    Intermittent administration of peracetic acid is a mild environmental stressor that elicits mucosal and systemic adaptive responses from Atlantic salmon post-smolts

    Background Fish encounter oxidative stress several times during their lifetime, and it has a pervasive influence on their health and welfare. One of the triggers of oxidative stress in fish farming is the use of oxidative disinfectants to improve rearing conditions, especially in production systems employing recirculation technology. Here we report the physiological and morphological adaptive responses of Atlantic salmon (Salmo salar L.) post-smolts to intermittent exposure to a potent oxidative agent peracetic acid (PAA). Fish reared in semi-commercial scale brackish water recirculating aquaculture system (RAS) were exposed to 1 ppm PAA every 3 days over 6 weeks. Mucosal and systemic responses were profiled before exposure, 22 and 45 days during the intermittent PAA administration. Results Oxidative stress was likely triggered as plasma antioxidant capacity increased significantly during the exposure period. Adaptive stress response to the periodic oxidant challenge was likewise demonstrated in the changes in plasma glucose and lactate levels. PAA-induced alterations in the transcription of antioxidants, cytokines, heat shock proteins and mucin genes showed a tissue-specific pattern: downregulation was observed in the gills and olfactory rosette, upregulation occurred in the skin, and no substantial changes in the liver. Further, PAA exposure resulted in histological changes in key mucosal organs (i.e. olfactory rosette, skin and gills); pathological alterations were predominant in the gills where cases of epithelial lifting, hypertrophy and clubbing were prevalent. In addition, intermittent PAA administration resulted in an apparent overproduction of mucus in the nasal mucosa. Lastly, PAA did not dramatically alter the ability of salmon to mount a physiological stress response in the presence of a secondary stressor, though some subtle interference was documented in the kinetics and magnitude of plasma cortisol and glucose response post-stress. Conclusions The present study collectively demonstrated that intermittent oxidant exposure was a mild environmental stressor that salmon could mount strong adaptive responses at systemic and mucosal levels.     Overcoming barriers to breeding for increased lice resistance in farmed Atlantic salmon: A case study from Norway

    Lice is a persistent and major problem in the salmon aquaculture sector with serious environmental impacts and reducing growth potential and income of the salmon industry. This article discusses whether there is an untapped potential in breeding for improved lice resistant of Atlantic salmon. To this end, three sets of factors that may impact the state of breeding for lice resistance are examined, using document analysis and key actor interviews. First, our data material indicates that market-based factors will hardly stimulate this type of breeding, as the benefits from breeding for lice resistance is predominantly a public good, and because the polygenic nature of lice resistance does not enable patenting as a powerful instrument to secure private goods or privatize the benefits of genetic improvement. Second, the regulation of gene editing technologies is in flux, and increase the risk of investments in technologies as selective breeding that could handle the polygenic challenges of lice resistance. Finally, policy instruments aimed at stimulating relevant innovation has been applied generously for other types of innovations to deal with the lice problem. For instance, new technologies for delousing have resulted in increased treatments and caused higher stress and mortality the recent years. However, none of these have targeted major root causes of the salmon lice problem (e.g. big monocultures or susceptible fish) or been exploited by breeding actors.     Variation in volatile organic compounds in Atlantic salmon mucus is associated with resistance to salmon lice infection

    Salmon lice are ectoparasites that threaten wild and farmed salmonids. Artificial selection of salmon for resistance to the infectious copepodid lice stage currently relies on in vivo challenge trials on thousands of salmon a year. We challenged 5750 salmon with salmon lice (Lepeophtheirus salmonis) from two distinct farmed strains of salmon in two separate trials. We found that volatile organic compounds (VOC), 1-penten-3-ol, 1-octen-3-ol and 6-methyl-5-hepten-2-one in the mucus of the salmon host after salmon lice infection, were significantly associated with lice infection numbers across a range of water temperatures (5 °C, 10 °C, 17 °C). Some VOCs (benzene, 1-octen-3-ol and 3,5,5-trimethyl-2-hexene) were significantly different between lines divergently selected for salmon lice resistance. In a combined population assessment, selected VOCs varied between families in the range of 47- 59% indicating a genetic component and were positively correlated to the salmon hosts estimated breeding values 0.59–0.74. Mucosal VOC phenotypes could supplement current breeding practices and have the potential to be a more direct and ethical proxy for salmon lice resistance provided they can be measured prior to lice infestation.publishedVersio

