Application of direct electric current to the corneal and conjunctival epithelia regulates the tight junctional assembly for ocular iontophoretic drug delivery
Objectives: In this study, we determined how iontophoresis (IP) affects tight junctions (TJs) in isolated rabbit corneas and conjunctiva.
Methods: Direct electric current in the range of 0.5–2.0 and 0.5–10 mA/cm2 were applied to the cornea and conjunctiva, respectively, for 30 min. The localization and expression levels of TJ-associated proteins were assessed before and after the application of the electric currents using immunostaining and western blotting.
Results: In both corneal and conjunctival epithelia, the localization of proteins, such as claudin-1, claudin-4, occludin, and ZO-1, was temporarily altered by anodal and cathodal IP; however, the protein relocalization was slower at higher currents. Additionally, in both anodal and cathodal IP, the expression levels of claudin-1 and occludin in the cornea and conjunctiva remained unchanged after the application of the electric currents compared with those before.
Conclusion: Our results indicated that the application of a direct electric current temporarily regulated TJ assemblies without altering the levels of TJ-associated proteins in both the cornea and conjunctiva. This temporary weakening of the paracellular barrier by the current may be responsible for the enhanced drug transport across the cornea and conjunctiva induced by ocular IP.

Validation of its efficacy with scientific investigation is a prerequisite of any prospective herbal resource. The plant is a rich source of various bioactive chemical compounds. The present review is an attempt to consolidates comprehensive information sourced from 100 different literatures, exploring the plant\u27s distribution, phytochemistry, traditional medicinal uses, and pharmacological activities specially focusing on some prevalent diseases of present day. The herb’s antimicrobial, anticancer, antiulcer and antidiabetic properties signify its potential in modern pharmacology. The review underscores B. ciliata\u27s relevance in drug development and nutraceutical formulations, bridging age-old wisdom with contemporary healthcare solutions. Further exploration into its bioactive compounds and clinical applications promises innovative avenues in therapeutic intervention.

Pharmacovigilance: A Necessary Tool for Drug Safety Monitoring Globally: Pharmacovigilance
Pharmacovigilance, which is like a parasol to define the procedures for monitoring and analysing adverse drug reactions (ADRs), is a crucial part of efficient clinical practise, public health initiatives, and drug control systems. The volume of data processed has increased as a result of the rise in reported adverse drug reactions (ADRs), and understanding pharmacovigilance requires a high degree of skill in order to quickly identify medication hazards and defend the product against an unwarranted removal. An impartial method of evaluation would strengthen the present global network of pharmacovigilance centres, which is managed by the Uppsala Monitoring Centre. This would take into account contentious and significant medication safety problems that might have a negative impact on public health across international borders. Pharmacovigilance has recently focused mostly on identifying previously unrecognised or poorly understood adverse medication occurrences. Pharmacovigilance is a crucial and essential component of clinical research, and it is now expanding in many nations. However, with the turn of the millennium, pharmacovigilance confronts enormous hurdles in aspect of improving safety and monitoring of medications. Today, several pharmacovigilance centres are engaged for drug safety monitoring in this global arena. In this overview, we\u27ll talk about medication safety, the function of international pharmacovigilance centres, the advantages and drawbacks of pharmacovigilance, and how it may be used in the future by the healthcare industry.

Evaluation of Some Hormones Total Antioxidant Capacity and Malondialdehyde Levels in Polycystic Ovarian Syndrome Women attending the gynecology Clinic at Nnewi: Hormones, Total Antioxidant Capacity and Malondialdehyde Levels in Polycystic Ovarian Syndrome Women
Polycystic ovarian syndrome (PCOS), marked by oxidative stress and hormonal imbalances, causes infertility, insulin resistance, and diverse health problems. It not only affects physical health but also can strain marriages and lead to divorce, posing a notable societal issue. The levels of hormones (Estradiol (E2), Dihydroepiandrosterone sulfate (DHEA), luteinizing hormone (LH), follicle-stimulating hormone (FSH), progesterone (PROG) Testosterone (TEST)), total antioxidant capacity (TAC) and Malondialdehyde (MDA) in PCOS Women attending the gynecology Clinic at Nnewi were investigated. This cross-sectional study had 45 participants with PCOS as a test group and 45 participants without PCOS as the control group between the ages of 18-50 recruited. All the biochemical parameters were determined by enzyme-linked immunoassay technique. Data was expressed as Mean ± standard deviation. The differences in parameters studied between the PCOS group (test) and the control group were evaluated using an independent t-test. Statistical significance was set at p-value < 0.05. Result showed significant higher differences in the mean serum levels of DHEA (87.40±65.90), LH (34.7±36.1), FSH (19.61±14.73) and Testosterone (3.04±1.36) in women with PCOS compared with the control (1.035±0.54),( 20.76±18.1), (13.2±10.19), and (3.04±1.36)(p< 0.05) respectively. A higher significant difference exists in the mean serum MDA values of the test group (women with PCOS compared with the control group(p<0.05). This study concluded that oxidative stress and hormone imbalance occurred among participants with PCOS attending the gynecology clinic of Nnamdi Azikiwe University Teaching Hospital in Nnewi.

