcomes and MeasuresThe primary aim of the study was axillary, locoregional, or distant recurrence rates; disease-free survival; and disease-specific survival. Number of axillary lymph nodes removed was also evaluated.ResultsA total of 976 patients (median age, 46 years [range, 21-80 years]) with cT1-4N1-3M0 disease underwent SLNB (n = 620) or TAD alone (n = 356). Most of the cohort had a mapping procedure with blue dye alone (645 [66.1%]) with (n = 177) or without (n = 468) TAD. Overall, no difference was found between patients treated with TAD and patients treated with SLNB in the median number of total lymph nodes removed (TAD, 4 [3-6] vs SLNB, 4 [3-6]; P = .09). Among patients with ypN-positive disease, those who underwent TAD were more likely to have a lower median lymph node ratio (TAD, 0.28 [IQR, 0.20-0.40] vs SLNB, 0.33 [IQR, 0.20-0.50]; P = .03). At a median follow-up of 39 months (IQR, 29-48 months), no significant difference was found in the rates of ipsilateral axillary recurrence (0.3% [1 of 356] vs 0.3% [2 of 620]; P ≥ .99) or locoregional recurrence (0.6% [2 of 356] vs 1.1% [7 of 620]; P = .50) between the TAD and SLNB groups, with an overall locoregional recurrence rate of 0.9% (9 of 976). The initial clinical tumor stage, pathologic complete response, and use of blue dye alone as a mapping procedure were not associated with the outcome. Even though patients with TAD demonstrated an increased disease-free survival rate compared with the SLNB group, this difference did not reach statistical significance (94.9% vs 92.6%; P = .07). Factors associated with decreased 5-year disease-specific survival were cN2-3 axillary stage (cN1, 98.7% vs cN2-3, 96.8%; P = .03) and nonluminal type tumor pathologic characteristics (luminal, 98.9% vs nonluminal, 96.9%; P = .007).Conclusions and RelevanceThe short-term results suggest very low rates of axillary and locoregional recurrence in a select group of patients with cN-negative disease after NAC treated with TAD alone or SLNB alone followed by regional nodal irradiation regardless of the SLNB technique or nodal pathology. Whether TAD might provide a clear survival advantage compared with SLNB remains to be proven in studies with longer follow-up.</jats:sec
Renewable Energy Consumption and the Ecological Footprint in Denmark: Assessing the Influence of Financial Development and Agricultural Contribution
The aim of this study is to empirically examine the relationship between renewable energy consumption and the ecological footprint (EF), using Denmark as a case study, based on data covering the period from 1990 to 2020. In examining this relationship, the roles of agricultural, forestry, and fisheries value-added; economic growth; and financial development are also explored. The analysis, conducted using fractional frequency Fourier approaches, considers the presence of structural breaks. The results reveal a negative relationship between renewable energy consumption and EF, while a positive relationship is found between agricultural, forestry, and fisheries value-added; economic growth; and financial development with the EF. According to the causality analysis, a unidirectional causality is detected from renewable energy consumption to the EF. These findings highlight the potential impact of renewable energy on EF and emphasize the importance of integrating green energy investments and renewable fuel usage into strategies aimed at reducing the EF.</jats:p
Analysis of Attitudes Toward Breastfeeding and Spiritual Life During Pregnancy in Türkiye: A Qualitative Study
Abstract It is crucial for nurses to understand the meaning of spirituality, which can become more pronounced during pregnancy, childbirth, and breastfeeding, and to take a supportive approach to breastfeeding attitudes during pregnancy. This study aimed to explore pregnant women’s attitudes and spiritual experiences in Türkiye regarding breastfeeding. A qualitative inductive content analysis design was employed. Seventeen pregnant women (36–39-week gestation) were admitted to a maternity hospital in Zonguldak, Türkiye. They were selected using purposive sampling. Data were collected through semi-structured, face-to-face, in-depth interviews and analyzed using an inductive content analysis approach. The mean age of the participants was 27.41 years, and ten participants were primiparous. Multiparous participants had previously breastfed for a mean duration of 19.1 months. Three main themes emerged: “Breastfeeding and life,” “Spiritual life and breastfeeding,” and “Cultural synthesis.” Considering the impact of spirituality on breastfeeding during pregnancy, these findings may help nurses recognize pregnant women’s spiritual needs and value the protective role of spirituality. Understanding these dimensions could improve the quality of support provided by healthcare professionals and potentially enhance breastfeeding outcomes.</jats:p
