    Heavy metals contamination disturb balance and diversity of microorganism in soil. Microorganisms which can able to survive in those conditions are bacteria capable of oxidizing heavy metals. Identification based on 16S rRNA was used to determine characteristics and phylogenetic relationship of bacteria which can oxidize iron and sulphur in tin mining areas. The aim of this research was able to determine the bacterias characteristics isolated from tin mining areas and determine the phylogenetic relation of iron-sulphur oxidizing bacteria on tin mining soil in Belitung based on 16S rRNA sequences. This research was done using descriptive method, including isolation, morphological characterization, and identification based on 16S rRNA sequences. Morphology characterization includes colony and cell morphology through Gram staining. Molecular characterization includes amplification of 16S rRNA gene (Polymerase Chain Reaction/ PCR), electrophoresis amplicon and sequencing. Bacteria identification was done by comparing the 16S rRNA gene sequence in GenBank. The result showed three bacterias were identified by 16S rRNA have a similarity with Bacillus anthracis strain Ames, Bacillus cereus ATCC 14579, Staphylococcus sciuri subsp. Sciuri strains DSM 20345 and Micrococcus luteus NCTC 2665

    Parasitic mites are known as the natural enemy of Aedes sp. that can potentially infect any life stages of the Aedes sp. The parasitic mites’ ability to infect Aedes sp. influence the distribution pattern of parasitic mites. The widespread distribution of Aedes sp. is expected to affect the distribution of parasitic mites. The aims of this study were to determine the taxonomic family of parasitic mites that infected Aedes sp. larvae and to determine the geographical distribution patterns of parasitic mites of Aedes sp. in the endemic area of Dengue Hemorrhagic Fever (DHF) in Banjarnegara District. This study used larvae of Aedes sp. taken from the endemic area of DHF in Banjarnegara District. The study sites were in the Parakancanggah Village, Kutabanjar Village, Krandegan Village, and Sokanandi Village. This study used survey method with purposive sampling technique. Variable observed in this research was the distribution pattern of the parasitic mite of Aedes sp. larvae. Parameter observed included the family and the individual number of parasitic mites on each mosquito larvae, the average value, and the variance. Data were analyzed using mean value and the variance so that the distribution pattern can be determined. The level of parasitic mites’ distribution was analyzed using K' index negative binomial distribution. Results showed that 30 individual of parasitic mite were found from 1429 samples of examined Aedes sp. larvae. The identification result were five families of parasitic mites: Pionidae, Histiostomatidae, Hydryphantidae, Hydrachnidae, and Arrenuridae. The distribution pattern of the parasitic mites was regular, and the highest value of K’ index negative binomial distribution was 1,3225, in the Krandegan Village

    Ceriops tagal are found at Segara Anakan Cilacap, Baluran National Park and Karimunjawa National Park. Information on the genetic diversity can be used for conservation policy. Genetic variation of C. tagal were analysed by using Random Amplified Polymorphic DNA (RAPD). This study aims to determine the genetic variation in each population and between populations C. tagal based on RAPD markers. The total DNA was extracted with CTAB extraction method. Amplification was performed using 10 primer (OPA-3, OPA-10, OPB-1, OPB-6, OPB-7, OPB-8, OPB-10, OPB-12, OPB-17 and OPB-18).  Amplification includes pradenaturation at 94°C for 5 min, followed by 45 cycles of denaturation at 94°C for 1 min, primer annealing at 37°C for 1 min, and primer elongation on 72°C for 1 min 30 sec, final extension at 72°C for 5 min and ends with holding at 4°C for 1 min. The analysis was done by changing the pattern of fragments to binary data (for bands appear = "1" and for bands do not appear = "0"). It is done using a program GenAlEx 2.1 and NTSYS-PC 6.1. A population that has the highest genetic variation (h) was Segara Anakan (0.308), followed by Baluran (0.295) and Karimunjawa (0.123). Baluran has the highest polymorphism (75.00%), Segara Anakan 69.23%, while Karimunjawa was 30.77%. The results of AMOVA, showed that the percentage of genetic variation within populations is greater (77%) than among populations (23%). The results of PCA analysis showed a similar pattern of genetic distance and UPGMA. Samples of Karimunjawa clustered with samples of the population, SA1 and SA3 likely close to the sample of Karimunjawa and SA2 are likely not one group with another sample of the Segara Anakan.     Pengembangan Indirect Dipstick ELISA untuk Deteksi Aflatoksin B1 pada Pakan dan Jagung

    Enzyme Linked Immunosorbent Assay (ELISA) is an immunological method can be used to analyze aflatoxin B1 (AFB1) in feed. ELISA technique must be done by using an instument (ELISA reader) which is not effective when used in the field. Therefore, the simple ELISA technique is needed such as indirect dipstick ELISA (d-ELISA). The aim of research is to develop AFB1 screening method using   d-ELISA. The research is focusing on  development and validation of indirect d-ELISA, and its application on sample of feed and corn. The results showed that the best coating time for antigen AFB1-BSA (0,4 ug/ml) is 24 hours, reaction time for antibody anti AFB1 (1/800) and AFB1 standard is 15 minutes, and reaction time for goat anti rabbit-HRPO conjugate 1/2500 and substrate of orthodiasianin (ODN) is 20 minutes. The results of indirect d-ELISA on 22 samples are found that 7 samples are contaminated by AFB1 with concentration above 20 ng/g and 7 samples are contaminated by AFB1 with concentration in the range of 0 – 20 ng/g. Finally, it is concluded that the indirect d-ELISA is applicable to be used in the fields