    Havfisk AS ønsker å forbedre kvaliteten på trålfanget fisk, og satte i 2018 i gang med å designe og bygge ny tråler. Utgangspunktet var å holde fangsten levende om bord, for å kunne slakte ut fisken på en kontrollert måte. Nofima ble forespurt om å bidra med kunnskap rundt design av levendefisk-tanker, kvalitet, samt selve levendelagringen av hvitfisk. Uttesting og evaluering ble gjennomfør på to tokt, oktober 2020 og oktober 2021. På grunn av lav pumpekapasitet – vannstrøm gjennom tankene - under gjennomføring på første tokt, varierende mengde fisk i de ulike tankene og høy dødelighet (opp mot 90 %), så er det uklart om vannfordelingen opp gjennom flat eller skråstilt bunnplate skiller seg fra hverandre. Etter første tokt ble pumpekapasiteten forbedret, men fortsatt var det høy dødelighet (40-80 %) under levendelagring. Under levendelagringen av torsk, økte fiskens blod-pH (7,1 til 7,5), etter 5 timer. Dette indikerer at torsken restituerer, når den får god tilgang på friskt sjøvann. Samtidig viser resultatene at rødfargen i muskel øker raskt de to første timene av levendelagring og selv etter 5 timer levendelagring, reduseres ikke rødfargen.     Observations from the Hydrolysis of the Green Sea Urchin (Strongylocentrotus droebachiensis)

    There is a large amount of coproduct generated by the sea urchin fisheries around the world as well as a growing interest in removing large quantities of undersize and low value sea urchins from barren areas in the northern Atlantic and Pacific coasts as well as other areas around the world. The authors believe there is scope to develop a hydrolysate product from this and this study gives preliminary observations on the characteristics of hydrolysate from the sea urchin Strongylocentrotus droebachiensis. The biochemical composition for S. droebachiensis is moisture 64.1%, protein 3.4%, oil 0.9%, and ash 29.8%. Amino acid composition, molecular weight distribution, lipid-class, and fatty acid composition are also presented. The authors suggest a sensory-panel mapping be undertaken on future sea urchin hydrolysates.     Optimalisering av prosessbetingelser for ekstraksjon av fosfolipider fra silderogn

    Umoden silderogn inneholder høye nivåer av fosfolipider og omega-3 fettsyrer og er et godt utgangspunkt for utvikling av produkter rettet mot kosttilskudd og helsekostmarkedet. Det er i prosjektet optimalisert en prosess for ekstraksjon av fosfolipider fra tørket rogn basert på etanol-vann ekstraksjon. Optimal vann% i etanolfasen er funnet å være 11,3 %. Det ble ikke funnet effekt av temperatur i området 20-60 °C. Høyeste oppnådde utbytte av fosfolipider ved bruk av ett ekstraksjonstrinn er 83 %. Etanolekstraksjon gir økende co-ekstraksjon av salt og protein med økende vann%, men ingen effekt på farge. Lipidekstraktet har høy viskositet som kan reduseres ved tilsetting av makrellolje. Direkte ekstraksjon av våt rogn ga 25 % lavere utbytte sammenlignet med spraytørket rogn. Nivå av organiske miljøgifter (POPs) og tungmetaller i lipidekstraktet er lave og godt under gjeldende grenseverdier.     Hensikten med dette prosjektet var å teste og evaluere om ultra-kald temperatur (underkjølt saltlake) kan være en egnet metode for å bedøve fisk i forkant av slakting. I lakseindustrien brukes det i dag el-bedøving eller slag, og selv om begge metodene er effektive (virker raskt) så er feilprosenten såpass høy at optimalisering av gjeldende metoder eller vurdering av ny metode har blitt etterspurt. Saltlake kan underkjøles langt under vannets normale frysepunkt, og en mettet NaClløsning kan teoretisk holdes flytende til -21 C, men i praksis holdes stabil ved -18 C. Eksperimenter i dette prosjektet viser at små laks (< 500g) roes ned svært raskt når de plasseres i en slik løsning. Håndteringen av fisken i forkant forårsaket noe stress og gav tydelige stressreaksjoner hos fisken. Denne responsen forsvant raskt etter at fisken ble plassert i den underkjølte laken. Det ble gjort observasjoner i en 30 sekunders periode der fisken var eksponert for denne behandlingen. Observasjonene viste at noen fisk ble slått ut (mistet likevekt og sluttet å bevege seg), mens majoriteten av fisken framstod som døsig med svake muskel- og gjellelokkbevegelser. Det er derimot ikke klart stadfestet om, og i hvilken grad fisken bedøves som følge av ultra-kald lake. Hvis fisken opplever umiddelbart ubehag av behandlingen, ville akutt fluktrespons være en forventet adferd. Fraværet av slike responser er en indikasjon på at fisken ikke opplever mye ubehag, noe som videre underbygges av målinger av stresshormoner. Nivået av kortisol ble forhøyet ved transport, men det ble ikke påvist noe forhøyning ved ultra-kald behandling i 30 sekunder. Siden dagens krav til hurtig bedøvelse av oppdrettsfisk er streng (virketid < 0,5 s), så vil bedøvelse ved ultra-lav temperatur foreløpig ikke være et alternativ for denne industrien. Det er derimot aspekter ved denne metoden som virker å være lovende, men det anbefales at videre dokumentasjon først og fremst rettes mot små individer (arter som sild, skalldyr) med sikte å kunne tilby bedøvelse for villfangst.Bedøvelse ved ultra-lav temperatur – Faglig sluttrapportUltra-low temperature stunningpublishedVersio
