    ラット歯髄幹細胞の長期in vitro培養による細胞老化の時間的変化

    大阪歯科大学Osaka Dental University博士(歯学)Rat dental pulp stem cells (DPSCs) can be used to elucidate mesenchymal stem cell (MSC) applications in regenerative medicine. However, information on rat DPSCs during long-term passage, which could lead to replicative senescence, is limited. In this study, we investigated the phenotypic changes in DPSCs after 3–26 passages (3P–26P). The results show that cell morphology and nuclear size increase proportionally with passage number. The phosphorylated histone H2A.X (γ-H2A.X) positive cells (indicating DNA damage) increased significantly earlier than the 4-Hydroxynonenal(4-HNE) stained cells (indicating an abundance of intracellular reactive oxygen species). Compared to the cells subjected to 3P and 5P, the cells subjected to 15P showed reduced proliferation despite being positive for Ki67. Furthermore, cell growth was completely arrested after 26P. The senescence markers, senescence-associated β-galactosidase (SA-β-gal) and p16, exhibited similar expression patterns that were not correlated with those of p21 and urokinase-type plasminogen activator receptor (uPAR). Nearly all cells expressed SA-β-gal and p16 after 26P, whereas only half expressed p21 and uPAR. These results will contribute to understanding the characteristics of DPSCs toward replicative senescence, which are applicable to elucidate mechanisms related to regenerative medicine and stem cell aging.doctoral thesi

    大阪歯科大学Osaka Dental University博士(歯学)Purpose: To perform vertical bone augmentation on rat parietal bone by coating the inner surface of dense polytetrafluoroethylene (d-PTFE) domes with hydroxyapatite (HA) using Erbium Yttrium Aluminum Garnet (Er:YAG) pulsed laser deposition in a rat model. Methods: The d-PTFE plate surface, α-tricalcium phosphate (α-TCP) coating, and HA coating were measured using scanning electron microscopy and X-ray diffraction to confirm the replacement of α-TCP with HA via high-pressure steam sterilization. The dome was glued to the center of the rat parietal bone and closed with periosteal and epithelial sutures. The rats were euthanized at 4, 8, and 12 weeks postoperatively, and the d-PTFE dome and surrounding tissues were harvested for radiological and histological evaluation. Results: In a group without HA coating (control group), a small amount of layered neosteum was observed in the d-PTFE dome. Conversely, a new balloon-shaped bone was observed in a HA-coated group (experimental group). Bone growth and number of bone beams were higher in the HA-coated group from 4 to 12 weeks postoperatively than those in the control group. However, no significant differences in bone mineral density values or bone bridge widths were observed. Conclusions: The HA coating enhanced bone augmentation without bone replacement. Histological findings indicated that the new bone comprised normal bone tissue with cartilage lumen and matrix. Therefore, our results indicate that bone augmentation with a HA coating may be mediated by endochondral ossification, a process similar to that of ectopic ossification.doctoral thesi

Periodontal surgical treatment is categorized as tissue attachment therapy, resective therapy, regeneration therapy and plastic surgery. Attachment therapy is one of the most popular treatments for periodontal disease, and is also currently the most commonly used treatment for periodontal disease. We conducted a post-operative questionnaire survey of periodontal surgery procedures using flap surgery and FGF-2 preparations, comparing and studying each item. We found that when FGF-2 preparations were used there was significant reduction in the number of days of swelling, discomfort, and difficulty eating. This suggests that FGF-2 can be useful in periodontal surgery.doctoral thesi

Surface treatment is essential for dental applications owing to its poor surface energy and wettability; however, no consensus on an effective treatment method has been achieved. In this study, we attempted to carboxylate PEEK sample surfaces via Friedel–Crafts acylation using succinic anhydride and AlBr3. The possibility of further chemical modifications using carboxyl groups was examined. The samples were subjected to dehydration–condensation reactions with 1H,1H-pentadecafluorooctylamine and N,N’-dicyclohexylcarbodiimide. Furthermore, the sample’s surface properties at each reaction stage were evaluated. An absorption band in the 3300–3500 cm-1 wavenumber region was observed. Additionally, peak suggestive of COOH was observed in the sample spectra. Secondary modification diminished the absorption band in 3300–3500 cm-1 and a clear F1s signal was observed. Thus, Friedel–Crafts acylation with succinic anhydride produced carboxyl groups on the PEEK sample surfaces. Further chemical modification of the carboxyl groups by dehydration-condensation reactions is also possible. Furthermore, TNF-α is an inflammatory mediator that is central in initiating immune responses. Interleukin-8 (IL-8) is a potent inflammatory mediator involved in the severity of periodontal tissue disease by attracting and activating neutrophils to inflammatory sites. We examined the response of the human oral epithelial cell line, HSC-3, to TNF-α stimulation. TNF-α stimulation of HSC-3 enhanced IL-8 production and phosphorylation of mitogen-activated protein kinases (MAPKs) p38 and extracellular-signal-regulated kinase (ERK)1/2. Pretreatment with various MAPK inhibitors attenuated IL-8 production, suggesting that TNF-α stimulation via the MAPK signaling pathway enhances the production of IL-8 and may exacerbate periodontal disease.doctoral thesi

However, few studies have investigated the biological influence of flowable composite resins commercially available in Japan. In this study, we compared cell viability among 5 products currently available in dental clinical practice. Using mouse-derived Balb/c 3T3 cells, DMEM containing 5% fetal calf serum was adopted as an assay medium. Initially, the flowable resins were dissolved in the assay medium in the presence of dimethyl sulfoxide (DMSO) be-fore photopoly-merization, and each solution was diluted multiple times to investigate cell viability. As a result, all products showed cell death at a dilution ratio of 2. At a dilution ratio of 4 or 8, there was a marked decrease in cell viability. At a dilution ratio of 16, Subsequently, cell viability tests after photopolymerization were performed. Sam-ples were irradiated with an LED-type light irradiator for 5 or 10 seconds, and cell viability was investigated using the three-dimensional culture method with colla-gen. Cell viability was evaluated using MTT assay. Cell viability in the 10-second irradiation group for all products was higher than in the 5-second irradiation group. Some products showed no significant difference in comparison with the negative control group. There was no relationship between the monomer composition or filler type of any product and cell viability. The results of this study showed that cell viability slightly differed among the 5 clinically available products. There was no product for which cell viability was particularly low.doctoral thesi