Ananas comosus (Pineapple): A Comprehensive Review of Its Medicinal Properties, Phytochemical Composition, and Pharmacological Activities
Herbal medicine, also known as phytomedicine, utilizes various plant parts like leaves, fruits, roots, bark, or flowers for their health benefits, standing as an alternative healthcare approach to conventional medicine. Traditional herbal remedies, integral to medical systems worldwide, notably in China and India, are gaining global recognition for their efficacy and fewer side effects. Approximately 80% of the world\u27s population relies on traditional herbal medicine for primary healthcare, as per the World Health Organization. Ananas comosus (L.) Merril, commonly known as pineapple, belongs to the Bromeliaceae family, encompassing about 50 genera and 200 species. Pineapple, a perennial herbaceous plant, exhibits unique physical attributes, with its main parts including the stem, leaves, peduncle, multiple fruit, crown, shoots, and roots. Pineapple contains various amino acids, including tyrosine and tryptophan, along with bromelain, an enzyme aiding digestion by breaking down proteins. This review outlines previous research on the phytochemical and pharmacological characteristics of Ananas comosus, highlighting its diverse medicinal properties. From antimicrobial and anticancer activities to antioxidant, anti-inflammatory, and hepatoprotective effects, pineapple demonstrates significant therapeutic potential across multiple health domains. Additionally, its nutritional value and traditional uses further underscore its importance as a versatile herbal remedy.

Phytochemical Analysis, Antioxidant and Anti Hyperlipidemic Activity of Nyctanthes arbor-tristis in Male Wistar Rats
The current study\u27s objective was to assess the antihyperlipidemic, in vitro antioxidant, and qualitative and quantitative phytochemical analysis of Nyctanthes arbor-tristis flowers hydroalcoholic extract at 200 and 400 mg/kg in rats that were made hyperlipidemic by a high-fat diet. The Folins Ciocalteau reagent and the aluminium chloride method were used for the quantitative measurement of flavonoids and phenolics, respectively. Through phytochemical investigation, alkaloids, carbohydrates, glycosides, tannins, phenolic compounds, triterpenoids, and steroids were found in the hydroalcoholic extracts of N. arbor-tristis flowers. The hydroalcoholic extract flowers had a total phenolic content of 126.000 mg/gm, which was followed by flavonoids at 103.104 mg/gm. Hydroalcoholic extracts of flowers showed concentration-dependent antioxidant activity in vitro against DPPH and superoxide radical scavenging test technique. In this investigation, rats were given a high-fat diet as part of an experimental induction procedure. By lowering LDL and VLDL cholesterol levels and raising HDL levels, the HANAT therapy and the medication atorvastatin dramatically reduce body weight. grew When hydroalcoholic flower extracts were given orally to animals that had been forced to become hyperlipidaemic due to a high cholesterol diet, the levels of HDL cholesterol, triglycerides, low density lipoproteins, and very low-density lipoproteins were all much lower. The p value (p<0.001) indicated that the results were significant. The triton-induced study results indicate that animals treated with HANAT at dose levels of 200 mg/kg and 400 mg/kg had considerably lower blood lipid parameters (p<0.01) than the control group; however, the 400 mg/kg of HANAT group animals showed significantly higher serum lipid parameters (p<0.001) than the control group. Additionally, a higher HDL level was noted at this time.

