Exploring cultivation strategies for the propagation of chicken parvovirus (ChPV)

    No full text
Chicken parvovirus (ChPV) has been investigated since the mid-eighties, particularly due to its suspected involvement in malabsorption syndrome (MAS) in broilers. However, no effective method for the in vitro isolation and propagation of ChPV has been reported to date. This study aimed to evaluate the replication potential of ChPV in vitro using various avian and mammalian cell lines, as well as in vivo through experimental inoculation of chicks. The challenge inoculum was prepared from ChPV-positive chicken liver and used to infect primary fibroblast cultures derived from chickens, geese, and pheasants, as well as established cell lines (CER, Vero, and MA-104). Across five consecutive passages, no cytopathic effect was observed, and qPCR analyses failed to detect ChPV DNA in any of the inoculated cells. In contrast, oral inoculation of one-day-old chicks resulted in a progressive increase in viral load in feces, accompanied by transient diarrhea between days 3 and 6 post-inoculation. ChPV DNA was also detected systemically, with the highest loads found in lymphoid tissues, especially the bursa of Fabricius and spleen. These results confirm that ChPV does not replicate in the tested cell cultures, whereas one-day-old chicks represent a viable in vivo model for viral propagation and further studies on ChPV biology and its role in MAS

Objective: To compare the preferences of children and their parents regarding the characteristics of the dental operator and the clinical environment for dental care. Material and Methods: This cross-sectional study was conducted from August 2022 to July 2023. A questionnaire was developed in English and then translated into Spanish, before being subjected to a trial and a pilot study. After obtaining informed consent, 152 parents and their children, selected by convenience sampling, participated in the survey. Bivariate statistical analysis was performed using Fisher\u27s exact and chi-square tests. Results: Statistically significant differences (p<0.05) were determined when comparing the preferences of children and parents for the variables gender, age, clothing, mask, floor, door, lighting, curtains, dental chair, play area, waiting area, and consulting room. Based on participants’ gender, it was determined that the highest percentage of boys (71.76%) and girls (86.57%) prefer to be seen by an operator of the same gender (p=0.001). Significant differences were identified among children regarding clothing preferences (p=0.001). Additionally, differences were observed in parents’ preferences for curtains and walls in the clinical scenario (p<0.05). Children showed varying preferences for the play area, waiting area, and office, whereas both parents and children had varying preferences for the reception area. Conclusion: Differences were observed when comparing the preferences of children and their parents concerning the dental operator and the clinical scenario

Objective: To describe digital intraoral scanning in five children with microcephaly caused by the Zika virus, detailing occlusal and dental characteristics using digital models. Material and Methods: An observational study assessed the dental occlusal profile of five 4-year-old children with Congenital Zika Virus Syndrome (CZS) using digital oral scanning. The children were treated at the Unichristus School of Dentistry Clinic. The Panda 2 intraoral scanner was utilized. A mouth opener made from wooden sticks, adhesive tape, and gauze was used individually for each patient, thereby minimizing the risk of trauma. The upper arches were scanned first, followed by the lower arches, and finally the arches in occlusion. Results: The 3D digital models revealed the relationship between the distal faces of the deciduous second molars, Baume\u27s arch type, tooth eruption chronology, anterior open bite, posterior crossbite, overbite, overjet, and deviated midline. Conclusion: Intraoral scanning in children with congenital Zika virus microcephaly proved to be a viable alternative for producing reliable digital study models, offering more comfort and safety to these patients and replacing conventional molding techniques

The aim of this article is to discuss what is the meaning of being a philosopher for María Zambrano, a reference in 20th century philosophical thought. To present her ideas on the subject, the works Filosofia y poesia (1939) and Hacia un saber sobre el alma (1950) are the main references. At first, we will discuss, as a way of introducing her to those who are not yet familiar with her books and ideas, who this Spanish thinker was, who lived in exile for almost her entire life. Next, we will discuss how she defines philosophy, a central question for understanding what it means to be someone who does philosophy. In this way, we will present the concept of the metaphor of the heart and the relationship between philosophy and poetry, a fundamental theme of her thought. Finally, in order to discover what she understood by being a philosopher, we will discuss her perspective dialoguing with central themes of her work and her own life experience.O objetivo deste artigo é discutir o que significava ser filósofa para María Zambrano, referência no pensamento filosófico do século XX.

    Get PDF
Objective: To verify the accuracy of deep learning models in detecting cellular alterations in histological images of oral mucosa. Material and Methods: The study compares three convolutional neural network (CNN) architectures for classifying histological images: EfficientNet-B3, MobileNet-V2, and VGG16. Efficient and focused on computer vision, each has specific advantages. A Kaggle database with 5192 images was used, divided into training (70%), validation (15%), and test (15%) sets. The CNNs were implemented using the Keras library, trained with pre-trained ImageNet weights, and evaluated using accuracy and AUC metrics. Results: The findings indicate that EfficientNet-B3 achieved the lowest training and validation losses at epoch 30, with the highest accuracy and stability during training. Evaluation metrics showed EfficientNet-B3 with 98% accuracy and 99% sensitivity for oral squamous cell carcinoma (OSCC) images, outperforming MobileNet-V2 and VGG16. MobileNet-V2 achieved 97% accuracy and 96% sensitivity, while VGG16 reached 94% accuracy and 93% sensitivity for OSCC images. All models exhibited high sensitivity and specificity in differentiating between normal and OSCC images, as demonstrated by ROC curves. EfficientNet-B3 had the highest AUC (0.982), followed by MobileNet-V2 (AUC=0.967) and VGG16 (AUC=0.937). These findings underscore the effectiveness of EfficientNet-B3 for accurately detecting cellular alterations in histological images of oral mucosa. Conclusion: Our study reveals the superior performance of CNNs, particularly EfficientNet-B3, in classifying histological images of OSCC