    Water quality variable to affect the productivity of Pacific white shrimp (L. vannamei) in the brackishwater ponds. Therefore the aimed of this research to study the water quality variable  to  influent on the productivity of Pacific white shrimp (L. vannamei) in the brackishwater ponds. The research was conducted in the brackishwater ponds area that were in fish farming operation in the brackishwater ponds in the brackishwater ponds of Gresik Regency, East Java Province by conducting interviews with respondents to obtained primary data in the brackishwater ponds productivity through the submission of questionnaires, while the technical data of water quality variable were obtained by direct measurement in situ. As independent variable were water quality variables, namely: temperature, dissolved oxygen (DO), salinity, pH, total suspended solid (TSS), turbidity, nitrate, nitrite, phosphate, and total organic matter (TOM) while the dependent variable was  the productivity of Pacific white shrimp. Regression model selection “best” method was based on the backward. The results showed that productivity of Pacific white shrimp in the range from 133.3 to 3,000 kg / ha with an average of  690.38  kg / ha. Water quality variables that a have positive effect on the productivity of Pacific white shrimp  were nitrate and phosphate, while water quality variables have a negative effect on the productivity of Pacific white shrimp. were pH and TSS

    Three treatments and four replications in randomised completely design were conducted to reveal the protease activity and body protein content on catfish, Clarias gareipinus, under starvation and refeeding conditions. The result showed that the protease activity has no significant different (P>.05) in first week, but has significant different (P<.05) in second week after treatment.  Body protein content has significant different (P<.05) in first and second weeks after treatment. We concluded that the protease activity and body protein content decreased at deprivation, but increased after two weeks refeeding

    There are three known kraits species of the Genus Bungarus (Elapidae) inhabit Java Island, Indonesia. Two species out of the three species, i.e., Bungarus candidus (Malayan kraits) and Bungarus fasciatus (Banded kraits) are commonly found in Daerah Istimewa Yogyakarta (DIY) Province. However, a longterm  research about herpetofauna diversity in the Southern slope of Mount Merapi since 2006 until 2011 shows no species records from genus Bungarus. A nocturnal survey on April 28th, 2012 was administerd around the Kaliurang recreational area. We found snake which was suggested belong to the genus Bungarus. Detailed identification showed that this snake was Bungarus fasciatus or banded kraits snake. This finding provide the new record of genus Bungarus in Southern slope of Merapi especially since the periodical research of herpetofauna diversity which was started in 2006. To complete the data, it needs intensive research to know the distribution and species abundance of Bungarus fasciatus in Southern slope of Mount Merapi

    Batik industries have negative impacts on the environment from the dye used. An alternative method for decolorization in batik wastewater using the fungi Pleurotus ostreatus was grown in baglog. Spent mushroom (baglog) still contains fungal mycelium of P. ostreatus and contaminant fungus such as Penicillium sp., and Aspergillus sp.). Those fungi have a potential to decoloring dyes in batik wastewater. The aim of this research was to determine the ability of fungal mycelium isolated from spent mushroom P. ostreatus at decoloring batik wastewater, and determine the most effective fungal mycelium to decoloring batik wastewater. The results showed that the mycelium isolated from sewage baglog P. ostreatus was able to decoloring batik wastewater. Penicillium sp. mycelium is most effective at decoloring batik wastewater

    This study reports on giant freshwater prawn fecundity with or without unilateral ablation treatment combined with alpha tocopherol (vit E) during 3 month culture. This exsperimental study applied 8 combination treatments of 4 different vit E dosages, i.e. 0, 200, 400, and 600 IU per kg feed with or without unilateral ablation. The results showed that treatments very highly significantly different (p<0,01) in fecundity with the 200 IU vit E dose combined with unilateral ablation was the best (45.228 ± 3.867 egg) optimum resulted in fecundity of the treated females. The overall fecundities averages were ablation group with 0 IU = 41.835 ± 837 eggs; 200 IU = 45.228 ± 3.867 eggs; 400 IU = 43394 ± 1523 eggs ; and the 600 IU = 43.718 ± 2.255 eggs, and for the non ablation with vit E group were 0 IU = 45.293 ± 2.899 eggs, 200 IU = 44.106 ± 1.759 eggs, 400 IU = 46.623 ± 2.556 eggs, and 600 IU=51.824 ± 1.132 eggs. The analysis results also proven that ablation factor gave very highly significantly (p<0,01) and vit E factor separately and in combination with ablation significantly (p<0,05) determined the females fecundity. In conclution, combination of unilateral ablation and tocopherol effective improved fecundity and the combination of unilateral ablation and vit E 200 IU the optimum resulted in giant freshwater prawn females fecundity

    The study to determine amylase activity and metabolic rate of catfish, Clarias gariepinus under starvation and refeeding conditions has been conducted with three treatments and four replications in randomized completely design. The result showed that the amylase activity was significant different (P<.05) only at second week after treatment  and metabolic rate was significant different (P<.050 at first and second week after treatment. Conclutions, the amylase activity and metabolic rate of catfish have been decreased at starvation, especially at second week, but it recovery at refeeding condition