Design and Evaluation of Gastro-Retentive Floating Tablets of Ibandronic acid
The objective of this study is Preparation and evaluation of gastro-retentive floating tablets of Ibandronic acid by direct compression method polymers or combination. In this study, Ibandronic acid, the most commonly used biphosphonate for treating osteoporosis, was formulated as gastro retentive dosage form (GRDF) tablets to enhance its oral bioavailability. GRDDs are an approach to prolong gastric residence time, there by targeting site-specific drug release in the upper GIT for local or systemic effect. GRDF tablets of Ibandronic acid (200 mg) were characterized with the effects of Carbopol 934P, HPMC 4KM and Na-CMC at various ratios on swelling, floating, physical integrity. A prolonged dissolution profile of Ibandronic acid GRDF tablets developed in this study was observed for its thickness, friability, hardness, drug content and in-vitro drug release. Formulated Floating tablets of Ibandronic acid gave satisfactory results for various physicochemical parameters like hardness, friability, thickness, weight variation and content uniformity. Sodium bicarbonate has predominant effect on the buoyancy lag time, while HPMC K4M has predominant effect on total floating time and drug release. From the study it is evident that a promising controlled release floating tablets of Ibandronic acid can be developed to increase gastric residence time and thereby increasing its bioavailability. Both doses significantly reduced the occurrence risk of new vertebral fractures by 50–52 percent when compared to the effects of the placebo drug.

Herbs as Skeletal Muscle Relaxant: An Updated Review
When compared to modern drugs, herbal medicines are usually accepted by doctors and patients for having greater therapeutic efficacy and fewer adverse effects.A diverse class of drugs known as skeletal muscle relaxants is frequently used to treat two distinct underlying disorders: spasticity resulting from upper motor neuron syndromes and muscular pain or spasms resulting from peripheral musculoskeletal problems. Skeletal muscle (SM) controls insulin sensitivity, glucose uptake, and blood glucose homeostasis, all of which are essential for energy and glucose metabolism. A progressive and widespread loss of skeletal muscle mass and strength that results in physical handicap, a low quality of life, and perhaps death is known as sarcopenia. Herbs are a valuable tool for illness prevention. These all have dysregulated skeletal muscle and sarcopenia. These medicinal herbs are being used over recent years. In this Review article we will keep an eye on an updated review on herbs as skeletal muscle relaxant. We will discuss about musculoskeletal disorders, phytochemicals role in disorders. Herbs like aloe barbedensis, curcuma longa, zingiber officinale,citrus aurantium,chamaemelum nobile,hibiscus rosa sinensis etc. will be studied for their role in skeletal muscle relaxation.

Chimeric Antigen Receptor T-Cells (CAR T-Cells): An Engineered Targeted Therapy for Treatment of Cancer
We have undertaken this review to explore the various developments and insights of CAR-T cell therapy during 1989-2023 and its advantages in the treatment of cancer and immune modulation. It is a chimeric antigen receptor T-cell therapy, which is an innovative form of immunotherapy that harnesses the power of the immune system to fight cancer. At first, T cells are extracted from the patient’s blood through a process called leukapheresis. Then the modification has been done in T cells by genetically engineered to express chimeric antigen receptors (CARs) on their surface. These receptors are designed to recognize specific proteins, or antigens, that are found on the surface of cancer cells. Many conventional therapies available in the market for the treatment of cancer and Immuno modulation but most of them having Adverse Drug Reaction (ADR). But CAR-T cells possess upper hand on these conventional Formulations. Once a sufficient number of CAR-T cells have been produced, they are infused back into the patient’s bloodstream. Once reach inside the body, the CAR-T cells recognize and bind to the cancer cells that express the specific antigen targeted by the CAR. This triggers the destruction of the cancer cells by the immune system. CAR-T cell therapy has shown remarkable success in treating certain types of blood cancers, such as acute lymphoblastic leukaemia (ALL), chronic lymphocytic leukaemia (CLL), and certain types of lymphoma. The content of this review will pave the way to work on CAR-T cell therapy.

Analytical method development and validation for the evaluation of related substances in Apalutamide by RP-HPLC
Apalutamide is an anti-cancer agent used for the management of prostate cancer. A new stability indicating RP-HPLC method (Gradient mode) has been developed for the estimation of Apalutamide and its related substances using Waters Alliance system (Model no. 2996 and 2695) with Inertsil ODS-3 (250 × 4.6 mm, 5μm) column (PDA detector) was used for the present study. A mixture of Ammonium phosphate buffer solution and Acetonitrile (30: 70, v/v) was used as the mobile phase for the chromatographic study (Flow rate: 1.0 mL/min; Detection wavelength: 243 nm). Stress degradation studies were performed and the method was validated as per ICH guidelines.
